RESUMO

Progeny born to primiparous sows (gilt progeny; GP) have lower birth, weaning and slaughter weights than sow progeny (SP). GP also have reduced gastrointestinal tract (GIT) development, as evidenced by lower organ weights. Therefore, the aim of this experiment was to quantify changes in GIT barrier function that occur in birth and weaning, representing two major challenges to the young piglet. The effects of parity (GP vs. SP) in GIT barrier integrity function were quantified at four timepoints: birth (~0 h), 24 h after birth (24 h), 1-d preweaning (PrW), and 1-d postweaning (PoW) in commercially reared piglets. Due to inherent differences between newborn and weanling pigs, the results were analyzed in two cohorts, birth (0 vs. 24 h, n = 31) and weaning (PrW vs. PoW, n = 40). Samples of the stomach, jejunum, ileum, and colon were excised after euthanasia and barrier integrity was quantified by measuring transepithelial resistance (TER), macromolecular permeability, the abundance of inflammatory proteins (IL-8, IL-1ß, and TNF-α) and tight junction proteins (claudin-2 and -3). Papp was characterized using a dual tracer approach comprising 4 KDa fluorescein isothiocyanate (FD4) and 150 kDa tetramethyl rhodamine isothiocyanate (T150)-labeled dextrans. Characteristic effects of the initiation of feeding and weaning were observed on the GIT with the initiation of feeding, such as increasing TER and reducing Papp at 24 h, consistent with mucosal growth (P = 0.058) This was accompanied by increased cytokine abundance as evidenced by elevations in TNF-α and IL-1ß. However, GP had increased IL-8 abundance (P = 0.011 and 0.063 for jejunum and ileum respectively) at birth than 24 h overall. In the weaning cohort, jejunal and ileal permeability to FD4 was higher in GP (P = 0.05 and 0.022, respectively) while only higher ileal T150 was observed in GP (P = 0.032). Ileal claudin-2 abundance tended to be higher in SP overall (P = 0.063), but GP ileal claudin-2 expression was upregulated weaning while no change was observed in SP (P = 0.043). Finally, other than a higher jejunal TNF-α abundance observed in SP (P = 0.016), no other effect of parity was observed on inflammatory markers in the weaning cohort. The results from this study indicate that the GIT of GP have poorer adaptation to early life events, with the response to weaning, being more challenging which is likely to contribute to poorer postweaning growth.


The progeny of primiparous sows (gilt progeny; GP) have poorer lifetime growth performance in comparison to progeny from multiparous sows (sow progeny; SP). Previous research suggests that there is an underlying biological basis for reduced growth performance which is attributed to differences in gastrointestinal tract (GIT) barrier development during early life. This study aimed to clarify the timeframes of when these differences are in effect by investigating GIT development during two major events of a piglet's life: birth and weaning. To do this, GIT tissue was collected from GP and SP at four time points; birth, 24 h after birth, 1-d preweaning, and 1-d postweaning and assessed for functional development. The main findings from this study indicate there are early signs of variation in GIT development within the first 24 h of life between GP and SP, and that these differences increase through the preweaning period, with GP entering weaning with poorer GIT development and function. Possible explanations for the reduced GI development may be reduced maternal nutrition during the suckling period.

Tail biting (TB) in pigs is a complex issue that can be caused by multiple factors, making it difficult to determine the exact etiology on a case-by-case basis. As such, it is often difficult to pinpoint the reason, or set of reasons, for TB events, Decision Support Tools (DSTs) can be used to identify possible risk factors of TB on farms and provide suitable courses of action. The aim of this review was to identify DSTs that could be used to predict the risk of TB behavior. Additionally, technologies that can be used to support DSTs, with monitoring and tracking the prevalence of TB behaviors, are reviewed. Using the PRISMA methodology to identify sources, the applied selection process found nine DSTs related to TB in pigs. All support tools relied on secondary information, either by way of the scientific literature or expert opinions, to determine risk factors for TB predictions. Only one DST was validated by external sources, seven were self-assessed by original developers, and one presented no evidence of validation. This analysis better understands the limitations of DSTs and highlights an opportunity for the development of DSTs that rely on objective data derived from the environment, animals, and humans simultaneously to predict TB risks. Moreover, an opportunity exists for the incorporation of monitoring technologies for TB detection into a DST.

