RESUMO

Pregnancy losses from fixed-time embryo transfer (FTET) to calving were evaluated in Bos indicus-influenced beef and dairy recipients. Data from 4366 FTET events were collected from Nelore × Angus recipient heifers, and from 38538 FTET events in Gir × Holstein recipient heifers and cows. In beef recipients, pregnancy losses were greater (P < 0.01) from FTET (day 7 of the experiment) to day 32 compared with day 32-100 and with day 100 to calving (58.7, 39.5, and 36.7%, respectively), and did not differ (P = 0.56) between these latter periods. Recipients that lost the pregnancy from FTET to day 32 gained less (P < 0.01) body condition score after FTET compared with recipients that maintained the pregnancy. Pregnancy losses from day 32 to calving were greater (P < 0.01) in recipients reared in drylots and moved to pastures on day 32 compared with recipients reared on pasture. In dairy recipients, pregnancy losses from FTET (day 7) to day 32 were greater (P < 0.01) than losses from day 32 to calving (50.4 and 29.4%). Pregnancy losses throughout gestation were greater (P < 0.01) when the FTET event was performed during the warm season, and greater (P < 0.01) in recipients with < 3/8 Gir influence. Recipients with ≥ 3/8 Gir influence experienced a lesser (P ≤ 0.05) increase in pregnancy losses during the warm season compared with recipients with < 3/8 Gir influence. Collectively, this experiment provides novel information about pregnancy losses in B. indicus-influenced herds receiving FTET.

This experiment evaluated pregnancy losses from day 30 of gestation to calving in Bos indicus females, and evaluated if serum haptoglobin concentrations during early gestation impacts subsequent pregnancy losses. A total of 4926 Nelore females were used, being 1802 nulliparous heifers inseminated as yearlings (precocious heifers), 1356 nulliparous heifers inseminated at 24 months of age (conventional heifers), 887 suckling primiparous cows, and 881 suckling multiparous cows. Cows were assigned to an ovulation synchronization + fixed-time artificial insemination (FTAI) protocol from day -11 to 0 of the experiment. Pregnancy status was verified using transrectal ultrasonography on days 30 and 60 after FTAI, via transrectal palpation on day 150 after FTAI, and according to calf birth. Blood samples were collected from all animals diagnosed pregnant on day 30 after FTAI, and analyzed for serum concentrations of haptoglobin and pregnancy associated glycoproteins (PAG). Pregnancy loss was greater (P < 0.01) from day 60-150 of gestation (10.2 %) compared with day 30-60 of gestation (6.0 %) and with day 150 of gestation to calving (7.4 %), and differed (P = 0.04) between these latter two periods. Pregnancy loss from day 30-60 of gestation did not differ (P = 0.26) among parities, whereas total pregnancy losses (day 30 to calving) were greater (P < 0.01) in precocious (28.4 %) and conventional (27.1 %) heifers compared with primiparous (16.4 %) and multiparous (13.0 %) cows. Serum PAG concentrations on day 30 after FTAI were less (P ≤ 0.03) in cows that lost the pregnancy (5.63 ng/mL) from day 30-60 of gestation, as well as those that lost the pregnancy from day 30 to calving (8.59 ng/mL) compared with cohorts that maintained the pregnancy (9.39 and 9.32 ng/mL, respectively). No differences in serum PAG concentrations on day 30 after FTAI were noted (P ≥ 0.23) according to pregnancy losses from day 60 to calving. Serum haptoglobin concentration on day 30 after FTAI also did not differ (P ≥ 0.48) between cows that maintained or lost the pregnancy. This experiment provides novel information about pregnancy losses after day 30 of gestation in B. indicus cattle, with most losses occurring as fetal mortality and not affected by systemic inflammation during early gestation. Pregnancy losses were nearly doubled in precocious and conventional heifers compared with parous cows, demonstrating the relevance of this reproductive failure to B. indicus replacement heifers.

