RESUMO

In this paper we show that the electronic properties of ultrathin tetrahedral amorphous carbon (ta-C) films are heavily dependent on their thickness. By using scanning tunnelling spectroscopy, Raman spectroscopy, and conductive atomic force microscopy, it was found that a decrease of ta-C thickness from 30 to 7 nm leads to (i) the narrowing of the band gap; (ii) appearance of shallower monoenergetic traps as well as the increase of their concentration; (iii) the increase of the equilibrium concentration of free charge carriers and their mobility; which were caused by (iv) the increase in the sp(2) fraction. However, beyond a certain ta-C thickness (7 nm) the electronic properties of the studied samples start to deteriorate, which is highly likely related to titanium oxide formation at the Ti/ta-C interface. The same tendency is observed for the sample with beforehand air-formed native titanium oxide at the interface. With respect to the last point, it is suggested that the ta-C layer has no uniform coverage if its thickness is small enough (less than 7 nm). The experimental results were rationalized by detailed atomistic simulations. By using the so-called "Tauc plot" we introduce the possibility of the coexistence of bulk and surface band gaps originating from the large increase in sp(2) bonded carbon atoms in the surface region compared to that in the bulk ta-C. The results from the simulations were found to be consistent with the experimental measurements. The previously stated variation in the electronic properties of the layers as a function of their thickness was also exhibited in the electrochemical properties of the samples. It appears that the thinner ta-C layers had more facile electron transfer kinetics as determined with a ferrocenemethanol (FcMeOH) outer sphere redox system. However, if the ta-C layer thickness was reduced too much, the films were not stable anymore.

RESUMO

When two physical systems share the quantum property of entanglement, measurements of one system appear to determine the state of the other. This peculiar property is used in optical, atomic, and electrical systems in an effort to exceed classical bounds when processing information. We extended the domain of this quantum resource by entangling the motion of a macroscopic mechanical oscillator with a propagating electrical signal and by storing one half of the entangled state in the mechanical oscillator. This result demonstrates an essential requirement for using compact and low-loss micromechanical oscillators in a quantum processor, can be extended to sense forces beyond the standard quantum limit, and may enable tests of quantum theory.

Macroscopic mechanical oscillators have been coaxed into a regime of quantum behaviour by direct refrigeration or a combination of refrigeration and laser-like cooling. This result supports the idea that mechanical oscillators may perform useful functions in the processing of quantum information with superconducting circuits, either by serving as a quantum memory for the ephemeral state of a microwave field or by providing a quantum interface between otherwise incompatible systems. As yet, the transfer of an itinerant state or a propagating mode of a microwave field to and from a storage medium has not been demonstrated, owing to the inability to turn on and off the interaction between the microwave field and the medium sufficiently quickly. Here we demonstrate that the state of an itinerant microwave field can be coherently transferred into, stored in and retrieved from a mechanical oscillator with amplitudes at the single-quantum level. Crucially, the time to capture and to retrieve the microwave state is shorter than the quantum state lifetime of the mechanical oscillator. In this quantum regime, the mechanical oscillator can both store quantum information and enable its transfer between otherwise incompatible systems.

What is the relationship between variability in ongoing brain activity preceding a sensory stimulus and subsequent perception of that stimulus? A challenge in the study of this key topic in systems neuroscience is the relative rarity of certain brain 'states'-left to chance, they may seldom align with sensory presentation. We developed a novel method for studying the influence of targeted brain states on subsequent perceptual performance by online identification of spatiotemporal brain activity patterns of interest, and brain-state triggered presentation of subsequent stimuli. This general method was applied to an electroencephalography study of human auditory selective listening. We obtained online, time-varying estimates of the instantaneous direction of neural bias (towards processing left or right ear sounds). Detection of target sounds was influenced by pre-target fluctuations in neural bias, within and across trials. We propose that brain state-triggered stimulus delivery will enable efficient, statistically tractable studies of rare patterns of ongoing activity in single neurons and distributed neural circuits, and their influence on subsequent behavioral and neural responses.

A cellulase-producing clone was isolated from a genomic library of the Erwinia rhapontici (Millard) Burkholder strain NCPPB2989. The corresponding gene, named celA, encodes an endoglucanase (EC 3.2.1.4) with the extremely low pH optimum of 3.4 and a temperature optimum between 40 and 50 degrees C. A single ORF of 999 nt was found to be responsible for the Cel activity. The corresponding protein, named CelA, showed 67% identity to the endoglucanase Y of E. chrysanthemi and 51.5% identity to the endoglucanase of Cellulomonas uda, and thus belongs to the glycosyl hydrolase family 8. The celA gene, or its homologue, was found to be present in all E. rhapontici isolates analysed, in E. chrysanthemi, and in E. amylovora. The presence of plant cell wall-degrading enzymes in the amylovora group of Erwinia spp. had not previously been established. Furthermore, the DNA of both E. rhapontici and E. amylovora was found to exhibit homology to genes encoding the type II (GSP) secretion pathway, which is known to be responsible for extracellular targeting of cellulases and pectinases in Erwinia spp. that cause soft rotting, such as E. carotovora and E. chrysanthemi. Secretion of the CelA protein by E. rhapontici could not be verified. However, the CelA protein itself was found to include the information necessary for heterologous secretion by E. chrysanthemi.

An intact C-terminus was previously shown to be required for stability and secretion of the polygalacturonase (PehA) in Erwinia carotovora ssp. carotovora. Here we have analyzed the effects of amino acid (aa) substitutions generated to five C-terminal positions of PehA. Conservation of two hydrophobic and one non-hydrophobic residue (V372, V374 and N371, respectively) was found to be essential for maintenance of the protein stability. As an exception, one of the mutants (V372G) did not show major effects on protein stability, as determined by immunoblots and enzyme activity assay, yet it prevented the secretion completely. We conclude that the C-terminus of PehA is directly involved in the formation or stabilization of a conformation-sensitive structure needed for recognition of the protein as secreted.

A set of gene fusions was constructed between the pehA gene encoding the secreted endopolygalacturonase (PehA) and the bla gene coding for a normally periplasmic beta-lactamase (Bla). The resulting hybrid proteins were specifically and actively routed out of the cells via the Out-terminal branch of the general secretory pathway (GSP) in Erwinia carotovora subsp. carotovora (Ecc), provided that no more than the last two amino acids (aa) of the PehA domain were excluded from the fusion. However, both PehA-Bla hybrid proteins and PehA variants lacking at least four aa from the C-terminus of the PehA accumulated in the periplasm. Also, overexpression of the gene fusions prevented extracellular targeting of the hybrid proteins. Site-directed mutagenesis of the codons -4 and -3 (encoding Asn-373 and Val-374, respectively) from the end of the pehA gene and analysis of the protein products suggested that the Val-374 was important both for the structure and secretion of PehA, while the Asn-373 proved to be insignificant. We conclude that: (i) the GSP of Ecc is capable of secreting heterologous proteins; (ii) as the PehA protein can accommodate C-terminal extensions, secretion can occur with no part of the proposed targeting signal lying within the C-terminal extremity of a secreted molecule; and (iii) residues within the C-terminus of PehA play a role in secretion, possibly through stabilization of a structure needed for proper exposition of the proposed targeting motif.
