RESUMO

An electrogenic bacterium was isolated from a marine coral, designated as strain JC435T and its taxonomic status examined by using a polyphasic approach. Results from the 16S rRNA gene sequence study showed that the isolate belonged to the genus Rhodococcus and formed a cluster with Rhodococcus ruber KCTC 9806T (99.5 % 16S rRNA gene sequence similarity) and Rhodococcus aetherivorans JCM 14343T (99.3 %), respectively. Genome relatedness based on DNA-DNA hybridization to the type strains of closest-related species was less than 30 % and the ΔTm of >7 °C, suggesting that strain represents a new species of the genus Rhodococcus. The major fatty acids were C16 : 0, C18 : 1ω9c, C18 : 010-methyl and C16 : 1ω6c and/or C16 : 1ω7c. The polar lipids of strain JC435T were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol mannoside, phosphatidylinositol, three unknown phospholipids and an unknown amino lipid. The major isoprenoid quinone was MK-8(H2), with 8 % of MK-7(H2) and 2 % of MK-9(H2) as minor components. Whole-cell hydrolysates contained meso-diaminopimelic acid, arabinose and galactose as the diagnostic diamino acid and sugars. Mycolic acids were detected. The genomic DNA G+C content of strain JC435T was 69.8 mol%. On the basis of phylogenetic genotypic, physiological and chemotaxonomic analysis, strain JC435T is considered to represent a novel species of the genus Rhodococcus for which the name Rhodococcuselectrodiphilus sp. nov. is proposed. The type strain is JC435T (=KCTC 39856T=LMG 29881T=MCC 3659T).

RESUMO

Gram-stain-positive, acid-fast-positive, rapidly growing, rod-shaped bacteria (designated as strains JC290T, JC430 and JC431) were isolated from paddy cultivated soils on the Western Ghats of India. Phylogenetic analysis placed the three strains among the rapidly growing mycobacteria, being most closely related to Mycobacterium tokaiense 47503T (98.8 % 16S rRNA gene sequence similarity), Mycobacterium murale MA112/96T (98.8 %) and a few other Mycobacterium species. The level of DNA-DNA reassociation of the three strains with M. tokaiense DSM 44635T was 23.4±4 % (26.1±3 %, reciprocal analysis) and 21.4±2 % (22.1±4 %, reciprocal analysis). The three novel strains shared >99.9 % 16S rRNA gene sequence similarity and DNA-DNA reassociation values >85 %. Furthermore, phylogenetic analysis based on concatenated sequences (3071 bp) of four housekeeping genes (16S rRNA, hsp65, rpoB and sodA) revealed that strain JC290T is clearly distinct from all other Mycobacteriumspecies. The three strains had diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol, phosphatidylinositolmannosides, unidentified phospholipids, unidentified glycolipids and an unidentified lipid as polar lipids. The predominant isoprenoid quinone for all three strains was MK-9(H2). Fatty acids were C17 : 1ω7c, C16 : 0, C18 : 1ω9c, C16 : 1ω7c/C16 : 1ω6c and C19 : 1ω7c/C19 : 1ω6c for all the three strains. On the basis of phenotypic, chemotaxonomic and phylogenetic data, it was concluded that strains JC290T, JC430 and JC431 are members of a novel species within the genus Mycobacterium and for which the name Mycobacterium oryzae sp. nov. is proposed. The type strain is JC290T (=KCTC 39560T=LMG 28809T).