The weaning of pigs in most commercial pork production systems is an abrupt event performed at a fairly young age, i.e., mostly between 2.5 and 5 weeks of age. This practice induces a stress response, and its impact on behavior, performance and the gastrointestinal tract has been well described. Historically, there has been a focus on pre- and post-weaning nutritional strategies and post-weaning housing conditions and medication to improve production and reduce mortality after weaning. However, alternative pre-weaning housing and management systems that promote the development of natural social behaviors of piglets before weaning have recently received more attention. Co-mingling of non-littermates before weaning is a strategy that aims to initiate social interactions prior to weaning. The separation of the litter from the sow in the period leading up to weaning, termed intermittent suckling, aims to enhance the gradual separation from the sow. In addition, these practices encourage the young pig to learn explorative nutrient sourcing. Altogether, they may reduce weaning-associated stress. In this review, these strategies are defined, and their effects on behavior, performance, mortality, gastrointestinal function and immunocompetence are described. Though these strategies may be adapted to a commercial setting, it also becomes clear that many factors can contribute to the success of these strategies.

Current interventions targeting antimicrobial resistance (AMR), a major impact on commercial pork production, focus on reducing the emergence of AMR by minimising antimicrobial usage through antimicrobial stewardship and a range of alternative control methods. Although these strategies require continued advancement, strategies that directly aim to reduce or eliminate existing antimicrobial resistant bacteria, specifically bacteria resistant to critically important antimicrobials (CIAs), need to be investigated and established. This study established an in vivo model for examining the effects of postbiotics, in the form of Lactobacillus acidophilus fermentation products (LFP) and Saccharomyces cerevisiae fermentation products (SFP), on the shedding of extended-spectrum cephalosporin (ESC)-resistant E. coli. The model was successful in demonstrating the presence of ESC-resistant E. coli as evidenced by its detection in 62 of 64 pigs. There was a strong trend (p = 0.065) for the SFP postbiotics to reduce the shedding of ESC-resistant E. coli, indicating positive impacts of this additive on reducing the carriage of bacteria resistant to CIAs. Overall, this in vivo model enables future evaluation of strategies targeting ESC-resistant E. coli while increasing our knowledge on the carriage of ESC-resistant E. coli in pigs.

Different cereal types, in combination with different protein sources, are fed to pigs after weaning, but their interactions and possible implications are not well researched. In this study, 84 male weaned piglets were used in a 21-day feeding trial to investigate the effects of feeding either medium-grain or long-grain extruded rice or wheat, in a factorial combination with protein sources of either vegetable or animal origin, on postweaning performance, shedding of ß-haemolytic Escherichia coli, and the coefficient of total tract apparent digestibility (CTTAD). Pigs fed either rice type performed the same (p > 0.05) as wheat-fed pigs after weaning. The use of vegetable protein sources reduced growth rate (p < 0.001) and feed intake (p = 0.007) and deteriorated the feed conversion ratio (p = 0.028) in weeks two and three compared to pigs fed animal protein sources. The number of antibiotic treatments given for clinical diarrhoea was similar (p > 0.05). However, the faecal E. coli score showed a trend for the main effect of protein source, with pigs fed animal proteins showing a higher E. coli score than pigs fed vegetable proteins (0.63 vs. 0.43, p = 0.057). There was also a tendency for an interaction (p = 0.069) between cereal type and protein source (p = 0.069), with this difference being associated with a greater faecal score in pigs fed diets with long-grain rice plus animal proteins and wheat plus animal proteins. Significant interactions occurred for the CTTAD when assessed in week three. In general, pigs fed diets with medium-grain rice or long-grain rice with animal proteins had a higher (p < 0.001) CTTAD for dietary components than pigs fed all other diets, and vegetable proteins depressed (p < 0.001) CTTAD compared to animal proteins (main effect of protein: p < 0.001). In summary, pigs tolerated the extruded rice-based diets well and performed equivalently to pigs fed wheat as the sole cereal, and the use of vegetable proteins decreased the E. coli score.

