RESUMO

Arginine (ARG) and Glutamine (GLN) have been reported to play significant roles in protein metabolism, immunity, and intestinal health in weanling pigs. The present study investigated the independent and interactive effect of supplementing ARG and GLN on pigs immune status and growth performance following an Escherichia coli F4 challenge. A total of 240 mixed-sex pigs (24 ±â€…2 d old; 7.3 ±â€…0.1 kg BW) were used in a 42-d experiment after selection for E. coli F4 susceptibility. The pigs were group-housed (3 pigs per pen), and pens were randomly assigned to five experimental treatments (N = 16 pens per treatment). Experimental treatments were: 1) a wheat-barley-soybean meal-based basal diet (CTRL), 2) a basal diet with 2500 mg/kg zinc oxide (ZnO), 3) a basal diet + 0.5% Glutamine (0.5% GLN), 4) basal diet + 0.5% Arginine (0.5% ARG), and 5) basal diet with 0.5% Glutamine + 0.5% Arginine (0.5% GLN + ARG). All Pigs were inoculated with E. coli F4 on days 7, 8, and 9 post-weaning. Rectal swabs were taken from each pig and plated on blood agar plates for E. coli F4 presence. Blood and fecal samples were taken to determine the acute phase response and selected fecal biomarkers for the immune response. Growth performance and fecal scores were recorded. Fecal swabs resulted in no positive pig for E. coli F4 before inoculation and 73.3% positive postinoculation. Diarrhea incidence during days 7 to 14 was significantly lower for the ZnO treatment (P < 0.05). The haptoglobin level on day 3 was lower than days 10 and 20, irrespective of treatment (P < 0.05). The albumin level was lower on day 20 compared to days 3 and 10 (P < 0.05). There was no treatment effect on albumin levels regardless of sampling day (P > 0.05). The PigMAP was lowest on day 3 and highest on day 10 (P < 0.05). We did not observe significant treatment differences (P > 0.05) in myeloperoxidase and calprotectin. Pancreatitis-associated protein was higher in the ZnO (P = 0.001) treatment than in the other treatments. Fecal IgA tended (P = 0.10) to be higher in the ZnO and 0.5% ARG treatments. There were no performance differences, except during days 0 to 7, where the ZnO treatment was lower in average daily gain and average daily feed intake (P < 0.001), while feed efficiency (G:F) FE was similar across treatments. In summary, no improved performance was observed with either ARG, glutamate, or both. The immune response results showed that the E. coli F4 challenge may have exacerbated the acute phase response; hence, the benefits of dietary treatments did not go beyond immune repair and reduction in inflammation.


The supplementation of functional amino acids such as arginine and glutamine has been reported to improve growth performance in weanling pigs. However, during periods of enteric Escherichia coli challenge, results have been inconsistent. Here, we investigated whether independent or combined arginine and glutamine supplementation could improve performance and immune response under an E. coli F4 challenge. The results showed no performance improvements but an improvement in the acute phase response and immune response status. Thus, the benefit of supplemental arginine and glutamine may be prioritized towards building an immune response rather than growth under conditions of an enteric immune challenge.

The present study investigated the impact of providing different supplemental essential amino acids (EAA) in a low crude protein (CP) diet on growth performance in weanling pigs. A total of 324 mixed-sex 24-d weaned piglets (initial BW 6.9 ± 0.34 kg) were used in a 27-d growth trial with six dietary treatments immediately post-weaning. The first two treatments were a control standard CP (19%) diet (positive control; PC) and a negative control (NC) diet with low CP (16%) and reduced Ile, Leu, and histidine levels. The rest of the treatments had low CP with varied EAA types and levels; T1 had similar Ile, Leu, and His levels as PC but with low CP (16%), while T2 had low CP and 10% higher His, Thr, Trp, and Met+Cys compared to PC. The T3 was a low CP diet with 10% supplemental Leu, Ile, and Val compared to PC, while T4 was a low CP diet with 10% supplementation with all the EAA except Lys compared to PC. The initial body weight (BW) was not statistically different (P > 0.05) among the treatments. Also, on d 6, no statistical differences in BW were observed among the treatments. The average BW recorded on d 13, 20, and 27 showed significant treatment differences where the PC had consistently higher BW than all the other treatments (P < 0.05). The average daily gain (ADG) of the PC was higher than the rest of the treatments. Between d 13 and 20, the average daily feed intake (ADFI) for PC was not different from NC and T1 (P > 0.05), but compared to T2, T3, and T4, the PC treatment showed a high ADFI (P < 0.05). Overall (d 0-27), the ADFI for PC was not different from T1 and was significantly higher than all other treatments. Overall, results showed that the gain to feed (G:F) ratio was higher (P < 0.05) for PC compared to other dietary treatments. In summary, although the treatments (T1-T4) consisted of varying levels of EAA above the recommended requirement levels for optimal performance, we did not see a significant impact on growth performance improvement, which may indicate that the targeted EAA (His, Val, Thr, lle, Leu, Trp, and Met) may not have been limiting in these diets. On the other hand, the phenylalanine (Phe) requirement may be limited in the current formulations, or perhaps the EAA: total N ratio in T1, T2, T3, and T4 may have been too high, resulting in the inefficiency of EAA utilization for growth.

