RESUMO

In the field of coordination and bioorganometallic chemistry, a notable shift is occurring. This mini-review explores a new generation of carefully 3D-crafted coordination and organometallic complexes that differ from conventional structures. Emphasizing disease intervention and microbial control, these compounds, incorporate noble and transition metals, and aim to enhance therapeutic efficacy while minimizing potential health risks. The mini-review covers diverse applications, showcasing their effectiveness against bacteria, viruses, fungi, and as potential tools in cancer treatment. Additionally, it sheds light on the inventive aspects of these complexes within biological systems. By highlighting advancements in bioorganometallic chemistry, the review offers insights and guidance for future developments in safer and more effective therapeutics.

Novel building materials were manufactured and analyzed for 226Ra, 232Th and 40K using an HPGe gamma-ray spectrometer. The results show that the highest value of 40K was 4530 Bq per kg which was measured in a sample containing fly ashes from olive stones. The highest values of 226Ra and 232Th activities were 181 and 185 Bq per kg, which were measured in a sample with fly ashes from the co-combustion of coal and coke, respectively. On the other hand, the lowest values of 40K, 226Ra and 232Th activities were obtained for samples incorporating mussel shells. The radiological health hazard parameters, such as radium equivalent activity (Raeq), activity concentration index (I), absorbed and effective dose rates, associated with these radionuclides were evaluated. These values are within the EU recommended limits in building materials, except for samples of concrete containing fly ashes from olive stones, coal and coke. This study has contributed to the inclusion of industrial wastes that have not been collected previously in the Naturally Occurring Radioactive Material (NORM) databases on radioactivity of building materials.

The inappropriate use of antibiotics and the inadequate control of infections have led to the emergence of drug-resistant strains. In recent years, metallo-pharmaceutics and metallic nanoparticles have been proposed as potential alternative antimicrobials due to their broad-spectrum antimicrobial properties. Moreover, recent findings have shown that combinations of transition metal compounds can exhibit synergistic antimicrobial properties. Therefore, the synthesis and design of bimetallic nanoparticles is a field worth exploring to harness the interactions between groups of metals and organic complex structures found in different microbial targets, towards the development of more efficient combinatorial antimicrobials composed of synergistic metals. In this study, we present a green synthesis of Ag-Fe bimetallic nanoparticles using an aqueous extract from the leaves of Gardenia jasminoides. The characterization of the nanoparticles demonstrated that the synthesis methodology produces homogenously distributed core-shell Ag-Fe structures with spherical shapes and average diameter sizes of 13 nm (± 6.3 nm). The Ag-Fe bimetallic nanoparticles showed magnetic and antimicrobial properties; the latter were evaluated against six different, clinically relevant multi-drug-resistant microbial strains. The Ag-Fe bimetallic nanoparticles exhibited an antimicrobial (bactericidal) synergistic effect between the two metals composing the bimetallic nanoparticles compared to the effects of the mono-metallic nanoparticles against yeast and both Gram-positive and Gram-negative multidrug-resistant bacteria. Our results provide insight towards the design of bimetallic nanoparticles, synthesized through green chemistry methodologies, to develop synergistic combinatorial antimicrobials with possible applications in both industrial processes and the treatment of infections caused by clinically relevant drug-resistant strains.

Notwithstanding evidence that tuberculosis (TB) is declining, one of the greatest concerns to public health is the emergence and spread of multi-drug resistant strains of Mycobacterium tuberculosis (MDR-TB). MDR-TB are defined as strains which are resistant to at least isoniazid (INH) and rifampicin, the two most potent TB drugs, and their increasing incidence is a serious concern. Recently, notable efforts have been spent on research to pursue novel treatments against MDR-TB, especially on synergistic drug combinations as they have the potential to improve TB treatment. Our research group has previously reported promising synergistic antimicrobial effects between transition-metal compounds and antibiotics in Gram-negative and Gram-positive bacteria. In this work, we evaluated antimycobacterial activity of transition-metals/antibiotics combinatorial treatments against first-line drug resistant strains of Mycobacterium tuberculosis. Our data showed that INH/AgNO3 combinatorial treatment had an additive effect (bactericidal activity) in an isoniazid-resistant clinical strain of Mycobacterium tuberculosis. Moreover, in vitro evaluation of cytotoxicity induced by both, the individual tratments of AgNO3 and INH and the combinatorial treatment of INH/AgNO3 in murine RAW 264.7 macrophages and human A549 lung cells; showed no toxic effects. Together, this data suggests that the INH/AgNO3 combinatorial treatment could be used in the development of new strategies to treat resistant strains of Mycobacterium tuberculosis.