Fragrances can cause allergic skin reactions, expressed as allergic contact dermatitis and reactions in the respiratory tract that range from acute temporary upper airway irritation to obstructive lung disease. These adverse health effects may result from the stimulation of a specific (adaptive) immune response. Th1 cells, which essentially produce interleukin-2 (IL-2) and interferon-γ (IFN-γ), play a key role in allergic contact dermatitis and also on allergic sensitization to common allergens (e.g., nickel and fragrance). It has been shown that fragrance allergy leads to Th2/Th22 production of IL-4, IL-5 and IL-13, controlling the development of IgE and mediating hypersensitivity reactions in the lung, such as asthma. Cytokines released during immune response modulate the expression of cytochrome P450 (CYPs) proteins, which can result in alterations of the pharmacological effects of substances in inflammatory diseases. The mechanisms linking environment and immunity are still not completely understood but it is known that aryl hydrocarbon receptor (AhR) is a sensor with conserved ligand-activated transcription factor, highly expressed in cells that controls complex transcriptional programs which are ligand and cell type specific, with CYPs as targeted genes. This review focuses on these important aspects of immune responses of the skin and respiratory tract cells, describing some in vitro models applied to evaluate the mechanisms involved in fragrance-induced allergy.

A new alternative for hydrodynamic cavitation-assisted pretreatment of sugarcane bagasse was proposed, along with a simultaneous saccharification and co-fermentation (SSCF) process performed in interconnected columns. Influential variables in the pretreatment were evaluated using a statistical design, indicating that an ozone flow rate of 10 mg min-1 and a pH of 5.10 resulted in 86 % and 72 % glucan and xylan hydrolysis yields, respectively, in the subsequent enzymatic hydrolysis process. Under these optimized conditions, iron sulfate (15 mg L-1) was added to assess Fenton pretreatment, resulting in glucan and xylan hydrolysis yields of 92 % and 71 %, respectively, in a material pretreated for 10 min. In SSCF, ethanol volumetric productivities of 0.33 g L-1 h-1 and of 0.54 g L-1 h-1 were obtained in batch and fed-batch operation modes, achieving 26 g L-1 of ethanol in 48 h in the latter mode.

INTRODUCTION: Low muscle mass is a common condition in the critically ill population and is associated with adverse clinical outcomes. The primary aim of this study was to analyze the prognostic significance of low muscle mass using computed tomography (CT) scans in COVID-19 critically ill patients. A second objective was to determine the accuracy and agreement in low muscle mass identification using diverse markers compared to CT as the gold standard. METHODS: This was a prospective cohort study of COVID-19 critically ill patients. Skeletal muscle area at the third lumbar vertebra was measured. Clinical outcomes (intensive care unit [ICU] and hospital length of stay [LOS], tracheostomy, days on mechanical ventilation [MV], and in-hospital mortality) were assessed. Phase angle, estimated fat-free mass index, calf circumference, and mid-upper arm circumference were measured as surrogate markers of muscle mass. RESULTS: Eighty-six patients were included (mean age ± SD: 48.6 ± 12.9; 74% males). Patients with low muscle mass (48%) had a higher rate of tracheostomy (50 vs 20%, p = 0.01), prolonged ICU (adjusted HR 0.53, 95%CI 0.30-0.92, p = 0.024) and hospital LOS (adjusted HR 0.50, 95% CI 0.29-0.86, p = 0.014). Bedside markers of muscle mass showed poor to fair agreement and accuracy compared to CT-assessed low muscle mass. CONCLUSION: Low muscle mass at admission was associated with prolonged length of ICU and hospital stays. Further studies are needed to establish targeted nutritional interventions to halt and correct the catabolic impact of COVID-19 in critically ill patients, based on standardized and reliable measurements of body composition.

Air pollution has been identified as one of the main environmental risks to health. Since exercise training seems to act as an anti-inflammatory modulator, our hypothesis is that exercise training prevents damage to respiratory and cardiovascular function caused by diesel exhaust particle (DEP) exposure. This study aimed to evaluate whether aerobic exercise training prior to DEP exposure prevents inflammatory processes in the pulmonary and cardiovascular systems. Therefore, BALB/C male mice were or were not submitted to a 10-week exercise training protocol (5×/week, 1 h/d), and after four weeks, they were exposed to DEP in a chamber with 24 µg/m3 PM2.5 or filtered air. Heart rate variability, lung mechanics and bronchoalveolar lavage fluid, cytokines and polymorphonuclear cells in the lung parenchyma were evaluated. Exposure to DEPs reduced heart rate variability and the elastance of the respiratory system and increased the number of cells in bronchoalveolar lavage fluid, as well as macrophages, neutrophils and lymphocytes, the density of polymorphonuclear cells and the proportion of collagen fibres in the lung parenchyma. Additionally, DEP-exposed animals showed increased expression of IL-23 and IL-12p40 (proinflammatory cytokines) and inducible nitric oxide synthase. Exercise training avoided the increases in all these inflammatory parameters, except the elastance of the respiratory system, the amount of collagen fibres and the expression of inducible nitric oxide synthase. Additionally, trained animals showed increased expression of the anti-inflammatory cytokine IL-1ra. Although our data showed a reduction in proinflammatory markers and an increase in markers of the anti-inflammatory pathway, these changes were not sufficient to prevent damage to the lung and cardiovascular function induced by DEPs. Based on these data, we propose that aerobic exercise training prevents the lung inflammatory process induced by DEPs, although it was not sufficient to avoid chronic damage, such as a loss of lung function or cardiovascular events.

