The present study aimed to develop a feeding strategy for pregnant sows that involved the prenatal administration of a mixture of pancreatic-like fungal enzymes, such as lipase, amylase, and protease, at (1) 1-115 days of gestation (group D1) and (2) 80-115 days of gestation (group D2) and to carry out a comparison with groups of sows that were not receiving such supplementation (negative control (NC) and positive control (PC)). It was found that the administration of the enzyme supplement resulted in a significant shortening of gestation (p ≤ 0.01). The pancreatic enzymes administered to sows had a significant effect on the number of liveborn piglets and weaned piglets, which was higher compared with the control groups that did not receive supplementation: D1-12.1 ± 1.1 and 11.12 ± 1.1 and D2-12.8 ± 1.3 and 11.75 ± 0.07 vs. the control groups KN-10.7 ± 1.0 and 9.62 ± 0.95 and KP-10.9 ± 1.2 and 10.15 ± 1.0 (p < 0.006), respectively. Significant changes in piglet growth were observed after weaning up to 70 days of age. During this period, the most favorable growth parameters were observed in groups D2 (420 ± 91 g) and PC (407 ± 103 g), in which piglets obtained a mixture of pancreatic enzymes (lipase, amylase, and protease) at 3 weeks of age, and significantly higher weight gain and feed intake were observed compared with groups NC (378 ± 114 g) and D1 (381 ± 96 g) (p ≤ 0.007). In contrast, insulin levels were significantly lower in groups D1 and D2, with values of 6.8 IU/mL and 6.7 IU/mL, respectively, compared with groups NC (14.6 IU/mL) and PC (16.6 IU/mL) (p ≤ 0.01). Piglets in group D2 had a significantly better feed conversion ratio (FCR) of 1.604 ± 0.10 compared with the other dietary groups: KN-1.986 ± 0.14; KP-1.704 ± 0.11; and D1-1.932 ± 0.15 (p ≤ 0.03). Histological imaging confirmed a significantly thicker intestinal epithelium and intestinal mesenteron in animals from groups D2 and PC (p ≤ 0.03). Animals from the groups KP, D1, and D2 receiving enzymes showed a highly significant increase in the surface area of pancreatic follicles and pancreatic surface area compared with the group without KN supplementation (p < 0.01). Furthermore, significantly higher activity of the brush border enzyme lactase was observed in groups D1, D2, and PC, with values of 32.90 ± 3.99, 30.00 ± 6.83, and 29.60 ± 29.60, respectively, compared with group NC, with a value of 21.80 ± 3.27 (p ≤ 0.01).

During the initial months of calves' lives, the young animals are exposed to bacterial and viral infections, and during this period, crucial physiological changes take place in their organisms. Offering calves feed additives that will have a beneficial influence on their organisms and improve their growth while reducing the morbidity rate is the optimal task of feeding. This is the first study to investigate the effect of experimental supplementation for calves with the combination of two feed additives­one containing Lentinula edodes enriched with selenium (Se), and the second containing pancreatic-like enzymes, fat-coated organic acids, sodium butyrate, and silicon dioxide nanoparticles­on the serum Se concentration, selected immune parameters, and the average daily gains in the calves. During the study, the serum Se concentration was examined by means of inductively coupled plasma mass spectrometry, and the immunoglobulin and cytokine concentrations with ELISA assays. The white blood cell (WBC) count with leukocyte differentiation was examined with the use of a hematological analyzer, and the percentages of subpopulations of T lymphocytes and monocytes, phagocytic activity, and oxidative burst of monocytes and granulocytes with the use of a flow cytometer. The average daily gains of the calves were also evaluated. In summary, the supplementation of the experimental calves with the combination of two feed additives resulted in significantly higher serum Se concentrations, and the immune systems of the calves were not suppressed while the examined feed additives were being delivered. Although not statistically significant, some positive effects on the calves were seen: a tendency towards the improvement of some of the immune parameters evaluated, and a tendency for higher average daily gains in the calves.

