RESUMO

AIMS: We evaluated the role of intergenerational paternal exercise on fibrosis, inflammatory profile, and redox status in the adipose tissue of male rat offspring fed with high-fat diet (HFD) and explored to what extent programming affects the systemic metabolic profile. MAIN METHODS: Adult wistar rats were randomly divided into two groups: sedentary fathers and trained fathers (8 weeks of resistance training (RT), three times per week). The offspring were obtained by mating with sedentary females. Upon weaning, male offspring were divided into four groups (7 animals per group): offspring of sedentary fathers exposed to either a control diet (SFO-C) or a high-fat diet (SFO-HF); offspring of trained fathers exposed to a control diet (TFO-C) or a high-fat diet (TFO-HF). KEY FINDINGS: Paternal RT was effective in attenuating body weight gain, adipocyte size, collagen deposition, as well as downregulating genes (CTGF, VEGF, C/EBPα SREBP1, MCP-1, and NF-kB), pro-inflammatory cytokine levels (Tumor Necrosis Factor alpha and Interleukin-1-beta), matrix metalloproteinase -2 activity, and ROS production in the epididymal adipose tissue of offspring fed with HFD (TFO-HF vs. SFO-HF; P < 0.05). Moreover, paternal RT increased adiponectin and superoxide dismutase (SOD) activity in the tissue. These beneficial effects were accompanied by the increase of antioxidant enzymes (SOD and α-Klotho), while decreasing pro-oxidant agents (F2-isoprostanes, protein carbonyls levels), and metabolic markers (insulin and leptin, HOMA-ß, and HOMA-IR) in the offspring blood circulation. SIGNIFICANCE: Our findings reveal protective effects of intergenerational paternal RT on adipose tissue remodeling and metabolic health of offspring fed with HFD.

RESUMO

Although some studies have shown that a high-fat diet (HFD) adversely affects muscle extracellular matrix remodeling, the mechanisms involved in muscle trophism, inflammation, and adipogenesis have not been fully investigated. Thus, we investigated the effects of 8 weeks of paternal resistance training (RT) on gene and protein expression/activity of critical factors involved in muscle inflammation and remodeling of fathers and offspring (offspring exposed to standard chow or HFD). Animals were randomly distributed to constitute sedentary fathers (SF; n = 7; did not perform RT) or trained fathers (TF n = 7; performed RT), with offspring from mating with sedentary females. After birth, 28 male pups were divided into four groups (n = 7 per group): offspring from sedentary father submitted either to control diet (SFO-C) or high-fat diet (SFO-HF) and offspring from trained father submitted to control diet (TFO-C) or high-fat diet (TFO-HF). Our results show that an HFD downregulated collagen mRNA levels and upregulated inflammatory and atrophy pathways and adipogenic transcription factor mRNA levels in offspring gastrocnemius muscle. In contrast, paternal RT increased MMP-2 activity and decreased IL-6 levels in offspring exposed to a control diet. Paternal RT upregulated P70s6k and Ppara mRNA levels and downregulated Atrogin1 mRNA levels, while decreasing NFκ-B, IL-1ß, and IL-8 protein levels in offspring exposed to an HFD. Paternal physical training influences key skeletal muscle remodeling pathways and inflammatory profiles relevant for muscle homeostasis maintenance in offspring submitted to different diets.

RESUMO

BACKGROUND: Repeated measures analysis of covariance and three-way analysis of variance with repeated measures are common statistical methods. For a valid interpretation of blood pressure (BP) response to exercise, a variety of additional statistical methods must be implemented. Four additional statistical methods are presented: technical error of measurement (SEM), smallest real difference (SRD), magnitude-based inference and mixed effect modeling technique (MEM). The aim of this perspective article is to demonstrate how to apply already known statistical analyses regarding BP responsiveness in order to improve interpretation and achieve higher reliability for future studies in exercise science. METHODS: A total of 27 hypertensive older women (aged 68.37 ± 5.55 years) participated in the present study. A whole-body resistance training (RT) program was performed on two nonconsecutive days per week for 10 weeks. BP was monitored during the 10-week RT intervention and after 15 weeks of detraining. First, individuals were classified as high and low responders, then statistical methods to analyze data included the use of SEM, SRD, magnitude-based inference and MEM. RESULTS: When magnitude-based inference was used to classify responsiveness, most participants displayed a trivial response. Decrements in SBP between 1 and 10 mmHg were not clinically meaningful but fell within the measurement error of the SBP measurements. Baseline SBP and time of training predicted post-SBP response. CONCLUSION: Changes over time and declines in SBP might not be a SRD and fell in the SEM. Moreover, SBP responsiveness was the result of inappropriate control of covariates such as period of training.

