BACKGROUND: Dry socket (DS) or fibrinolytic osteitis is a relatively common complication that can occur following tooth extraction. This study aimed to determine the prevalence of DS and identify its associated predictive and mediating variables. MATERIAL AND METHODS: This study is classified as prospective observational, cross-sectional, and multicenter. Patients were consecutively selected in accordance with established criteria for tooth extraction. Data on patient demographics, surgical procedures and postoperative outcomes were collected. Nominal variables were analyzed using the Chi-Square Test, while associations involving ordinal values or considering counts or layers were examined using the Kendall's Tau-B Test or Mantel-Haenszel Test for trend. The GLM Mediation Model was employed to investigate potential mediation or indirect effects or potential underlying mechanisms of predictive variables on the development of DS. Two-tailed significance level of p ≤0.05 was considered statistically significant. RESULTS: A total of 1,357 patients undergoing routine dental extractions were included. DS was observed in 13 patients (prevalence of 1%). DS was associated with younger patients (under 50 years old), longer procedures, and the presence of surgical accidents, but only when mediated by surgical complexity. Smoking, particularly in combination with complex surgeries and surgical accidents, was associated with DS. Postoperative pain for more than two days and reported at moderate to high levels, emerged as a potential warning sign for DS. The use of antibiotics was found to significantly reduce the risk of DS (RR reduction of 36% and absolute risk reduction of 0.63%). CONCLUSIONS: Routine dental extractions revealed a 1% prevalence of dry socket. The obtained results suggests that DS is a multifactorial condition influenced by various factors, including gender, age, smoking, antibiotic prescription and surgical factors such as length, technique and accidents, nevertheless, those associations were observed mainly considering the influence of one variable on another.

The production of beef cattle in the Atlantic Forest biome mostly takes place in pastoral production systems. There are millions of hectares covered with pastures in this biome, including degraded pasture (DP), and only small area of the original Atlantic Forest has been preserved in tropics, implying that actions must be taken by the livestock sector to improve sustainability. Intensification makes it possible to produce the same amount, or more beef, in a smaller area; however, the environmental impacts must be assessed. Regarding climate change, the C dynamics is essential to define which beef cattle systems are sustainable. The objectives of this study were to investigate the C balance (t CO2e./ha per year), the intensity of C emission (kg CO2e./kg BW or carcass) and the C footprint (t CO2e./ha per year) of pasture-based beef cattle production systems, inside the farm gate and considering the inputs. The results were used to calculate the number of trees to be planted in beef cattle production systems to mitigate greenhouse gas (GHG) emissions. The GHG emission and C balance, for 2 years, were calculated based on the global warming potential (GWP) of AR4 and GWP of AR5. Forty-eight steers were allotted to four grazing systems: DP, irrigated high stocking rate pasture (IHS), rainfed high stocking rate pasture (RHS) and rainfed medium stocking rate pasture (RMS). The rainfed systems (RHS and RMS) presented the lowest C footprints (-1.22 and 0.45 t CO2e./ha per year, respectively), with C credits to RMS when using the GWP of AR4. The IHS system showed less favorable results for C footprint (-15.71 t CO2e./ha per year), but results were better when emissions were expressed in relation to the annual BW gain (-10.21 kg CO2e./kg BW) because of its higher yield. Although the DP system had an intermediate result for C footprint (-6.23 t CO2e./ha per year), the result was the worst (-30.21 CO2e./kg BW) when the index was expressed in relation to the annual BW gain, because in addition to GHG emissions from the animals in the system there were also losses in the annual rate of C sequestration. Notably, the intensification in pasture management had a land-saving effect (3.63 ha for IHS, 1.90 for RHS and 1.19 for RMS), contributing to the preservation of the tropical forest.

