RESUMO

Epithelial homeostasis is fundamental for the physiological functions of colon tissue. Dysregulation of colon epithelial structure leads to abnormal immune responses and diseases such as inflammatory bowel disease. In this work we found long non-coding RNA DANCR was a novel regulator to colon epithelial homeostasis. Silencing DANCR resulted in decreased expression of epithelial barrier proteins and enhanced susceptibility to TNFα stimulation, which was accompanied by hyperactivation of the NF-κB pathway. Mechanistical studies revealed DANCR modulated the expression of a protein methyltransferase SET7 to suppress responses to TNFα, as well as the activity of NF-κB pathway. In summary, DANCR regulated colon epithelial homeostasis through modulating the TNFα/NF-κB axis. These findings cast light on the discovery of novel regulators to colon epithelial homeostasis and added new evidence to the physiological functions of DANCR.

Leydig cell failure (LCF) caused by gene mutations leads to testosterone deficiency, infertility and reduced physical function. Adeno-associated virus serotype 8 (AAV8)-mediated gene therapy shows potential in treating LCF in the Lhcgr-deficient (Lhcgr-/-) mouse model. However, the gene-treated mice still cannot naturally sire offspring, indicating the modestly restored testosterone and spermatogenesis in AAV8-treated mice remain insufficient to support natural fertility. Recognizing this, we propose that enhancing gene delivery could yield superior results. Here, we screened a panel of AAV serotypes through in vivo transduction of mouse testes and identified AAVDJ as an impressively potent vector for testicular cells. Intratesticular injection of AAVDJ achieved markedly efficient transduction of Leydig cell progenitors, marking a considerable advance over conventional AAV8 vectors. AAVDJ-Lhcgr gene therapy was well tolerated and resulted in significant recovery of testosterone production, substantial improvement in sexual development, and remarkable restoration of spermatogenesis in Lhcgr-/- mice. Notably, this therapy restored fertility in Lhcgr-/- mice through natural mating, enabling the birth of second-generation. Additionally, this treatment led to remarkable improvements in adipose, muscle, and bone function in Lhcgr-/- mice. Collectively, our findings underscore AAVDJ-mediated gene therapy as a promising strategy for LCF and suggest its broader potential in addressing various reproductive disorders.

BACKGROUND: D-type cyclins (CYCD) regulate the cell cycle G1/S transition and are thus closely involved in cell cycle progression. However, little is known about their functions in rice. RESULTS: We identified 14 CYCD genes in the rice genome and confirmed the presence of characteristic cyclin domains in each. The expression of the OsCYCD genes in different tissues was investigated. Most OsCYCD genes were expressed at least in one of the analyzed tissues, with varying degrees of expression. Ten OsCYCD proteins could interact with both retinoblastoma-related protein (RBR) and A-type cyclin-dependent kinases (CDKA) forming holistic complexes, while OsCYCD3;1, OsCYCD6;1, and OsCYCD7;1 bound only one component, and OsCYCD4;2 bound to neither protein. Interestingly, all OsCYCD genes except OsCYCD7;1, were able to induce tobacco pavement cells to re-enter mitosis with different efficiencies. Transgenic rice plants overexpressing OsCYCD2;2, OsCYCD6;1, and OsCYCD7;1 (which induced cell division in tobacco with high-, low-, and zero-efficiency, respectively) were created. Higher levels of cell division were observed in both the stomatal lineage and epidermal cells of the OsCYCD2;2- and OsCYCD6;1-overexpressing plants, with lower levels seen in OsCYCD7;1-overexpressing plants. CONCLUSIONS: The distinct expression patterns and varying effects on the cell cycle suggest different functions for the various OsCYCD proteins. Our findings will enhance understanding of the CYCD family in rice and provide a preliminary foundation for the future functional verification of these genes.

