RESUMO

Direct white light emitting phosphors play a significant role in the display industry due to their ability to improve the quality, efficiency, and versatility of lighting sources used in most of the displays. The currently investigated phosphor SrZr2 CaLa2 O8 :Eu3+ was prepared by a conventional solid-state reaction method. It has been observed that the stoichiometric ratio of all precursors plays an important role in determining the characteristics of the final phosphor. From X-ray diffraction (XRD) analysis, the phosphor was observed to have a hexagonal phase and a crystal size of ~28 nm. Scanning electron microscopy (SEM) observations revealed a cluster of rod-like structures with an average diameter of ~0.2 µm. The excitation peak maximum observed at 280 nm is due to charge transfer between Eu3+ -O2- ions. The energy transitions 7 F0 → 5 L6 and 7 F0 → 5 D2 are responsible for the appearance of other excitation peaks at ultraviolet (UV) (395 nm), blue (~467 nm), green (~540 nm), orange (~590 nm), and red (~627 nm) attributed to 5 D0 → 7 FJ (J = 0-4) transitions of europium ion (Eu3+ ). The Commercial International de I'Eclairage (CIE) chromaticity coordinates were estimated to be (0.37, 0.0.33) and (0.67, 0.33) for the emissions corresponding to 395 and 590 nm, respectively. The characteristic emissions of Eu3+ ions allow this novel phosphor to be used to generate direct white light in light-emitting diodes (LEDs), which is otherwise difficult to achieve in single-component systems.

RESUMO

Hybrid rice technology offers great promise to further enhance rice production and productivity for global food security. Improving hybrid rice parental lines is the first step in developing heterotic rice hybrids. To improve resistance against blast disease, a maintainer line DRR 9B was fortified with a major broad-spectrum blast resistance gene Pi2 through marker-assisted selection. The rice blast caused by Magnaporthe oryzae is a major disease and can cause severe yield losses upto 100%. The NILs of Samba Mahsuri namely BA-23-11-89-12-168 possessing Pi2 was utilized as a donor parent. The PCR-based molecular marker tightly linked to Pi2 gene was used for the foreground selection at BC1F1 generation. The molecular marker tightly linked to the major fertility restorer gene Rf4 was used for negative selection (i.e., selection of plants possessing non fertility restoring alleles) at BC1F1 generation to identify maintainer lines. The positive plants with Rf4 gene were added to the restorer pool for restorer line development. At each stage, MAS for Pi2 coupled with stringent phenotypic selection for agro-morphological and grain quality traits were exercised. At BC1F3 generation, one hundred families were screened against blast disease at uniform blast nursery (UBN) and selected resistant lines were advanced to next generations. In the BC1F5 generation plants were subjected to agro-morphological evaluation for yield and yield-contributing traits. The selected plants at BC1F5 generation were crossed with DRR 9A to assess the maintainer ability of blast resistance lines and for further CMS line conversion for hybrid rice breeding for developing blast resistance rice hybrids.

RESUMO

A doubled haploid (DH) population consisting of 125 DHLs derived from the popular rice hybrid, KRH-2 (IR58025A/KMR3R) was utilized for Quantitative Trait Loci (QTL) mapping to identify novel genomic regions associated with yield related traits. A genetic map was constructed with 126 polymorphic SSR and EST derived markers, which were distributed across rice genome. QTL analysis using inclusive composite interval mapping (ICIM) method identified a total of 24 major and minor effect QTLs. Among them, twelve major effect QTLs were identified for days to fifty percent flowering (qDFF12-1), total grain yield/plant (qYLD3-1 and qYLD6-1), test (1,000) grain weight (qTGW6-1 and qTGW7-1), panicle weight (qPW9-1), plant height (qPH12-1), flag leaf length (qFLL6-1), flag leaf width (qFLW4-1), panicle length (qPL3-1 and qPL6-1) and biomass (qBM4-1), explaining 29.95-56.75% of the phenotypic variability with LOD scores range of 2.72-16.51. Chromosomal regions with gene clusters were identified on chromosome 3 for total grain yield/plant (qYLD3-1) and panicle length (qPL3-1) and on chromosome 6 for total grain yield/plant (qYLD6-1), flag leaf length (qFLL6-1) and panicle length (qPL6-1). Majority of the QTLs identified were observed to be co-localized with the previously reported QTL regions. Five novel, major effect QTLs associated with panicle weight (qPW9-1), plant height (qPH12-1), flag leaf width (qFLW4-1), panicle length (qPL3-1) and biomass (qBM4-1) and three novel minor effect QTLs for panicle weight (qPW3-1 and qPW8-1) and fertile grains per panicle (qFGP5-1) were identified. These QTLs can be used in breeding programs aimed to yield improvement after their validation in alternative populations. RESUMO