Fifteen New Zealand adult rabbits were randomly allocated into three groups: Sham-operated (group A), Ischemia and Reperfusion (group B) and Carolina Rinse Solution (CRS) (group C). Groups B and C were subjected to one hour of ischemia and two hours of reperfusion. In group C, ten minutes before reperfusion, the bowel lumen was filled with CRS, and the segment immersed in CRS. Necrosis and loss of integrity of the villi were visible in groups B and C. Edema of the submucosa and circular muscle was observed in all groups. Hemorrhage was observed in different layers for groups B and C, but group C showed more severe hemorrhage in different layers during reperfusion. All groups showed polymorphonuclear leukocyte infiltration on the base of the mucosa, submucosa, and longitudinal muscle, in addition to polymorphonuclear leukocytes margination in the mucosal and submucosal vessels. Necrosis of enterocytes, muscles, crypts of Lieberkühn and myenteric plexus was observed in groups B and C during reperfusion. Topical and intraluminal Carolina Rinse Solution did not attenuate the effects of ischemia and reperfusion in the small intestine of rabbits.(AU)


Quinze coelhos da raça Nova Zelândia foram alocados em três grupos: instrumentado (grupo A), isquemia e reperfusão (grupo B) e solução de Carolina rinse (CRS) (grupo C). Os grupos B e C foram submetidos a uma hora de isquemia e a duas horas de reperfusão. No grupo C, 10 minutos antes da reperfusão, o segmento isolado foi imerso e teve seu lúmen preenchido com CRS. Os grupos B e C apresentaram necrose e perda progressiva da integridade das vilosidades. Foi observado edema na submucosa e na camada muscular circular em todos os grupos. Nos grupos B e C, foi observada hemorragia em diferentes camadas, mas, no grupo C, a hemorragia foi mais intensa durante a reperfusão. Todos os grupos apresentaram infiltrado de PMN na base da mucosa, na submucosa e na camada muscular longitudinal e marginação de PMN nos vasos da mucosa e da submucosa. Durante a reperfusão, foi observada necrose dos enterócitos, das camadas musculares, das criptas de Lieberkühn e do plexo mioentérico nos grupos B e C. O uso tópico e intraluminal de CRS não atenuou os efeitos da isquemia e da reperfusão no intestino delgado de coelhos.(AU)

A ultrassonografia é um exame complementar não invasivo ainda pouco utilizado na avaliação prepucial e sem descrições detalhadas da parte livre do pênis (PLP) de bovinos. Este estudo objetivou padronizar a ultrassonografia prepucial em bovinos hígidos da raça Nelore com um ano de idade e, para tanto, descreveu o aspecto da lâmina interna prepucial (LIP) e da PLP, determinando as principais janelas acústicas, a viabilidade da utilização de contraste no lúmen da cavidade prepucial (LCP) e o melhor posicionamento do animal para a realização do exame. O escaneamento foi realizado com os animais na posição quadrupedal e em decúbito lateral direito. Foram determinadas cinco janelas acústicas a partir do óstio até a identificação do recesso prepucial. Os escaneamentos foram realizados nos planos longitudinais, transversais e dorsais, antes e após a infiltração de solução fisiológica a 0,9% no LCP. Como conclusões, o exame ultrassonográfico permitiu identificar a LIP e a PLP em todos os planos e em todas as janelas acústicas propostas. O melhor ponto de referência é a identificação da glande. O posicionamento quadrupedal é mais adequado para a realização do exame e o uso de contraste é recomendado para delimitação topográfica das estruturas.(AU)


Ultrasonography is a noninvasive complementary exam that is still rarely used in the preputial evaluation and without detailed descriptions of the bovine free end of the penis. The purpose of this study was to standardize preputial ultrasonography in one-year-old healthy Nelore cattle, describing the main acoustic windows, the viability of using contrast in the preputial cavity and the best positioning of the animal for performing the ultrasound examination. The scanning was performed with the animals in standing and decubitus position. Five acoustic windows were determined from the preputial ostium to the preputial fornix, at the free end of the penis. The scans were performed in longitudinal, transverse and dorsal planes, before and after infiltration of 0.9% saline solution into the preputial cavity. As conclusions, the ultrasound examination is able to identify the internal layer and the free part of the penis in all proposed planes and acoustic windows. The best landmark is the identification of the glans. Furthermore, standing positioning is the most appropriate to perform the exam and the use of contrast is recommended for topographic delimitation of the structures.(AU)