OBJECTIVE: To describe the current situation of Yingyangbao feeding in rural areas of Sichuan, and to explore the mechanism among micronutrient powder cognition, self-efficacy and feeding behavior. METHODS: The multi-stage cluster sampling method was adopted. In October 2019, in 2 Han counties, 2 Tibetan counties, and 2 Yi counties in Sichuan Province, a total of 108 villages in 36 townships in 6 sample counties were included in the cluster. Caregivers of infants and young children aged 6-24 months were selected as the research subjects, and a face-to-face interview survey was conducted using a self-designed structured questionnaire. RESULTS: A total of 765 caregivers of infants and young children aged 6-24 months were included in this study. The effective feeding rate of the Yingyangbao was 54.43%. The average scores of caregivers Yingyangbao cognition, self-efficacy and feeding behavior were 3.80±1.16, 7.96±1.53 and 0.83±0.38, and there was a significant positive correlation between them. Self-efficacy played a partial mediating effect between caregivers' Yingyangbao cognition and feeding behavior, and the mediating effect accounted for 23.37% of the total effect. CONCLUSION: Yingyangbao cognition directly affects feeding behavior and indirectly affects feeding behavior through self-efficacy.

In order to generate high-quality single-photon emission computed tomography (SPECT) images under low-dose acquisition mode, a sinogram denoising method was studied for suppressing random oscillation and enhancing contrast in the projection domain. A conditional generative adversarial network with cross-domain regularization (CGAN-CDR) is proposed for low-dose SPECT sinogram restoration. The generator stepwise extracts multiscale sinusoidal features from a low-dose sinogram, which are then rebuilt into a restored sinogram. Long skip connections are introduced into the generator, so that the low-level features can be better shared and reused, and the spatial and angular sinogram information can be better recovered. A patch discriminator is employed to capture detailed sinusoidal features within sinogram patches; thereby, detailed features in local receptive fields can be effectively characterized. Meanwhile, a cross-domain regularization is developed in both the projection and image domains. Projection-domain regularization directly constrains the generator via penalizing the difference between generated and label sinograms. Image-domain regularization imposes a similarity constraint on the reconstructed images, which can ameliorate the issue of ill-posedness and serves as an indirect constraint on the generator. By adversarial learning, the CGAN-CDR model can achieve high-quality sinogram restoration. Finally, the preconditioned alternating projection algorithm with total variation regularization is adopted for image reconstruction. Extensive numerical experiments show that the proposed model exhibits good performance in low-dose sinogram restoration. From visual analysis, CGAN-CDR performs well in terms of noise and artifact suppression, contrast enhancement and structure preservation, particularly in low-contrast regions. From quantitative analysis, CGAN-CDR has obtained superior results in both global and local image quality metrics. From robustness analysis, CGAN-CDR can better recover the detailed bone structure of the reconstructed image for a higher-noise sinogram. This work demonstrates the feasibility and effectiveness of CGAN-CDR in low-dose SPECT sinogram restoration. CGAN-CDR can yield significant quality improvement in both projection and image domains, which enables potential applications of the proposed method in real low-dose study.

In plants, MAP65 preferentially cross-links the anti-parallel microtubules (MTs) and plays an important role for cytokinesis. However, the functions of MAP65 isoforms in rice (Oryza sativa. L) are largely unknown. Here, we identified two MAP65-3 homologs in rice, OsMAP65-3.1 and OsMAP65-3.2. We found that both OsMAP65-3.1 and OsMAP65-3.2 were similar in dimerization and location to AtMAP65-3, and the expression of either rice genes driven by the AtMAP65-3 promoter suppressed the cytokinesis failure and growth defect of atmap65-3. However, OsMAP65-3.1 with native promoter also recovered the atmap65-3, but OsMAP65-3.2 with its own promoter had no effects. OsMAP65-3.1 but not OsMAP65-3.2 was actively expressed in tissues enriched with dividing cells. R1R2R3-Myb (MYB3R) transcription factors directly bound to the OsMAP65-3.1 promoter but not that of OsMAP65-3.2. Furthermore, osmap65-3.2 had no obvious phenotype, while either osmap65-3.1 or osmap65-3.1(+/-) was lethal. The eminent MTs around the daughter nuclei and cytokinesis defects were frequently observed in OsMAP65-3.1-defective plants. Taken together, our findings suggest that OsMAP65-3.1, rather than OsMAP65-3.2, plays essential roles in rice cytokinesis resulting from their differential expression which were passably directly regulated by OsMYB3Rs.