RESUMO

A reddish-brown-pigmented, phototrophic bacterium, designated strain JA877T, was isolated from a brown algae mat sample collected from Jalandhar beach, Gujarat, India. On the basis of the 16S rRNA gene sequence, strain JA877T belongs to the class Alphaproteobacteria and is closely related to the type strains Rhodovulum viride JA756T (99.0 %), Rhodovulum sulfidophilum Hansen W4T (98.9 %), Rhodovulumvisakhapatnamense JA181T (98.8 %),Rhodovulum kholense JA297T (97.5 %) and Rhodovulum salis JA746T (97.0). However, strain JA877T showed only 20-45 % relatedness with its phylogenetic neighbours and had a ∆Tm between 5.8 and 7.0 °C. The major respiratory quinone was ubiquinone-10 (Q10), and the polar lipid profile was composed of the major components phosphatidylglycerol, phosphatidylethanolamine, an unidentified phospholipid, two unidentified sulfolipids and five unidentified lipids. The major fatty acids were C18 : 1ω5c, C18 : 1ω7c/C18 : 1ω6c, C16 : 0 and C18 : 0. The DNA G+C content was 64.5 mol%. On the basis of 16S rRNA gene sequence analysis, physiological data, and chemotaxonomic and molecular differences, strain JA877T is significantly different from other species of the genus Rhodovulum and represents a novel species, for which the name Rhodovulum algae sp. nov. is proposed. The type strain is JA877T (=LMG 29228T= KCTC 42963T).

RESUMO

A Gram-staining-positive, non-spore-forming actinobacterium, strain JC311T, isolated from marine green alga of the genus Ulva was studied to examine its taxonomic position. On the basis of the 16S rRNA gene sequence similarity studies, strain JC311T was shown represent a member of the genus Ornithinimicrobium and to be closely related to Ornithinimicrobium pekingense LW6T (98.6 %), Ornithinimicrobium kibberense K22-20T (98.3 %) and Ornithinimicrobium humiphilum HKI 0124T (98.1 %). However, strain JC311T showed less than 22 % DNA reassociation value (based on DNA-DNA hybridization) with O. pekingense JCM14001T, O. kibberense JCM12763T and O. humiphilum KCTC19901T. The predominant menaquinone of strain JC311T was MK-8(H4). The peptidoglycan contained l-ornithine as the diagnostic diamino acid. The polar lipid profile consisted of the lipids diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, glycophospholipid, aminophospholipid, phospholipid and two unidentified lipids. The major fatty acids iso-C16 : 0, iso-C15 : 0, iso-C17 : 1ω9c and iso-C17 : 0 were consistent with the fatty acid patterns reported for members of the genus Ornithinimicrobium. The distinct genomic, morphological, physiological and chemotaxonomic differences from the previously described taxa support the classification of JC311T as a representative of a novel species of the genus Ornithinimicrobium, for which we propose the name Ornithinimicrobium algicola sp. nov., with the type strain JC311T ( = KCTC 39559 T = LMG 28808T).

RESUMO

A red-pigmented bacterium producing a metallic green sheen, designated strain JC333T, was isolated from a sand sample collected from Shivrajpur-Kachigad beach, Gujarat, India. Phylogenetic analyses based on the 16S rRNA gene sequence of strain JC333T showed highest sequence similarity to Zooshikella ganghwensis JC2044T (99.24 %) and less than 91.94 % similarity with other members of the class Gammaproteobacteria. DNA-DNA hybridizations between JC333T and Z. ganghwensis JC2044T showed low relatedness values of 19 ± 1.3 % (reciprocal 21 ± 2.2 %). The major respiratory quinone was ubiquinone-9 (Q9) and the polar lipid profile was composed of the major components diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, an unidentified aminophospholipid and an unidentified lipid. The presence of C16 : 1ω7c/C16 : 1ω6c, C16 : 0, C18 : 1ω7c and C12 : 0 as major fatty acids supported the affiliation of strain JC333T to the genus Zooshikella. Prodigiosin, cycloprodigiosin and eight other prodigiosin analogues were the pigments of JC333T. Characterization based on 16S rRNA gene sequence analysis, physiological parameters, pigment analysis, ubiquinone, and polar lipid and fatty acid compositions revealed that JC333T represents a novel species of the genus Zooshikella, for which the name Zooshikella marina sp. nov. is proposed. The type strain is JC333T ( = KCTC 42659T = LMG 28823T).