The objective of this study was to evaluate the effect of biotin supplementation on milk yield and the reproductive efficiency in Girolando cows. The study was conducted on a dairy farm located in central Brazil, between April 2012 and December 2016. Thirty-six Girolando cows in their first lactation were used. The cows were distributed in two treatment groups, each with equivalent weight distributions. Control treatment (CT) cows (n = 18) received a diet without any supplemental biotin, whereas biotin treatment (BT) cows (n = 18) received a diet supplemented with 20 mg/day of biotin during lactation. Biotin supplementation caused a significant increase (p = 0.001) in milk yield in the second lactation, and a trend (p = 0.09) toward higher average production during the three lactations was evaluated. There was no statistically significant difference between the treatments in terms of reproductive efficiency. In conclusion, biotin supplementation (20 mg/day) during lactation for Girolando cows reared in a tropical climate is able to increase milk yield, but does not improve reproductive efficiency.

The objective of this study was to evaluate the influence of different amounts of rumen-undegradable protein (RUP) on intake, N balance, performance, mammary gland development, carcass traits, and hormonal status of Holstein heifers at different physiological stages (PS). Sixteen prepubertal (PRE) heifers (initial BW = 106 ± 7.6 kg; age = 4.3 ± 0.46 mo) and 16 pubertal (PUB) heifers (initial BW = 224 ± 7.9 kg; age = 12.6 ± 0.45 mo) were used in an experiment over a period of 84 d. Four diets with increasing RUP contents (38, 44, 51, and 57% of dietary crude protein) and heifers at 2 PS (PRE or PUB) were used in a 4 × 2 factorial arrangement of treatments in a completely randomized design. Throughout the experiment, 2 digestibility trials were performed over 5 consecutive days (starting at d 36 and 78) involving feed and ort sampling and spot collections of feces and urine. At d 0 and 83, body ultrasound images were obtained for real-time carcass trait evaluation. The mammary gland was ultrasonically scanned at d 0 and every 3 wk during the experiment. Blood samples were taken at d 0 and 84 to determine serum concentrations of progesterone, estrogen, insulin-like growth factor I (IGF-I), and insulin. No interaction between PS and the level of RUP was found for any trait. Apparent digestibility of dry matter, organic matter, and neutral detergent fiber corrected for ash and protein was not affected by RUP level but was lower for PRE compared with PUB heifers. Sorting against neutral detergent fiber corrected for ash and protein (tendency only) and for crude protein was greater for PUB than PRE heifers. Pubertal heifers had greater average daily gain (905 vs. 505 g/d) and N retention (25.9 vs. 12.5 g/d) than PRE heifers. In addition, average daily gain and N retention were greatest at 51% RUP of dietary protein. Mammary ultrasonography indicated no effects of RUP amounts on mammary gland composition, whereas PRE heifers had greater pixel values than PUB, indicating higher contents of fat rather than protein in the mammary glands of PRE heifers. Serum progesterone and IGF-I concentration was affected only by PS, and PRE heifers had greater values of progesterone and IGF-I concentrations than PUB heifers. Serum insulin concentration was unaffected by PS but tended to be higher at 51% of RUP. In conclusion, an RUP level of 51% increases body weight, average daily gain, feed efficiency, and N retention in heifers regardless of the PS. In addition, PRE heifers have a lower sorting ability and reduced intake, total-tract digestibility, and N retention. They also have higher amounts of fat in their mammary glands, even at moderate growth rates.

This study was carried out to evaluate the effects of four levels of intensification of grazing systems: 1) degraded pasture - DP; 2) irrigated pasture with high stocking rate - IHS; 3) dryland pasture with high stocking rate - DHS; 4) dryland pasture with moderate stocking rate - DMS; on growth, muscle development and meat quality of Nellore steers (271±2.2kg of live body weight - BW; 15months old) during two consecutive periods (17 and 15months). The final BW, the average daily BW gain, the hot carcass weight and the dress percentage were greater (P<0.0001), and the ribeye area tended to be greater (P=0.085), in the intensified systems compared to the degraded system. Animals in all systems presented similar back fat. Muscle development increased with the intensification of the grazing systems and meat quality was not affected.