Consumption of pigmented corn has been associated with health benefits due to its flavonoid contents (mainly anthocyanins) and antioxidant ability. Therefore, the objective of this study was to determine the effect of blue corn tortilla (BM) consumption on memory and learning ability adult rats. Eighteen adult female rats were divided into three groups and fed for 38 consecutive days with commercial food (Control group) or the commercial food plus 6 g/day of either blue corn (BM) or white corn (WM) tortillas. Memory and learning capabilities were assessed using Barnes's labyrinth at the end of the feeding period. Short-and long-term memory was improved in the BM group, showing that consumption of blue maize tortillas improves learning and memory capabilities in adult rats.

BACKGROUND: In their origin, abdominal aortic aneurysms (AAAs) are related to an inflammatory reaction within the aortic wall, which can lead to weakness and degeneration of this structure. One of the most widely accepted treatment modalities for AAAs is the placement of stent grafts. Nevertheless, in some patients blood re-enters the aneurysm sac, creating so-called leaks, which constitute a renewed risk of rupture and death.This study explores the possibility of filling aneurysm sacs treated by endovascular aneurysm repair with adipose tissue-derived mesenchymal stem cells (ASCs) in a porcine model. METHODS: We developed a porcine model using 22 animals by creating an artificial AAA made with a Dacron patch. AAAs were then treated with a coated stent that isolated the aneurysm sac, after which we introduced allogeneic ASC into the sac. Animals were followed-up for up to 3 mo. The experiment consisted of the aforementioned surgical procedure performed first, followed by computed tomography and echo-Doppler imaging during the follow-up, and finally, after sacrificing the animals, histologic analysis of tissue samples from the site of cell implantation by a blinded observer and the detection of implanted cells by immunofluorescence detection of the Y chromosome. RESULTS: Our findings demonstrate the survival of ASCs over the 3 mo after implantation and histologic changes associated with this treatment. Treated animals had less acute and chronic inflammation throughout the study period, and we observed increasing fibrosis of the aneurysm sac, no accumulation of calcium, and a regeneration of elastic fibers in the artery. CONCLUSIONS: The combination of endovascular aneurysm repair and cell therapy on AAAs has promising results for the stabilization of the sac, resulting in the generation of living tissue that can secure the stent graft and even showing some signs of wall regeneration. The therapeutic value of such cell-based therapy will require further investigation.

A series of mono- and bistriazine derivatives were selectively prepared in high yields using microwave irradiation. Donor substituents were attached on the triazine ring, including pyrazolyl-substituted anilines and o-, m-, and p-phenylenediamine as π-conjugated spacers. This method was used to build σ-π-σ-A-σ-D systems for monotriazines and D-σ-A-σ-π-σ-A-σ-D systems for bistriazines. A study of the optoelectronic properties was performed by UV-vis and fluorescence spectroscopy and cyclic voltammetry. The monotriazines do not show any emission, but the bistriazines are blue emitters and show an interesting solvatochromic effect with large Stokes shifts of more than 10,000 cm(-1) in some cases and quantum yields up to 23%. The optoelectronic properties depend on the conjugation and the position and donor character of the substituents and spacers. Cyclic voltammetry was used to determine the energy levels (HOMO and LUMO) in the bistriazines. An increase in the energy of the HOMO and a decrease in the energy of the LUMO were observed upon extending the conjugation. The title compounds showed interesting properties for use in optoelectronic devices, especially as blue emitters.