With this study, we aimed to evaluate the effect of two multicomponent feed additives given to animals from 10 to 70 d with milk replacer on the production parameters of reared calves: serum selenium level and histology of duodenum and rumen. The first additive was based on nanosilica (3000 mg) containing pancreatic enzymes (protease (18 mg), lipase (45 mg)), a mixture of fat-coated organic acids (2000 mg) (fumaric, malic, citric, and sorbic acids), and sodium butyrate (10,000 mg) (nanosilica/E/OA/SB), whereas the second was based on the mycelium of L. edodes (7 g) enriched in selenium (0.058 mg/g d.w.) (L. edodes/Se). The study was conducted on 18 male crossbred Holstein dairy calves from birth to 70 days of age, which were randomly assigned to control and experimental groups (six animals in each). From each group, three animals were sacrificed at 70 days of age, and histological analysis of the digestive tract (abdominal rumen and small intestine) was performed. It was observed that the additives used did not have any effect on growth performance and hematological or biochemical parameters. However, higher levels of selenium were found in serum in the calves fed with modified L. edodes mycelium on days 35 and 70 (44.33 and 51.33 µg/L in the control group and 132 and 93 µg/L in the L. edodes/Se group, respectively; p < 0.001). Moreover, we noticed lower average daily milk replacer intake at 35−42 d, which increased significantly from 42 to 70 d in the animals receiving L. edodes/Se.

Every year, thousands of tons of fruit seeds are discarded as agro-industrial by-products around the world. Fruit seeds are an excellent source of oils, monounsaturated fatty acids, and n-6 and n-3 polyunsaturated essential fatty acids. This study aimed to develop a novel technology for extracting active substances from selected seeds that were obtained after pressing fruit juices. The proposed technology involved sonification with the use of ethyl alcohol at a low extraction temperature. Seeds of four species-blueberry (Vaccinium myrtillus L.), raspberry (Rubus idaeus), cranberry (Vaccinium macrocarpon), and cuckooflower (Cardamine pratensis)-were used for extraction. Following alcohol evaporation under nitrogen, the antioxidant activity, chemical composition, and volatile compounds of the obtained extracts were analyzed using chromatographic methods, including gas chromatography (GC)-mass spectrometry (MS) (GC-MS/MS), and high-performance liquid chromatography-MS. We analyzed physicochemical properties, fatty acid, and volatile compounds composition, sterol and tocochromanol content of blueberry, cranberry, raspberry, and cuckooflower seed oils obtained by sonication. This method is safe and effective, and allows for obtaining valuable oils from the seeds.

Pancreatic enzyme replacement therapy (PERT) and fat predigestion are key in ensuring the optimal growth of patients with cystic fibrosis. Our study attempted to highlight differences between fat predigestion and conventional PERT on body composition of young pigs with exocrine pancreatic insufficiency (EPI). EPI and healthy pigs were fed with high-fat diet for six weeks. During the last two weeks of the study, all pigs received additional nocturnal alimentation with Peptamen AF (PAF) and were divided into three groups: H-healthy pigs receiving PAF; P-EPI pigs receiving PAF+PERT; and L-EPI pigs receiving PAF predigested with an immobilized microbial lipase. Additional nocturnal alimentation increased the body weight gain of EPI pigs with better efficacy in P pigs. Humerus length and area in pigs in groups L and P were lower than that observed in pigs in group H (p value 0.005-0.088). However, bone mineral density and strength were significantly higher in P and L as compared to that of H pigs (p value 0.0026-0.0739). The gut structure was improved in P pigs. The levels of neurospecific proteins measured in the brain were mainly affected in P and less in L pigs as compared to H pigs. The beneficial effects of the nocturnal feeding with the semielemental diet in the prevention of EPI pigs' growth/development retardation are differently modified by PERT or fat predigestion in terms of growth, bone properties, neurospecific protein distribution, and gut structure.

INTRODUCTION: The rearing of calves is a difficult period for farmers due to health problems to which the animals are prone this time. Since the use of antibiotics as growth promoters has been forbidden, various innovative feed additives have been tested in many countries around the world. MATERIAL AND METHODS: In this study, experimental (E) calves were supplemented with a novel feed additive consisting of the pancreatic-like enzymes protease and lipase, a fat-coated mixture of organic fumaric, malic, citric and sorbic acids, sodium butyrate and silicon dioxide nanoparticles. Control (C) calves received feed without additive. During the supplementation, white blood cell (WBC) counts with leukocyte differentiation, percentages of B lymphocytes and T lymphocytes and their subpopulations, phagocytic activity and oxidative burst of circulating monocytes and granulocytes were examined. Body weight (b.w.) gains of the calves were also monitored. RESULTS: The WBC counts in the E and C calves were within the reference ranges throughout the study. In the analysis of the percentages of the lymphocyte subpopulations, phagocytic activity and oxidative burst, no statistically significant differences were reported between the E and C groups. However, higher average daily body weight gains were obtained for the E calves. CONCLUSION: The study revealed that the examined feed additive did not modulate the immune response of the calves significantly. The tendency to higher daily average b.w. gains in the E calves than in the C calves suggests a beneficial effect of this feed additive.