Serum creatinine (SCr) in humans has proven to be a reliable biomarker of body protein breakdown and/or muscle mass change. This study set out to investigate the potential of SCr to indicate a loss in sow muscle mass over lactation, validated against 3 methyl histidine (3MH) and blood urea nitrogen (BUN), markers of dietary and/or body protein breakdown. A total of 40 sows were allocated to four treatment groups aimed to induce body weight changes by restrictively feeding sows using a stepwise percentage reduction model. Data were pooled and reallocated into three groups representing the 25th , 50th and 75th percentiles based on body weight change over lactation in the range -22.3 to -4.1% (treatment 25), -4.0 to 6.2% (Treatment 50), and 6.3-15.2% (Treatment 75). Indirect measures for the prediction of protein (3MH, BUN) or fat change (caliper, P2) were taken on entry into the farrowing house, day 5 of lactation, and at weaning. Serum was collected on these days, and SCr, 3MH and BUN were analysed. Piglet weaning weight and average daily feed intake did not differ between treatments (p > .05). There were no changes (p > .05) in indirect measures of body composition (sow caliper score, P2) and analytes (SCr, 3MH, BUN) over lactation. By day 20, those sows in treatment 25 had higher (p < .05) 3MH concentrations whilst changes from day 5 to 20 were not different (p > .05) and did not correlate with SCr change (p > .05) but were highly correlated to BUN change (R2 = 0.691, p < .001). The data suggested that concentrations of SCr and BUN may have been the result of dietary and/or body protein breakdown and/or changes in muscle mass. In the current testing conditions, SCr was not a reliable marker of changes in muscle mass.

Post weaning diarrhea (PWD) caused by enterotoxigenic strains of E. coli (ETEC) remains a major problem in the industry, causing decreases in performance and survival of weaned pigs. Traditionally, antimicrobials have been used for its mitigation/control. This study tested the hypothesis that a combination of two organic acid (OA)-based commercial feed additives, Presan FX [an OA, medium-chain fatty acid (MCFA) and phenolic compound-based product] and Fysal MP (free and buffered OA based on formic acid), would reduce PWD and improve post-weaning performance in pigs challenged with an F4-ETEC. This combination was assessed against a Negative control diet without any feed additives and a diet containing amoxicillin. Combined with a reduction in temperature during the infection period, inoculation with F4-ETEC resulted in 81% of pigs developing diarrhea, but with no differences between treatments (P > 0.05). However, between days 14 to 20 of the study and due to colonization by Salmonella serovars, pigs fed the combination of Presan FX and Fysal MP showed less (P = 0.014) diarrhea commensurate with a lower (P = 0.018) proportion of Salmonella numbers relative to total bacterial numbers. This caused less (P = 0.049) therapeutic antibiotic administrations relative to the diet with amoxicillin during this time. The diversity of bacteria within amoxicillin-treated pigs was lower (P = 0.004) than the diversity in control or Presan FX + Fysal MP-treated pigs (P = 0.01). Pair-wise comparisons showed that amoxicillin-treated pigs had altered (P < 0.001) fecal microbial communities relative to both Presan FX + Fysal MP-treated pigs and control pigs. Amoxicillin-treated pigs were characterized by an increased abundance of bacterial families generally linked to inflammation and dysbiosis in the gastrointestinal tract (GIT), whereas Presan FX + Fysal MP-treated pigs had an increased abundance of bacterial families considered beneficial commensals for the GIT. Control pigs were characterized by an increased abundance of Spirochaetaceae associated with healthy piglets, as well as bacterial families associated with reduced feed intake and appetite. The combination of two OA-based feed additives did not reduce the incidence of F4 ETEC-associated diarrhea nor enhance performance. However, the combination markedly reduced diarrhea caused by Salmonella that occurred following the ETEC infection, commensurate with less therapeutic administrations relative to the diet with amoxicillin.