RESUMO

Environmental problems due to utilization of fossil-derived materials for energy and chemical generation has prompted the use of renewable alternative sources, such as lignocellulose biomass (LB). Indeed, the production of biomolecules and biofuels from LB is among the most important current research topics aiming to development a sustainable bioeconomy. Yet, the industrial use of LB is limited by the recalcitrance of biomass, which impairs the hydrolysis of the carbohydrate fractions. Hydrodynamic cavitation (HC) and Advanced Oxidative Processes (AOPs) has been proposed as innovative pretreatment strategies aiming to reduce process time and chemical inputs. Therefore, the underlying mechanisms, procedural strategies, influence on biomass structure, and research gaps were critically discussed in this review. The performed discussion can contribute to future developments, giving a wide overview of the main involved aspects.

INTRODUCTION: Entry into prison involves adapting to a prison culture that is sometimes altered by the effect of imprisonment. Prisons are overcrowded and hold large numbers of inmates suffering from mental disorders and difficulties of adaptation, who affect the delicate equilibrium of the prison environment and can worsen the relational climate. MATERIAL AND METHOD: Several bibliographical databases on the influence of the mental health of adult inmates on the prison relationship climate and existing interventions in this regard that have been published in the last 15 years were reviewed. This data was complemented by other information obtained from the online bibliographic indexes of the Ministry of the Interior. RESULTS: There is little literature on the influence of mental health on the relational climate of prisons and existing interventions. However, what literature there is does respond to a current prison reality where inmates with mental health problems have an increased risk of victimization and also a greater predisposition to penitentiary misconduct that causes violations of rules and the consequent application of disciplinary sanctions that lead to segregation. DISCUSSION: The dysfunctional behaviour of such inmates, as well as the stigmatized treatment they receive, negatively impact the prison social climate, generating pathological relational styles and distorting the prison environment. This creates a need for an adequate number of programs and interventions of sufficient quality to prevent and mitigate their consequences.

OBJECTIVES: Sarcopenia is a debilitating condition affecting millions of individuals worldwide and is defined with different criteria. The objective of this study was to determine the prevalence of sarcopenia in older Canadians using three internationally accepted criteria. DESIGN: Observational cohort study. SETTINGS AND PARTICIPANTS: Data from 12,592 subjects [6,314 males (50.1%), 6,278 females (49.9%)] ≥65 years old in the Canadian Longitudinal Study on Aging were included. MEASUREMENTS: Appendicular lean mass (ALM; kg) and appendicular lean mass index (ALM kg/height in m2) were collected from dual X-ray absorptiometry measurements. Physical performance was assessed using the 4-m gait speed test, and muscle strength was measured by hand dynamometry. Sarcopenia was defined according to criteria put forth by the International Working Group on Sarcopenia (IWGS), Foundation for the National Institutes of Health (FNIH) Sarcopenia Project, and revised European Working Group on Sarcopenia in Older People (EWGSOP). Positive and negative percent agreements and Cohen's kappa (κ) described the agreement among sarcopenia definitions. RESULTS: Among the evaluated criteria, gait speed ≤ 1.0 m/s (IWGS definition of slowness) was the most frequently identified deficit (56.8% males, 57.2% females). The prevalence of sarcopenia ranged from 1.4 to 5.2% in males and 1.6 to 7.2 % in females among the different definitions. Positive percent agreement values among criteria were generally low (range: 1.5 - 55.3%) and corresponded to κ indicating none to minimal agreement (0.01 - 0.23). Negative percent agreement values were ≥ 95%. CONCLUSION: Sarcopenia prevalence was relatively low in older Canadian adults and current definitions had poor agreement in diagnosing individuals as sarcopenic.

