Aerobic vaginitis (AV) is a recently defined vaginal recurring infection, which is treated with antibiotics. However, excessive and prolonged use of antibiotics disrupts healthy vaginal microflora and leads to the emergence of antibiotic resistance among pathogens. This situation has directed researchers to explore alternative antimicrobials. The current study describes in vitro and in vivo antimicrobial efficacy and pharmaceutical interactions between plant essential oils (EOs) and five lactic acid bacteria (LABs), isolated from the healthy vagina, against E. faecalis, one of the major etiological agents of AV. In vitro experiments confirm good antimicrobial activity of both plant EOs and cell free supernatant (CFS) from LABs. Based on high antimicrobial efficacy, Moringa essential oil (MO) was selected to determine its nature of interaction with CFS of five LAB strains. Synergism was recorded between MO and CFS of L. reuteri (MT180537). To validate in vitro findings, prophylactic responses of individual and synergistic application of MO and L. reuteri (MT180537) were evaluated in an E. faecalis (MW051601) induced AV murine model. The prophylactic efficacy was evidenced by a reduction in intensity of clinical symptoms, E. faecalis (MW051601) count per vaginal tissue along with a reduction in AV associated changes in histological markers of infection in animals receiving Moringa essential oil and L. reuteri (MT180537) alone or in combination. However, significant synergism between Moringa essential oil and L. reuteri (MT180537) could not be observed. Our data confirms the importance of in vivo experiments in deducing pharmacological interactions.

RESUMO
This article describes the environmental impacts of producing a single seedling in forest nurseries of selected districts (i.e., Haripur, Abbottabad, and Mansehra) of Hazara Division of Khyber Pakhtunkhwa, Pakistan using the life cycle assessment (LCA) approach. This study was based on the cradle-to-gate approach which begins with the pre-nursery stage and progresses toward the main nursery before transplanting seedlings into the plantation site. Data or life cycle inventory (LCI) of seedling production were collected through questionnaire surveys and personal meetings with forest nurseries managers and workers regarding consumption of different inputs such as electricity, diesel, fertilizers, herbicides, and polyethylene bags, organic manure, and water consumption. The SimaPro software version 8.5 and the CML2000 v2.05 environmental model was applied to perform life cycle impact assessment (LCIA) for a single seedling production in forest nurseries in the study area. In line with the objectives of the study, primary data regarding inputs and outputs of the nurseries were collected from 35 nurseries in the study area by using a random questionnaire method. In addition, secondary data were taken from online databases such as Eco-invent v.3.2 CORRIM and peer-reviewed published literature. For this study, a functional unit of a single seedling was considered. Production weighted average data were modeled in the latest environmental modeling software i.e., SimaPro v.8.5 for ten US-EPA most wanted environmental impacts, such as global warming potential (GWP), abiotic depletion (AD), eutrophication potential (EP), acidification potential (AP), freshwater aquatic eco-toxicity (FAE), marine water eco-toxicity (MWE), terrestrial eco-toxicity (TE), ozone layer depletion (OLD), photochemical oxidation (PO), and human toxicity (HT). The results showed that the highest environmental impact posed by a single seedling was marine aquatic eco-toxicity (11.31360 kg 1,4-DB eq), followed by global warming potential (0.02945 kg CO2 eq) and (0.01227 kg 1,4-DB eq) human toxicity. The primary reason for these environmental burdens was the use of synthetic fertilizers in forest nurseries and the consumption of fossil fuels in nursery mechanization and transportation activities. The total cumulative energy demand for a single seedling was (0.800 MJ) with more than 90% contribution from fossil fuel energy resources such as petrol and diesel. It is therefore highly recommended to use renewable energy resources and organic fertilizers instead of chemical fertilizers in forest nurseries to avoid and minimize greenhouse gas emissions (GHS) and other toxic emissions in the study area.