Recent predictions on climate change indicate that high temperature episodes are expected to impact rice production and productivity worldwide. The present investigation was undertaken to assess the yield stability of 72 rice hybrids and their parental lines across three temperature regimes over two consecutive dry seasons using the additive main effect and multiplicative interaction (AMMI), genotype and genotype × environment interaction (GGE) stability model analysis. The combined ANOVA revealed that genotype × environment interaction (GEI) were significant due to the linear component for most of the traits studied. The AMMI and GGE biplot explained 57.2% and 69% of the observed genotypic variation for grain yield, respectively. Spikelet fertility was the most affected yield contributing trait and in contrast, plant height and tiller numbers were the least affected traits. In case of spikelet fertility, grain yield and other yield contributing traits, male parent contributed towards heat tolerance of the hybrids compared to the female parent. The parental lines G74 (IR58025B), G83 (IR40750R), G85 (C20R) and hybrids [G21 (IR58025A × KMR3); G3 (APMS6A × KMR3); G57 (IR68897A × KMR3) and G41 (IR79156A × RPHR1005)] were the most stable across the environments for grain yield. They can be considered as potential genotypes for cultivation under high temperature stress after evaluating under multi location trials.

RESUMO

Genetic improvement of rice for grain micronutrients, viz., iron (Fe) and zinc (Zn) content is one of the important breeding objectives, in addition to yield improvement under the irrigated and aerobic ecosystems. In view of developing genetic resources for aerobic conditions, line (L) × tester (T) analysis was conducted with four restorers, four CMS lines and 16 hybrids. Both hybrids and parental lines were evaluated in irrigated and aerobic field conditions for grain yield, grain Fe and Zn content. General Combining Ability (GCA) effects of parents and Specific Combining Ability (SCA) effects of hybrids were observed to be contrasting for the micronutrient content in both the growing environments. The grain Fe and Zn content for parental lines were negatively correlated with grain yield in both the contrasting growing conditions. However, hybrids exhibited positive correlation for grain Fe and Zn with grain yield under limited water conditions. The magnitude of SCA mean squares was much higher than GCA mean squares implying preponderance of dominance gene action and also role of complementary non-allelic gene(s) interaction of parents and suitability of hybrids to the aerobic system. The testers HHZ12-SAL8-Y1-SAL1 (T1) and HHZ17-Y16-Y3-Y2 (T2) were identified as good combiners for grain Zn content under irrigated and aerobic conditions respectively.

RESUMO

Hereditary ovarian tumour syndromes are a diverse group of hereditary syndromes characterised by the development of specific histotypes of ovarian neoplasms. While BRCA syndromes are exclusively associated with high-grade serous carcinomas, patients with Lynch syndrome show a preponderance of endometrioid subtype of ovarian and endometrial carcinomas. Distinct non-epithelial phenotypes, such as sex cord stromal tumours with annular tubules, Sertoli-Leydig cell tumours, and small cell carcinoma of the hypercalcaemic type occur in patients with Peutz-Jeghers, DICER1, and rhabdoid tumour predisposition syndromes, respectively. Gorlin-Goltz syndrome is characterised by the development of bilateral, multiple ovarian fibromas in 14-24% of patients. Ovarian steroid cell tumours and broad ligament papillary cystadenomas are characteristically found in women with von Hippel-Lindau syndrome. Recent studies have allowed the characterisation of tumour genetics and associated oncological pathways that contribute to tumourigenesis. Implications of the diagnosis of these syndromes on screening, management, and prognosis are discussed.

RESUMO

The study was undertaken to identify the quantitative trait loci (QTLs) governing yield and its related traits using a recombinant inbred line (RIL) population derived from the popular rice hybrid, KRH-2 (IR58025A/KMR3R). A genetic map spanning 294.2 cM was constructed with 126 simple sequence repeats (SSR) loci uniformly distributed across the rice genome. QTL analysis using phenotyping and genotyping information identified a total of 22 QTLs. Of these, five major effect QTLs were identified for the following traits: total grain yield/plant (qYLD3-1), panicle weight (qPW3-1), plant height (qPH12-1), flag leaf width (qFLW4-1) and panicle length (qPL3-1), explaining 20.23-22.76% of the phenotypic variance with LOD scores range of 6.5-10.59. Few genomic regions controlling several traits (QTL hotspot) were identified on chromosome 3 for total grain yield/plant (qYLD3-1) and panicle length (qPL3-1). Significant epistatic interactions were also observed for total grain yield per plant (YLD) and panicle length (PL). While most of these QTLs were observed to be co-localized with the previously reported QTL regions, a novel, major QTL associated with panicle length (qPL3-1) was also identified. SNP genotyping of selected high and low yielding RILs and their QTL mapping with 1,082 SNPs validated most of the QTLs identified through SSR genotyping. This facilitated the identification of novel major effect QTLs with much better resolution and precision. In-silico analysis of novel QTLs revealed the biological functions of the putative candidate gene (s) associated with selected traits. Most of the high-yielding RILs possessing the major yield related QTLs were identified to be complete restorers, indicating their possible utilization in development of superior rice hybrids.