Because of its rather low melt strength, polylactide (PLA) has yet to fulfill its promise as advanced biobased and biodegradable foams to replace fossil-based polymer foams. In this work, PLA vitrimers were prepared by two-step reactive processing from commercial PLA thermoplastics, glycerol, and diphenylmethane diisocyanate (MDI) using Zn(II)-catalyzed addition and transesterification chemistry. The transesterification reaction of PLA and glycerol occurs with zinc acetate as the catalyst, and chain scission will take place due to the alcoholysis of the PLA chains by the free hydroxyl groups from the glycerol. Long-chain PLA with hydroxyl groups can be obtained and then cross-linked with MDI. Rheological analysis shows that the formed cross-linked network can significantly improve melt strength and promote strain hardening under extensional flow. PLA vitrimers still maintain the ability of thermoplastic processing via extrusion and compression. The enhanced melt strength and the rearrangement of network topology facilitate the foaming processing. An expansion ratio as large as 49.2-fold and microcellular foam with a uniform cell morphology can be obtained for PLA vitrimers with a gel fraction of 51.8% through a supercritical carbon dioxide foaming technique. This work provides a new way with the scale-up possibility to enhance the melt strength of PLA, and the broadened range of PLA applicability brought by PLA vitrimers is truly valuable in terms of the realization of a sustainable society.

OBJECTIVE: To investigate the status quo and influencing factors of the first-time complementary food addition behavior of caregivers in the multi-ethnic background in the western rural areas. METHODS: In 2019, a multi-stage stratified cluster random sampling was used to selecting research subjects in western rural areas. A structured questionnaire was designed by ourselves with literature review and expert consultation method, and information such as sociodemographic characteristics, feeding knowledge, and complementary food addition of 1290 caregivers and infants were collected, ordered multi-classification Logistic regression was used to analyze the influencing factors of the first complementary food addition behavior of the Han nationality and the minority nationality. RESULTS: In the western rural areas, only 8.22% of the caregivers had better behavior of first complementary food addition, and 16.31% of the caregivers in the Han group had a good behavior of first complementary food addition. In the minority population, only 3.64% of infant caregivers had a good behavior of first complementary food addition. In Han, parents, as caregivers, had better first complementary food addition behavior than grandparents(OR=1.7829, 95% CI 1.1651-2.7283). Among ethnic minorities, education(OR=1.753, 95%CI 1.190-2.581), family fixed assets(OR=3.870, 95%CI 1.959-7.645)and feeding knowledge(OR=3.396, 95%CI 2.749-4.195) were the promoting factors for the first complementary food addition behavior. CONCLUSION: In western rural areas, caregivers' behavior of adding complementary food for the first time is generally poor.

BACKGROUND: Granulomatous amoebic encephalitis (GAE) is a rare central nervous system infection caused by the Balamuthia mandrillaris or Acanthamoeba species. Diagnosis is challenging because of the non-specific clinical presentation, cerebrospinal fluid analysis, and radiological features. There is no effective treatment for GAE to date. CASE PRESENTATION: A 54-year-old male was admitted to hospital after experiencing acute onset of numbness and weakness on his left limb. Due to the initial consideration of intracranial tumor, surgical removal of the right parietal lesion was performed. However, the patient had a headache accompanied by diplopia, difficulty walking and a new lesion was found in the left occipital-parietal lobe two weeks after the first operation. High-throughput next-generation sequencing (NGS) detected the presence of high copy reads of the B. mandrillaris genome sequence in the patient's blood, cerebral spinal fluid (CSF), and brain tissue. Pathological investigation of the brain tissue showed granulomatous changes and amoebic trophozoite scattered around blood vessels under high magnification. The patient was re-operated due to developing progressive confusion caused by subfalcine herniation of the left cerebral hemisphere. The lesions of the right parietal lobe were obviously decreasing in size after the first surgery, and the lesions of the left occipital lobe and the sunfalcine herniation didn't ameliorate two months after the second surgery. The patient was transferred to local hospital for continuous treatment with sulfamethoxazole and azithromycin. After five months of the second surgery, the patient showed good recovery with mild headache. CONCLUSIONS: This is the first report of a patient with B. mandrillaris encephalitis initially confirmed by NGS and have experienced two excisions, responding favorably to the combination of surgeries and medications. Early surgical resection of intracranial lesions combined with drug treatment may offer the chance of a cure.