AIM: To investigate the shaping and cleaning performance of the Clearsonic ultrasonic tip as a supplementary tool during the preparation of long oval-shaped root canals through micro-computed tomographic (micro-CT) imaging technology. The Reciproc M-Wire R40 instrument was used as a reference instrumentation technique for comparison. METHODOLOGY: Twenty mandibular incisors with single, straight and long oval-shaped canals were selected and pair-matched by micro-CT scanning. The root canals were prepared with Reciproc R25 and R40 instruments, scanned again in a micro-CT device and randomly allocated into one of the two experimental groups (n = 10), according to the supplementary debridement protocol used: Clearsonic or Reciproc R40 used in a brushing motion against buccal and lingual walls. Noninstrumented canal areas, accumulation of hard-tissue debris (AHTD) and volume of removed dentine were assessed using micro-CT imaging following the supplementary debridement protocols. Data were analysed statistically in two phases using a t-test with a significance level of 5%. RESULTS: In phase-1, there was a significant similarity between the samples regarding the evaluated micro-CT baseline parameters (P > 0.05), validating the anatomical similarity between the tooth pairs. In phase-2, there was a significant difference between the groups for canal volume, surface area, noninstrumented root canal walls and the amount of removed dentine for both absolute and percentage values (P < 0.05), but not for AHTD (P = 0.759). CONCLUSIONS: The Clearsonic tip outperformed the Reciproc R40 instrument as a supplementary debridement protocol as it was associated with significantly greater reduction of noninstrumented root canals walls and consequently a larger amount of removed dentine. Both groups performed similarly regarding the AHTD.

INTRODUCCIÓN: La entrada en prisión supone la necesidad de adaptación a una cultura penitenciaria que, en ocasiones, se encuentra alterada por el efecto de la prisionización. La sobreocupación de los reclusos con trastornos mentales y sus dificultades adaptativas en las instalaciones obstaculizan el equilibrio del ambiente carcelario e implica el empeoramiento del clima relacional de prisiones. MATERIAL Y MÉTODO: Se han revisado distintas bases de datos bibliográficas acerca de la influencia de la salud mental de los reclusos adultos en el clima relacional de prisiones y las intervenciones existentes al respecto publicadas en los últimos 15 años. Estos datos se han completado con los obtenidos en los índices bibliográficos online del Ministerio del Interior. RESULTADOS: Hay poca literatura acerca de la influencia de la salud mental en el clima relacional de prisiones y las intervenciones existentes. Aunque es cierto que responde a la realidad penitenciaria actual, donde los reclusos con problemas de salud mental poseen un mayor riesgo de estigmatización, pero también una mayor predisposición a la realización de malas conductas penitenciaras que ocasionan infracciones de reglas y la consecuente aplicación de sanciones disciplinarias que tienden a la segregación. DISCUSIÓN: El comportamiento disfuncional de este tipo de reclusos, así como el trato estigmatizado que reciben, impactan negativamente en el clima social penitenciario, generando estilos relacionales patológicos y distorsionando el ambiente de la prisión. Este hecho pone de relieve la necesidad de alcanzar una cantidad y calidad adecuada de programas e intervenciones que puedan prevenir y mitigar sus consecuencias

O presente artigo é um relato sobre uma fêmea de D. novemcinctus que foi capturada de forma imprevista, durante um projeto de avaliação sanitária de animais silvestres sinantrópicos do município de Toledo/PR (autorização Sisbio/Ibama n. 55109-1). Durante a avaliação física, amostras de fezes foram coletadas após defecação espontânea e encaminhadas para análise. O material foi submetido aos métodos de flutuação simples em solução hipersaturada de cloreto de sódio e centrífugo-flutuação em sacarose. Na leitura das lâminas obtidas pelo método de centrífugo-flutuação em sacarose, foram observadas estruturas com comprimento de aproximadamente 10µm, forma bicônica e um tampão em cada extremidade, compatíveis com esporocistos de Monocystis sp. Protozoários do filo Apicomplexa, os Monocystis sp., são geralmente parasitas de anelídeos, como as minhocas. Esse organismo é comumente correlato a pseudoparasitismo em vários animais, porém não foram encontrados relatos de pseudoparasitismo desse agente em tatus-galinhas, logo essa é a primeira descrição de Monocystis sp. em espécime de D. novemcinctus.(AU)