RESUMO

BACKGROUND AND AIM: Growth of elderly population is a worldwide phenomenon that impacts public health. The objective of this study was to compare the pain levels, strength, and quality of life among elderly obese with diabetes or hypertension. MATERIALS AND METHODS: The study cohort comprised 52 obese elderly subjects with hypertension (n = 35) and diabetes (n = 17). The parameters measured were anthropometric features, handgrip strength, visual analog scale for pain, and quality of life using the World Health Organization questionnaire. RESULTS: The level of pain reported by obese hypertensive elderly subjects (5.3 ± 3.4) was lower than reported by obese diabetic elderly subjects (7.4 ± 2.4). Obese hypertensive elderly scored higher on quality of life (sensory functioning and past, present, and future [PPF] activities) than obese diabetic elderly. No differences were observed for the other parameters. Strength, pain, anthropometrics, and hemodynamics were not correlated to quality of life. CONCLUSIONS: Obese elderly diabetics exhibit worse pain scores, sensorial abilities, and PPF activities than obese hypertensive elderly individuals. RELEVANCE FOR PATIENTS: The difference in pain and quality of life aspects between obese elderly individuals with hypertension and diabetes should be accounted for in health-care programs designed for these individuals.

RESUMO

ABSTRACT Introduction Aging is a natural process and marked by changes and adaptations of both a biological and physiological nature. As regards adaptations, there are numerous works that address these responses following various types of training programs. Resistance training (RT) can be assessed by biochemical parameters such as creatine kinase (CK), which is a major marker of stress in the skeletal muscles. C-reactive protein (CRP) is a biochemical marker used to assess damage to the cardiac muscles. Objective To evaluate the influence of CK on CRP in elderly female subjects undergoing RT. Methods An experimental study was carried out with 10 elderly women (61 ± 1.8 years). Peripheral venous blood was collected for the CK and CRP analysis both before and 24 hours after 8 weeks of RT. Anthropometric measurements involved BMI (Body Mass Index), WHR (waist-to-hip ratio) and body composition. The RT involved combined series - Bi-Set. For statistical analysis, the Shapiro-Wilk normality test was conducted first and presented p >0.05, confirming the use of parametric tests. Group variables were presented as mean and standard deviation. To compare the load-dependent samples, the repeated measures one-way ANOVA was performed first, followed by Tukey's post hoc test. For CK and CRP variables, we conducted the paired Student's t-test for the timepoints pre- and post-eight weeks of RT along with the one-way ANOVA test, also conducting Tukey's post-test when necessary. The level of significance was set at p ≤0.05. Results There was a statistically significant decrease in both serum CK and CRP, which indicated a reduction of 73.14% and 75%, respectively. Conclusion Long-term RT promoted influences among biomarkers assessed through skeletal muscle (CK) and cardiac muscle (CRP) damage, determining adaptation and muscle remodeling in any age group. Level of evidence II, Investigation of treatment results.

Abstract The detraining effects on cardiovascular parameters in the elderly women (EW) were evaluated. EW underwent 8 weeks of resistance training, followed by eight weeks of detraining. 9 EW were evaluated (age 62 ± 2.30). Peripheral venous blood collections and body composition evaluation were performed before training, after training and after the detraining phase. There was no significant increase in CRP in detraining phase, but we observed negative changes for total cholesterol and body composition, represented by fat weight. The results suggest that 8 weeks of detraining did not significantly increase CRP, but had a negative influence on other parameters related to cardiovascular risks, as anthropometric and biochemical values represented by total cholesterol and fat mass, respectively. In fact, continuity of physical training is essential to acquiring and maintaining good health, otherwise the beneficial adaptations achieved will return to initial values.