This study evaluated the effect of sodium butyrate (SB) on performance, expression of immune-related genes in the cecal tonsils, and cecal microbiota of broiler chickens when dietary energy and amino acids concentrations were reduced. Day-old male Ross 708 broiler chicks were fed dietary treatments in a 3 × 2 factorial design (8 pens per treatment) with 3 dietary formulations (control diet; reduction of 2.3% of amino acids and 60 kcal/kg; and reduction of 4.6% of amino acids and 120 kcal/kg) with or without the inclusion of 0.1% of SB. Feed intake (FI), body weight gain (BW gain), and feed conversion ratio (FCR) were recorded until 28 d of age. From 14 to 28 d, there was an interaction of nutrient density by SB (P = 0.003) wherein BW gain of birds fed SB was impaired less by the energy/amino acids reduction than unsupplemented birds. A similar result was obtained from 1 to 28 d (P = 0.004). No interaction (P < 0.05) between nutrient density by SB was observed for FCR. Nutritional density of the diets and SB modified the structure, composition, and predicted function of the cecal microbiota. The nutritionally reduced diet altered the imputed function performed by the microbiota and the SB supplementation reduced these variations, keeping the microbial function similar to that observed in chickens fed a control diet. The frequency of bacterial species presenting the butyryl-CoA: acetate CoA-transferase gene increased in the microbiota of chickens fed a nutritionally reduced diet without SB supplementation, and was not changed by nutrient density of the diet when supplemented with SB (interaction; P = 0.01). SB modulated the expression of immune related genes in the cecal tonsils; wherein SB upregulated the expression of A20 in broilers fed control diets (P < 0.05) and increased IL-6 expression (P < 0.05). These results show that SB had positive effects on the productive performance of broilers fed nutritionally reduced diets, partially by modulating the cecal microbiota and exerting immune-modulatory effects.

The present study aimed to evaluate the effect of different levels of ethanolic extract of propolis (EEP) and raw propolis (RP) on broiler performance and on selected bacterial groups in caecal microbiota using fluorescent in situ hybridization (FISH) measured by fluorescent activated cell sorting. Two experiments were conducted with 120 male chicks from 1 to 21 days of age for each, raised in cages and distributed in a completely randomized experimental design; there were five replicates with four birds per experimental unit and six treatments for each experiment (trial 1 - EEP - 0, 1000, 2000, 3000, 4000 and 5000 ppm and trial 2 - RP - 0, 100, 200, 300, 400 and 500 ppm). Fluorescent probes were used against the bacterial groups in caecal samples collected at 21 days of age. The data were subjected to one-way anova followed by Tukey's and regression analyses were used to assess the relationship between dietary levels of EEP or RP on performance and intestinal microbiota (p < 0.05). In the trial 1, results showed that the EEP did not cause any significant (p > 0.05) modification in the performance and caecal microbiota. In the trial 2, RP inclusion did not affect the performance but changed the bacterial composition (p < 0.05). Clostridiaceae, Gammaproteobacteria excluding Enterobacteriaceae and Lactobacillus spp. showed a quadratic response (p < 0.05), with the lowest value predicted to occur at 240 ppm, 221 ppm and 213 ppm of RP respectively. The proportion of Bacteroidaceae and Gammaproteobacteria did not differ (p > 0.05) among the experimental groups. The inclusion of ethanolic extract of propolis did not affect the performance and intestinal microbiota, whereas the supplementation of raw propolis modulates the caecal microbiota composition without any effects on chicken performance.

