OBJECTIVES: To evaluate the impact of COVID-19 pandemics on clinical and surgical practice, educational activities, health and lifestyle behavior of Brazilian urology residents after 1 year of socio-economic restrictions. MATERIALS AND METHODS: An electronic survey was e-mailed to all postgraduate (PG) students registered by the Brazilian Society of Urology. The survey inclu-ded an assessment of socio-demographic, clinical practice, educational, health-related and behavior parameters. We also evaluated which subareas of urology were predominantly affected. A similar survey was adapted and sent to the directors of all urology residency programs. RESULTS: COVID-19 pandemic has severely impacted the clinical, surgical, and educational activities of urology residents in Brazil. Urology residents reported >50% decrease in multiple surgical modalities. We highlight kidney transplantation surgeries (66.2%), minor surgeries (62.3%), endoscopic surgeries (42.6%) and reconstructive surgeries (38.8%). This could represent a critical skills gap that residents may face beyond the COVID-19 pandemic. Furthermore, PG students faced stressful situations that caused worsening of mental and physical health, such as getting redirected to assistance of COVID-19 patients (66.9%), and high rate of infection by SARS-CoV-2 (58.2%). CONCLUSIONS: The COVID-19 pandemic has severely impacted the clinical, surgical, and educational activities of urology residents in Brazil. This could represent a critical skills gap that residents may face beyond the COVID-19 pandemic. PG students faced stressful situations that caused worsening of mental and physical health such as redirection to assistance of COVID-19 patients, concern about their own contamination and of family members.

Arboviruses are etiological agents in an extensive group of emerging diseases with great clinical relevance in Brazil, due to the wide distribution of their vectors and the favorable environmental conditions. Among them, the Mayaro virus (MAYV) has drawn attention since its emergence as the etiologic agent of Mayaro fever, a highly debilitating disease. To study viral replication and identify new drug candidates, traditional antiviral assays based on viral antigens and/or plaque assays have been demonstrating low throughput, making it difficult to carry out larger-scale assays. Therefore, we developed and characterized two DNA-launched infectious clones reporter viruses based on the MAYV strain BeAr 20290 containing the reporter genes of firefly luciferase (FLuc) and nanoluciferase (NLuc), designated as MAYV-firefly and MAYV-nanoluc, respectively. The viruses replicated efficiently with similar properties to the parental wild-type MAYV, and luminescence expression levels reflected viral replication. Reporter genes were also preserved during passage in cell culture, remaining stably expressed for one round of passage for MAYV-firefly and three rounds for MAYV-nanoluc. Employing the infectious clone, we described the effect of Rimantadine, an FDA-approved Alzheimer's drug, as a repurposing agent for MAYV but with a broad-spectrum activity against Zika virus infection. Additionally, we validated MAYV-nanoluc as a tool for antiviral drug screening using the compound EIDD-2749 (4'-Fluorouridine), which acts as an inhibitor of alphavirus RNA-dependent RNA polymerase.