RESUMO

Antimicrobial resistance (AMR) is one of the significant clinical challenges and also an emerging area of concern arising from nosocomial infections of ESKAPE pathogens, which has been on the rise in both the developed and developing countries alike. These pathogens/superbugs can undergo rapid mutagenesis, which helps them to generate resistance against antimicrobials in addition to the patient's non-adherence to the antibiotic regimen. Sticking to the idea of a 'one-size-fits-all' approach has led to the inappropriate administration of antibiotics resulting in augmentation of antimicrobial resistance. Antimicrobial peptides (AMPs) are the natural host defense peptides that have gained attention in the field of AMR, and recently, synthetic AMPs are well studied to overcome the drawbacks of natural counterparts. This review deals with the novel techniques utilizing the bacteriolytic activity of natural AMPs. The effective localization of these peptides onto the negatively charged bacterial surface by using nanocarriers and structurally nanoengineered antimicrobial peptide polymers (SNAPPs) owing to its smaller size and better antimicrobial activity is also described here.

RESUMO

AIM: The aim of this study was to compare the effectiveness of hand file (K-file), rotary file (HERO shaper), and passive ultrasonic irrigation (PUI; U-file) in removing Ca(OH)2 from the root canal. MATERIALS AND METHODS: Thirty single-rooted teeth were collected and decoronated to standardize the length to 14 mm. Cleaning and shaping were conducted using HERO shaper rotary files (up to no. 25, 4% taper). Ca(OH)2 powder was mixed with normal saline and filled into the canals using lentulo spiral and the orifice was sealed with zinc oxide eugenol. After 7 days of incubation, samples were divided into three groups of 10 samples each: Group I (hand files)-no. 20 K-file; Group II (rotary files)-no. 25, 4% HERO shaper; Group III (PUI)-no. 20 U-file. Ca(OH)2 paste was removed using 2 mL of 3% sodium hypochlorite solution followed by 1-minute activation of the respective file system. All the samples were finally irrigated with 17% ethylenediaminetetraacetic acid and flushed with distilled water. RESULTS: Ultrasonic group was more effective in removing Ca(OH)2 followed by HERO shaper and hand file. CONCLUSION: It was concluded that PUI had the highest ability to remove Ca(OH)2 from the root canal walls.

RESUMO

This paper reports a novel optical ballistocardiography technique, which is non-invasive, for the simultaneous measurement of cardiac and respiratory activities using a Fiber Bragg Grating Heart Beat Device (FBGHBD). The unique design of FBGHBD offers additional capabilities such as monitoring nascent morphology of cardiac and breathing activity, heart rate variability, heart beat rhythm, etc., which can assist in early clinical diagnosis of many conditions associated with heart and lung malfunctioning. The results obtained from the FBGHBD positioned around the pulmonic area on the chest have been evaluated against an electronic stethoscope which detects and records sound pulses originated from the cardiac activity. In order to evaluate the performance of the FBGHBD, quantitative and qualitative studies have been carried out and the results are found to be reliable and accurate, validating its potential as a standalone medical diagnostic device. The developed FBGHBD is simple in design, robust, portable, EMI proof, shock proof and non-electric in its operation which are desired features for any clinical diagnostic tool used in hospital environment.

RESUMO

This study was carried out to improve the RPHR-1005, a stable restorer line of the popular medium slender grain type rice hybrid, DRRH-3 for bacterial blight (BB) and blast resistance through marker-assisted backcross breeding (MABB). Two major BB resistance genes, Xa21 and Xa33 and a major blast resistance gene, Pi2 were transferred to RPHR-1005 as two individual crosses. Foreground selection for Xa21, Xa33, Pi2, Rf3 and Rf4 was done by using gene-specific functional markers, while 59 simple sequence repeat (SSR) markers polymorphic between the donors and recipient parents were used to select the best plant possessing target resistance genes at each backcross generation. Backcrossing was continued till BC2F2 and a promising homozygous backcross derived line possessing Xa21+ Pi2 and another possessing Xa33 were intercrossed to stack the target resistance genes into the genetic background of RPHR-1005. At ICF4, 10 promising lines possessing three resistance genes in homozygous condition along with fine-grain type, complete fertility restoration, better panicle exertion and taller plant type (compared to RPHR-1005) were identified.