Genetic selection of pigs over recent decades has sought to reduce carcass fat content to meet consumer demands for lean meat in many countries (e.g., Australia). Due to the impacts of genetic changes, it is unknown whether the carcass fat measures are still responsive to energy intake. Thus, the present experiment aimed to quantify the relationship between tissue composition and dietary energy intake in finisher pigs selected for low carcass backfat. Intact male and female pigs (n = 56 for each sex; Primegro Genetics, Corowa, NSW, Australia) were fed seven different amounts of an amino acid adequate wheat-based diet containing 14.3 MJ digestible energy (DE)/kg to provide the following daily DE intakes- 25.8, 29.0, 32.6, 35.3, 38.5, 41.5, and 44.2 (ad libitum) MJ DE/d for males, and 25.8, 28.9, 32.0, 35.6, 38.3, 40.9, and 44.5 (ad libitum) MJ DE/d for females between 60 and 108 kg live weight. Body composition of anesthetized pigs was measured using the dual energy X-ray absorptiometry (DXA) method when individual pigs reached 108 kg, and protein, fat, and ash deposition rates were calculated. Pigs were slaughtered on the second day post-DXA scan for carcass backfat measurement. The results showed that the carcass backfat thickness (standardized at 83.7 kg carcass) increased by 0.125 mm for every MJ increase in daily DE intake in male pigs (P = 0.004; R2 = 0.130), but carcass backfat of female pigs (standardized at 85.1 kg carcass) was not responsive to daily DE intake. Whole-body fat composition and fat deposition rate increased linearly (both P < 0.01) in male pigs but quadratically (both P < 0.01) in female pigs in response to DE intake. Every MJ increase of daily DE intake increased the rate of daily protein deposition by 3.8 g in intact male pigs (P < 0.001; R2 = 0.781) and by 2.5 g in female pigs (P < 0.001; R2 = 0.643). In conclusion, the selection for low backfat thickness over the last two decades has altered the response of fat deposition and backfat thickness to energy intake, particularly in female pigs. Despite this change, the linear relationship between DE intake and protein deposition rate was maintained in these modern genetics.

The aim of this experiment was to determine whether administration of an anti-inflammatory compound to sows prior to farrowing would, via reduced pain and inflammation, increase piglet survival and growth. At day 114 of gestation, multiparous sows were randomly allocated to one of the following treatments: Control (n = 43), which received 10 mL saline, NSAID (n = 55) which received 0.4 mg/kg meloxicam and SAID (n = 54) which received 0.1 mg/kg dexamethasone. Treatments were applied again on day 116 if farrowing had not occurred. There was no treatment effect on piglets born alive or dead from parity two to four sows but in those of parity five and older, NSAID administration reduced the number of piglets born alive and increased the number of piglets born dead (p < 0.05). Sow rectal temperature and incidence of mastitis were unaffected by treatment (p > 0.05). Lactation day two plasma concentrations of cortisol, prostaglandin F2 alpha metabolite and haptoglobin did not differ among treatments (p > 0.05). Treatment effects were not observed in liveborn piglet mortality at any age, or litter weight at day 21 (p > 0.05). Average feed intake during lactation was increased by both NSAID and SAID treatments (p = 0.001). The use of meloxicam prior to farrowing should be avoided as it reduced the number of piglets born alive and did not improve piglet survival and growth.

Strains of enterotoxigenic Escherichia coli (ETEC) causing post-weaning diarrhoea (PWD) in piglets have a widespread and detrimental impact on animal health and the economics of pork production. Traditional approaches to control and prevention have placed a strong emphasis on antimicrobial use (AMU) to the extent that current prevalent porcine ETEC strains have developed moderate to severe resistance. This complicates treatment of ETEC infection by limiting therapeutic options, increasing diagnostic costs and increasing mortality rates. Management factors, the use of supra-physiological levels of zinc oxide and selected feed additives have all been documented to lower the incidence of ETEC infection in pigs; however, each intervention has its own limitations and cannot solely be relied upon as an alternative to AMU. Consequently, treatment options for porcine ETEC are moving towards the use of newer antimicrobials of higher public health significance. This review focuses on microorganisms and microbial-derived products that could provide a naturally evolved solution to ETEC infection and disease. This category holds a plethora of yet to be explored possibilities, however studies based around bacteriophage therapy, probiotics and the use of probiotic fermentation products as postbiotics have demonstrated promise. Ultimately, pig producers and veterinarians need these solutions to reduce the reliance on critically important antimicrobials (CIAs), to improve economic and animal welfare outcomes, and to lessen the One Health threat potentiated by the dissemination of AMR through the food chain.