A transcriptome analysis of the venom glands of the spider Loxosceles laeta, performed by our group, in a previous study (Fernandes-Pedrosa et al., 2008), revealed a transcript with a sequence similar to the human complement component C3. Here we present the analysis of this transcript. cDNA fragments encoding the C3 homologue (Lox-C3) were amplified from total RNA isolated from the venom glands of L. laeta by RACE-PCR. Lox-C3 is a 5178 bps cDNA sequence encoding a 190kDa protein, with a domain configuration similar to human C3. Multiple alignments of C3-like proteins revealed two processing sites, suggesting that Lox-C3 is composed of three chains. Furthermore, the amino acids consensus sequences for the thioester was found, in addition to putative sequences responsible for FB binding. The phylogenetic analysis showed that Lox-C3 belongs to the same group as two C3 isoforms from the spider Hasarius adansoni (Family Salcitidae), showing 53% homology with these. This is the first characterization of a Loxosceles cDNA sequence encoding a human C3 homologue, and this finding, together with our previous finding of the expression of a FB-like molecule, suggests that this spider species also has a complement system. This work will help to improve our understanding of the innate immune system in these spiders and the ancestral structure of C3.

Background Sphingomyelinase D is the main toxin present in the venom of Loxosceles spiders. Several isoforms present in these venoms can be structurally classified in two groups. Class I Sphingomyelinase D contains a single disulphide bridge and variable loop. Class II Sphingomyelinase D presents an additional intrachain disulphide bridge that links a flexible loop with a catalytic loop. These classes exhibit differences in their toxic potential. In this paper we address the distribution of the structural classes of SMase D within and among species of spiders and also their evolutionary origin by means of phylogenetic analyses. We also conducted tests to assess the action of natural selection in their evolution combined to structural modelling of the affected sites. Results The majority of the Class I enzymes belong to the same clade, which indicates a recent evolution from a single common ancestor. Positively selected sites are located on the catalytic interface, which contributes to a distinct surface charge distribution between the classes. Sites that may prevent the formation of an additional bridge were found in Class I enzymes. Conclusions The evolution of Sphingomyelinase D has been driven by natural selection toward an increase in noxiousness, and this might help explain the toxic variation between classes

The human complement system is composed of more than 30 proteins and many of these have conserved domains that allow tracing the phylogenetic evolution. The complement system seems to be initiated with the appearance of C3 and factor B (FB), the only components found in some protostomes and cnidarians, suggesting that the alternative pathway is the most ancient. Here, we present the characterization of an arachnid homologue of the human complement component FB from the spider Loxosceles laeta. This homologue, named Lox-FB, was identified from a total RNA L. laeta spider venom gland library and was amplified using RACE-PCR techniques and specific primers. Analysis of the deduced amino acid sequence and the domain structure showed significant similarity to the vertebrate and invertebrate FB/C2 family proteins. Lox-FB has a classical domain organization composed of a control complement protein domain (CCP), a von Willebrand Factor domain (vWFA), and a serine protease domain (SP). The amino acids involved in Mg2+ metal ion dependent adhesion site (MIDAS) found in the vWFA domain in the vertebrate C2/FB proteins are well conserved; however, the classic catalytic triad present in the serine protease domain is not conserved in Lox-FB. Similarity and phylogenetic analyses indicated that Lox-FB shares a major identity (43%) and has a close evolutionary relationship with the third isoform of FB-like protein (FB-3) from the jumping spider Hasarius adansoni belonging to the Family Salcitidae