RESUMO

AIM: The aim of this in vitro study was to evaluate and compare the microleakage of restorations using low shrinkage silorane-based dental composite and methacrylate-based dental composites in Class II cavity at the occlusal and gingival margins. MATERIALS AND METHODS: Sixty mandibular molars were collected and divided into three experimental groups and one negative control group. Class II slot cavity was prepared on the mesial surface. Experimental groups were restored with Group I: silorane-based microhybrid composite, Group II: methacrylate-based nanohybrid composite, and Group III: Methacrylate-based microhybrid composite, respectively. Group IV: negative control. The samples were thermocycled, root apices were sealed with sticky wax and coated with nail varnish except 1 mm around the restoration. This was followed by immersion in 2% Rhodamine-B dye solution under vacuum at room temperature for 24 h. Then, the samples were sectioned longitudinally in the mesiodistal direction and evaluated under stereomicroscope ×40 magnification. Scoring was done according to the depth of dye penetration in to the cavity. Statistical analysis of the data was done. RESULTS: The results were that no statistically significant difference in the microleakage at the occlusal margin for all the restorative materials, whereas at the gingival margin, silorane-based microhybrid composite showed less microleakage than the methacrylate-based nano- and micro-hybrid composites. CONCLUSION: In general, silorane-based microhybrid composite had less microleakage among the other materials used in this in vitro study.

RESUMO

RPHR-1005, the stable restorer line of the popular medium slender (MS) grain type rice hybrid, DRRH-3 was improved in this study for resistance against bacterial blight (BB) and blast diseases through marker-assisted backcross breeding (MABB). In this study, four major resistance genes (i.e., Xa21 and Xa33 for BB resistance and Pi2 and Pi54 for blast resistance) have been transferred to RPHR-1005 using RPBio Patho-1 (possessing Xa21 + Pi2), RPBio Patho-2 (possessing Xa21 + Pi54) and FBR1-15EM (possessing Xa33) as the donors. Foreground selection was carried out using PCR-based molecular markers specific for the target resistance genes and the major fertility restorer genes, Rf3 and Rf4, while background selection was carried out using a set of parental polymorphic rice SSR markers and backcrossing was continued uptoBC2 generation. At BC2F2, plants possessing the gene combination- Xa21 + Pi2, Xa21 + Pi54 and Xa33 in homozygous condition and with >92% recovery of the recurrent parent genome (RPG) were identified and intercrossed to combine all the four resistance genes. Twenty-two homozygous, pyramid lines of RPHR-1005 comprising of three single-gene containing lines, six 2-gene containing lines, eight 3-gene containing lines, and five 4-gene containing lines were identified among the double intercross lines at F3 generation (DICF3). They were then evaluated for their resistance against BB and blast, fertility restoration ability and for key agro-morphological traits. While single gene containing lines were resistant to either BB or blast, the 2-gene, 3-gene, and 4-gene pyramid lines showed good level of resistance against both and/or either of the two diseases. Most of the 2-gene, 3-gene, and 4-gene containing pyramid lines showed yield levels and other key agro-morphological and grain quality traits comparable to the original recurrent parent and showed complete fertility restoration ability, with a few showing higher yield as compared to RPHR-1005. Further, the experimental hybrids derived by crossing the gene-pyramid lines of RPHR-1005 with APMS6A (the female parent of DRRH-3), showed heterosis levels equivalent to or higher than DRRH-3. The results of present study exemplify the utility of MABB for targeted improvement of multiple traits in hybrid rice.

RESUMO

Surface defect or damage is one of the critical factors leading to the failure of engineering materials and structures. The methodologies for the measurement of surface shape and feature or defect have been extensively explored and developed over the past few decades, including both contact and non-contact methods. Speckle pattern interferometry, as a non-contact optical method, has been demonstrated to effectively contour the surface shape through adjusting the illumination vector. However, few studies have been made to investigate the effect of the initial position of the illumination source as well as the source translation direction. In this paper, we report to carry out a study of measuring the surface form and feature using digital speckle pattern interferometry system via a slight translation of illumination source. Through theoretically analyzing the sensitivity factor along with the experimental validation, it is shown that the contouring fringe is more sensitive to the surface height with an off-axis illumination than the paraxial illumination. It is also found that translating the source along axial and lateral direction can be both used for the surface shape re-construction.