Background and Objectives: it is extremely important and necessary to assess epidemiological events through the analysis of measures adopted at the time of crises, especially those with a health impact as a way of improving the system for future events, with testing being a gold standard to be assessed during an epidemic. This study aimed to analyze tests for COVID-19 diagnosis, with a view to detecting possible false negative results, in Parnaíba, Piauí, from March to December 2020. Methods: a statistical analysis of the data reported and made available by the Municipal Health Department using the IBM SPSS Statistics® 21.0 software, in which the variables type of test, date of symptom onset and date of material collection were crossed to obtain the results. Results: a total of 9,473 tests were negative, of which 11.1% were carried out using the RT-PCR methodology, 6.5% using rapid antigen tests, and 82.3% using a rapid antibody test. The analysis revealed that only 0.47% RT-PCR tests and 1.7% rapid antigen tests had been carried out within the ideal testing interval. On the other hand, the rapid antibody test had 0.14% performed outside the range. Conclusion: the most successful diagnostic test was the rapid antibody test, but it is the least specific and not suitable for determining health crisis management policies, especially for isolation measures for infected people, which suggests improvements in testing systems and development of tests with longer and more accurate testing intervals.(AU)
Justificativa e Objetivos: é de suma importância e necessidade a avaliação dos eventos epidemiológicos por meio da análise das medidas adotadas no momento das crises, em especial aqueles de impacto sanitário como forma de melhor o sistema para eventos futuros, sendo a testagem um padrão-ouro a ser avaliado durante uma epidemia. O objetivo deste estudo foi analisar os testes para o diagnóstico de COVID-19, na perspectiva de detectar possíveis resultados falsos negativos, em Parnaíba, Piauí, de março a dezembro de 2020. Métodos: análise estatística dos dados notificados e disponibilizados pela Secretaria Municipal de Saúde a partir do software IBM SPSS® Statistics 21.0, em que as variáveis tipo de teste, data do início dos sintomas e data da coleta do material foram cruzadas para obtenção dos resultados. Resultados: 9.473 testes resultaram negativo, em que 11,1% foram realizados pela metodologia RT-PCR, 6,5%, pelos testes rápidos de antígeno, e 82,3%, por teste rápido de anticorpo. A análise revelou que apenas 0,47% testes por RT-PCR e 1,7% testes rápidos de antígeno haviam sido realizados dentro do intervalo ideal de testagem. Por outro lado, o teste rápido de anticorpo teve 0,14% realizados fora do intervalo. Conclusão: o teste com maior sucesso de diagnóstico foi o teste rápido de anticorpo, porém é o menos específico e não adequado para determinação de políticas de gerenciamento de crise sanitária, em especial para medidas de isolamento de infectados, o que sugere melhorias em sistemas de testagem e desenvolvimento de testes com intervalos de testagem maior e precisos.(AU)
Justificación y Objetivo: es sumamente importante y necesario evaluar los eventos epidemiológicos a través del análisis de las medidas adoptadas en el momento de las crisis, especialmente aquellas con impacto en la salud, como una forma de mejorar el sistema para eventos futuros, siendo las pruebas un estándar de oro a ser evaluado durante una epidemia. El objetivo de este estudio fue analizar pruebas para el diagnóstico de COVID-19, con miras a detectar posibles resultados falsos negativos, en Parnaíba, Piauí, de marzo a diciembre de 2020. Métodos: análisis estadístico de los datos reportados y puestos a disposición por la Secretaría de Salud Municipal mediante el software IBM SPSS® Statistics 21.0, en el cual se cruzaron las variables tipo de prueba, fecha de inicio de síntomas y fecha de recolección del material para obtener los resultados. Resultados: un total de 9.473 pruebas resultaron negativas, de las cuales el 11,1% se realizaron mediante la metodología RT-PCR, el 6,5% mediante pruebas rápidas de antígenos y el 82,3% mediante prueba rápida de anticuerpos. El análisis reveló que sólo el 0,47% de las pruebas RT-PCR y el 1,7% de las pruebas rápidas de antígenos se habían realizado dentro del intervalo de prueba ideal. Myrcene (ß-myrcene), found in essential oils from plant species such as hops and cannabis, has many advantageous properties, but its use is limited due to volatility and low solubility in water. One way to circumvent these limitations is to encapsulate the essential oils in a polymer matrix. However, these hydrophobic molecules are difficult to quantify when dispersed in water. Seeking to study the release of this terpene in drug release tests from polymeric matrices, this work aimed to develop an easy and cheap UV spectrophotometric method for the quantification of ß-myrcene in aqueous medium. To achieves this goal, samples were prepared in 0.05% (w/v) polysorbate 80 solution, with concentrations of ß-myrcene ranging from 0.01% to 0.1% (v/v), and were analyzed at 226 nm. Each sample was analyzed in triplicate and repeated on three different days, to evaluate the repeatability of the results. The results were subjected to Q, F and Student's t-tests. The regression parameters obtained for ß-myrcene were above 0.99 and through statistical analysis, it was possible to confirm the repeatability for the results. The values of the limits of detection and quantification indicated that the method is not affected by intrinsic factors of the equipment. The results of accuracy, robustness and selectivity showed recovery rates within acceptable limits. This demonstrates that the quantification of ß-myrcene in aqueous medium by UV spectrophotometry is feasible.

The evaluation of the ecotoxicological effects of the effluent after treatment with peracetic acid is relevant to help establish reference concentrations for the disinfection process and waste recovery. Therefore, the objective of this work was to evaluate the ecotoxicity of effluent from a bovine slaughterhouse treated with peracetic acid on Girardia tigrina. The toxicity bioassays for planaria were the acute test (LC50) and chronic assays: locomotion, regeneration, reproduction and fertility. The results showed that the effluent treated with peracetic acid showed less toxicity than the effluent without application of peracetic acid. The effluent after peracetic acid application showed a chronic toxic effect in the reduction of locomotor speed in all studied disinfectant concentrations (0.8, 1.6, 3.3 and 6.6 µg L-1 of peracetic acid) and a delay in the formation of G. tigrina photoreceptors at the concentration of 6.6 µg L-1 of peracetic acid. Peracetic acid concentrations of 0.8, 1.6 and 3.3 µg L-1 were not toxic for blastema regeneration, photoreceptor and auricle formation, fecundity and fertility. In addition, this study assists in defining doses of peracetic acid to be recommended in order to ensure the wastewater disinfection process without causing harm to aquatic organisms.