Runting-stunting syndrome (RSS) in poultry has been known for more than 40 years, but the precise etiology remains unknown and a licensed vaccine is consequently not currently available. In order to mitigate the symptoms associated with RSS, a series of experiments was performed to investigate whether a combined bacteriotherapeutic treatment consisting of probiotics, prebiotics, and organic acids could influence the outcome of this disease. Initially two groups of commercial broiler chickens were either left uninoculated or inoculated with filtrate from homogenized intestines of RSS-affected broiler chickens. One group from each of these two challenge groups was treated, with a bacteriotherapeutic regimen. After 12 days chickens were euthanatized, the body weight was measured, and duodenal lesions were enumerated. Five consecutive broiler chicken flocks were then raised either on litter from RSS-affected birds or on fresh wood shavings. Treatment had no beneficial effect on the number and severity of intestinal lesions. There appeared to be a significant build-up of RSS agent(s) in poultry litter, with each consecutive flock placement, independent of bacteriotherapeutic treatment, as more individuals exhibited intestinal lesions on built-up litter in RSS-affected houses (28.9% vs. 44%). While treatment did not appear to consistently reduce intestinal lesions, it did significantly improve the mean body weights (P<0.05) and uniformity of 12-day-old chickens placed on reused litter in houses in which RSS-infected birds were previously raised. A combination of litter management and bacteriotherapy may be needed to ameliorate the adverse effects of RSS on intestinal health and body weight in broiler chickens.

Ozone (O3) is the most abundant tropospheric oxidant as well as an important component of photochemical pollution. Once inside the plant, ozone can produce reactive oxygen species that change the antioxidative pool and the carbohydrate metabolism. The current study aimed to analyze whether the contents and the composition of the fructan, the ascorbate peroxidase activity, and the H2O2 accumulation were changed in Lolium multiflorum ssp. italicum cv. Lema plants as response to short-term exposure to ozone and/or to different meteorological conditions, in two contrasting seasons (winter and summer). Results showed that higher solar radiation tends to decrease fructose content and, along with temperature, increases the ascorbate peroxidase (APX) activity. Such activity and levels of fructans practically did not vary during the time the experiment was being done, but APX daylight variation was modified by the ozone. Thus, the higher levels of this pollutant decreased the APX activity and increased fructose content, as well as changed the size of the fructan chains. Hydrogen peroxide (H2O2) accumulation was higher in plants that were fumigated with ozone when compared to the control, and it decreased throughout the day. As a conclusion, fructan contents increased when the APX activity decreased. It suggested that fructans could also help the defense system when there is a reduction on the APX activity in the plant.

Ascaris lumbricoides is the most prevalent soil-transmitted helminth (STH) infection of human beings worldwide. Chemotherapy with synthetic anthelmintics such as albendazole, mebendazole, and pyrantel pamoate is the current method of treatment; however, the emergence of anthelmintic resistance could substantially decrease the efficacy of such treatments and the sustainability of STH control programs. Additionally, benzimidazoles are not recommended for pregnant women or children under age one. A blinded, controlled study was conducted to evaluate the efficacy of two microencapsulated, plant-based essential oil blends, TTN1013 (α-pinene, linalyl acetate, p-cymene, and thymol octanoate) and TTN1014 (α-pinene, linalyl acetate, p-cymene, and thymol acetate) as functional foods against Ascaris suum infection in pigs, an important pathogen that closely resembles human infections with A. lumbricoides. Four groups of 16 female, 21-24 day old, Yorkshire-cross pigs were treated daily with 0.5 or 1.0mg/kg TTN1013, 1.0mg/kg TTN1014, or 1.0mg/kg equivalent of empty capsules, delivered inside a cream-filled sandwich cookie for 14 weeks. Three days after the initiation of daily treatments, pigs were inoculated daily with A. suum eggs for four weeks. Pigs were weighed weekly and fecal egg counts (FEC) were conducted weekly starting five weeks after initial inoculation with A. suum eggs. Fourteen weeks after first infection with eggs, pigs were necropsied and worms were recovered, counted and separated according to sex. TTN1013 administered daily at a dose of 1.0mg/kg yielded a statistically significant reduction in total worm counts (76.8%), female worm counts (75.5%), FEC (68.6%), and worm volume (62.9%) when compared to control group. Reduction of total and female worm numbers and FEC were not significant for TTN1014 or at the 0.5mg/kg dose of TTN1013. All treatments were well-tolerated by all pigs and did not cause any adverse reactions. All pigs remained clinically normal and showed no signs of reduced intestinal health for the duration of treatment. Based on these results, TTN1013 shows promise as a daily supplement to reduce infection burdens of soil transmitted helminths in both pigs and human beings.