Este artigo descreve os impactos ambientais da produção de uma única muda em viveiros florestais de distritos selecionados (Haripur, Abbottabad e Manshera) da divisão Hazara, de Khyber Pakhtunkhwa, Paquistão, usando a abordagem de avaliação do ciclo de vida (ACV). Este estudo baseou-se na abordagem "do berço ao portão", que se inicia na fase pré-viveiro e progride em direção ao viveiro principal antes do transplante das mudas para o local de plantio. Os dados ou inventário de ciclo de vida (ICV) da produção de mudas foram coletados por meio de questionários e reuniões pessoais com o gerente e os trabalhadores dos viveiros florestais sobre o consumo de diferentes insumos, como energia elétrica, diesel, fertilizantes, herbicidas, sacos de polietileno, adubo orgânico e consumo de água. O software SimaPro, versão 8.5, e o modelo ambiental CML2000, v2.05, foram aplicados para realizar a avaliação de impacto do ciclo de vida (AICV) de uma única produção de mudas em viveiros florestais na área de estudo. Em consonância com os objetivos do estudo, os dados primários relativos às entradas e saídas dos viveiros foram coletados de 35 viveiros na área de estudo por meio de um método de questionário aleatório. Além disso, dados secundários foram obtidos de bancos de dados online, como Eco-invent v.3.2 CORRIM e literatura publicada revisada por pares. Para este estudo, foi considerada uma unidade funcional de uma única plântula. Os dados médios ponderados de produção foram modelados no software de modelagem ambiental mais recente, ou seja, SimaPro, v.8.5, para 10 impactos ambientais mais desejados pela US-EPA, como potencial de aquecimento global (PAG), depleção abiótica (DA), potencial de eutrofização (PE), potencial de acidificação (PA), ecotoxicidade de água doce (EAD), ecotoxicidade da água marinha (EAM), ecotoxicidade terrestre (ET), destruição da camada de ozônio (DCO), oxidação fotoquímica (OF) e toxicidade humana (TH). Os resultados mostraram que o maior impacto ambiental causado por uma única muda foi a ecotoxicidade da água marinha (11,31360 kg 1,4-DB eq), seguido pelo potencial de aquecimento global (0,02945 kg CO2 eq) e toxicidade humana (0,01227 kg 1,4-DB eq) eq). A principal razão para esses ônus ambientais foi o uso de fertilizantes sintéticos em viveiros florestais e o consumo de combustíveis fósseis nas atividades de mecanização e transporte de viveiros. A demanda total acumulada de energia para uma única muda foi de 0,800 MJ, com mais de 90% de contribuição de recursos energéticos de combustíveis fósseis, como gasolina e diesel. Portanto, é altamente recomendável usar recursos de energia renovável e fertilizantes orgânicos em vez de fertilizantes químicos em viveiros florestais para evitar e minimizar as emissões de gases de efeito estufa (GEE) e outras emissões tóxicas na área de estudo.

However, multiple abiotic stresses especially high and low-temperature proved to be a substantial threat for rice production ultimately risks for food security. To overcome various types of abiotic stress; seed priming is among the effective approaches to improve the rice seed germination and growth vigor. Therefore, the present study was planned to evaluate physiological and biochemical modifications in Chinese and Pakistani rice varieties by Qiangdi 863 biological assistant growth apparatus nano treated water (NTW), Osmopriming Calcium chloride (CaCl2), redox priming hydrogen peroxide (H2O2) and hormonal priming by Salicylic acid (SA) under temperature stress conditions. The experiment was performed with completely randomize design conditions. Five rice varieties, nomenclature as Zhongzoa 39, (Chinese rice variety) KSK 133, KS 282, Super basmati and PK 1121 aromatic (Pakistani rice variety) were sown under low temperature (LT) (17ºC), optimal temperature (OT) 27ºC and high temperature (HT) 37ºC conditions. The present study indicated that nanopriming were the most effective treatments increased Germination Energy Percentage (GEP) (96.1, 100, 100%), Speed of Germination (SG) (27.2, 35.45, 37.1), Final Germination Percentage (FGP) (98.2, 99.1, 99.4%), Seedling Dry Weight Biomass (DWB) (0.1, 0.137, 0.14g), Total Chlorophyll Content (0.502, 13.74, 15.21), antioxidant enzymes Superoxide Dismutase (SOD)(3145, 2559, 3345 µg-1FWh-1), Catalase (CAT) (300, 366, 3243 µg-1FWh-1) and decreased Malondialdehyde (MDA) (6.5, 12.2, 6.5 µmol g-1 FW) for Zhongzao 39 and KSK 133 rice varieties under low (LT+NTW), optimal temperature (OP+NTW) and high temperature (HT+NTW) stress., Therefore, nano-priming is recommended to cope with the high and low-temperature stress conditions along with improved productivity of rice.