RESUMO

A Gram-stain-negative, aerobic, pale orange, rod-shaped, non-motile bacterial strain, designated JC289T was isolated from a rhizosphere soil sample of the plant Oryza sativa (L.). 16S rRNA gene sequence analysis clearly allocated strain JC289T to the Flectobacillus cluster, showing highest sequence similarities to Flectobacillus roseus GFA-11T (99.5 %), Flectobacillus major ATCC 29496T (96.7 %) and Flectobacillus lacus CL-GP79T (94.6 %). Major (>5 %) fatty acids of strain JC289T were C16 : 1ω5c, iso-C15 : 0, C15 : 0 2-OH, iso-C15 : 0 3-OH and C16 : 1ω7c/C16 : 1ω6c, with minor amounts (>1- < 5 %) of C14 : 0, C16 : 0, anteiso-C15 : 0, C16 : 0 3-OH, iso-C17 : 0 3-OH, anteiso-C17 : 1B and/or iso-C17 : 1I. Menaquinone-7 was the major quinone of strain JC289T, and the polar lipid profile contained phosphatidylethanolamine, five unidentified aminophospholipids, two unidentified phospholipids, two unidentified aminolipids, two unidentified glycolipids and five unidentified lipids. DNA-DNA relatedness values of strain JC289T with Flectobacillus roseus LMG 24501T and Flectobacillus major LMG 13163T were 24.6 % (31.4 %, reciprocal analysis) and 16.2 % (22.4 %), respectively. On the basis of 16S rRNA gene sequence analysis, physiological and biochemical test results, and chemotaxonomic analysis, strain JC289T can be differentiated from its closest relatives in the genus Flectobacillus. Based on the data presented, it is concluded that strain JC289T represents a novel species of the genus Flectobacillus, for which the name Flectobacillus rhizosphaerae sp. nov. is proposed. The type strain is JC289T ( = KCTC 42575T = LMG 28712T).

RESUMO

Two strains (JC83, JC84(T)) of obligately anaerobic, H2S-producing bacteria were isolated from estuarine sediment samples collected from Gangasagar, West Bengal, India. Cells were Gram-stain-negative, non-motile rods. Both strains were positive for oxidase, negative for catalase, hydrolysed casein, reduced nitrate and utilized citrate. Both strains grew chemoorganoheterotrophically with optimal pH of 7-8 (range 7-10) and at 30 °C (range 25-37 °C). C16 : 1ω7c, C18 : 1ω7c, C16 : 0 and C12 : 0 were the major fatty acids of both strains with minor amounts of C14 : 0, C12 : 0 3-OH and C18 : 0. Polar lipids of both strains included diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylserine, phosphatidylcholine, phosphatidylinositol, an unidentified aminolipid (AL2), an unidentified phospholipid (PL2) and an unidentified lipid (L3). MK-6 was the major respiratory quinone. The DNA G+C content of strains JC83 and JC84(T) was 25.0 and 24.6 mol%, respectively. The strains showed DNA reassociation >85 % (86.0±0.5 %) (based on DNA-DNA hybridization). Based on 16S rRNA gene sequence analysis, both strains were identified as belonging to the family Campylobacteraceae of the class Epsilonproteobacteria with Arcobacter marinus CL-S1(T) (95.4 % sequence similarity) as their closest phylogenetic neighbour. On the basis of morphological, physiological and chemotaxonomic characteristics as well as phylogenetic analysis, strains JC83 and JC84(T) are considered to represent a novel species, for which the name Arcobacter anaerophilus sp. nov. is proposed. The type strain is JC84(T) ( = KCTC 15071(T) = MTCC 10956(T) = DSM 24636(T)). An emended description of the genus Arcobacter is provided.

RESUMO

The carotenoid, neurosporene, is of commercial importance due to its use as an antioxidant and UV-B radiation protector. It is usually obtained from fruits and vegetables. Although some bacterial mutants can accumulate neurosporene, these are unstable. A recently-isolated, natural and stable neurosporene accumulating, phototrophic purple non-sulfur bacterium Rhodobacter viridis JA737 is described that accumulates neurosporene as the major (98%) carotenoid up to ~7 mg g dry cell(-1) and at 7.3 mg l(-1).