Hemicellulose is widely available in nature, is a sustainable resource and has a wide range of applications. Among them, adsorption stands out for the removal of potentially toxic ions. Thus, in the study, the adsorption of Cd2+, Pb2+ and Hg2+ ions in two hemicellulose matrices were elucidated through computational simulations using density functional theory. Molecular electrostatic potential and frontier molecular orbitals demonstrated whether the interactions could happen. Four interaction complexes were highlighted due to the interaction energy criteria, ΔEBind, ΔH and ΔG < 0.00 kcal mol-1, that is: Hm1 Pb (1); Hm2 Pb (3); Hm2 Cd (4) and Hm2 Hg (4) and the results show that they occur through physisorption. In structural parameter studies, interaction distances smaller than 3000 Å were identified, which ranged from 2.253 Å to 2.972 Å. From the analysis of the topological parameters of QTAIM, it was possible to characterize the intensities of the interactions, as well as their nature, which were partially covalent or electrostatic in nature. Finally, based on the theoretical results, it can be affirmed that the hemicellulose can interact with Cd2+, Pb2+ and Hg2+ ions, evidencing that this study can support further experimental essays to remove contaminants from effluents.

RESUMO
In this work, silica nanospheres were used as support for gold nanoparticles and applied for bisphenol A electrochemical detection. The development of new silica-supported materials has attracted increasing attention in the scientific world. One approach of interest is using silica nanospheres as support for gold nanoparticles. These materials have a variety of applications in several areas, such as electrochemical sensors. The obtained materials were characterized by solid-state UV-vis spectroscopy, electron microscopy, X-ray diffraction, and electrochemical techniques. The electrode modified with AuSiO2700/CHI/Pt was applied as an electrochemical sensor for BPA, presenting an oxidation potential of 0.842 V and a higher peak current among the tested materials. The AuSiO2700/CHI/Pt electrode showed a logarithmic response for the detection of BPA in the range of 1-1000 nmol L-1, with a calculated detection limit of 7.75 nmol L-1 and a quantification limit of 25.8 nmol L-1. Thus, the electrode AuSiO2700/CHI/Pt was presented as a promising alternative to an electrochemical sensor in the detection of BPA.

Collagen is considered a nutraceutical, and its consumption has been expanding due to the increased life expectancy, rising per capita income, and increased consumer awareness of health care. This study aimed to evaluate consumers' perceptions, knowledge, attitudes, and practices about the consumption of collagen-based products by using an online questionnaire and to correlate them with socio-economic data. A market survey (pharmacy stores and online) was also conducted to evaluate the available products. In total, 275 participants answered the survey, 73.3% from the Southeast region, mostly female (84.0%). Most participants reported three months as the period of collagen intake (31.6%), and the consumption period was associated with the perception of the health benefits (p < 0.001). Furthermore, the participants' knowledge and perceptions regarding collagen intake are frequently associated with dermatological and orthopedic changes. Collagen-based products supplementation is a growing market with a broad target audience (genders, age groups, and socio-economic levels). The commercial presentation of collagen has been diversified over the years, and powder collagen is the most consumed (52.7%) and cheapest compared with capsules, pills, or gummies. The results of the present study demonstrate that most consumers of this type of supplement associate its benefits with aesthetic care such as skin, hair, and nails, although the scientific literature has shown its effects in treating osteoarticular diseases, for example. Undoubtedly, the correct dose prescription, treatment time, and choice of product presentation must be analyzed carefully, as they significantly impact treatment results.

Health care-associated infections (HAIs) contribute to a significant rate of morbidity, mortality, and financial burden on health systems. These infections are caused by multidrug-resistant bacteria that produce biofilm as the main virulence factor. This study aimed to evaluate the effect of the copper-based metallic compounds [Cu(phen)(pz)NO2]Cl (I), [Cu(bpy)(pz)(NO2)]Cl (II), and [Cu(phen)(INA)NO2]Cl (III), where phen = phenanthroline, bpy = bipyridine, pz = pyrazinamide, and INA = isonicotinic acid, against planktonic cells and biofilms formation of Staphylococcus aureus, Staphylococcus epidermidis, and Escherichia coli. The susceptibility of the microorganisms was evaluated by minimum inhibitory concentration (MIC), minimum bacterial concentration (MBC), and time-kill curve assay on planktonic cells. The biofilm formation was evaluated by biomass quantification through staining with crystal violet (CV), colony-forming units (CFUs) quantification, and biofilm metabolic activity determination by XTT assay. The compounds showed bacteriostatic and bactericidal activity on all microorganisms analyzed. Regarding the antibiofilm activity, all metallic compounds were able to reduce significantly the biofilm biomass, colony-forming units, and the metabolic activity of remaining cells, varying the efficient concentration according to the strain analyzed. Interestingly, compounds (I), (II) and (III) did not exhibit DNA degradation activity even with up to 100 µM of these metal complexes. On the other hand, complexes (I) and (III) showed a remarkable capacity to cleave DNA upon addition of glutathione, a reducing agent (CuII/CuI) that leads to reactive oxygen species (ROS) formation. The results presented in this study showed promising antimicrobial and antibiofilm effects.