The objectives of this study were to determine if midlactation dairy cows (Bos taurus L.) grazing intensively managed elephantgrass would have their protein requirement met exclusively with the pasture and an energy concentrate, making the use of protein ingredients unnecessary, as well as to determine the dietary crude protein (CP) content that would optimize the efficiency of N utilization (ENU). Thirty-three Holstein and crossbred (Holstein × Jersey) midlactation dairy cows, producing approximately 20 kg/d, were grouped within breed into 11 blocks according to milk yield and days in milk. Within blocks, cows were randomly assigned to 1 of 3 treatments and remained in the study for 11 wk. The control treatment contained only finely ground corn, minerals, and vitamins, and it was formulated to be 8.7% CP. Two higher levels of CP (formulated to be 13.4 and 18.1%) were achieved by replacing corn with solvent-extracted soybean meal (SSBM). Pasture was fertilized with 50 kg of N/ha after each grazing cycle and averaged 18.5% CP (dry matter basis). No differences were observed in milk yield or milk fat, protein, and casein content or casein yield. In addition, pasture intake was not different among treatments. Milk urea N increased linearly as the concentrate CP content increased. Cows fed the 8.7% CP concentrate had higher ENU. In another experiment, 4 ruminally cannulated Holstein dry cows were used in a metabolism trial designed in a 4×4 Latin square. Cows were fed the same treatments described as well as a fourth treatment with 13.4% CP in the concentrate, in which urea replaced SSBM as the main N source. Ruminal volatile fatty acid concentration and microbial synthesis were not affected by levels or sources of N in the concentrate. Ruminal NH(3)N content increased as the concentrate CP content increased. Inclusion of SSBM in the concentrate did not increase production and decreased the ENU of midlactation dairy cows grazing on tropical forage. Supplementation of an 8.7% CP concentrate, resulting in a diet with CP levels between 15.3 and 15.7% of dry matter, was sufficient to meet the protein requirements of such milk production, with the highest ENU (18.4%).

This study aimed to evaluate the effectiveness of 3 commercial bacterial inoculants at controlling Escherichia coli O157:H7 in corn silages during ensiling and feedout phases of silage production. A second objective was to determine whether the inoculants exhibited and transferred antibacterial activity against E. coli O157:H7 to the silages. Chopped corn forage was ensiled after treatment with the following: distilled water (control); 5 x 10(5) cfu/g of E. coli O157:H7 (EC); EC and 1 x 10(6) cfu/g of Pediococcus pentosaceus and Propionibacterium freudenreichii (EC+BII); EC and 1 x 10(6) cfu/g of Lactobacillus buchneri (EC+LB); and EC and 1 x 10(6) cfu/g of L. buchneri and P. pentosaceus (EC+B500). Each treatment was ensiled in triplicate in mini silos for 3, 7, 31, and 82 d and analyzed for pH and E. coli O157:H7 counts. Samples from d 82 were also analyzed for volatile fatty acids, lactate, and aerobic stability. Antibacterial activity of inoculants and silages was determined by the Kirby-Bauer disc diffusion test. The pH of silages from all treatments decreased below 4 within 3 d of ensiling and remained low until d 82. Therefore, E. coli O157:H7 was not detected in silages after any of the ensiling durations. Applying inoculants containing L. buchneri resulted in less lactate, more acetate, and greater aerobic stability compared with the control. Applying EC+BII containing P. freudenreichii did not increase propionate or aerobic stability. Subsamples of d 82 silages were reinoculated with 1 x 10(5) cfu/g of E. coli O157:H7 either immediately after silo opening on d 82 or after 144h of aerobic exposure (d 88), and E. coli were enumerated 24h later. All silages reinoculated with the pathogen on d 82 had similar, low pH values (<4) and no E. coli were detected 24h later. Control, EC, and EC+BII silages reinoculated with the pathogen after 144h of aerobic exposure had relatively greater pH values (4.71, 5.67, and 6.03, respectively) and E. coli counts (2.87, 6.73, and 6.87 log cfu/g, respectively) 24h later, whereas those treated with L. buchneri had low pH values (<4) and undetectable (EC+B500) or 10-fold lower (1.97, cfu/g; EC+LB) E. coli counts. All pure cultures of commercial bacterial inoculants exhibited antibacterial activity independent of pH against E. coli O157:H7, but the pH-independent activity did not persist in the treated silages, suggesting that E. coli elimination from silages was mediated by pH reduction.