The purpose of this study was to investigate the expression of heat shock protein (HSP70) in oral non-dysplastic leucoplakia and in relation to the clinical and pathological features of oral lichen planus. The expression of HSP70 was assessed in the epithelial compartment of normal mucosa (n = 5), oral lichen planus (n = 28) and benign leucoplakia (n = 11) using an immunohistochemical method. The immunostaining intensity distribution (IID) index was used to quantify the positivity of the staining. There was no association between HSP70 overexpression and clinical presentation of oral lichen planus. Oral lichen planus patients showed no statistically significant differences in the depth of the inflammatory infiltrate when expression of HSP70 was considered (X(i)- X(j) = 42.30; 95% confidence interval (95% CI) = -120.87-205.48). No statistically significant differences were identified in terms of HSP70 expression between oral lichen planus and normal buccal mucosal specimens (X(i)- X(j) = 4.07; 95% CI = -0.53-8.67). The IID index score for HSP70 expression in leucoplakia specimens was significantly higher than the one of the oral lichen planus group (X(i) - X(j)= 5.11; 95% CI = 1.73-8.48). It is concluded that there are no statistically significant differences in HSP70 expression between oral lichen planus and normal buccal mucosal specimens, suggesting that HSP70 does not play an obvious part in the pathogenesis of oral lichen planus. The expression of HSP70 was significantly higher in oral leucoplakia than in oral lichen planus, possibly because of differences in cellular activity or cell proliferation.

OBJECTIVES: To investigate the artefacts in oral biopsies taken by general dental practitioners (GDP) and oral and maxillofacial surgeons (O&MF) and to assess the concordance of clinical and pathological diagnoses. DESIGN: Cross-sectional investigation. SETTING: Primary care and hospital practice. MATERIAL AND METHODS: A total of 354 randomly selected oral biopsy samples studied by the same pathologist. MAIN OUTCOME MEASURES: Clinical and pathological diagnoses - prevalence and location of surgical handling artefacts: crush, splits, haemorrhage and fragmentation. RESULTS: Artefacts identified in GDPs biopsies: crush 27.1%; haemorrhage 19.8%; splits 11.3%; and fragmentation 6.2%. O&MSs biopsies showed: crush 10.2%; splits 13%; fragmentation 2.3%; and haemorrhage 8.5%. No differences in split or fragmentation were identified between GDPs and O&MSs. Crush (chi2 = 16.76; P = 0.000) and haemorrhage (chi2 = 9.31; P = 0.003) were more frequently identified in GDPs biopsies. Concordance between clinical and pathological diagnoses was excellent ( > 0.75) for both groups in all disorders considered. CONCLUSIONS: The elicited clinical knowledge, the quality of the samples sent to the pathologists seem to suggest the advisability for the GDPs to perform diagnostic biopsies.

Objetivo: Conocer la prevalencia de la beta-talasemia en nuestra área sanitaria, la cual no se había estudiado previamente y detectar los portadores para control y consejo genético. Material y métodos: Estudiamos la prevalencia de esta patología en nuestra área sanitaria, de la cual no existían datos previos. Realizamos un estudio prospectivo de 10237 hemogramas de pacientes analizados en nuestro laboratorio (3.95 por ciento de la población total de nuestro área) siendo un 35 por ciento hombres y un 65 por ciento mujeres. La selección de las muestras de los pacientes se realizó a partir del volumen corpuscular medio de los hematies (VCM) <=70 fL; a todos ellos se les determinó la ferritina, hemoglobina A2 y morfología eritrocitaria óptica. Conclusiones: Detectamos un 1.39 por ciento de la microcitosis, de las cuales el 0.38 por ciento fueron diagnosticadas de beta-talasémicas y un 0.88 por ciento de ferropenias; el 0.13 por ciento restante eran secundarias a otras patologías (AU)

The aim of this study is to compare the artefacts ascribed to the technique of incisional biopsy using a punch or scalpel and the influence of suture use for traction and delivery of the specimen. A total of 160 samples were obtained from 10 fresh pig tongues by four experienced oral surgeons. Handling artefacts (squeeze artefacts): crush, splits, fragmentation and pseudocysts were assessed. No differences were identified in terms of crush, fragmentation or pseudocysts between samples obtained with a punch or scalpel. Splits were more frequent in those biopsies taken with a scalpel (chi2 = 9.26; P= 0.0023). Artefacts in the punch biopsy group were significantly less than in the group that combined punch and suture traction (P < 0.01). The scalpel and suture traction group showed significantly more artefacts than the group without suture. It is concluded the use of a stitch for traction in small incisional biopsies causes squeeze artefacts, so its use should be restricted to specimen orientation.