Gilt progeny (GP) exhibit poorer growth compared with sow progeny (SP), particularly in the pre-weaning and post-weaning period. Late gestation/lactation sow diets and weaner diets were supplemented with 0.5% Polygain (POL), a sugarcane extract rich in polyphenols, to collectively improve GP growth in these periods. Gilts (n = 60) and sows (n = 68, parities 2 and 3) were fed a control or POL diet. Weaned GP (n = 79) and SP (n = 92) born to these dams were also fed either a CON or POL diet. Gilts litters weighed less than sow litters at birth and 21 days (p < 0.001 for both) and were not improved by POL (p = 0.80 and 0.54, respectively). GP were lighter than SP at day 7 and day 14 post-weaning (p < 0.001 for both) and were not improved by the POL diet at these timepoints (p = 0.61 and 0.97, respectively). Plasma interleukin-1ß (IL-1ß) was increased at weaning despite POL supplementation (p = 0.022) and GP had reduced IL-1ß overall (p = 0.021). Overall, POL was unable to improve growth in GP and the attenuated immune response seen in GP could be contributing to their poor growth performance.

Dietary tryptophan (Trp) is a precursor for serotonin, a neuromediator involved in stress responses. Tryptophan competes with other large neutral amino acids (LNAA: tyrosine, isoleucine, leucine, valine, and phenylalanine) to cross the blood-brain barrier; therefore, the regulation of circulating LNAA can influence Trp availability in the cortex and serotonin biosynthesis. The hypothesis examined in this study was that increased supplementation of dietary Trp and a reduction in LNAA for weaned pigs experimentally infected with enterotoxigenic Escherichia coli (ETEC; F4) will increase Trp availability in plasma and reduce indices of the stress response, which will translate to reduced production losses. At 21 ± 3 d of age (mean ± SEM), 96 male pigs (Large White × Landrace) weighing 6.3 ± 0.98 kg (mean ± SEM) were individually penned and allocated to a 4 × 2 factorial arrangement of treatments, with respective factors being 1) four dietary standardized ileal digestible (SID) Trp and LNAA contents, being HTrpHLNAA (Low Trp-High LNAA; 0.24% SID Trp: 5.4% SID LNAA), HTrpHLNAA (Low Trp-Low LNAA; 0.24% SID Trp: 4.6% SID LNAA), HTrpHLNAA (High Trp-High LNAA; 0.34% SID Trp: 5.4% SID LNAA), and HTrpHLNAA (High Trp-Low LNAA; 0.34% SID Trp: 4.6% SID LNAA), and 2) without/with ETEC infection. Pigs were orally infected with 0.8 mL (3.6 × 109 CFU/mL) ETEC at days 7 and 8 after weaning. Pigs fed diets high in Trp irrespective of the level of LNAA (HTrpHLNAA and HTrpLLNAA) had higher plasma Trp concentrations (P < 0.001) and a Trp:LNAA ratio (P < 0.001) before infection and 6 d after infection. Following infection, noninfected pigs had higher plasma Trp (P = 0.03) and a Trp:LNAA ratio (P = 0.004) compared with pigs infected with ETEC. Plasma cortisol levels after infection were higher in ETEC-infected pigs (P = 0.05) and altering dietary Trp and LNAA concentrations did not influence (P > 0.05) plasma cortisol. Pigs fed diet HTrpLLNAA had higher serum serotonin levels 24 h after infection (P = 0.02) compared with pigs fed diets LTrpLLNAA and HTrpHLNAA. Similarly, pigs fed diet HTrpLLNAA had a higher (P = 0.02) average daily gain during the 3-wk study. Overall, average daily feed intake tended to be higher in pigs fed an HTrpLLNAA diet compared with the other diets (P = 0.08). These results suggest that the increased supplementation of dietary Trp with reduced LNAA increased circulating Trp levels that, in turn, likely caused higher serum serotonin levels, irrespective of infection with ETEC, and improved aspects of post-weaning performance.