RESUMO

A reddish-brown bacterium, designated strain JA318(T), was purified from a photoheterotrophic enrichment culture obtained from the rhizosphere soil of paddy. Cells of strain JA318(T) are spiral shaped, Gram-stain-negative and motile by means of amphitrichous flagella. Strain JA318(T) has no NaCl requirement for growth but can tolerate up to 1.5 % (w/v) NaCl. Internal photosynthetic membranes are present as lamellar stacks. Photoorganoheterotrophy is the only growth mode observed. Strain JA318(T) contains bacteriochlorophyll a, lycopene and rhodopin as major carotenoids. Thiamine, niacin and para-aminobenzoic acid (PABA) are required as growth factors. Major fatty acids are C18 : 1ω7c and C16 : 0. Ubiquinone-8 and rhodoquinone-8 are the observed quinones. Diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and an unidentified aminolipid are the major polar lipids in strain JA318(T). Phylogenetic analysis based on 16S rRNA gene sequences showed that the strain JA318(T) clustered with species of the genus Rhodospirillum which belongs to the class Alphaproteobacteria. The highest sequence similarity of strain JA318(T) was found with Rhodospirillum sulfurexigens JA143(T) (99.9 %). The DNA-DNA reassociation values of strain JA318(T) with Rsp. sulfurexigens JA143(T) and Rhodospirillum photometricum DSM 122(T) were 52 ± 2 % and 45 ± 1 %, respectively. The genomic DNA G+C content of strain JA318(T) was 60.2 mol%. Based on the morphological, physiological, chemotaxonomical and molecular evidence, strain JA318(T) is significantly different from the type strains of species of the genus Rhodospirillum, of the family Rhodospirillaceae, and it is proposed that the strain be classified as a representative of a novel species for which the name Rhodospirillum oryzae sp. nov. is proposed. The type strain is JA318(T) (= KCTC 5960(T) = NBRC 107573(T)).

RESUMO

A rod-shaped, non-motile, Gram-stain-positive, catalase-positive, starch-hydrolysing strain, JC16(T), was isolated from a semi-arid tropical soil from India. Phylogenetic analysis based on 16S rRNA gene sequence showed that strain JC16(T) clustered with the type species of the genus Chryseomicrobium, Chryseomicrobium imtechense MW 10(T), a member of the family Planococcaceae within the phylum Firmicutes with 99.3 % sequence similarity. Major (>10 %) fatty acids of strain JC16(T) were iso-C15 : 0 and iso-C16 : 0. Minor (<10 and >1 %) amounts of C16 : 0, iso-C14 : 0, anteiso-C15 : 0, iso-C17 : 0, anteiso-C17 : 0, iso-C17 : 1ω10c and C16 : 1ω11c are present in strain JC16(T). Polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phospholipids (PL2-4), aminolipids (AL1, 2) and an unknown lipid. Cell wall peptidoglycan was of the type l-Orn-D-Glu. The quinone system was composed of MK-7, MK-8 and MK-6. Genomic DNA G+C content of strain JC16(T) was 57.6 mol%. Distinct physiological, chemotaxonomic and genotypic differences (37 % reassociation based on DNA-DNA hybridization) from Chryseomicrobium imtechense MW 10(T) support the classification of strain JC16(T) as a representative of a novel species in the genus Chryseomicobium, for which the name Chryseomicrobium amylolyticum sp. nov. (type strain JC16(T) = DSM 23442(T) = NBRC 105215(T)) is proposed.