In order to test recombinant Toxoplasma as adjuvant and live vaccine carrier in the infectious disease model of murine experimental leishmaniasis, we engineered the attenuated, temperature-sensitive Toxoplasma gondii strain ts-4 to express the heterologous Leishmania antigen kinetoplastid membrane protein-11 (KMP-11). Transgenic ts-4 clones were obtained which express KMP-11 as cytoplasmatic protein or target it to the secretory pathway of the tachyzoites. Immunization of BALB/c mice with these stably transformed parasites elicited proliferative responses to both T. gondii antigen and recombinant KMP-11. When challenged with Leishmania major, we observed significant protection in animals that had been vaccinated with the KMP-11-expressing ts-4 mutants. The adjuvant attenuated only the onset of the Leishmania infection, but animals were ultimately not able to control the disease. Thus, our findings demonstrate that recombinant Toxoplasma has the potential to serve as an efficient vaccine carrier for cutaneous leishmaniasis. Furthermore, they establish a protective role for the antigen KMP-11 when given in such a vaccine formulation.

The involvement of the vascular endothelium in a large number of diseases supports the importance of vascular-specific gene delivery for their treatment. The hereditary hemorrhagic telangiectasia type 1 is an example of a vascular inherited disease (OMIM 187300). This is an autosomal dominant vascular disorder originated by mutations in the endoglin gene and associated with frequent epistaxis, telangiectases, gastrointestinal bleedings, and arteriovenous malformations in brain, lung and liver. Here, we address for the first time the possibility of using in vivo gene transfer to target endoglin expression to the vasculature. The promoter of the endothelial gene, ICAM-2, was used to generate transgenic animals which demonstrated endothelial expression of endoglin. Next, the promoters of the human endothelial genes, endoglin and ICAM-2, were inserted upstream of the human endoglin cDNA, and the resulting constructs were systemically or locally delivered, demonstrating endoglin expression in the vessel walls of liver, lung and skin. These gene transfer experiments represent an initial step in the treatment of the hereditary hemorrhagic telangiectasia type 1 by gene therapy, and suggest that endoglin and ICAM-2 promoters can be used to deliver other genes to the endothelium specifically.

Planteamiento: se ha estudiado la inducción de las proteínas de choque térmico (hsp) 70 y 27 en 35 pacientes con cáncer de mama comparando con tumores malignos.Material y métodos: Se han hecho estudios con inmunohistoquímica sobre cortes de parafina de los 35 pacientes con cáncer de mama empleando anticuerpos frente a las proteínas hsp70 y hsp27. Al mismo tiempo se han analizado los tumores, pero no sucede lo mismo con hsp27. La expresión de hsp70 es alta en todos los casos, con más del 60 por ciento de células teñidas por campo en cada tumor. La expresión de hsp27 es menor en los casos dende hay reacción positiva. La expresión de hsp70 parece estar relacionada con los procesos de proliferación en tejido mamario, mientras que en tumores malignos hay una localización nuclear de hsp70.Conclusiones: Se puede decir que los resultados del trabajo apoyan el empleo de hsp70 como marcador de malignidad en el cáncer de mama, dada su expresión aumnetada y translocación nuclear relacionada con el cáncer (AU)

Parasitologic confirmation of cutaneous leishmaniasis is obligatory before chemotherapy can be considered. Direct microscopic examination of scrapings taken from indurated borders of ulcers has been routinely used as primary method of diagnosis. In this report we compared the sensitivity of examination of dermal scrapings taken from the bottoms of ulcers (BDS) with that of dermal scrapings taken from indurated active margins of lesions (MDS) in a total of 115 patients. The sensitivities of the microscopic examination were 90.4 and 78.3% for BDS and MDS samples, respectively. When the PCR method was used with a group of 40 patients, we also observed a higher sensitivity when BDS samples were examined (80.8% in BDS samples versus 57.7% in MDS samples). The improvement of the diagnostic sensitivity in the BDS samples appears to be related to the higher parasite load and more easily detectable morphology of amastigotes in the centers of the ulcers. Other parasitologic diagnostic methods, such as culture and histopathologic examination of biopsies, are less sensitive (67.5 and 64.3%, respectively). Aspirate culture, however, was shown to be the most sensitive method for the diagnosis of patients with chronic ulcers. When microscopic examinations of both MDS and BDS samples are combined, the sensitivity of diagnosis may rise up to 94%. We therefore recommend this method as a primary routine procedure for diagnosis of cutaneous leishmaniasis.