Our objective was to evaluate disordered eating and associated factors in university students in the early period of the COVID-19 pandemic in Brazil. This is an analytical cross-sectional observational study, and data collection took place between May and June 2020, using an online semi-structured questionnaire. In total, 936 students were evaluated. Present signs/symptoms of the flu syndrome (OR = 1.605), negative mood changes (OR = 1.628), weight gain (OR = 1.739), obesity (OR = 3.089), follow fitness/health profile on social media (OR = 2.050), having inadequate body perception (OR = 2.416), and body dissatisfaction (OR = 2.612) increased the chances of presenting a higher score on the disordered eating scale.

Introdução - Alinhada à vertente dos estudos críticos da saúde global, esta tese explora a interface entre o conceito de segurança humana e a saúde do trabalhador no contexto da pandemia de covid-19. Baseada em revisão de literatura narrativa, pesquisa documental e pesquisa empírica, a tese elegeu como recorte geográfico as cidades de São Paulo (Brasil) e Medellín (Colômbia). Objetivo - Apresentar a análise da percepção da segurança humana em relação à realização do trabalho de Manicure em São Paulo e Medellín. Método - O estudo de caso enquanto método de investigação qualitativo foi realizado com manicures. Foram realizadas dez entrevistas semiestruturadas em São Paulo no mês de dezembro de 2021, com profissionais manicures, de forma voluntária, atuando em salão de beleza, residência própria e/ou atendimento em domicílio. Em Medellín, foram aplicados dois questionários no formato online, em função da impossibilidade de deslocamento por conta das incertezas decorrentes da pandemia de covid-19 e suas variantes. Resultados - A partir dos dados coletados, foram analisadas as respostas obtidas nas entrevistas realizadas com as manicures, a fim de avançar na identificação de variáveis geradoras de possíveis inseguranças no ambiente de trabalho. Conclusão - Concluiu-se pela pertinência das hipóteses de que a saúde do trabalhador é um elemento fundamental da percepção da segurança ou insegurança humana pelas trabalhadoras entrevistadas; o histórico de experiências laborais passadas interfere na percepção de segurança humana quando da realização de um trabalho ou atividade; e a percepção de segurança humana pelo indivíduo é influenciada pelo contexto do ambiente laboral no qual ele está inserido.
Introduction - In line with critical studies of global health, this thesis explores the interface between the concept of human safety and workers' health in the context of the covid-19 pandemic. Based on a narrative literature review, documental research and empirical research, the thesis chose the cities of São Paulo (Brazil) and Medellín (Colombia) as a geographic focus. Objective - To present the analysis of the perception of human safety in relation to the performance of manicure work in São Paulo and Medellín. Method - The case study as a qualitative research method was carried out with Manicures. Ten semi-structured interviews were carried out in São Paulo in December 2021, with Manicure professionals, on a voluntary basis, working in a beauty salon, their own residence and/or home care. In Medellín, two online questionnaires were applied, due to the impossibility of travel due to the uncertainties arising from the covid-19 pandemic and its variants. Results - From the collected data, the answers obtained in the interviews with the manicurists were analyzed, achieving a better understanding of the variables that generate possible insecurities in the work environment. Conclusion - it was concluded that the hypotheses that the worker's health is a fundamental element of the perception of human safety or insecurity by the interviewed workers were relevant; that the history of past work experiences interferes with the perception of human safety when carrying out a job or activity; and that the perception of human security by the individual is influenced by the context of the work environment in which he is inserted.

OBJECTIVE: Assessing the regular consumption of ultra-processed foods by children at 24 months of age from the 2015 Pelotas Birth Cohort and the main demographic, socioeconomic, and behavioral factors related to the consumption of these products. METHODS: Population-based cohort in the city of Pelotas, RS, where 4,275 children were assessed at birth and 95.4% of them were followed up until 24 months of age. Food consumption was assessed by a questionnaire on regular consumption of ultra-processed foods, which collected information regarding sex, household income, maternal skin color, schooling level, and age, the child attending day care and having siblings, breastfeeding status, and obesity. The outcome was the sum of ultra-processed foods regularly consumed by a child. A multivariate Poisson regression analysis was used to calculate the association between the regular consumption of ultra-processed foods and exposure variables. RESULTS: The mean number of ultra-processed foods consumed was 4.8 (SD = 2.3). The regular consumption of ultra-processed foods was positively associated with black skin color and having siblings, and negatively associated with household income and maternal schooling level and age. CONCLUSION: The mean regular consumption of ultra-processed foods by children from the 2015 Pelotas Birth Cohort is high, which can negatively affect the children's diet. The risk of consuming this kind of food was higher among children from families of lower socioeconomic status, whose mothers present lower education level, black skin color, and younger age.