RESUMO

Five strains (JA325, JA389, JA473, JA563 and JA582) of Gram stain-negative, vibrioid to spiral shaped, phototrophic purple bacteria were isolated from solar salterns of India. All strains contained bacteriochlorophyll-a and carotenoids of the spirilloxanthin series as photosynthetic pigments. C(18:1)ω7c, C(18:1)ω7c 11-methyl and C(16:0) were the major fatty acids of all strains. Diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), ornithine lipid (OL), an unidentified phospholipid (PL), and an unidentified aminolipid (AL) were the major polar lipids of all the strains. According to 16S rRNA gene sequences, all strains clustered phylogenetically with the only species of the genus Rhodothalassium (99.8-99.3% sequence similarity) but only strains JA325 and JA563 were distinctly related (60+1.5% DNA-DNA hybridization [DDH]) to the type strain Rhodothalassium salexigens DSM 2132(T). However, the genotypic data of strains JA325 and JA563 was not supported because of a large number of phenotypic differences compared to the type strain, therefore, it is proposed that all five newly isolated strains were R. salexigens-like strains. In addition, phylogenetically, the Rhodothalassium clade represented a distinct lineage and formed a deep branch with less than 90% 16S rRNA gene sequence similarity to other orders of the Alphaproteobacteria, and characteristic phenotypic properties also distinguished these bacteria from other purple non-sulfur bacteria. Therefore, the novel family Rhodothalassiaceae fam. nov. and the novel order Rhodothalassiales ord. nov. are proposed for the distinct phyletic line represented by the genus Rhodothalassium.

Strain JA737(T) contained bacteriochlorophyll a, and the major carotenoid was neurosporene. The major quinone was Q-10 and the polar lipids were phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, an unidentified aminolipid, two unidentified phospholipids and five unidentified lipids. Phylogenetic analysis showed that the strain clustered with members of the genus Rhodobacter belonging to the family Rhodobacteraceae of the class Alphaproteobacteria. Based on 16S rRNA gene sequence analysis, strain JA737(T) had highest sequence similarity with Rhodobacter capsulatus ATCC 11166(T) (98.8 %), Rhodobacter maris JA276(T) (97 %), Rhodobacter aestuarii JA296(T) (96.7 %) and other members of the genus Rhodobacter (<96 %). However, strain JA737(T) showed 22-55 % DNA-DNA relatedness with the above type strains. On the basis of phenotypic, chemotaxonomic and molecular genetic evidence, strain JA737(T) represents a novel species of the genus Rhodobacter, for which the name Rhodobacter viridis sp. nov. is proposed. The type strain is JA737(T) ( = KCTC 15167(T) = MTCC 11105(T) = NBRC 108864(T)).

RESUMO

Two strains (JA266(T) and JA333) of Gram-negative, rod-shaped, phototrophic, purple non-sulfur bacteria were isolated from a freshwater fish pond and an industrial effluent. Both strains were capable of phototrophic and chemotrophic growth. Bacteriochlorophyll a and carotenoids of the spirilloxanthin series were present as photosynthetic pigments. The major fatty acid for both strains was C(18 : 1)ω7c (>65 %), with minor amounts of 11-methyl C(18  : 1)ω7c, C(16 : 0), C(16 : 1)ω7c and C(18 : 0) also present. Both strains have the lamellar type of intracellular photosynthetic membranes. Ubiquinone-10 (Q(10)) and rhodoquinone-10 (RQ(10)) were present as primary quinone components. Diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and phosphatidylcholine were the major polar lipids, while minor amounts of amino lipids (AL1, AL2) and an unidentified lipid (L1) were common to both strains. The DNA G+C contents of strains JA266(T) and JA333 were 71.3 and 69.9 mol%, respectively. Phylogenetic analysis on the basis of 16S rRNA gene sequences showed that both strains clustered with members of the genus Rhodoplanes in the class Alphaproteobacteria. Strains JA266(T) and JA333 had gene sequence similarity of 98.7 and 98.9 % with Rhodoplanes serenus TUT3530(T), 96.4 and 96.5 % with Rhodoplanes elegans AS130(T), respectively, and less than 96 % with other members of the genus Rhodoplanes. 16S rRNA gene sequence similarity between the two strains was 99.3 % and they exhibited high (84.7 %) relatedness based on DNA-DNA hybridization. Furthermore, both strains had less than 65 % DNA-DNA relatedness with the type strain R. serenus TUT3530(T). On the basis of phenotypic and genotypic data, it is proposed that strain JA266(T) be classified as a novel species of the genus Rhodoplanes, with the species name Rhodoplanes piscinae sp. nov. The type strain of the proposed novel species is JA266(T) ( = JCM 14934(T) = KCTC 5627(T)), while strain JA333 ( = NBRC 107574 = KCTC 5962) is an additional strain.

