RESUMO

The human skin is constantly exposed to external factors that affect its integrity, UV radiation being one of the main stress factors. The repeated exposure to this radiation leads to increased production of Reactive Oxygen Species (ROS) which activate a series of processes involved in photoaging. Excessive UV exposure also exacerbates melanin production leading to a variety of pigmentation disorders. Xanthones are reported to exhibit properties that prevent deleterious effects of UV exposure and high levels of ROS in the organism, so in this work a wide library of xanthones with different patterns of substitution was synthesized and tested for their inhibitory activity against the skin enzymes tyrosinase, elastase, collagenase and hyaluronidase, many of which were evaluated for the first time. Most of the compounds were tyrosinase inhibitors, with the best one (xanthone 27) presenting an IC50 of 1.9 µM, which is approximately 6 times lower than the IC50 of the positive control kojic acid. Concerning the other enzymes, only one compound presented IC50 lower than 150 µM in elastase inhibition (xanthone 14 = 91.8 µM) and none in collagenase and hyaluronidase inhibition. A QSAR model for tyrosinase inhibitory activity was built using six molecular descriptors, with a partial negative surface area descriptor and the relative number of oxygen atoms being positively contributing to the tyrosinase inhibitory activity. Docking using AutoDock Vina shows that all the tested compounds have more affinity to mushroom tyrosinase than kojic acid. Docking results implied that the tyrosinase inhibitory mechanisms of xanthonic derivatives are attributed to an allosteric interaction. Taken together, these data suggest that xanthones might be useful scaffolds for the development of new and promising candidates for the treatment of pigmentation-related disorders and for skin whitening cosmetic products.

The aim of this study was to estimate genetic parameters and genetic trends for reproductive traits in Wistar rats. A total of 1,167 data records from 283 females over six generations of monogamous mating pairs was used. Heritability and genetic correlation were estimated through Bayesian inference and genetic trends were calculated by linear regression of breeding values over generations. Heritability estimates for litter size at birth (LS), calving interval (CI), pup mortality (PM) and maternal cannibalism (CAN) presented low magnitude, ranging from 0.01 to 0.13. CAN presented high and positive genetic correlation with LS and PM (0.77 and 0.78, respectively). On the other hand, all the other estimated genetic correlations were not significant. Genetic trend was positive for LS (+0.0900 pups per generation), and negative for PM and CAN (-1.0085 and -0.5217 pups per generation, respectively). For CI the genetic trend was not significant. It is recommended to increase selection intensity on dams in this Wistar rat population in order to accelerate the genetic progress.(AU)


O objetivo deste estudo foi estimar os parâmetros genéticos e as tendências genéticas de características reprodutivas em ratos Wistar. Foram analisados 1.167 registros coletados em 283 fêmeas ao longo de seis gerações de pares de acasalamentos monogâmicos. Herdabilidade e correlação genética foram estimadas por meio de inferência bayesiana, e as tendências genéticas foram calculadas pela regressão linear dos valores genéticos em função das gerações. As estimativas de herdabilidades para as características número de filhotes nascidos (LS), intervalo de parto (CI), mortalidade de filhotes (PM) e canibalismo materno (CAN) foram de baixa magnitude (0,01 a 0,13). CAN apresentou correlação genética alta e positiva com LS e PM, 0,77 e 0,78, respectivamente. As demais correlações genéticas estimadas foram não significativas. A tendência genética foi positiva para LS (+0,0900 filhote por geração) e negativa para PM e CAN (-1,0085 e -0,5217 filhote por geração, respectivamente). A tendência genética não foi significativa para CI. Recomenda-se aumentar a intensidade de seleção nas fêmeas nessa população de ratos Wistar, a,fim de acelerar o progresso genético.(AU)

AIMS: The importance of bacterioferritin in the virulence and pathogenicity of the genus Mycobacterium is still unclear. The aim of this study was to analyse if the expression of a recombinant bacterioferritin from M. tuberculosis (Mtb) by Mycma could improve the capacity of this bacillus to resist the host defence mechanisms. METHODS AND RESULTS: Recombinant Mycma, expressing bacterioferritin (Rv1876) from Mtb, was developed by transformation with pMIP12_Rv1876. To determine bacterioferritin influence on Mycma physiology and virulence, the mycobacteria growth was analysed in vitro and in vivo. It was observed that the expression of bacterioferritin improved the growth rate of recombinant Mycma_BfrA under iron excess and oxidative stress, as compared to the wild type. Furthermore, in the murine model of infection, it was observed that Mycma_BfrA-infected mice had higher bacillary load and a more pronounced lesion in the lungs when compared with the wild type. CONCLUSION: This study showed that bacterioferritin confers additional resistance to stress conditions, resulting in increased pathogenicity of Mycma during mice infection. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provides new insights about the importance of bacterioferritin in the virulence and pathogenicity of the Mycobacterium genus.