PURPOSE: The purpose of the present study was to identify the variability of blood pressure response to a 10-week resistance training (RT) program in hypertensive and normotensive elderly women. PARTICIPANTS AND METHODS: Twenty-seven untrained hypertensive and 12 normotensive elderly women participated in the present study. A whole-body RT program was performed on two nonconsecutive days per week for 10 weeks. The responsiveness of resting systolic blood pressure (SBP) was determined based on the percent decline between the pre- and post-training time points T1 and T4. The term responders were used to describe subjects who exhibited a percent SBP decline ≥-2.58% and the term nonresponders for subjects who exhibited a percent SBP decline <-2.58%, respectively. RESULTS: Both the responders and nonresponders in the hypertensive group presented significant changes in SBP (-7.83 ± 5.70 mmHg vs 3.78 ± 7.42 mmHg), respectively. Moreover, the responders and nonresponders in the normotensive group presented significant changes in SBP as well (-8.58 ± 5.52 mmHg vs 5.71 ± 3.84 mmHg). CONCLUSION: SBP presents a heterogeneous response to a controlled RT program in hypertensive and normotensive elderly women. A different modality of training and additional therapies should be used for nonresponders in order to decrease resting SBP.

RESUMO

Prestes, J, Nascimento, DdC, Neto, IVS, Tibana, RA, Shiguemoto, GE, Perez, SEA, Botero, JP, Schoenfeld, BJ, and Pereira, GB. The effects of muscle strength responsiveness to periodized resistance training on resistin, leptin, and cytokine in elderly postmenopausal women. J Strength Cond Res 32(1): 113-120, 2018-The aim of this study was to evaluate muscle strength responsiveness and to determine whether interleukin 4 (IL-4), leptin, and resistin would be affected by the individual pattern of response to a resistance training (RT) periodization program. Twenty-six elderly postmenopausal women participated in the present study (mean age = 62.57 ± 6.69 years and body mass index = 28.09 ± 4.83 kg·m). Four-month longitudinal training program was performed consisting of 2 whole-body sessions per week with increasing intensity and decreasing volume by using 6-14 repetitions maximum (RM). Two acute whole-body RT sessions (before and after chronic training) were also performed comprising 3 sets of 12-14RM. The responsiveness was determined based on their relative muscle strength gains in 45° leg press. High responders were defined as relative muscle strength gains ≥32% and low responders <32% (n = 13 in each group). Muscle strength increased by a higher amount in the high-responsive group as compared with the low-responsive group in the 45° leg press and bench press, whereas muscle strength increased for both groups when compared with pretraining evaluation (p = 0.001). Low responders displayed higher plasma leptin levels when compared with high responders at baseline (p = 0.001), and after 16 weeks of training, leptin levels were significantly lower when compared with baseline (pretraining period). Both groups displayed a decrease in baseline resistin values after 16 weeks of RT, but only a statistically simple main effect was observed for low responders. In addition, there were no effect of time and no significant interaction between the responsiveness and time on IL-4 concentration. In conclusion, RT is effective in improving upper and lower limb muscle strength in elderly women, with higher magnitudes of increase seen for those classified as high responders. Resistin and leptin displayed a decrease over time, regardless of responsiveness classification.

RESUMO

Abstract The aim of the present study was to compare differences in heart rate (HR) response during and following exercise in obese older women with different percent body fat levels. Ninety older, obese women aged 60-87 years participated in the study, were categorized, and enrolled to one of two groups based on a lower percent body fat (LPBF ≤ 41.10 %) or higher percent body fat (HPBF > 41.10 %) as measured by dual-energy x-ray absorptiometry. The peak HR during exercise and in the first and second minutes of recovery period were compared between groups. The HPBF group presented a lower peak HR during exercise (p =.001) and an impaired HR recovery (p =.001) when compared to LPBF group. The present study demonstrated that older women who were in exceedingly obese level have an impaired heart rate response during exercise and in the recovery period, indicating possible autonomic dysfunction.