The Leishmania infantum Mat-1 gene--recently described in L. major as a highly stage-specific, metacyclogenesis-associated transcript--has been cloned. The 420-bp Mat-1 coding region is conserved with respect to the L. major gene (82% sequence homology). Analysis of the predicted amino-acid sequence reveals structural motifs showing homology with the class of leucine-zipper transcription factors. Southern-blot hybridization analysis suggests that Mat-1 is a low-copy-number gene, probably consisting of two gene copies. The recombinant Mat-1 protein expressed in fusion with the Escherichia coli maltose-binding protein shows a tendency to form dimers in the presence of the leucine-rich C-terminal domain. Bacteria expressing the Mat-1 open reading frame are highly growth-attenuated and tend to delete or modify the insert, which suggests that expression of Mat-1 is toxic for the bacteria.

To investigate 1) the correlation between the proliferative activity of epididymal epithelium plus myoid cells and the increase in the number of these cells and 2) the role of the basal epithelial cells in the renewal of epididymal epithelium, a quantitative evaluation of the proliferation of both epithelial cells and periductal myoid cells in the different epididymal regions (caput, corpus, and cauda) has been carried out during postnatal development of the rat by immunohistochemical evaluation of BrdU-labeling indices. These data were correlated with cell numbers and counted by the optical dissector method. The presence of bcl-2 protein was immunohistochemically detected and evaluated. No significant differences in BrdU indices were observed among epididymal regions in any stage studied. Cell proliferation decreased from the prepubertal period to adulthood in both epithelial and myoid cells in the three regions of the epididymis, suggesting a close relationship between epithelial and mesenchymal components. The numbers of both cell types were significantly higher in the caput than in the corpus and cauda in all stages studied, suggesting functional differences between regions. A negative linear correlation between proliferative activity and cell numbers was noted that might be related to regulation of the cell population size. Basal cells showed a lower proliferation rate than principal cells, but most of the immunoreactive bcl-2 protein, in pubertal and adult epididymides, was observed in basal cells. Therefore, these cells might comprise a low-proliferating and apoptosis-resistant population.

PURPOSE: To examine experimentally the feasibility of transfemoral endoluminal repair of aneurysms containing the ostia of essential branch arteries. METHODS: In a canine model (n = 4), suprarenal aortic aneurysms were created by suturing an artificial patch onto an anterior arteriotomy. Following a 2-week recovery period, the dogs underwent endovascular exclusion of their aneurysms using an aortic stent-graft with separate renal artery branch grafts. Outcome was evaluated using angiography, intravascular ultrasound (IVUS), Doppler flow, invasive pressure monitoring, and autopsy, respectively. RESULTS: Successful creation and subsequent endovascular exclusion of the aneurysm using aortic stent-grafts and separate bilateral renal artery stent-grafts was achieved in all trials. Angiographically, all aneurysms were excluded from aortic flow and all renal arteries were patent at completion of the procedure. With IVUS, good graft apposition and absence of perigraft flow were demonstrated in all animals. Mean pressure in the aneurysmal sac at completion of the procedure was 40 +/- 7 mmHg, compared to a mean systemic blood pressure of 105 +/- 8 mmHg (p < 0.05). At autopsy, no gross intimal damage was seen in the aorta or the renal arteries, and intact aortic grafts and branch grafts without twisting, coiling, or kinking were found in all trials. CONCLUSIONS: In an acute animal model, suprarenal aortic aneurysms can be excluded from the circulation with preservation of renal flow using an endoluminally placed aortic stent-graft with separate branch grafts.