The present study investigated the effects of supplementing a low protein (LP) diet supplemented with key essential amino acids (AA) to broilers on growth performance, intestinal tract function, blood metabolites, and nitrogen excretion when the animals were maintained under various sanitary conditions for 35 D after hatching. Three hundred eighty-four one-day-old male broilers (Ross 308) were randomly allotted to groups that received one of 6 dietary treatments in a 2 × 3 factorial arrangement (i.e., 2 environmental conditions and 3 dietary treatments) to give 8 replicates per treatment. Broilers were challenged with 2 environmental conditions (sanitary vs. poor sanitary). The dietary treatments were (1) high protein (HP) diet, (2) LP diet, and (3) LP diet with synthetic key essential AA (LPA): the LP diet was supplemented with synthetic AA up to the required levels for broilers. On day 14, birds consumed the LP diet impaired growth performance compared with those fed the HP diet, while the average daily weight gain-to-feed conversion ratio of birds fed the LPA diet improved to the level of birds fed the HP diet under poor sanitary conditions (P < 0.05). Broilers raised under poor sanitary conditions and fed the LP diet displayed higher (P < 0.05) zonula occludens (ZO-1) expression on day 14 than broilers fed either the HP or LPA diet. Under sanitary conditions, birds fed HP and LPA diets showed higher villus height and crypt depth compared with those of broilers fed the LP diet on day 35. Moreover, broilers raised in the poor sanitary environment had higher (P < 0.05) serum endotoxins than those raised in the sanitary environment. Broilers fed the LPA diet showed reduced (P < 0.05) nitrogen excretion on days 14 and 35 compared with those fed the LP and HP diets independent of the environment. In conclusion, the LPA diet did not impair growth performance under poor sanitary conditions for 14 D after hatch while resulting in lower nitrogen excretion in any environment conditions throughout the experiment.

An infection model with enterotoxigenic Escherichia coli (ETEC) harboring the F4 fimbriae can be used to assess the impacts that various challenges associated with weaning (e.g., dietary, psychological, environmental) have on the expression of postweaning diarrhea. The objective of this study was to develop a novel inoculation method for administering an ETEC culture that would induce a higher proportion of ETEC-F4 diarrhea, in pigs that genetically showed ETEC-F4 susceptibility or resistance. The study was designed as a factorial arrangement of treatments with the factors being 1) partially susceptible or resistant to ETEC-F4 based on genetic testing, and 2) 4 challenge treatments, being a) a conventional liquid broth method using a drenching gun [Positive control (PC)], b) a Syringe method, c) a Capsule method, and d) Negative control [pigs not challenged (NC)]. At 21 ± 3 d of age (mean ± SEM), 48 male castrate pigs (Large White × Landrace) weighing approximately 7.0 ± 1.18 kg were allocated to 4 treatment groups in 2 replicate pens (6 pigs per pen). Initial ETEC-F4 susceptibility was based on a DNA marker test and each treatment group had 9 partially susceptible and 3 resistant pigs. On days 7 and 8 after weaning, pigs were challenged with ETEC (serotype O149:K88; toxins LT1, ST1, ST2, and EAST). On each inoculation day the PC pigs were orally dosed with 9 mL 7.12 × 109 colony-forming unit (CFU), the Syringe pigs with 0.8 mL 6.72 × 109 CFU, the Capsule pigs were orally administered 2 capsules containing 0.8 mL 3.28 × 109 CFU, and the NC pigs 1 mL of phosphate-buffered saline (PBS) solution. Approximately 72 h after infection, 44, 22, 78, and 0% of partially susceptible pigs in the PC, the Syringe, the Capsule, and the NC group had developed ETEC-F4 diarrhea (P = 0.007). Partially susceptible pigs had a higher diarrhea index (DI) compared to resistant pigs (31.5 vs. 4.8, P < 0.001). The NC group had a lower DI compared to the PC and Capsule pigs (3.9, 38.1, and 40.3, respectively, P < 0.005). Following infection, genetically resistant pigs in the Capsule group had a DI of zero and the partially susceptible pigs had a DI of 55.6 (P = 0.014). This study showed that genetically screening pigs and using a Capsule to deliver ETEC-F4 can increase cases of diarrhea and the efficiency of the challenge model. Taken together, these methods have the potential to reduce the number of pigs needed in future experimental infection studies.