The presence of selenium in European soil is low and this causes its deficiency in livestock and, in consequence, in humans. This study aimed to obtain Lentinula (L.) edodes mycelium with the maximum content of selenium. This species was used for experiment based on its documented medicinal properties. Calves were fed with selenium-enriched L. edodes mycelium, and serum selenium concentration, average daily weight gains and selected immune parameters were estimated. The selenium-enriched mushroom was found to be safe based on cytotoxicity tests (MTT and LDH tests) and for this reason it was used for further experiments. The mean quantity of selenium in the serum of calves fed with selenium-enriched L. edodes mycelium was significantly higher than that of control calves. Additionally, the calves fed with selenium-enriched L. edodes mycelium had higher body weight gains than those of control calves. White blood cell counts and subpopulations of lymphocytes in the experimental and control calves were within the reference range. The administration of L. edodes enriched with selenium had a beneficial effect on state of health of the calves.

Heme is an efficient dietary iron supplement applied in humans and animals to prevent iron deficiency anemia (IDA). We have recently reported that the use of bovine hemoglobin as a dietary source of heme iron efficiently counteracts the development of IDA in young piglets, which is the common problem in pig industry. Here, we used maternal Polish Large White and terminal sire breed (L990) pigs differing in traits for meat production to evaluate the long-term effect of split supplementation with intramuscularly administered small amount of iron dextran and orally given hemoglobin on hematological indices, iron status, growth performance, slaughter traits, and meat quality at the end of fattening. Results of our study show that in pigs of both breeds split supplementation was effective in maintaining physiological values of RBC and blood plasma iron parameters as well as growth performance, carcass parameters, and meat quality traits. Our results prove the effectiveness of split iron supplementation of piglets in a far-reach perspective.

Nanoparticles are increasingly popular in numerous fields including electronics, optics and medicine (vaccines, tissue engineering, microsurgery, genomics and cancer therapies). The most widely used nanoparticles in biomedical applications are those designed by man. Scientists have obtained many types of silica nanoparticles with defined shape and chemical composition, but different properties and applications. Nanoparticles include particles with at least one dimension ranging from 1-100 nm. Silica nanoparticles (Sn), reaching values from several dozen to several hundred m2/g, have unique physicochemical properties due to their porous structure and well-developed specific surface. Currently, the use of Sn in animal nutrition, with a focus on gastrointestinal tract function, is of great interest.

INTRODUCTION: The standard therapy for exocrine pancreatic insufficiency (EPI) is porcine-derived pancreatic enzyme replacement therapy (PERT). In the present study we tested a new approach with a mixture of pancreatic-like enzymes of microbial origin (PLEM) in a 1-week efficacy study in EPI pigs. In addition to the conventionally used coefficient of fat and nitrogen absorption (CFA and CNA), parameters that more accurately reflect the nutritional and health status, such as changes in the lipemic index (LI), plasma triglyceride (TG) and non-esterified fatty acid (NEFA) levels, and somatic growth, were determined. MATERIAL AND METHODS: A PLEM dose containing 120 000 active lipase units, 80 000 active protease units and 12 000 active amylase units (all from Sigma, St. Louis, MO) was given as a powder, twice daily with a meal (40 g fat/meal) to 8 EPI pigs for 7 days. Ten healthy pigs were used as a comparator. RESULTS: The PLEM enhanced fat and protein digestion, and reversed growth impairment in EPI pigs. With treatment, CFA and CNA increased by 59% and 43% (p < 0.05), respectively. Although fat and protein absorption were lower than in the comparator, the postprandial blood lipid profile was normal as in healthy pigs. The mucosal thickness significantly increased by 27%, 50% and 26%, in the proximal, middle, and distal jejunum (p < 0.05) with treatment and resembled that of healthy animals. CONCLUSIONS: Pancreatic-like enzymes of microbial origin supported somatic growth and normalized the postprandial lipid profile. As a measure of efficacy, postprandial LI, TG and NEFA are viable endpoints to be explored in human trials.

Heme is an efficient source of iron in the diet, and heme preparations are used to prevent and cure iron deficiency anemia in humans and animals. However, the molecular mechanisms responsible for heme absorption remain only partially characterized. Here, we employed young iron-deficient piglets as a convenient animal model to determine the efficacy of oral heme iron supplementation and investigate the pathways of heme iron absorption. The use of bovine hemoglobin as a dietary source of heme iron was found to efficiently counteract the development of iron deficiency anemia in piglets, although it did not fully rebalance their iron status. Our results revealed a concerted increase in the expression of genes responsible for apical and basolateral heme transport in the duodenum of piglets fed a heme-enriched diet. In these animals the catalytic activity of heme oxygenase 1 contributed to the release of elemental iron from the protoporphyrin ring of heme within enterocytes, which may then be transported by the strongly expressed ferroportin across the basolateral membrane to the circulation. We hypothesize that the well-recognized high bioavailability of heme iron may depend on a split pathway mediating the transport of heme-derived elemental iron and intact heme from the interior of duodenal enterocytes to the bloodstream.