RESUMO

Four strains (JA310(T), JA531(T), JA447 and JA490) of red to reddish brown pigmented, rod-shaped, motile and budding phototrophic bacteria were isolated from soil and freshwater sediment samples from different geographical regions of India. All strains contained bacteriochlorophyll a and carotenoids of the spirilloxanthin series. The major cellular fatty acid of strains JA310(T) and JA531(T) was C(18:1)ω7c, the quinone was Q-10 and polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, an aminohopanoid and an unidentified aminolipid. Phylogenetic analysis based on 16S rRNA gene sequences showed that all strains clustered with species of the genus Rhodopseudomonas in the class Alphaproteobacteria. Strains JA531(T), JA447 and JA490 were genotypically (>80% related based on DNA-DNA hybridization) and phenotypically closely related to each other and the three strains were distinct from strain JA310(T) (33% related). Furthermore, all four strains had less than 48% relatedness (DNA-DNA hybridization) with type strains of members of the genus Rhodopseudomonas, i.e. Rhodopseudomonas palustris ATCC 17001(T), Rhodopseudomonas faecalis JCM 11668(T) and Rhodopseudomonas rhenobacensis DSM 12706(T). The genomic DNA G+C contents of strains JA310(T) and JA531(T) were 63.8 and 62.4 mol%, respectively. On the basis of phenotypic, chemotaxonomic and molecular genetic evidence, it is proposed that strains JA310(T) ( = NBRC 106083(T) = KCTC 5839(T)) and JA531(T) ( = NBRC 107575(T) = KCTC 5841(T)) be classified as the type strains of two novel species of the genus Rhodopseudomonas, Rhodopseudomonas parapalustris sp. nov. and Rhodopseudomonas harwoodiae sp. nov., respectively. In addition, we propose that strain DSM 123(T) ( = NBRC 100419(T)) represents a novel species, Rhodopseudomonas pseudopalustris sp. nov., since this strain differs genotypically and phenotypically from R. palustris ATCC 17001(T) and other members of the genus Rhodopseudomonas. An emended description of R. palustris is also provided.

RESUMO

A Gram-stain-positive, oxidase-negative, starch-hydrolysing, actinobacterium (strain JC82(T)) was isolated from a soda lake in Lonar, India. Based on 16S rRNA gene sequence similarity studies, strain JC82(T) belonged to the genus Georgenia and was most closely related to Georgenia muralis 1A-C(T) (96.8 %) and other members of the genus Georgenia (<96.5 %). The DNA G+C content of strain JC82(T) was 73.4 mol%. The cell-wall amino acids were alanine, glutamic acid and lysine with peptidoglycan type A4α. Polar lipids included diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannosides, an unidentified lipid (L1) and an unidentified glycolipid (GL3). The predominant isoprenoid quinone was menaquinone MK-8(H(4)). Anteiso-C(15 : 0) was the predominant fatty acid and significant proportions of iso-C(14 : 0), C(14 : 0), C(16 : 0), iso-C(15 : 0) and iso-C(16 : 0) were also detected. Strain JC82(T) produced thermostable alkaline amylase. The results of physiological and biochemical tests allowed a clear phenotypic differentiation of strain JC82(T) from all other members of the genus Georgenia. Based on these data, strain JC82(T) represents a novel species of the genus Georgenia, for which the name Georgenia satyanarayanai sp. nov. is proposed. The type strain is JC82(T) ( = KCTC 19802(T) = NBRC 107612(T)).