Gilt progeny (GP) often have restricted growth performance and health status in comparison to sow progeny (SP) from birth, with the underlying mechanisms responsible for this yet to be fully understood. The present study aimed to compare differences in growth and development between GP and SP in the first 24 h after birth and in the periweaning period. Two cohorts of pigs including 36 GP and 37 SP were euthanized at 1 of 4 time points: a birth cohort (at birth before suckling, 0 h; and 24 h after birth, 24 h; n = 33) and a weaning cohort (at approximately 29 d of age; "pre-weaning," PrW; and 24 h after weaning; "post-weaning," PoW; n = 40). Pigs were individually weighed at 0 h, 24 h, PrW, and PoW up until the point of euthanasia, at which time the weights of selected tissues and organs were recorded and analyzed relative to BW. The length of the small intestine (SI), femur, and body were also measured, and a serum sample was collected and analyzed for IgG concentration. Samples of jejunal and ileal mucosa were collected and analyzed for total protein and specific activity of lactase. Euthanized GP were lighter (P < 0.01) than SP at all time points. At all time points, the ratios of quadriceps weight to femur length, BW to body length, spleen to BW (all P < 0.05), and SI weight to length (P < 0.10) were lower in GP than in SP. There was no difference (P ≥ 0.05) in stomach or heart to BW ratios between GP and SP in either cohort. The brain to liver weight ratio was greater (P = 0.044) in GP than in SP in the birth cohort, and the brain to BW ratio was greater (P < 0.01) in GP in both the birth and weaning cohorts. The liver to BW ratio was similar (P = 0.35) at birth but greater (P = 0.014) in GP around weaning. Total mucosal protein content in the jejunum and ileum was lower (P = 0.007) in GP at 24 h compared with SP, and specific activity of lactase was greater (P = 0.022) in GP in the birth cohort, whereas there were no differences in the weaning cohort (P ≥ 0.10). Gilt progeny had lower (P < 0.001) serum IgG concentration compared with SP at 24 h, but there was no difference (P ≥ 0.10) in the weaning cohort. Collectively, these findings suggest that the early development of GP may be delayed compared with SP and that a number of the anatomical differences between GP and SP that exist after birth are also present at weaning.

1,3-Butanediol (BD) is a ketogenic substance that can improve piglet growth and survival and potentially increase performance in gilt progeny when provided as a dietary supplement during late gestation. Gilts (n = 77; parity 1) and sows (n = 74; parities 2 and 3) were fed either a standard commercial gestation diet or a diet supplemented with 4% BD from day 90 of gestation until farrowing. Dams fed with diets supplemented with BD had higher plasma beta-hydroxybutyrate (p = 0.01) and lower non-esterified fatty acid concentrations (p < 0.001). The percentage of progeny that were light-for-age (<1.1 kg) at birth was decreased by BD (18.2 vs. 13.5%, p < 0.006), particularly in gilts (24.0 vs. 18.3%, p < 0.034). Individual birth weights and litter weights birth weights tended to be increased by the BD diet (p = 0.085 and 0.078; respectively) although these effects were not maintained to weaning. Pre-weaning mortality was greater in gilt than in sow progeny and was not altered by dietary BD. Feeding BD in late gestation can improve birth weight, but further work is needed to see if these effects are carried through subsequent stages of growth, particularly in gilt progeny.