Nutrient kinetic data and the timing of nutrient release along the gastrointestinal tract (GIT), are not yet widely used in current feed formulations for pigs and poultry. The present review focuses on interactions between fermentable substrates (e.g., starch, fiber, and protein) and selected minerals on nutrient digestion and absorption to determine nutritional solutions to maximize animal performance, principally in the grower-finisher phase, with the aim of minimizing environmental pollution. For phosphorus (P), myo-inositol 1,2,3,4,5,6-hexakis (dihydrogen phosphate) (InsP6), copper (Cu), and zinc (Zn), no standardized methodologies to assess in vitro mineral digestion exist. The stepwise degradation of InsP6 to lower inositol phosphate (InsP) forms in the GIT is rare, and inositol phosphate4 (InsP4) might be the limiting isomer of InsP degradation in diets with exogenous phytase. Furthermore, dietary coefficients of standardized total tract digestibility (CSTTD) of P might be underestimated in diets with fermentable ingredients because of increased diet-specific endogenous P losses (EPL), and further clarification is required to better calculate the coefficients of true total tract digestibility (CTTTD) of P. The quantification of fiber type, composition of fiber fractions, their influence on digestion kinetics, effects on digesta pH, and nutrient solubility related to fermentation should be considered for formulating diets. In conclusion, applications of nutrient kinetic data should be considered to help enhance nutrient digestion and absorption in the GIT, thereby reducing nutrient excretion.

Body protein losses in lactating sows have a negative impact on sow and litter performance. Improving dietary amino acid utilization may limit protein mobilization. The effects of dietary protein kinetics on sow body condition loss, blood plasma metabolites, and plasma insulin-like growth factor-1 (IGF-1), and also on litter gain during lactation, were investigated in this study. In total, 57 multiparous sows were fed one of three lactation diets with the same crude protein level: low level of slow protein diet (LSP) (8% slowly degradable protein of total protein), medium level of slow protein diet (MSP) (12% slowly degradable protein of total protein), or high level of slow protein diet (HSP) (16% slowly degradable protein of total protein) in a complete block design. Our results showed that HSP sows lost the least body weight compared to MSP and LSP sows (11.9 vs. 17.3 and 13.5 kg, respectively; p = 0.01), less body protein than MSP sows (1.0 vs. 2.1 kg; p = 0.01), and tended to lose less loin muscle thickness than LSP sows (1.7 vs. 4.9 mm; p = 0.09) between Day 2 to Day 21 post-farrowing. LSP sows had greatest plasma urea level on Day 6 than MSP and HSP sows (4.9 vs. 3.6 and 3.1 mmol/L, respectively; p < 0.01) and on Day 13 (5.6 vs. 4.1 and 3.7 mmol/L, respectively; p < 0.01). HSP sows had the lowest plasma urea level at Day 20 compared to LSP and MSP sows (4.0 vs. 5.5 and 4.9 mmol/L, respectively; p < 0.01). The average plasma urea level of Days 6, 13, and 20 post-farrowing was negatively correlated with slow protein intake (r = -0.49, p < 0.01). Litter gain, milk composition, and nitrogen output to the environment did not differ significantly among the treatment groups. Therefore, the dietary protein kinetics affected mobilization of maternal reserves in multiparous sows during lactation, with a high fraction of slow protein-sparing protein mobilization.

RESUMO

Every year 25 000 Australians experience a cardiac arrest in our community, but only 12% survive. The faster cardiopulmonary resuscitation and defibrillation, known as basic life support (BLS), is commenced, the greater the chance of survival. Currently, only half of the Australian adults are trained in BLS. The Australian Resuscitation Council and key stakeholder organisations believe that the best way to ensure all Australians know how to save a life is by mandating BLS education and training in our schools. This 'Aussie KIDS SAVE LIVES' position statement outlines our strategy to help facilitate the introduction of a programme of regular BLS training into the Australian school curriculum.