The present article is an account of a female of D. novemcinctus which was captured during a sanitary evaluation project of synanthropic wild animals of the municipality of Toledo/PR (license SISBIO/ICMBIO n°. 55109-1). During a physical evaluation, samples of feces were collected, after spontaneous defecation, and sent for analysis. The material was subjected to simple flotation methods in hypersaturated sodium chloride solution and centrifugal-flotation solutions in sucrose. In a test reading by the centrifugal-flotation method in sucrose, structures with approximately 10µm, biconical shape and one plate at each end were observed, compatible with sporocysts of Monocystis sp. Protozoa of the phylum Apicomplexa, like Monocystis sp., it is common parasitic annelids, such as worms. This organism is a correlate of pseudoparasitism in several animals but was not found to describe this product as an agent in nine banded armadillos, so this is a first description of Monocystis sp. in specimen of D. novemcinctus.(AU)

OBJECTIVE: To identify the prevalence of sarcopenic obesity, a phenotype of low muscle mass and high adiposity, in adults with end-stage knee osteoarthritis (OA). Various diagnostic criteria, including assessment of muscle/fat mass, muscle strength and physical function, were used to identify patients with and without sarcopenic obesity, and to compare outcomes of pain, function and quality of life. DESIGN: Cross-sectional clinical study including adults with a body mass index (BMI) ≥30 kg/m2 and knee OA. Body composition was measured by dual-energy X-ray absorptiometry (DXA). Assessments included gait speed, handgrip strength, six minute walk test, and self-reported pain, physical function, and health-related quality of life using the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) and EuroQol Foundation (EQ-5D). RESULTS: 151 adults (59% female) aged 65.1 ± 7.9 years, mean BMI 37.1 ± 5.5 kg/m2, were included. Prevalence of sarcopenic obesity using diagnostic cut-offs of appendicular skeletal muscle mass (ASM) relevant to height2, weight and BMI varied from 1.3% (95% confidence interval (CI): 0.2-4.7%) to 14.6% (9.4-21.2%) and 27.2% (20.2-35%), respectively. A combined diagnostic approach including low ASM with either low strength or low function yielded a prevalence of 8.6% (4.7-14.3%). Sarcopenic obesity influenced walking speed, endurance, strength, and patient-reported difficulty with self-care activities, regardless of diagnostic approach. CONCLUSION: Prevalence of sarcopenic obesity varied depending on diagnostic criteria. Given the impact of this condition and OA on physical function, we suggest a combined diagnostic approach be used to clarify expected prevalence and enable early clinical identification and management of sarcopenic obesity in patients with knee OA.

RESUMO

Development of new technologies for pretreatment of lignocellulosic biomass is a current research challenge. In this way, hydrodynamic cavitation (HC) was used to assist alkaline hydrogen peroxide pretreatment of sugarcane bagasse (SCB) in sequential batches (SB-HC), semi-continuous (SC-HC) and continuous (C-HC) processes. Pretreatment resulted in compositional modifications in the material, mainly regarding the cellulose and lignin contents. The released sugars after enzymatic hydrolysis resulted, on average, in 42 g and 32-35 g of glucose per 100 g of SCB for samples treated in B-HC (10 min of process) and SC-HC process (7.5 min residence time), respectively. In C-HC process, with an average residence time of 7.5 min and 3.75 min, 38-46 g and 32-38 g of glucose per 100 g of SCB were obtained respectively in enzymatic hydrolysis step. HC technology was shown as a promising alternative for pretreatment of lignocellulosic biomass in all evaluated configurations aiming to produce high value bioproducts.

The roles of obestatin and adropin in paediatric obesity are poorly understood. We compared obestatin and adropin concentrations in younger (n = 21) and older children (n = 14) with Prader-Willi syndrome (PWS) and age and BMI-z-matched controls (n = 31). Fasting plasma obestatin and adropin were higher in younger children with PWS than controls; adropin was also higher in older children with PWS. Growth hormone treatment had no effects on obestatin or adropin in PWS. The ratio of ghrelin to obestatin declined from early to late childhood but was higher in older PWS than older controls. Adropin correlated with fasting glucose in the PWS group only. Changes in the ratio of ghrelin to obestatin may suggest changes in the processing of preproghrelin to ghrelin and obestatin during development and differential processing of preproghrelin in PWS.