BACKGROUND: This study investigated the validity of the DNA-marker based test to determine susceptibility to ETEC-F4 diarrhoea by comparing the results of two DNA sequencing techniques in weaner pigs following experimental infection with F4 enterotoxigenic Escherichia coli (ETEC-F4). The effects of diet and genetic susceptibility were assessed by measuring the incidence of piglet post-weaning diarrhoea (PWD), faecal E. coli shedding and the diarrhoea index. RESULTS: A DNA marker-based test targeting the mucin 4 gene (MUC4) that encodes F4 fimbria receptor identified pigs as either fully susceptible (SS), partially or mildly susceptible (SR), and resistant (RR) to developing ETEC-F4 diarrhoea. To further analyse this, DNA sequencing was undertaken, and a significantly higher proportion of C nucleotides was observed for RR and SR at the XbaI cleavage site genotypes when compared to SS. However, no significant difference was found between SR and RR genotypes. Therefore, results obtained from Sanger sequencing retrospectively allocated pigs into a resistant genotype (MUC4-), in the case of a C nucleotide, and a susceptible genotype (MUC4+), in the case of a G nucleotide, at the single nucleotide polymorphism site. A total of 72 weaner pigs (age ~ 21 days), weighing 6.1 ± 1.2 kg (mean ± SEM), were fed 3 different diets: (i) positive control (PC) group supplemented with 3 g/kg zinc oxide (ZnO), (ii) negative control (NC) group (no ZnO or HAMSA), and (iii) a diet containing a 50 g/kg high-amylose maize starch product (HAMSA) esterified with acetate. At days five and six after weaning, all pigs were orally infected with ETEC (serotype O149:F4; toxins LT1, ST1, ST2 and EAST). The percentage of pigs that developed diarrhoea following infection was higher (P = 0.05) in MUC4+ pigs compared to MUC4- pigs (50% vs. 26.8%, respectively). Furthermore, pigs fed ZnO had less ETEC-F4 diarrhoea (P = 0.009) than pigs fed other diets, however faecal shedding of ETEC was similar (P > 0.05) between diets. CONCLUSION: These results confirm that MUC4+ pigs have a higher prevalence of ETEC-F4 diarrhoea following exposure, and that pigs fed ZnO, irrespective of MUC4 status, have reduced ETEC-F4 diarrhoea. Additionally, sequencing or quantifying the single nucleotide polymorphism distribution at the XbaI cleavage site may be more reliable in identifying genotypic susceptibility when compared to traditional methods.

Feeding conjugated linoleic acid (CLA) or medium-chain fatty acids (MCFA) to dams has been shown to improve progeny growth and survival, and hence may be particularly advantageous to gilt progeny. Primiparous (n = 129) and multiparous sows (n = 123; parities 3 and 4) were fed one of four diets from day 107 of gestation (107.3 ± 0.1 days) until weaning (day 27.2 ± 0.1 of lactation): (i) control diet; (ii) 0.5% CLA diet; (iii) 0.1% MCFA diet; and (iv) equal parts of (ii) and (iii). Progeny performance data were collected and, from a subset of sows (n = 78) and their piglets (n = 144), a colostrum (day 0), milk (day 21), and piglet serum sample (day 3) were analyzed for immunoglobulin G and several selected metabolites. Liveborn pre-weaning mortality tended to be lowest (p = 0.051) in piglets from sows fed 0.5% CLA. However, sows fed the CLA diet had more (p = 0.005) stillbirths than those on the other diets. There were few effects of diet or the dam parity x diet interaction (p ≥ 0.05) on other parameters. Overall, feeding CLA or MCFA did not improve the performance of primiparous sows, multiparous sows, or their progeny.

It is important to understand the biological factors influencing the poorer lifetime performance of gilt progeny in comparison to sow progeny and determine whether this may be partially due to differences in lactation performance between primiparous and multiparous sows. It was hypothesized that primiparous sows would have lower levels of immunoglobulin G (IgG) in colostrum and milk compared to multiparous sows, and lower levels of other energetic components. Differences in colostrum and milk composition between ten primiparous and ten multiparous sows (parities 3 and 4) from a commercial herd were examined throughout lactation (day 0, 1, 2, 3, 7, 14, and 21). Overall, there were no (p ≥ 0.05) parity differences in total IgG, fat, protein, lactose, and net energy (NE) concentrations. Primiparous sows had higher lactose levels at day 2 (parity by timepoint interaction; p = 0.036) and lower NE at day 3 (p = 0.091), and multiparous sows had higher lactose levels at days 14 and 21. Results suggest that shortcomings of gilt progeny are unlikely due to insufficient nutrient levels in colostrum and milk, and more likely to reduced colostrum and milk intake and their capacity to digest and absorb each component.