ABSTRACT Objective Patients infected with the human immunodeficiency virus (HIV) have an increased risk of metabolic disorders and alterations on irisin levels. Therefore, the purpose of the current investigation was to quantify the circulating irisin concentration in HIV-infected subjects under highly active antiretroviral therapy and to determine possible correlations between irisin levels with fat mass, fat-free mass, body mass index (BMI), and muscle strength. Subjects and methods Cross-sectional study of 10 men (36.7 ± 11.3 years) and 10 women (42.5 ± 10.3 years) infected with HIV, recruited from the Specialized Service Center in the State Center of Reference for High and Medium Complexity. Blood samples were collected to determine plasma irisin levels, glucose, HDL, total cholesterol, triglycerides, and LDL. Body composition (fat mass, fat-free mass) and anthropometrics (body mass index; BMI) were measured by bioelectrical impedance. Muscle strength was assessed using a mechanic hand dynamometer and one maximum repetition tests. Results Irisin levels correlated positively with fat mass (r = 0.67; p = 0.001) and BMI (r = 0.48; p = 0.036). In contrast, there was an inverse correlation between irisin levels and fat-free mass (r = -0.41; p = 0.008) and five strength parameters: right hand grip (r = -0.46; p = 0.044); left hand grip (r = -0.50; p = 0.027), relative hand grip (r = -0.79; p = 0.001), bench press (r = -0.58; p = 0.009), leg press (r = -0.40; p = 0.085), and biceps curl (r = -0.059; p = 0.009). Conclusion Irisin levels correlated positively with body fat and negatively with fat-free mass and strength parameters in HIV-infected patients. Female patients infected with HIV receiving highly active antiretroviral therapy have higher levels of irisin compared with men in a similar circumstance.

RESUMO

The aim of this study was to investigate the effects of different resistance training (RT) volumes on MMP activity in skeletal muscle, visceral adipose tissue and circulation. 21 Wistar rats were randomly divided into 3 groups (n=7 per group): sedentary control (SC); RT with 4 ladder climbs (RT-4; 50, 75, 90 and 100% of their maximal carrying capacity) and RT with 8 ladder climbs (RT-8 with 2 sets for each load). The 8-week RT consisted of climbing a 1.1-m vertical ladder with weights secured to the animals' tails. MMP-2 and -9 activity were analyzed by zymography. RT-8 displayed higher active MMP-2 activity as compared with SC and RT-4 in skeletal muscle (p<0.05). There was no significant difference between groups for pro and intermediate-MMP-2 activity in visceral adipose tissue, while RT-8 presented lower active MMP-2 activity as compared with SC (p<0.05). Plasma pro and active MMP-2 and MMP-9 activity was lower in RT-8 as compared with RT-4 (p<0.05). These results suggest that higher volume RT up-regulates MMP-2 activity in skeletal muscle, while down-regulating MMP-2 in visceral adipose tissue. Moreover, it induces a decrease of MMP-2, 9 activity in circulation. Different tissue and circulatory MMP responses to RT may result in specific remodeling.

RESUMO

BACKGROUND: Deregulation of glycemic and glycated hemoglobin (HbA1) levels accelerate the progression of cardiovascular complications in type 1 diabetes mellitus (T1DM). The aim of this study was to investigate the association between HbA1 and changes in blood pressure of children and adolescents with T1DM. METHODS: A total of 60 children and adolescents were recruited and allocated into two groups (prehypertension and control group). Blood pressure and HbA1 were measured by the oscillometric method and high-performance liquid chromatography, respectively. RESULTS: The prehypertensive group had (P < 0.05) higher disease duration, body weight, Z score for body weight, systolic blood pressure (SBP), diastolic blood pressure (DBP) and a higher HbA1 when compared with the control children and adolescents. Multiple regression to predict alterations in DBP from HbA1 adjusted for age, disease duration, and body mass index demonstrated a positive relationship with DBP (P < 0.05). A 1 % increase in HbA1 was associated with 1.73 mmHg increase in DBP. CONCLUSIONS: High levels of HbA1 may be associated with increased blood pressure in T1DM. RESUMO

