RESUMO

In the lymphocytes of 20 healthy subjects and 17 patients with chronic lymphatic leukaemia (CLL) the cAMP level was determined before and at different time periods during cell incubation with mitogens (phytohaemagglutinin--PHA, concanavalin A--Con A, and pokeweed mitogen PWM). The experiments demonstrated that PHA-induced lymphocyte proliferation was associated in healthy subjects during the first 10--20 minutes of incubation with a rise in the intracellular cAMP concentration, while the level of cAMP decreased systematically during 30 minutes of incubation with Con A and PWM. Incubation of lymphocytes of the patients with CLL during 30 minutes with PHA, Con A and PWM caused no changes in the low cAMP level in these cells. The obtained results suggest that PHA activation of lymphocytes in healthy subjects is connected with changes in the intracellular concentration of cAMP.

RESUMO

Deoxyribonucleic acid (DNA) synthesis was investigated in vitro in T and B lymphocyte populations isolated from peripheral blood. The synthesis was determined by measurements of 3H-thymidine incorporation. Only T lymphocytes isolated from peripheral blood of healthy subjects as well as patients with chronic lymphocytic leukemia (cll) showed proliferative ability after stimulation with phytohemagglutinin (PHA). DNA synthesis rate in normal T lymphocytes was enhanced by exogenous cyclic adenosine monophosphate (cAMP) added to lymphocyte culture together with PHA in concentrations of 10(-5) and 10(-4)M. Higher concentrations (10(-3) and 10(-2)M) of this nucleotide inhibited DNA synthesis in the lymphocytes of healthy subjects as well as cell patients. Impairment of 3H-thymidine incorporation observed in cll (in non-isolated cell populations) seems to be due to the presence of non-proliferating lymphocyte B population.

The level of cAMP was determined in isolated cells from bone marrow and peripheral blood of healthy subjects and patients with proliferative syndrome (acute and chronic myeloid leukaemia and chronic lymphatic leukaemia). In the investigations tritiated cAMP (3H-cAMP) was used and for binding of endogenous as well as exogenous cAMP protein isolated from bovine muscles was used. The mean cAMP level in peripheral blood granulocytes of healthy subjects was 27.90+/-3.82 pmol/10(7) cells and in normal lymphocytes it was from 11 to 18 pmol/10(7) cells. Much higher concentrations of cAMP: 56.4+/-16.25 and 52.7+/-11.02 pmol/10(7) cells were observed in myelocytes and metamyelocytes isolated from the bone marrow of healthy subjects. Lowering of cAMP concentration (below 4 pmol/10(7) cells) was observed in the lymphocytes of patients with chronic lymphatic leukaemia, while a higher cAMP concentration (above 90 pmol/10(7) cells) was found in the myeloblasts of patients with acute myeloid leukaemia.

RESUMO

Using a closed to-and-from respiratory system and measuring the COHb level in capillary blood the rise in endogenous carbon monoxide level in the blood was determined in 15 patients with haemolytic anaemia and in 6 healthy subjects. An evident rise in CO concentration with raised COHb level in the blood was found in all patients with haemolytic anaemia.

RESUMO

Serum levels of IgE in malignant lymphogranulomatosis (Hodgkin's disease) were lowered in patients who had been intensively treated, without eosinophilic granulocytes, and in less advanced forms of the disease. High levels were observed in untreated patients with eosinophilic granulocytes in their peripheral blood, and in clinically advanced cases.

RESUMO

The authors carried out a clinical analysis of problems connected with splenectomy in elderly patients treated surgically at the Institute of Haematology in the years 1965-1972. Out of 24 patients aged above 60 years 12 were analysed clinically (8 females and 4 males). Splenectomy was done in these patients from haematological indications (haemolytic anaemia, thrombocytopenia, pancytopenia). In all these cases indications to splenectomy, the course of operation and postoperative period as well as late therapeutic results were assessed. The analysis of these elderly patients showed that the criteria of indications for splenectomy were the same as in other age groups. The choice of surgical method depended in these cases on the clinical and haematological findings, a lack of permanent improvement after conservative treatment, contraindications to immunosuppressive treatment and results of isotope investigations (particularly the values of the spleen-liver index). In most splenectomized patients there were no complications during the operation and in the postoperative period. In 2 cases only acute circulatory failure was observed immediately after the operation. Evaluation of late results showed that the effects of the operation were similar to those in other age groups.