An elevated level of serum uric acid-hyperuricemia, is strongly associated with the development of gout and chronic kidney disease (CKD) which is often accompanied by a significantly reduced glomerular filtration rate (GFR). In the present study, we investigated the extra-renal elimination of uric acid via the intestine in a healthy pig model and the effect of oral uricase therapy on plasma uric acid concentrations in pigs with induced hyperuricemia and CKD. The experiment was conducted on eleven, ten-week-old pigs (n = 11). The porcine model of CKD was developed by performing 9/10 nephrectomy surgery on eight pigs. A stable model of hyperuricemia was established in only five of the eight nephrectomized pigs by frequent injections of uric acid (UA) into the jugular vein. All pigs (three healthy pigs and five CKD pigs) were operated for implantation of jugular vein catheters and the three healthy pigs also had portal vein catheters inserted. Blood uric acid concentrations were measured spectrophotometrically, using the Uric Acid Assay Kit (BioAssay Systems, Hayward, USA). The piglets with CKD received orally administered uricase (treatment) and served as their own controls (without uricase supplementation). Oral uricase therapy significantly decreased plasma uric acid concentrations in pigs with CKD, whereas hyperuricemia was observed in the pigs whilst not being treated with uricase. Urinary uric acid excretion was similar during both the treatment and control periods during the first 8 h and 24 h after UA infusions in the CKD pigs. To demonstrate the elimination of UA via the intestine, the healthy pigs were infused with UA into the jugular vein. The blood collected from the jugular vein represents circulating UA concentrations and the blood collected from the portal vein represents the concentration of UA leaving the intestine. The final (after 2 h) concentration of UA was significantly lower in blood collected from the portal vein compared to that collected from the jugular vein (3.34 vs. 2.43 mg/dL, respectively, p = 0.024). The latter allows us to suggest that UA is eliminated from the blood via the gut tissue.

The presence of biologically active substances in feed mixture is discussed to have beneficial effect on animals' health and products. The purpose of the study was to determine the effect of dietary supplementation with dried apple, chokeberry, black currant, strawberry and carrot pomaces on production parameters and meat quality in fattening pigs. The use of dried pomaces of chokeberry showed tendencies for increased feed intake and reduced fattening period. The dried pomaces had no impact on saturated and monounsaturated fatty acids profile in meat, however in some groups an elevated level of polyunsaturated fatty acids from n-3 family and a decline in total cholesterol level was observed (P≤0.05). The highest oxidative stability and vitamin E content was found after supplementation with black currant (P≤0.05). Summarizing, the used dried pomaces improved several parameters related to meat quality, what might positively influence consumers' health.

RESUMO

Moderate wine drinking is associated with reduced risk of cardiovascular, cerebrovascular and peripheral vascular disease, and reduced risk of cancer. This phenomenon is called the "French paradox", since it was observed for the first time in France--a country famous for its wine production. In the literature, the cardioprotective effect of wine is very well described and attributed mainly to contained therein resveratrol. Recently, it has been demonstrated that resveratrol extends the lifespan of yeast through activation of the SirT1 longevity gene, which is also responsible for the longevity caused by caloric restriction. Furthermore, resveratrol exhibits high biological activity, affecting cell structures and contributing to their protection. This paper summarizes the available reports on functional and molecular aspects of resveratrol, wines and grapes as a result of the activation of longevity genes.

Among livestock, domestic pig (Sus scrofa) is a species, in which iron metabolism has been most intensively examined during last decade. The obvious reason for studying the regulation of iron homeostasis especially in young pigs is neonatal iron deficiency anemia commonly occurring in these animals. Moreover, supplementation of essentially all commercially reared piglets with iron entails a need for monitoring the efficacy of this routine practice followed in the swine industry for several decades. Since the discovery of hepcidin many studies confirmed its role as key regulator of iron metabolism and pointed out the assessment of its concentrations in biological fluids as diagnostic tool for iron-related disorder. Here we demonstrate that urine hepcidin-25 levels measured by a combination of weak cation exchange chromatography and time-of-flight mass spectrometry (WCX-TOF MS) are highly correlated with mRNA hepcidin expression in the liver and plasma hepcidin-25 concentrations in anemic and iron-supplemented 28-day old piglets. We also found a high correlation between urine hepcidin level and hepatic non-heme iron content. Our results show that similarly to previously described transgenic mouse models of iron disorders, young pigs constitute a convenient animal model to explore accuracy and relationship between indicators for assessing systemic iron status.