Avaliou-se a infecção experimental por Salmonella Enteritidis fagotipo 4 (SEpt4) em embriões de frango de corte para averiguar a habilidade de penetração através da casca e o efeito da inoculação no albúmen, considerando-se a mortalidade, a eclodibilidade e a colonização intestinal dos pintos eclodidos. Foram realizados dois experimentos, distribuídos em quatro tratamentos (T) cada, com 200 e 194 ovos incubáveis das linhagens Ross e ISA Label, respectivamente. Utilizaram-se ovos não sanitizados e inoculados na casca com Salmonella Enteritidis (T1) ou com placebo (T2); ou inoculados no albúmen com Salmonella Enteritidis (T3) ou com placebo (T4). Imediatamente após a inoculação, os ovos foram incubados, e a mortalidade embrionária avaliada após 96, 432 e 528 horas. Salmonella Enteritidis inoculada na casca manteve-se viável na casca e nas membranas durante todo o período de incubação e migrou para o interior dos ovos, entretanto não afetou os parâmetros de incubação. O patógeno inoculado no albúmen determinou mortalidade embrionária tardia nas linhagens Ross, 17,0 por cento, e ISA Label, 13,0 por cento, e originou pintos com maior freqüência de colonização intestinal por Salmonella Enteritidis, 76,7 por cento e 26,7 por cento para Ross e ISA Label, respectivamente.


The experimental infection of Salmonella Enteritidis phagotype 4 (SEpt4) was evaluated in broilers embryos to verify the ability of penetration through eggshell and the effect of albumen inoculation considering embryo mortality, hatchability, and intestinal colonization of hatched chicks. Two trials were conducted using in four treatments each, totaling 200 and 194 fertile eggs of Ross and ISA Label lineages, respectively. Non-sanitized eggs were inoculated on the shell with Salmonella Enteritidis (Treatment 1) or placebo (Treatment 2); and eggs were inoculated in albumen with Salmonella Enteritidis (Treatment 3) or placebo (Treatment 4). Immediately after inoculation, the eggs were incubated and embryo mortality was evaluated after 96, 432, and 528 hours. The results showed that Salmonella Enteritidis inoculated on egg shell remained active there and in shell membranes during all incubation period and migrated to the interior of the eggs; however, it did not affect the incubation parameters. It was observed that Salmonella Enteritidis inoculated in albumen caused late embryo mortality in Ross (17.0 percent) and ISA Label (13.0 percent) lineages, and originated chicks with high frequency of intestinal colonization by Salmonella Enteritidis, being 76.7 percent and 26.7 percent for Ross and ISA Label, respectively.

This study was conducted to relate the performance of broiler chickens fed diets containing growth-promoting antibiotics to changes in the intestinal microbiota. The technique of denaturing gradient gel electrophoresis (DGGE) of amplicons of the region V3 of 16S rDNA was used to characterize the microbiota. Two experiments were conducted, one with broilers raised in battery cages and the other with broilers raised in floor pens. Antibiotics improved the performance of the chickens raised in floor pens only. Avilamycin, bacitracin methylene disalicylate, and enramycin induced changes in the composition of the intestinal bacterial community of the birds in both experiments. The number of bacterial genotypes found in the intestinal tract of chickens was not reduced by the antibiotics supplemented in either environment. However, the changes in the composition of the intestinal bacterial community induced by antibiotics may be related to improvement in growth performance. This was indicated by the suppression of 6 amplicons and the presence of 4 amplicons exclusive to the treatment that had the best performance in the floor pen experiment.