ABSTRACT Purpose: The purpose of this study was to investigate the postoperative outcomes and evaluate the success predictors of phacoemulsification with Kahook Dual Blade goniotomy for cataract and glaucoma management in eyes with primary open-angle glaucoma. Methods: This was a retrospective, non-comparative; inter ventional case series in which all patients with primary open-angle glaucoma who underwent phacoemulsification with Kahook Dual Blade goniotomy between June 2018 and April 2019 were enrolled. All the participants had a minimum follow-up period of 6 months. Preoperative and postoperative intraocular pressure values (at 1, 3, and 6 months), number of antiglaucoma medications, best-corrected visual acuity, surgical complications, and any subsequent related events or procedures were recorded. A logistic regression analysis was performed to investigate the association between the different variables and surgical outcomes. Results: A total of 47 patients (57 eyes) were included (mean age, 70.5 ± 7 years). The mean intraocular pressure was reduced from 15.5 ± 4.2 mmHg to 12.2 ± 2.4 mmHg at the last follow-up visit (p<0.001). The mean number of antiglaucoma medications decreased significantly from 1.9 ± 1.0 to 0.6 ± 1.0 during the same period (p<0.001). On the basis of the predefined criterion (intraocular pressure reduction ≥20% and/or reduction ≥1 medication), the 6-month success rate was 86%. A higher preoperative intraocular pressure value (odds ratio [OR]= 2.01; p=0.016) and greater percentage of initial (30 days) intraocular pressure reduction (OR= 1.02; p=0.033) were sig nificantly associated with surgical success. Conclusion: Our findings suggest that phacoemulsification with Kahook Dual Blade goniotomy is an effective and safe alternative for cataract management in eyes with primary open-angle glaucoma that positively impacts intraocular pressure control and medication burden. Eyes with higher baseline intraocular pressure and a more pronounced initial response to the procedure appeared to present better outcomes at 6 months. Further studies are needed to evaluate the long-term efficacy and safety profile of the procedure.
RESUMO Objetivo: Investigar os resultados pós-operatórios e avaliar os preditores de sucesso da facoemulsificação combinada à goniotomia com o Kahook Dual Blade para o tratamento da catarata e do glaucoma em olhos com glaucoma primário de ângulo aberto. Métodos: Série de casos retrospectivos, não comparativos e intervencionistas, em que todos os pacientes com glaucoma primário de ângulo aberto submetidos ao procedimento de facoemulsificação combinada à goniotomia com o Kahook Dual Blade entre junho de 2018 e abril de 2019 foram inscritos. Todos os participantes tiveram um acompanhamento mínimo de 6 meses. Foram registrados os valores de pressão intraocular pré e pós-operatória (em 1, 3 e 6 meses), número de medicamentos antiglaucomatosos, melhor acuidade visual corrigida, complicações cirúrgicas e quaisquer eventos ou procedimentos subsequentes relacionados. A análise de regressão logística foi usada para investigar a associação entre diferentes variáveis e resultados cirúrgicos. Resultados: Um total de 57 olhos de 47 pacientes foram incluídos (média de idade, 70,5 ± 7 anos). A pressão intraocular média reduziu de 15,5 ± 4,2 mmHg para 12,2 ± 2,4 mmHg na última visita de acompanhamento (p<0,001). O número médio de medicamentos antiglaucomatosos diminuiu significativamente de 1,9 ± 1,0 para 0,6 ± 1,0 durante o mesmo período (p<0,001). Com base no critério predefinido (redução da pressão intraocular ≥20% e/ou redução de ≥1 medicamento), a taxa de sucesso em 6 meses foi de 86%. Um valor de pressão intraocular pré-operatório mais alto (OR= 2,01; p=0,016) e maior porcentagem de redução da pressão intraocular inicial (30 dias) (OR= 1,02; p=0,033) foram significativamente associados ao sucesso cirúrgico. Conclusão: Nossos resultados sugerem que o procedimento de facoemulsificação combinada à goniotomia com o Kahook Dual Blade é uma alternativa eficaz e segura para o manejo da catarata em olhos com glaucoma primário de ângulo aberto, impactando positivamente no controle da pressão intraocular e no número de medicamentos. Olhos com pressão intraocular basal mais alta e resposta inicial mais pronunciada ao procedimento parecem apresentar melhores resultados em 6 meses. Mais estudos são necessários para avaliar a eficácia em longo prazo e o perfil de segurança.