Traditional devices, including conventional rigid electronics and machines, as well as emerging wearable electronics and soft robotics, almost all have a single mechanical state for particular service purposes. Nonetheless, dynamic materials with interchangeable mechanical states, which enable more diverse and versatile applications, are urgently necessary for intelligent and adaptive application cases in the future electronic and robot fields. Here, we report a gel-like material composed of a crosslinking polymer network impregnated with a phase changing molten liquid, which undergoes an exceptional stiffness transition in response to a thermal stimulus. Vice versa, the material switches from a soft gel state to a rigid solid state with a dramatic stiffness change of 105 times (601 MPa versus 4.5 kPa) benefiting from the liquid-solid phase change of the crystalline polymer once cooled. Such reversibility of the phase and mechanical transition upon thermal stimuli enables the dynamic gel mechanical transformation, demonstrating potential applications in an adhesive thermal interface gasket (TIG) to facilitate thermal transport, a high-temperature warning sensor and an intelligent gripper. Overall, this dynamic gel with a tunable stiffness change paves a new way to design and fabricate adaptive smart materials toward intelligent control of versatile devices.

Acute kidney injury (AKI), as a common oxidative stress-related renal disease, causes high mortality in clinics annually, and many other clinical diseases, including the pandemic COVID-19, have a high potential to cause AKI, yet only rehydration, renal dialysis, and other supportive therapies are available for AKI in the clinics. Nanotechnology-mediated antioxidant therapy represents a promising therapeutic strategy for AKI treatment. However, current enzyme-mimicking nanoantioxidants show poor biocompatibility and biodegradability, as well as non-specific ROS level regulation, further potentially causing deleterious adverse effects. Herein, the authors report a novel non-enzymatic antioxidant strategy based on ultrathin Ti3 C2 -PVP nanosheets (TPNS) with excellent biocompatibility and great chemical reactivity toward multiple ROS for AKI treatment. These TPNS nanosheets exhibit enzyme/ROS-triggered biodegradability and broad-spectrum ROS scavenging ability through the readily occurring redox reaction between Ti3 C2 and various ROS, as verified by theoretical calculations. Furthermore, both in vivo and in vitro experiments demonstrate that TPNS can serve as efficient antioxidant platforms to scavenge the overexpressed ROS and subsequently suppress oxidative stress-induced inflammatory response through inhibition of NF-κB signal pathway for AKI treatment. This study highlights a new type of therapeutic agent, that is, the redox-mediated non-enzymatic antioxidant MXene nanoplatforms in treatment of AKI and other ROS-associated diseases.

Body area networks (BANs) are configured with a great number of ultra-low power consumption wearable devices, which constantly monitor physiological signals of the human body and thus realize intelligent monitoring. However, the collection and transfer of human body signals consume energy, and considering the comfort demand of wearable devices, both the size and the capacity of a wearable device's battery are limited. Thus, minimizing the energy consumption of wearable devices and optimizing the BAN energy efficiency is still a challenging problem. Therefore, in this paper, we propose an energy harvesting-based BAN for smart health and discuss an optimal resource allocation scheme to improve BAN energy efficiency. Specifically, firstly, considering energy harvesting in a BAN and the time limits of human body signal transfer, we formulate the energy efficiency optimization problem of time division for wireless energy transfer and wireless information transfer. Secondly, we convert the optimization problem into a convex optimization problem under a linear constraint and propose a closed-form solution to the problem. Finally, simulation results proved that when the size of data acquired by the wearable devices is small, the proportion of energy consumed by the circuit and signal acquisition of the wearable devices is big, and when the size of data acquired by the wearable devices is big, the energy consumed by the signal transfer of the wearable device is decisive.