Lactose plays a crucial role in the growth performance of pigs at weaning because it is a palatable and easily digestible energy source that eases the transition from milk to solid feed. However, the digestibility of lactose declines after weaning due to a reduction in endogenous lactase activity in piglets. As a result, some lactose may be fermented in the gastrointestinal tract of pigs. Fermentation of lactose by intestinal microbiota yields lactic acid and volatile fatty acids, which may positively regulate the intestinal environment and microbiome, resulting in improved gastrointestinal health of weanling pigs. We hypothesize that the prebiotic effect of lactose may play a larger role in weanling pig nutrition as the global feed industry strives to reduce antibiotic usage and pharmacological levels of zinc oxide and supra-nutritional levels of copper. Evidence presented in this review indicates that high dietary lactose improves growth performance of piglets, as well as the growth of beneficial bacteria, particularly Lactobacillus, with the positive effects being more pronounced in the first 2 weeks after weaning. However, the risk of post-weaning diarrhea may increase as pigs get older due to reduced lactase activity, high dietary lactose concentrations, and larger feed intakes, all of which may lead to excessive lactose fermentation in the intestine of the pig. Therefore, dietary lactose levels exert different effects on growth performance and gastrointestinal physiological functions in different feeding phases of weanling pigs. However, no formal recommendation of lactose for weanling pigs has been reported. A meta-analysis approach was used to determine that diets fed to swine should include 20%, 15%, and 0 lactose from d 0-7, d 7-14, and d 14-35 post-weaning, respectively. However, sustainable swine production demands that economics must also be taken into account as lactose and lactose containing ingredients are expensive. Therefore, alternatives to lactose, so called "lactose equivalents" have also been studied in an effort to decrease feed cost while maintaining piglet performance with lower dietary lactose inclusions. In summary, the present review investigated dose-response effects of dietary lactose supplementation to exert positive responses and begin to elucidate its mechanisms of action in post-weaning pig diets. The results may help to replace some or all lactose in the diet of weanling pigs, while improving production economics given the high cost of lactose and availability in some swine production markets.

This study investigated the effects of a proprietary commercial feed additive (FA) comprised of a blend of fatty acids, organic acids, and phytochemicals; a hydroxychloride copper (MA); as well as a water acidification product (WA), alone and in combination, on growth performance in nonvaccinated broiler chickens raised in an antibiotic-free production system. The test treatments were FA; WA; FA and WA combined (FA + WA); and FA, WA, and MA combined (FA + WA + MA). The efficacy of these treatments was compared with a negative control (CON) and a medicated feeding program (bacitracin, antibiotic growth promoter [AGP]). Ross 708 cockerels (n = 2376) were subject to a 3-phase commercial feeding program, namely, starter (0-20 days), grower (21-28 days), and finisher (28-35 days), with no coccidiostats or additional medications added to the feed. On day 14, birds were subjected to an in-feed Clostridium perfringens challenge and a subset of animals were euthanized and the ileal digesta was collected for C. perfringens enumeration. Prior to pathogen challenge (day 14), birds fed the FA + WA and F + WA + MA treatments had significantly higher body weights (+2.6%-3.5%) than those fed CON and similar body weights to birds fed the AGP. These early growth advantages were not sustained postchallenge. Clostridia counts in ileal digesta were dramatically reduced in birds fed the AGP compared with all treatments. The FA (-2.5 log), FA + WA (-2.0 log), and FA + WA + MA (-2.3 log) treatments had significantly lower clostridia counts than the CON treatment. Together, these findings support the use of combined in-feed and in-water strategies for reducing clostridia, while maintaining growth, in antibiotic-free production systems.

Feed additives that promote gastrointestinal health may complement coccidiosis vaccination programs in antibiotic-free broiler production systems. This study examined the effects of a commercial feed additive blend (FA) on intestinal histomorphology and inflammatory biomarkers in vaccinated Ross 708 cockerels (N = 2,160). The study was a randomized complete block design (12 blocks) with 3 dietary treatments: CON (negative control), AGP (positive control: 55 ppm of bacitracin methylene disalicylate), and FA (1.5 kg/MT in starter; 1.0 kg/MT in grower; and 0.5 kg/MT in finisher). Birds were reared on re-used litter and fed a 3-phase feeding program (starter, 0 to 14 D; grower, 15 to 28 D; finisher, 29 to 36 D). One master batch of basal feed for each feeding phase was prepared and final experimental diets were manufactured by mixing the basal feed with the respective test ingredient prior to pelleting. Growth measurements, including pen body weight and feed intakes, and fresh fecal samples were taken throughout the study. On day 20, samples of intestinal tissue were collected from a subset of birds (n = 72, 1 block) for histomorphology and mRNA expression of tight junction and inflammatory genes. In the duodenum, the ratio of villi length to crypt depth was significantly lower in FA (and AGP) fed birds than those consuming the CON diet. Relative mRNA expressions of iNOS, IFNƔ, and claudin-1 were upregulated in the jejunum of FA and AGP treatment groups compared to those in the CON group; the response in the FA was of lesser magnitude than AGP. Together, these results demonstrated that the FA treatment altered the microstructure of the duodenum and affected the expression of inflammatory genes in the jejunum. The timing of these changes coincided with peak oocyte shedding in feces and an observed reduction in feed efficiency in all dietary treatment groups.