Dengue fever is a clinical manifestation of dengue virus (DENV) infection well defined by the intense host immune response with the development of high fever, anorexia, headache and muscle pain. Several immune mediators are involved in the pathophysiology of DENV infection, in which polymorphisms in immune molecule genes contribute with the susceptibility and severity of the infection. Several meta-analyses are available with significant findings in the association between genetic variants in immune-mediator genes and dengue, though the results may be false positive. Hence, to solve this issue, we have performed a systematic revaluation with Bayesian approaches to verify the false positive rate in these results. A systematic search was performed for meta-analytic studies on the aforementioned issue. The calculations of false positive report probability (FPRP) and the Bayesian false-discovery probability (BFDP) at the prior probability of 10-3 and 10-6 have been performed. To verify the methodological quality of the studies included, the evaluation by the Venice criteria was applied. In addition, gene-gene and protein-protein networks were designed. As results, seven meta-analyses on genetic variants in several immune-inflammatory mediator genes and DENV infection comprise the results. Only the polymorphisms in the TNF, MICB, PLCE1, VDR, CD32 and HLA-A genes were considered as noteworthy. There was a heterogeneity profile for the results on Venice criteria indicating variability in the methodological quality. The gene-gene and protein-protein networks showed these immune mediators as relevant players in the disease. We suggest these polymorphisms as potential biomarkers for the pathogenesis and immune response against DENV.

The study aimed to estimate the prevalence of delay in the three doses of quadrivalent vaccine (DTP+Hib) in children 12 to 23 months of age in Brazil, based on data from the Brazilian National Health Survey (PNS) of 2013 and to analyze the delay in each of the doses according to sociodemographic variables and use of health services and public health interventions. The data are from the PNS a cross-sectional study performed in 2013. The outcome was delay in at least one of the three doses of the quadrivalent vaccine. Delay was defined as a dose received at least 30 days after the recommended date according to information on the child's vaccination card. Prevalence of delay was analyzed according to sociodemographic variables and use of health services. A descriptive analysis was performed to obtain absolute and relative frequencies and their respective 95% confidence intervals. Of the 2,016 children with information collected, 1,843 were analyzed. The prevalence of delay in at least one dose of the vaccine was 44%. There was a delay of 14.8% in the first dose, 28.8% in the second, and 45.4% in the third, and 10% of the children had delays in all three doses. Higher prevalence of delay was associated with male gender, brown skin color, the poorest income quintile, and residence in rural areas and the North of Brazil. The study revealed high prevalence of delay with the quadrivalent vaccine (DTP+Hib) in children 12 to 23 months of age in Brazil, with the highest delay in the third dose.
O objetivo foi estimar a prevalência do atraso nas três doses da vacina tetravalente (DTP+Hib) em crianças de 12 a 23 meses de idade, no Brasil, por meio dos dados da Pesquisa Nacional de Saúde (PNS) de 2013 e descrever o atraso em cada uma das doses segundo variáveis sociodemográficas, utilização de serviços e intervenções públicas de saúde. Foram utilizados dados da PNS, estudo transversal realizado em 2013. O desfecho foi o atraso pelo menos em uma das três doses da vacina tetravalente. Considerou-se como atraso a dose recebida pelo menos 30 dias após a data preconizada, segundo informação da caderneta de vacinação. A prevalência do atraso foi descrita segundo variáveis sociodemográficas e utilização de serviços de saúde. Realizou-se análise descritiva obtendo-se frequências absolutas e relativas e seus respectivos intervalos de 95% de confiança. Das 2.016 crianças com informações coletadas, 1.843 foram analisadas. A prevalência de atraso de pelo menos uma dose da vacina foi de 44%. Observou-se atraso de 14,8% na primeira, 28,8% na segunda e 45,4% na terceira dose, sendo que 10% das crianças tiveram atraso nas três doses. Maiores prevalências de atraso foram encontradas em crianças do sexo masculino, de cor da pele parda, pertencentes ao quintil mais pobre de riqueza, moradores da zona rural e da Região Norte do Brasil. Evidenciou-se alta prevalência de atraso na vacina tetravalente (DTP+Hib) em crianças de 12 a 23 meses do Brasil, sendo maior na terceira dose.
El objetivo fue estimar la prevalencia del atraso en las tres dosis de la vacuna tetravalente (DTP+Hib) en niños de 12 a 23 meses de edad, en Brasil, mediante los datos de la Encuesta Nacional de Salud (PNS) de 2013 y describir el retraso en cada una de las dosis, según variables sociodemográficas, utilización de servicios e intervenciones públicas de salud. Se trata de un estudio transversal, realizado en 2013, con datos de la PNS. El resultado fue el retraso por lo menos en una de las tres dosis de la vacuna tetravalente. Se consideró como un atraso la dosis recibida por lo menos 30 días tras la fecha prefijada, según la información de la cartilla de vacunación. La prevalencia del atraso fue descrita según variables sociodemográficas y utilización de servicios de salud. Se realizó un análisis descriptivo, obteniéndose frecuencias absolutas y relativas, así como sus respectivos intervalos de 95% de confianza. De los 2016 niños con información recogida, se analizaron 1843. La prevalencia de atraso de por lo menos una dosis de la vacuna fue de un 44%. Se observó un retraso de 14,8% en la primera, un 28,8% en la segunda y un 45,4% en la tercera dosis, siendo que un 10% de los niños sufrieron atraso en las tres dosis. Las mayores prevalencias de atraso se encontraron en niños de sexo masculino, mestizos, pertenecientes al quintil más pobre de riqueza, habitantes de la zona rural y de la Región Norte de Brasil. Se evidenció una alta prevalencia de atraso en la vacuna tetravalente (DTP+Hib) en niños de 12 a 23 meses de Brasil, siendo mayor en la tercera dosis.