Spectrum defragmentation can improve spectrum utilization for an elastic optical network (EON). However, most of the existing studies have focused on defragmentation for working lightpaths, which may affect upper-layer network services. This paper considers protection lightpath-based hitless spectrum defragmentation for distance adaptive elastic optical networks. Without affecting working lightpaths, but defragmenting spectra for protection lightpaths, we expect to achieve truly hitless spectrum defragmentation for an EON. Shared backup path protection (SBPP) technique is employed as a representative network protection technique to evaluate the benefit of the proposed defragmentation scheme. To smooth the network spectra for future arriving lightpath requests so as to reduce bandwidth blocking probability (BBP), we propose two defragmentation triggering mechanisms, namely, defragmentation upon blocking (BTD) and batch defragmentation (BD). For each of them, we also propose two spectrum defragmentation algorithms, namely, defragmentation with sequentially releasing and re-establishing protection lightpaths (SR-D) and defragmentation with jointly releasing and re-establishing protection lightpaths (JR-D). The performances of these proposed algorithms are evaluated from perspectives of BBP and average number of reconfigurations per successfully established lightpath service (ANR). Simulation results show that compared to the case without defragmentation, the proposed scheme is effective to reduce BBP, which trades off with ANR.

Facing the increasing demands and challenges in the area of chronic disease care, various studies on the healthcare system which can, whenever and wherever, extract and process patient data have been conducted. Chronic diseases are the long-term diseases and require the processes of the real-time monitoring, multidimensional quantitative analysis, and the classification of patients' diagnostic information. A healthcare system for chronic diseases is characterized as an at-hospital and at-home service according to a targeted environment. Both services basically aim to provide patients with accurate diagnoses of disease by monitoring a variety of physical states with a number of monitoring methods, but there are differences between home and hospital environments, and the different characteristics should be considered in order to provide more accurate diagnoses for patients, especially, patients having chronic diseases. In this paper, we propose a patient status classification method for effectively identifying and classifying chronic diseases and show the validity of the proposed method. Furthermore, we present a new healthcare system architecture that integrates the at-home and at-hospital environment and discuss the applicability of the architecture using practical target services.

Stepwise key spectrum selection (SKSS) was introduced to resolve batch overlapping peaks from gas chromatography-mass spectrometry (GC-MS) analysis of ten batch tobacco flavoring samples in different storage times. Resolution was implemented on a software platform that embedded the SKSS method. The data from GC-MS analysis of the samples were saved and prepared in ASCII files and then were inputted into the software platform for visual inspections. The data segment with overlapping peaks was precut for subsequent analysis. Spectral background in the data was removed using a linear fitting of the baseline. Four components in the overlapping peaks were automatically detected by the SKSS method. The resolution of the concentration profiles and spectra of the four components was conducted by setting only one parameter, the negative area ratio, as 0.01. The fixed size moving window evolving factor analysis and evolving factor analysis were applied to validate the resolved concentration profiles. The resolved mass spectra were validated by the searched standard through library search at the pure component regions revealed by the resolved concentration profiles. The results showed that the SKSS method could be a simple but powerful tool in resolving batch chromatographic overlapping peaks.

The reliability and scalability of large-scale based optical fiber sensor networks (AOFSN) are considered in this paper. The AOFSN network consists of three-level hierarchical sensor network architectures. The first two levels consist of active interrogation and remote nodes (RNs) and the third level, called the sensor subnet (SSN), consists of passive Fiber Bragg Gratings (FBGs) and a few switches. The switch architectures in the RN and various SSNs to improve the reliability and scalability of AOFSN are studied. Two SSNs with a regular topology are proposed to support simple routing and scalability in AOFSN: square-based sensor cells (SSC) and pentagon-based sensor cells (PSC). The reliability and scalability are evaluated in terms of the available sensing coverage in the case of one or multiple link failures.

RESUMO

A new kind of 3(n) feed-forward programmable optical fiber true delay line was proposed. Theoretical analysis was presented on its delay performance and expandability. Experimental demonstration was given to show the implementation of such delay lines using SOAs and Farady rotation mirrors. Delay step as small as 0.5 ps with precision of about 0.03 ps was achieved. Measurement was performed to verify the feasibility and results are given.