BACKGROUND AND AIM: The aim of the present study was to examine nitrite concentration responses following eccentric resistance exercise (ERE) in elderly obese women. We also investigated the existence of high (HR) and low responders (LR) for nitrite and the possible differences with respect to creatine kinase (CK) response, metabolic and body composition variables. METHODS: Forty-nine elderly obese women completed an ERE session utilizing knee extensor exercise. LR for serum nitrite were defined as a ∆nitrite ≤ 20th percentile or 24.1 µmol/L and HR as a ∆nitrite > 20th percentile. RESULTS: Ten subjects were classified as LR and the remaining as HR (n = 39). The HR group displayed greater nitrite concentration at 0 h, 3 h, 24 h and 48 h following the ERE as compared with the LR (p < 0.05), and CK increased after 24 h and 48 h only for the HR group following the ERE (p < 0.05). Peak nitrite concentration was higher in the HR group versus the LR group (p < 0.05), while there was no difference between groups for pre-exercise nitrite values. The LR group displayed higher (p < 0.05) body fat, cholesterol, LDL and lower upper limb fat-free mass as compared with the HR group. The LR had lower (p < 0.05) upper limb fat-free mass than the HR group. CONCLUSIONS: Elderly obese women classified as HR displayed higher nitrite responses to ERE. Thus, researchers should be aware of the presence of different responsiveness of nitrite to acute exercise to avoid misinterpretation of data and to identify the higher cardiovascular risk factor of those classified as LR. RELEVANCE FOR PATIENTS: The elevated NO up to 48 h following an ERE session may suggest an important protective cardiovascular effect. The higher body fat, cholesterol, LDL and lower upper limb fat-free mass in the LR group might represent a deleterious effect of lower serum levels of nitrite.

RESUMO

Although aging compromises the functionality of macrophages (MΦ) and lymphocytes (LY), and dietary restriction (DR) and exercise partially counterbalance immunosenescence, it is unknown what effects of both strategies have on the functionality of these immune cells. Rats were randomly distributed into adult control (AD), older group (OLD), older submitted to 50% of DR (DR) and older submitted to swimming (EX) (n = 10 in each group). The function of immune cells (proliferative index, phagocytic capacity and H2O2 production), the weight and protein content of lymphoid organs (thymus and spleen), plasma glutamine concentration, interleukins (IL-1, IL-2, IL-6) and, immunoglobulins (IgA and IgG) were analysed. There was an increase of 74% in body weight in aged animals as compared with the AD group, while body weight reduced 19% in the DR as compared with the OLD group. Swimming training stimulated MΦ phagocytosis, while the EX group presented a decrease of the proliferative capacity of LY from the mesenteric lymph nodes (44% and 62%, respectively), when stimulated with ConA and LPS as compared with the old rats. These data demonstrated that DR and exercise affects differentially MΦ and LY function.

RESUMO

BACKGROUND: The purpose of this study was to verify the effects of eight weeks of resistance training (RT) on 24 hour blood pressure (BP) in patients with and without metabolic syndrome (MetS). METHODS: Seventeen women volunteered to participate in this study, 9 with MetS (37.0 ± 8.7 yrs; body mass 77.3 ± 9.7 kg; body mass index 30.3 ± 4.2 kg · m(-2)) and 8 without MetS (35.1 ± 7.2 yrs; body mass 61.3 ± 8.1 kg; body mass index 24.2 ± 2.5 kg · m(-2)). Individuals were subjected to eight weeks (3 times/week) of whole body RT comprised of one exercise for each main muscle group with three sets of 8-12 repetitions of each subject's maximal load . A rest interval of one minute was allowed between sets and exercises. Twenty-four hour BP was measured by ambulatory blood pressure monitoring. RESULTS: Mean and diastolic night-time BP decreased (-3.9 mmHg, p = 0.04; -5.5 mmHg, p = 0.03, respectively) after eight weeks of training in MetS patients. This decrease was observed at 11:00 pm, 02:00 am (only diastolic), 07:00 am, and 6:00 pm. There was no training effect on BP in women without MetS. CONCLUSIONS: Considering the elevation of BP as a contributor to the pathogenesis of MetS, and also to the increase of cardiovascular risk, this study supports RT as a non-pharmacological therapy in the management of BP control for MetS.