Baccharis dracunculifolia (Asteraceae), the most important plant source of the Brazilian green propolis (GPE), displayed in vitro activity against Leishmania donovani, with an IC(50) value of 45 microg/mL, while GPE presented an IC(50) value of 49 microg/mL. Among the isolated compounds of B. dracunculifolia, ursolic acid, and hautriwaic acid lactone showed IC(50) values of 3.7 microg/mL and 7.0 microg/mL, respectively. Uvaol, acacetin, and ermanin displayed moderate antileishmanial activity. Regarding the antiplasmodial assay against Plasmodium falciparum, BdE and GPE gave similar IC(50) values (about 20 microg/mL), while Hautriwaic acid lactone led to an IC(50) value of 0.8 microg/mL (D6 clone).

OBJECTIVES: To study the incidence of galactosaemia in the state of São Paulo and the benefit/cost (B/C) ratio of the introduction of neonatal screening for galactosaemia, comparing it with a selective approach. METHODS: An enzymatic-colorimetric assay was used for the screening of total galactose (TG) in a sample of 10% of the births in São Paulo in one year and positive cases were confirmed by the activity of galactose-1-phosphate uridyltransferase (GALT). Detected and referred cases were genotyped using enzyme restriction studies for Q188R, N314D and S135L mutations of the GALT gene. The economic analysis was determined by calculating the B/C ratio and by analysis of sensitivity as a function of the incidence of the disease detected and the variation of the interest rate in the economy. RESULTS: 59 953 newborns were screened for TG, with 3 cases of galactosaemia being identified (0.26% false positives), corresponding to a frequency of 1:19 984 liveborns (95% confidence interval: 1:7494 to 1:59 953). One classical case and one Duarte 2 variant referred to as a selective approach were confirmed. With an incidence of 1:19 984, the B/C ratio was 1.04 for the 11.75% interest rate in effect in Brazil, with values already decapitalized. With a maximum possible incidence of 1:7494, the B/C ratio was 2.79. DISCUSSION: There is an economic advantage in introducing neonatal screening for galactosaemia in the national neonatal screening programme. This advantage could increase with a reduction of the current interest rates in the economy.

The prevalence of virulence genes expressing fimbriae, production of hemolysin, colicin and aerobactin, was determined in Escherichia coli isolates from healthy cows genital tract not showing clinical signs of infection. The presence of fimbriae expression genes (pap, sfa, afa) was assayed using specific primers in a polymerase chain reaction; none were detected in any of the isolates. Yet, a prevalence of 90.4 percent, 69.8 percent, and 28.5 percent of virulence factors for colicin, hemolysin and aerobactin respectively, was detected in the isolates. Analysis of the bacterial pathogenicity of isolates from the bovine genital tract may contribute towards the understanding of E. coli behavior.


Determinou-se a prevalência dos genes de virulência expressando fimbrias, produção de hemolisina, colicina e aerobactina em cepas de Escherichia coli obtidas do trato genital de vacas saudáveis que não apresentam sinais clínicos indicativos de infecção. A presença dos genes responsáveis pela expressão de fimbrias (pap, sfa, afa) foi avaliada através de reação em cadeia da polimerase utilizando primers especificos para cada um dos genes, nenhum deles foi detectado em qualquer uma das cepas isoladas. A prevalência dos fatores de virulência foi de 90,4 por cento, 69,8 por cento, 28,5 por cento para colicina, hemolisina e aerobactina, respectivamente. A análise da patogenicidade das cepas do trato genital pode contribuir para o entendimento do comportamento das cepas de E. coli.

Intracellular development of Cystoisospora belli was demonstrated in 4 different mammalian cell lines. Human ileocecal adenocarcinoma (HCT-8), epithelial carcinoma of lung (A549), Madin-Darby bovine kidney (MDBK), and African green monkey kidney (VERO) were exposed in vitro to C. belli sporozoites, which had been isolated from the feces of HIV-AIDS patients. Parasites invaded all the cellular types between 4 and 12h after exposure and multiplication was demonstrated after 24 h. Grater number of merozoites formed in VERO cells, followed by HCT-8. In the MDBK and HCT-8 cells, the parasitophorous vacuole was less evident and immobile merozoites were observed in the cytoplasm. In VERO cells, one or several parasitophorous vacuoles contained up to 16 mobile sporozoites. No oocysts were found in any of the cell types used. VERO cells may be suitable for studies of the interaction between parasite and host cells.