RESUMO

YAlO3: Sm(3+) phosphor has been synthesized by the solid state reaction method with calcium flouride used as a flux. The resulting YAlO3: Sm(3+) phosphor was characterized by X-ray diffraction (XRD) technique, Fourier transmission infrared spectroscopy (FTIR), photoluminescence . . PL excitation spectrum was found at 254,332,380,400,407, 603 and 713 nm. Under excitation of UV(713 nm) YAlO3: Sm(3+) (0-3 %) broad band emission were observed from 400 to 790 nm with a maximum around 713 nm of YAlO3 host lattice accompanied by weak emission of Sm(3+) ((4)G5/2 - (6)H5/2, (6)H7/2,(6)H9/2) transitions. The results of the XRD show that obtained YAlO3: Sm(3+) phosphor has a orthorhombic structure. The study suggested that Sm(3+) doped phosphors are potential luminescence material for laser diode pumping and inorganic scintillators.

RESUMO

This work is focused on sol-gel synthesis of silver and strontium co-doped TiO2 nanoparticles and their utilization as photo-catalysts in degradation of two textile dyes. Effect of pH, intensity of light, amount of photo-catalyst, concentration of dye, sensitizers, etc., were studied to optimize conditions for obtaining enhanced photo-catalytic activity of synthesized nanoparticles. XRD, BET, HR-TEM, EDAX and UV-Vis (diffused reflectance mode) techniques were used to characterize the nanoparticles. Interestingly, band gap of Sr and Ag co-doped TiO2 nanoparticles showed considerable narrowing (2.6 eV) when compared to Ag doped TiO2 (2.7 eV) and undoped TiO2 (3.17 eV) nanoparticles. Incorporation of Ag and Sr in the lattice of TiO2 could bring isolated energy levels near conduction and valence bands thus narrowing band gap. The XRD analysis shows that both Ag and Sr nanoparticles are finely dispersed on the surface of titania framework, without disturbing its crystalline structure. TEM images indicate that representative grain sizes of Ag-doped TiO2 & Sr and Ag co-doped TiO2 nanoparticles are in the range of 8-20 nm and 11-25 nm, respectively. Effective degradation of Direct Green-6 (DG-6) and Reactive Blue-160 (RB-160) under UV and visible light has been achieved using the photo-catalysts. Sr and Ag co-doped TiO2 photo-catalysts showed higher catalytic activity during degradation process in visible region when compared to Ag-doped and undoped TiO2 nanoparticles which could be attributed to the interactive effect caused by band gap narrowing and enhancement in charge separation. For confirming degradation of the dyes, total organic carbon (TOC) content was monitored periodically.

RESUMO

Bacillus halodurans MTCC 865 was employed for decolorization of textile azo dye, Acid Black-24 (AB-24). Thousand mgl⁻¹ of AB-24 was decolorized with 90% efficiency by the strain within 6 hrs at pH 9 and 37 °C with 5% NaCl under static conditions in screening medium. Decolorization was evaluated by measuring the periodic decrease in absorbance at 557 nm (λ(max)). Biodegradation of Acid Black-24 was determined by FTIR and HPLC. The FTIR spectrum of the AB-24 dye suggests the presence of azo bond (-N = N-) peak at 1618.28 cm⁻¹. Absence of the azo bond in the degraded sample spectrum indicates biodegradation of the dye. Formation of metabolites with different retention times in HPLC analysis further confirmed degradation of the azo dye, Acid Black-24 by Bacillus halodurans.

RESUMO

The ability of halophiles to survive in the extreme salt concentrations has gained them the importance of being used in the treatment of industrial waste waters. A moderately halophilic bacterial strain with the ability to degrade the complex azo dye Direct Blue-1 (DB-1) was isolated from sea water and identified as Marinobacter sp. strain HBRA. Complete decolorization of DB-1 (100 mg L(-1)) was achieved in 6h at 37 °C, pH 8 and with 70 g L(-1) NaCl. Decolorization was analyzed by UV-vis spectrophotometer. The FT-IR spectrum revealed that Marinobacter sp. strain HBRA specifically targeted azo bond (NN) at 1631 cm(-1) to break down Direct Blue-1. Formation of metabolites at different retention times in HPLC indicated degradation. Biotransformation pathway for DB-1 was proposed based on LC-MS. Phytotoxicity study revealed the less toxic nature of the metabolites compared to the dye. Genotoxicity with Allium cepa confirmed the cytotoxic nature of DB-1 by inducing several chromosomal abnormalities compared to the negligible effects of degraded metabolites. The current study is the first report on the detoxification of DB-1 by Marinobacter sp. strain HBRA.