RESUMO

The aim was to investigate and compare the effects of proprioceptive neuromuscular facilitation (PNF) and static stretching (SS) on maximal voluntary contraction (MVC) and muscle activation in indoor soccer players. Thirty-three young adult men were divided into two groups: (i) sedentary and (ii) trained. Each group completed three different experimental trials: SS, PNF and no stretching (NS). The MVC of knee extension was evaluated before and immediately after each condition along with electromyography from the vastus lateralis (VL) and rectus femoris (RF) muscles of the dominant leg. PNF or SS techniques induced no decrease on MVC and muscle electromyographical activity in indoor soccer players (P>0·05). The electromyography of the RF and VL was lower after SS only in the sedentary group (P≤0·05). Short-duration PNF or SS has no effect on isometric MVC and muscle activity in indoor soccer players.

RESUMO

BACKGROUND: The purpose of the present study was to examine the effects of eight weeks of resistance training (RT) on anthropometric, cardiovascular and biochemical risk factors of metabolic syndrome (MetS), and neuromuscular variables on overweight/obese women. METHODS: Fourteen middle-aged (33.9 ± 8.6 years) overweight/obese women (body mass index - BMI 29.6 ± 4.1 kg/m2) underwent 24 sessions (3 times/week) of a whole body RT program with 3 sets of 8-12 repetitions maximum (RM). The following variables were evaluated: maximum strength on chest press and frontal lat pull-down; isometric hand-grip strength; biceps brachii (BB) and rectus femoris (RF) muscle thickness, body mass; BMI; body adiposity index (BAI); waist, hip and neck circumferences; visceral fat volume; blood glucose; glycated hemoglobin (HbA1c); insulin; HDL-C and triglycerides. RESULTS: There was an increase of chest press (from 52.9 ± 9.7 to 59.8 ± 7.7 kg; P = 0.02) and front lat pull-down (from 51.5 ± 7.5 to 57.6 ± 9.2 kg; P = 0.01) muscle strength, isometric handgrip (P = 0.02) and RF muscle thickness (from 42.2 ± 8.5 to 45.1 ± 7.3 mm; P = 0.02) after the 8 week RT program. There were no statistically significant alterations on plasma glucose, HbA1c, insulin, triglycerides, HDL-C, anthropometric indexes and BB muscle thickness (p > 0.05). CONCLUSIONS: A RT program without caloric restriction promotes an increase on muscle thickness and strength, with no effects on risk factors of MetS in overweight/obese women.

RESUMO

Exercise modulates both glucose and glutamine metabolism which influences lymphocyte function. We investigated the influence of chronic moderate exercise on glucose and glutamine metabolism in lymphocytes, the associated influence on proliferation, and cytokine and immunoglobulin production. Male Wistar rats (8 weeks old) were placed in an exercise training group (N = 15, 1 h day(-1) at 60 % VO2max, 5 days week(-1)) for 8 weeks of exercise, or a sedentary control group. Twenty-four hours following the final training session, lymphocytes were separated, and the incorporation of [U-14C]-glucose, [U-14C]-glutamine, and [2-14C]-thymidine from the supernatant was measured. The activity of glucose-6-phosphate dehydrogenase, hexokinase, and glutaminase was measured. Lymphocytes were stimulated with ConA and LPS and incubated with the Mycobacterium bovis bacille Calmette-Guerin (BCG) vaccine and plasma IgG and IgE were measured. Glutamine metabolism increased in both T and B lymphocytes in the trained group. In the trained group, proliferative capacity increased T lymphocytes under ConA stimulation, and increased B lymphocytes with LPS. There was a significant increase in IL-2 production and decrease in IL-4 in the trained group compared with sedentary controls. IL-2R and TNFR increased in trained rats while IL-4R decreased and were more pronounced in T lymphocytes compared with B lymphocytes. In both lymphocyte subsets, exercise training significantly increased the expression of CD54+ and CD30+ cell markers. Exercise training increased plasma IgG compared with the sedentary group. In conclusion, moderate exercise training improves immune function and metabolism in T and B lymphocytes, reflecting an increased ability to respond to immune challenges.