RESUMO

An oval to rod-shaped, Gram-stain-negative, phototrophic bacterium, strain JA738(T), was isolated from a sediment sample collected from a pink pond. Strain JA738(T) was non-motile and had vesicular-type intracellular photosynthetic membranes. Bacteriochlorophyll a and carotenoids of the spheroidene series were present as the major photosynthetic pigments. Strain JA738(T) required thiamine and pantothenate for growth. The major cellular fatty acids were C(18 : 1)ω7c, C(18 : 1)ω5c, C(18 : 0) and C(18 : 1)ω7c11-methyl; minor amounts of C(10 : 0) 3-OH and C(16 : 0) were also present. The major quinone was Q-10 and major polar lipids were phosphatidylglycerol, phosphatidylethanolamine and two unidentified sulfolipids (SL1-2). Phylogenetic analysis on the basis of 16S rRNA gene sequences showed that strain JA738(T) clustered with species of the genus Rhodovulum in the class Alphaproteobacteria. Strain JA738(T) was most closely related to Rhodovulum adriaticum DSM 2781(T) (96.4 % 16S rRNA gene sequence similarity) and other members of the genus Rhodovulum (<96.1 %). On the basis of phenotypic and molecular genetic evidence, it is proposed that strain JA738(T) should be classified as a novel species of the genus Rhodovulum for which the name Rhodovulum bhavnagarense sp. nov. is proposed. The type strain is JA738(T) ( = DSM 24766(T) = KCTC 15110(T)).

RESUMO

Two strains (JA492(T) and JA590) of spiral-shaped, anaerobic, Gram-stain-negative, motile, purple non-sulfur bacteria were isolated from aquatic sediments from a bird sanctuary and a stream, respectively, and were characterized by a polyphasic taxonomic approach. Bacteriochlorophyll a and carotenoids (rhodopin, lycopene, hydroxylycopene glucoside and dihydroxylycopene diglucoside) were present as photosynthetic pigments. Intracellular photosynthetic membranes were of the stacked type. The major fatty acids were C(18 : 1)ω7c, C(16 : 0) and summed feature 3 (C(16 : 1)ω6c and/or C(16 : 1)ω7c) in both strains. Ubiquinones and menaquinones were present as major quinone components. The genomic DNA G+C contents of strains JA492(T) and JA590 were 63.8 and 61.5 mol%, respectively. Both strains were closely related (mean DNA-DNA hybridization >70 %). Phylogenetic analysis showed that the strains clustered with species of the genus Phaeospirillum of the family Rhodospirillaceae, class Alphaproteobacteria. Based on 16S rRNA gene sequence analysis, both strains showed highest sequence similarity with Phaeospirillum oryzae JA317(T) (97.2-97.4 %), Phaeospirillum molischianum DSM 120(T) (96.5-96.7 %), Phaeospirillum fulvum DSM 113(T) (96.7-96.9 %) and Phaeospirillum chandramohanii JA145(T) (96.5-96.7 %). DNA-DNA relatedness between strain JA492(T) and its closest relative in the genus Phaeospirillum was less than 42 %. It is evident from phenotypic, chemotaxonomic and molecular genetic data that strain JA492(T) represents a novel species of the genus Phaeospirillum, for which the name Phaeospirillum tilakii sp. nov. is proposed; the type strain is JA492(T) ( = NBRC 107650(T) = KCTC 15012(T)).

RESUMO

Three strains (JA349(T), JA553(T), JA439) of phototrophic sulphur bacteria were isolated from marine habitats of India. 16S rRNA gene sequence of the three strains clustered phylogenetically with members of the genus Marichromatium of the family Chromatiaceae belonging to the class Gammaproteobacteria. All the strains shared highest sequence similarity with the type strains of Marichromatium spp. (96-99% sequence similarity) and the new strains were characterized based on polyphasic taxonomy. Strains JA349(T) and JA553(T) can be distinguished from closest relative species of the genus Marichromatium with respect to distinct differences in cellular polar lipids, fatty acids and carbon/nitrogen sources utilization. Both strains were distinctly related (<50% based on DNA-DNA hybridization) with the type strains of the genus Marichromatium. Multilocus Sequence Analysis (MLSA) of the concatenated five protein coding genes (fusA, pufM, dnaK, recA, soxB) along with internal transcribed spacer (ITS; 16S-23S rRNA) had sequence similarity of less than 92% with the type strains of Marichromatium spp. Distinct phenotypic, chemotaxonomic and molecular differences allow the separation of strains JA349(T) and JA553(T) into new species of the genus Marichromatium for which, we propose the names Marichromatium litoris sp. nov. and Marichromatium chrysaorae sp. nov., respectively.
