Between August and September, 2021, this study included 605 SARS-CoV-2 natural infection cases and 589 SARS-CoV-2 breakthrough cases from Nanjing and Yangzhou, as well as 690 inactivated COVID-19 vaccine recipients from Changzhou, China. In SARS-CoV-2 natural infection cases, the age range was 19-91 years (median age: 66 year), and the medians(Q1,Q3) of IgG titers were 0.19 (0.06-1.31), 3.70 (0.76-69.48), 15.31 (2.59-82.16), 4.41 (0.99-31.74), 2.31 (0.75-13.83), 2.28 (0.68-9.94) and 2.80 (1.00-9.53) at one to seven weeks after SARS-CoV-2 infection, respectively. In SARS-CoV-2 breakthrough cases, the age range was 18-76 years (median age: 45 year), and the medians(Q1,Q3)of IgG titers were 1.93 (0.34-26.67), 38.87 (7.90-121.0), 75.09 (11.85-123.70), 21.97 (5.20-95.58), 13.97 (3.47-46.82), 9.56 (2.48-33.38) and 4.38 (1.87-11.00) at one to seven weeks after SARS-CoV-2 infection, respectively. In inactivated COVID-19 vaccine recipients, the age range was 18-87 years (median age: 47 years), and the medians(Q1,Q3)of IgG titers were 16.22 (15.84-33.42), 5.35 (2.96-13.23), 3.30 (2.18-6.18), 3.14 (1.16-5.70), 2.77 (1.50-4.52), 2.72 (1.76-4.36), 2.01 (1.27-3.51) and 1.94 (1.35-3.09) at one to eight months after SARS-CoV-2 infection, respectively. The results suggested that IgG antibodies increased gradually within two weeks after SARS-CoV-2 infection, then declined gradually at three to seven weeks in SARS-CoV-2 natural infection cases. In SARS-CoV-2 breakthrough cases, IgG antibodies increased rapidly within two weeks, then declined gradually at three to seven weeks after SARS-CoV-2 infection. Additionally, IgG antibodies decreased rapidly within three months, then decreased gradually and remained at a low level within three months after immunization.

Nitric oxide (NO), an endothelium-derived relaxing factor, is synthesized from l-arginine by endothelial nitric oxide synthase (eNOS) in the endothelium. The objective of the present study was to preliminarily illuminate the expression of the eNOS gene in hypoxic adaptation of chicken embryonic development. The eNOS expression profiles between the Tibet and Shouguang chickens incubated under both normoxic and hypoxic conditions were detected by TaqMan real-time PCR. In this study, the chicken eNOS gene was found by both in silico cloning and RACE approaches. From the eNOS gene, we obtained a 3,310-bp mRNA sequence and a 10,666-bp DNA sequence and discovered that it was located on chicken chromosome 2 and had 7 unique transcripts. eNOS mRNA was detected in abundant amounts in some chick embryo organs (i.e., heart, liver, chorio-allantoic membrane, and lung), and expressed stably with the lowest levels in the brain. We observed that when exposed to hypoxia (13% O(2)) different embryo organ tissues had various sensitivities to hypoxia as determined by their eNOS expression profiles. Compared with the Shouguang chicken, the eNOS expression in the Tibet chicken was higher in the lung and liver, lower in the heart, and similar in the brain. In chorio-allantoic membranes, eNOS expression was higher in the Shouguang chicken than the Tibet chicken under hypoxic conditions, but not markedly different under normoxic conditions. The differences of eNOS expression between the 2 breeds may be relative to the hypoxic adaptation ability in Tibet chickens during embryonic development. This work will provide reference for future studies on the role of eNOS in hypoxic adaptation and response.