Most of the patients infected with Chikungunya virus (CHIKV) develop chronic manifestations characterized by pain and deformity in joints, impacting their quality of life. The aminoadamantanes, in their turn, have been exploited due to their biological activities, with amantadine and memantine recently described with anti-CHIKV activities. Here we evaluated the antiviral activity of rimantadine hydrochloride (rtdH), a well-known antiviral agent against influenza A, its platinum complex (Pt-rtd), and the precursor cis-[PtCl2(dmso)2], against CHIKV infection in vitro. The rtdH demonstrated significant antiviral activity in all stages of CHIKV replication (29% in pre-treatment; 57% in early stages of infection; 60% in post-entry stages). The Pt-rtd complex protected the cells against infection in 92%, inhibited 100% of viral entry, mainly by a virucidal effect, and impaired 60% of post-entry stages. Alternatively, cis-[PtCl2(dmso)2] impaired viral entry in 100% and post-entry steps in 60%, but had no effect in protecting cells when administered prior to CHIKV infection. Collectively, the obtained data demonstrated that rtdH and Pt-rtd significantly interfered in the early stages of CHIKV life cycle, with the strongest effect observed to Pt-rtd complex, which reduced up to 100% of CHIKV infection. Moreover, molecular docking analysis and infrared spectroscopy data (ATR-FTIR) suggest an interaction of Pt-rtd with CHIKV glycoproteins, potentially related to the mechanism of inhibition of viral entry by Pt-rtd. Through a migration retardation assay, it was also shown that Pt-rtd and cis-[PtCl2(dmso)2] interacted with the dsRNA in 87% and 100%, respectively. The obtained results highlight the repurposing potential of rtdH as an anti-CHIKV drug, as well as the synthesis of promising platinum(II) metallodrugs with potential application for the treatment of CHIKV infections. Importance Chikungunya fever is a disease that can result in persistent symptoms due to the chronic infection process. Infected patients can develop physical disability, resulting and high costs to the health system and significant impacts on the quality of life of affected individuals. Additionally, there are no licensed vaccines or antivirals against the Chikungunya virus (CHIKV) and the virus is easily transmitted due to the abundance of viable vectors in epidemic regions. In this context, our study highlights the repurposing potential of the commercial drug rimantadine hydrochloride (rtdH) as an antiviral agent for the treatment of CHIKV infections. Moreover, our data demonstrated that a platinum(II)-rimantadine metallodrug (Pt-rtd) poses as a potent anti-CHIKV molecule with potential application for the treatment of Chikungunya fever. Altogether, rtdH and Pt-rtd significantly interfered in the early stages of CHIKV life cycle, reducing up to 100% of CHIKV infection in vitro.

Contamination by heavy metal ions, particularly in water resources, is a severe environmental problem. In this study, the interaction of metal ions, namely, Cadmium Cd(II), Mercury Hg(II), and Lead Pb(II), on lignin matrices was investigated based on theoretical calculations. Binding energy (ΔEBind) values proved that the Pb(II) interacted better with lignin matrices than Cd(II) or Hg(II), having energy values between -8.4 kcal mol-1 to -20.2 kcal mol-1. The Gibbs energy (ΔG) and enthalpy (ΔH) values for Pb(II) were <0, indicating that the process was spontaneous and released heat. However, the lignin matrices studied in this work did not interact efficiently with Cd(II) and Hg(II) ions because almost all ΔEBind, ΔG, and ΔH values were positive. The bond length of the interaction proved that the Pb ions yielded the smallest values, ratifying the values for the interaction energy. Analyses based on the quantum theory of atoms in molecules showed that the interactions between Pb(II) and the matrices were partially covalent, whereas the interactions of Cd(II) and Hg(II) were predominantly electrostatic, justifying the positive values of ΔEBind, ΔG, and ΔH. The natural bond orbital results showed that the ligand orbitals of the matrix interacted with the lone pair antibonding orbital (LP*) of the metal ions. The theoretical results of the study show the possibility of applying lignin to remove heavy metal ions, especially Pb, and providing information for research related to wastewater treatment.