Low CD8(+) T lymphocyte numbers have contributed to deciphering the genotype/phenotype discrepancies found in hereditary hemochromatosis (HH) patients genotyped for the Hfe mutations, C282Y and H63D. In this study, we extend the analysis of T lymphocytes in HH to the T cell receptor (TcR) repertoire. Thirty-two HH patients (C282Y homozygous) and 274 Hfe genotyped healthy subjects were studied. The following TcR chains were analyzed: Valpha2.3, Vbeta5.1, Vbeta5.2, Vbeta5.3, Vbeta6.7, Vbeta8, and Vbeta12 among the CD4(+) and CD8(+) populations. Lymphopenias and absence of expansions of the Vbeta5.2 and Vbeta12 chains in the CD8(+) pool were seen in controls heterozygous for the C282Y mutation. Expansions in the control group were seen within the CD8(+) pool and were rare/absent within the CD4(+) pool. TcR expansions were found more frequent in patients with iron overload related pathology than in patients without pathology. 9/16 of the patients with pathology have at least one expansion among the CD8(+) pool a number significantly higher compared with patients without pathology (1/16). These findings suggest that Hfe has an effect in the shaping of T-cell populations either directly, as indicated by the lymphopenia seen in the two chains in C282Y heterozygous without iron overload, or indirectly by contributing to iron overload pathology.

BACKGROUND: Despite heavy alcohol consumption, only a low percentage of heavy drinkers develop liver disease. Imbalances in T-cell subsets and iron metabolism parameters are common findings in heavy drinkers, yet the possible role played by discrete T-lymphocyte subsets under heavy alcohol consumption remains unclear. METHODS: To gain new insights into the possible role played by T lymphocytes during alcohol consumption, characterization of CD28 expression and TcR repertoire in peripheral blood CD4+ and CD8+ T cells by two and three-color flow cytometry was performed. A group of heavy alcohol drinkers (AHD, n = 71) and a group of age-matched controls (n = 81), both HLA-phenotyped and HFE-genotyped, constituted the groups under study. RESULTS: Marked expansions of CD28- T cells within the CD8+ but not the CD4+ T-cell pool were observed in AHD compared with controls. These CD8+CD28- expansions were paralleled by expansions of CD8+ T cells bearing specific TcR Valpha/beta chains, namely VP5.2. Moreover, AHD, but not controls, carrying the H63D mutation in the HFE gene showed significantly higher percentages of CD28- T cells within the CD8+ T-cell pool than AHD carrying the normal HFE gene. Finally, high numbers of CD8+CD28- T cells in AHD were associated with lower levels of the liver-related enzymes ALT and GGT. CONCLUSIONS: This study showed that under active ethanol consumption, expansions of discrete CD8+ T-cell subsets occur within the CD8+ T-cell pool, that molecules of the MHC-class I locus seem to influence the extent of the expansions, and that high numbers of CD8+CD28- T cells are associated with low levels of liver enzymes in AHD.

In this study, the results obtained in a control programme of schistosomiasis in Ravena (Sabará, Minas Gerais) between 1980 and 1992 are evaluated. Control measures used in this programme were: specific treatment of the people infected with Schistosoma mansoni at four year-intervals (1980/84/88) and the supply of tap water to 90% of the residences in 1980. A significant reduction of the prevalence (36.7% to 11.5%, p < or = 0.05) and of the intensity of the infection (228.9 eggs per gram of feces (epg), s = 3.7 to 60.3 epg, s = 3.5, p < or = 0.05) was observed. No cases of the severe form of the disease were diagnosed in the area. Factors independently associated with the infection were in 1980 daily sand extraction and the lack of tap water in residences and in 1992 daily sand extraction and fishing and weekly swimming. Concluding, the supply of tap water together with quadrennial treatments significantly diminished both the prevalence and intensity of the S. mansoni infection, with the additional gain of persistent low indices even after four-year intervals between the treatments.

The reproduction of the snail Biomphalaria glabrata, one of the intermediate hosts of Schistosoma mansoni, was evaluated under laboratory conditions by measuring egg production, number of ovipositions, number of eggs per oviposition, and hatching rate using a Latin square design with five different temperature treatments. This permitted the elimination of occasional variation in oviposition due to manipulation or a priori group. 2. Egg production and spawning rates were higher at temperatures between 20.0 and 27.5 degrees C than at 17.5 degrees C. Number of eggs per spawning and hatching rate did not vary with temperature. 3. During a 3-day adaptation phase, a short-term thermal effect appeared in which some indices of reproductive rates differed significantly from those of the experimental phase.

RESUMO

Callithrix penicillata is a good model for dermatotropic strains of some New World Leishmania species (L. mexicana mexicana, L. m. amazonensis, and L. braziliensis guyanensis). Both amastigotes and promastigotes readily infect these primates, with a relative short period of incubation. The developed lesions parallel those found in man, and despite the self healing presented by some marmosets, cell and serum immunological responses are presented. Skin alterations are confined to the site of inoculum, and visceralization is very rarely found. A good response to standard antimonial treatment makes the model useful for preclinical experiments with promising compounds.

RESUMO

For a period of three years, gelatin was used as a vehicle for administration of food, vitamin complex and drugs to laboratory maintained Callithrix penicillata and Callithrix jacchus marmosets and a breeding colony of C. penicillata. In both circumstances, results were satisfactory. The animals survived for long periods of time, with adult, young and juvenile primates in the breeding colony showing good physical condition.