Rats exposed chronically to mild cold (5 degrees C/41 degrees F) develop hypertension. This cold-induced hypertension (CIH) is an environmentally induced, non-surgical, non-pharmacological and non-genetic model for studying hypertension in rats. The blood renin angiotensin system (RAS) appears to play a role in both initiating and maintaining the high blood pressure in CIH. The goal of the present study was to evaluate the role of brain angiotensin type 1 and type 2 receptors (AT1R and AT2R) in CIH. Sprague-Dawley adult male rats were used. Thirty-six rats were kept in a cold room at 5 degrees C and the other 36 were kept at 24 degrees C as controls. Systolic blood pressure (SBP) was recorded by tail cuff. The SBP was elevated in rats exposed to cold within 1 week (n=12, P>0.05), significantly increased at 3 weeks (P<0.05) and reached a maximum (125%) at 5 weeks (P<0.01). Three subgroups of the cold-treated and the controls were sacrificed at 1, 3 and 5 weeks. Specific brain sections were removed, either for reverse transcription polymerase chain reaction (RT-PCR) to measure mRNA, or for autoradiography to measure receptor binding for AT1R and AT2R. The AT1R mRNA was increased significantly in hypothalamus and brainstem after the first week in cold-treated rats and was maintained throughout the time of exposure to cold (n=6, P<0.01). AT1R binding significantly increased initially in hypothalamus and thereafter in brainstem. The mRNA and the receptor binding for AT2R decreased significantly (P<0.01, n=6) in nucleus of inferior olive and locus coeruleus of brainstem in cold-treated rats after exposure to cold. The experiments show differential regulation of RAS components, AT1R and AT2R, in different brain areas in cold-exposed rats and provide evidence that up-regulated AT1R and down-regulated AT2R in different brain areas are involved in CIH. The opposing directions of expression of AT1R and AT2R suggest that they play counterbalancing roles in brain function.

Phospholipase D (PLD) is a ubiquitous enzyme involved in many important cellular functions. Rat brain PLD isoforms have recently been cloned and characterized, but the expression of this gene has not been well studied. We thus examined the developmental expression of PLD2 in postnatal brains by Northern blotting employing a non-radioactive RNA probe. Two PLD2 mRNA transcripts of 3.9 and 10.8 kb were detected in different brain regions and various tissues of the rat. In postnatal brains, the level of PLD2 mRNA transcripts are low 1 day after birth, increase progressively during development, and reach the maximum level in the adult brain. We thus conclude that rat brain PLD2 mRNA is developmentally regulated.

Rats exposed chronically to mild cold (5 degrees C/41 degrees F) develop hypertension and cardiac hypertrophy. This provides a unique model of hypertension that is environmentally induced. The blood renin-angiotensin system (RAS) has been shown to play a role in both initiating and maintaining the high blood pressure (BP) in cold-induced hypertension. The mechanism also appears to involve both the tissue and brain RAS because there is increased mRNA for angiotensinogen (AGT) and angiotensin type 1 (AT1) receptors in brain and peripheral tissues, an increased spontaneous drinking response, and an increased dipsogenic response to acute administration of angiotensin II (Ang II) in cold-treated rats. Antisense oligodeoxynucleotides (AS-ODN), targeted to the RAS, have been shown to reduce BP in spontaneously hypertensive rats. Therefore, we injected AS-ODN in rats with cold-induced hypertension to test whether antisense inhibition was effective in reducing this nongenetic nonsurgical hypertension. Sprague-Dawley rats were made hypertensive by cold exposure and injected intracerebroventricularly with AS-ODN to AGT mRNA (n=6) or AT1 receptor mRNA (n=6). Systolic BP was recorded by tail cuff 24 hours later for 2 or 7 days, respectively. Systolic BP decreased significantly in response to AGT-AS-ODN (40+/-6 mm Hg, P<0.01) within 1 day after injection and to AT1 receptor-AS-ODN (P<0.05) for 3 days after injection. The maximum decrease was 41+/-10 mm Hg. Systolic BP then gradually increased to the preinjection level. The spontaneous drinking response to cold treatment also decreased significantly (P<0.05) after AGT-AS-ODN or AT1 receptor-AS-ODN intracerebroventricular injection. Intracardiac injection of AT1-AS-ODN (n=6) reduced systolic BP by 36+/-8 mm Hg (P<0.05) and decreased AT1 receptor as measured by autoradiography in aorta, adrenal glands, and kidneys 24 hours after injection. These data show that AS-ODN reduces BP in cold-induced hypertension and that the hypertension involves both peripheral tissues and central RAS in addition to blood-borne RAS mechanisms.