RESUMO

Rapeseed, strawberry and raspberry seed oils are a rich source of polyunsaturated fatty acids and antioxidants such as tocols, bioflavonoids and phytosterols. The aim of the study was to determine changes in the blood lipid profile of rats fed with rapeseed, strawberry and raspberry seed oils and their effects on selected parameters of oxidative status. The experiment was carried out on male Wistar rats. The oils were administered by oral gavage for 5 weeks once daily at the dose of about 0.8 ml per rat. Blood samples were taken before and after supplementation period. The activity of superoxide dismutase (SOD) and glutathione peroxidase (cGPx) was assessed in erythrocytes and contents of triglycerides (TG), total cholesterol, low-density fraction of cholesterol (LDL) and high-density fraction of cholesterol (HDL) were assessed in plasma. The experiment shows that oils supplemented in the diet for 5 weeks had no significant effect on the level of triglyceride (TG), total cholesterol as well as HDL and LDL fractions. Reduced activity of cGPX and SOD in the group of rats receiving raspberry and strawberry seed oils suggests that these native oils may contribute to oxidative stability (improves antioxidant status). Thus, strawberry and raspberry seed oils can be considered as special biological oils, which constitute potential nutraceuticals reducing oxidative stress.

The aim of the study was to establish an optimized protocol of iron dextran administration to pig neonates, which better meets the iron demand for erythropoiesis. Here, we monitored development of red blood cell indices, plasma iron parameters during a 28-day period after birth (till the weaning), following intramuscular administration of different concentrations of iron dextran to suckling piglets. To better assess the iron status we developed a novel mass spectrometry assay to quantify pig plasma levels of the iron-regulatory peptide hormone hepcidin-25. This hormone is predominantly secreted by the liver and acts as a negative regulator of iron absorption and reutilization. The routinely used protocol with high amount of iron resulted in the recovery of piglets from iron deficiency but also in strongly elevated plasma hepcidin-25 levels. A similar protocol with reduced amounts of iron improved hematological status of piglets to the same level while plasma hepcidin-25 levels remained low. These data show that plasma hepcidin-25 levels can guide optimal dosing of iron treatment and pave the way for mixed supplementation of piglets starting with intramuscular injection of iron dextran followed by dietary supplementation, which could be efficient under condition of very low plasma hepcidin-25 level.

The aim of the study was to evaluate the effect of semen storage on ferrous ion induced luminescence of boar spermatozoa and to determine the relationship between parameters of this luminescence and lipid peroxidation as measured by malondialdehyde (MDA) contents. Boar semen samples were diluted in Biosolwens extender and stored for 12 days at 15 degrees C. Luminescence and MDA were measured directly after dilution (day 0) and at 6 and 12 days of semen storage. Luminescence was measured at 20 degrees C using a luminometer equipped with a cooled photomultiplier with a spectral response range from 370 to 620 nm. Emission was induced by adding FeSO4 solution (final concentration 0.05 mM). MDA content was measured by the HPLC method. The day of storage had a significant effect on some luminescence parameters and MDA content in spermatozoa. A significant correlation was observed between luminescence parameters and MDA concentration. The results of the study confirm that induced luminescence is strictly related to lipid peroxidation in spermatozoa that occur during boar semen storage. Parameters of luminescence treated as a holistic response of cells to oxidative stress can be useful for monitoring spermatozoa quality during semen preservation.

The influence of adding 2% CLA to the morning feed of a group of 20 crossbred fatteners from 70 to 130 kg on meat quality, eating quality and dietetic value of m. longissimus dorsi was evaluated compared with a group of 20 controls. Addition of 2% CLA in feed did not influence fattening and slaughter results and also some traits of meat and eating quality. The only significant differences were observed in dry matter content, in meat flavour and L(∗), b(∗), and hue angle value. Meat from fatteners from the experimental group had a statistically significant higher content of CLA (P⩽0.01) and lower content of unsaturated fatty acids especially C18: 2, C18: 1 and C20: 4, PUFA fatty acids (P⩽0.01) and a reduction in the ratio of n-6: n-3 PUFA (P⩽0.01). No statistically significant influence of CLA on the content of cholesterol was observed. The results of the current histochemical study showed that dietary CLA supplementation significantly increased both the number and diameter of white fibres, and correspondingly reduced the number and diameter of red fibres. The percentage and diameter of intermediate fibres was unaffected by CLA.