OBJECTIVE: The aim was to evaluate the antibacterial and antibiofilm activity of natural (n-CNSL) and technical (t-CNSL) cashew nut shell liquid against streptococci and enterococci related to dental caries and chronic apical periodontitis, respectively. MATERIAL AND METHODS: Minimum inhibitory concentrations (MIC) and minimal bactericidal concentration (MBC) were determined to assess the antimicrobial effect of both CNSLs (n-CSNL and t-CNSL) against S. oralis ATCC 10557, S. sobrinus ATCC 6715, S. parasanguinis ATCC 903, S. mutans UA 159 and E. faecalis ATCC 19433. The antibiofilm activity was evaluated by total biomass quantification, colony forming unit (CFU) counting and scanning electron microscopy (SEM). Furthermore, cytotoxic effect of the substances was evaluated on L929 and HaCat cell lines by MTS assay. RESULTS: The n-CNSL and t-CNSL showed inhibitory and bactericidal effect against all strains tested in this study, with MIC and MBC values ranging from 1.5 to 25 µg/mL. Overall, both CNSLs showed significant reduction in biomass quantification and enumeration of biofilm-entrapped cells for the strains analyzed, in biofilm formation and preformed biofilms (p < 0.05). In biofilm inhibition assay, the t-CNSL and n-CNSL showed reduction in biomass and CFU number for all bacteria, except in cell viability of S. parasanguinis treated with t-CNSL (p > 0.05). Indeed, SEM images showed a reduction in the amount of biomass, bacterial cells and changes in cellular morphology of S. mutans. CONCLUSION: In conclusion, both substances showed effective antibacterial and antibiofilm activity against the strains used in the study, except in viability of S. parasanguinis cells treated with t-CNSL.

ABSTRACT OBJECTIVE Assessing the regular consumption of ultra-processed foods by children at 24 months of age from the 2015 Pelotas Birth Cohort and the main demographic, socioeconomic, and behavioral factors related to the consumption of these products. METHODS Population-based cohort in the city of Pelotas, RS, where 4,275 children were assessed at birth and 95.4% of them were followed up until 24 months of age. Food consumption was assessed by a questionnaire on regular consumption of ultra-processed foods, which collected information regarding sex, household income, maternal skin color, schooling level, and age, the child attending day care and having siblings, breastfeeding status, and obesity. The outcome was the sum of ultra-processed foods regularly consumed by a child. A multivariate Poisson regression analysis was used to calculate the association between the regular consumption of ultra-processed foods and exposure variables. RESULTS The mean number of ultra-processed foods consumed was 4.8 (SD = 2.3). The regular consumption of ultra-processed foods was positively associated with black skin color and having siblings, and negatively associated with household income and maternal schooling level and age. CONCLUSION The mean regular consumption of ultra-processed foods by children from the 2015 Pelotas Birth Cohort is high, which can negatively affect the children's diet. The risk of consuming this kind of food was higher among children from families of lower socioeconomic status, whose mothers present lower education level, black skin color, and younger age.
RESUMO OBJETIVO Avaliar o consumo habitual de alimentos ultraprocessados aos 24 meses de idade por crianças pertencentes à Coorte de Nascimentos de Pelotas de 2015 e os principais fatores demográficos, socioeconômicos e comportamentais relacionados ao consumo desses produtos. MÉTODOS Coorte de base populacional na cidade de Pelotas-RS, onde foram avaliadas 4.275 crianças ao nascimento, das quais 95,4% foram acompanhadas até os 24 meses. O consumo alimentar foi avaliado por meio de um questionário de consumo habitual de alimentos ultraprocessados, coletando informações sobre sexo, renda familiar, cor da pele, escolaridade e idade da mãe, frequentar creche, ter irmãos, status de amamentação e obesidade. O desfecho foi a somatória de alimentos ultraprocessados consumidos habitualmente pela criança. Análise multivariada por regressão de Poisson foi utilizada para estimar a associação entre consumo habitual de alimentos ultraprocessados e as variáveis de exposição. RESULTADOS O número médio de alimentos ultraprocessados consumidos habitualmente foi de 4,8 (DP = 2,3). O consumo habitual de alimentos ultraprocessados foi associado positivamente à cor da pele preta e ter irmãos e negativamente associado com renda familiar, escolaridade e idade materna. CONCLUSÕES A média de consumo habitual de alimentos ultraprocessados por crianças pertencentes à Coorte de Nascimentos de 2015 da cidade de Pelotas é elevada, o que pode causar um efeito negativo na dieta das crianças. O risco de consumo desses alimentos foi maior entre crianças de famílias de menor posição socioeconômica, filhas de mães de baixa escolaridade, de cor da pele preta, mais jovens e de baixa renda.