To overcome various types of abiotic stress; seed priming is among the effective approaches to improve the rice seed germination and growth vigor. Therefore, the present study was planned to evaluate physiological and biochemical modifications in Chinese and Pakistani rice varieties by Qiangdi 863 biological assistant growth apparatus nano treated water (NTW), Osmopriming Calcium chloride (CaCl2), redox priming hydrogen peroxide (H2O2) and hormonal priming by Salicylic acid (SA) under temperature stress conditions. The experiment was performed with completely randomize design conditions. Five rice varieties, nomenclature as Zhongzoa 39, (Chinese rice variety) KSK 133, KS 282, Super basmati and PK 1121 aromatic (Pakistani rice variety) were sown under low temperature (LT) (17ºC), optimal temperature (OT) 27ºC and high temperature (HT) 37ºC conditions. The present study indicated that nanopriming were the most effective treatments increased Germination Energy Percentage (GEP) (96.1, 100, 100%), Speed of Germination (SG) (27.2, 35.45, 37.1), Final Germination Percentage (FGP) (98.2, 99.1, 99.4%), Seedling Dry Weight Biomass (DWB) (0.1, 0.137, 0.14g), Total Chlorophyll Content (0.502, 13.74, 15.21), antioxidant enzymes Superoxide Dismutase (SOD)(3145, 2559, 3345 µg-1FWh-1), Catalase (CAT) (300, 366, 3243 µg-1FWh-1) and decreased Malondialdehyde (MDA) (6.5, 12.2, 6.5 µmol g-1 FW) for Zhongzao 39 and KSK 133 rice varieties under low (LT+NTW), optimal temperature (OP+NTW) and high temperature (HT+NTW) stress., Therefore, nano-priming is recommended to cope with the high and low-temperature stress conditions along with improved productivity of rice.
Resumo Cultivo e consumo de arroz ocorrem em grandes quantidades na Ásia, particularmente no Paquistão e na China. No entanto, vários estresses abióticos, especialmente de alta e baixa temperatura, provaram ser uma ameaça considerável para a produção de arroz, em última análise, riscos para a segurança alimentar. Para superar vários tipos de estresse abiótico, o priming de sementes está entre as abordagens eficazes que melhoram a germinação e o vigor de crescimento das sementes de arroz. Portanto, o presente estudo foi planejado para avaliar as modificações fisiológicas e bioquímicas em variedades de arroz chinês e paquistanês por Qiangdi 863, aparelho assistente biológico de crescimento com água nanotratada (NTW), Osmopriming cloreto de cálcio (CaCl2), peróxido de hidrogênio redox (H2O2) e priming hormonal por ácido salicílico (SA) em condições de estresse de temperatura. O experimento foi realizado em condições de delineamento inteiramente ao acaso. Cinco variedades de arroz, nomenclaturas como Zhongzoa 39 (variedade de arroz chinês), KSK 133, KS 282, Super basmati e PK 1121 aromático (variedade de arroz do Paquistão) foram semeadas sob baixa temperatura (LT) (17 ºC), temperatura ótima (OT) 27 ºC e condições de alta temperatura (HT) 37 ºC. O presente estudo indicou que nanocondicionamento foi o tratamento mais eficaz: aumento da porcentagem de energia de germinação (GEP) (96,1%, 100%, 100%), velocidade de germinação (SG) (27,2, 35,45, 37,1), porcentagem de germinação final (FGP) (98,2%, 99,1%, 99,4%), biomassa de peso seco de mudas (DWB) (0,1 g, 0,137 g, 0,14 g), conteúdo total de clorofila (0,502, 13,74, 15,21), enzimas antioxidantes superóxido dismutase (SOD) (3145, 2559, 3345 µg-1FWh- 1), catalase (CAT) (300, 366, 3243 µg-1FWh-1) e malondialdeído diminuído (MDA) (6,5, 12,2, 6,5 µmol g-1 FW) para as variedades de arroz Zhongzao 39 e KSK 133 sob baixo (LT + NTW), temperatura ótima (OP + NTW) e estresse de alta temperatura (HT + NTW). Portanto, o nanopriming é recomendado para lidar com as condições de estresse de alta e baixa temperatura, juntamente com a produtividade aprimorada do arroz.
RESUMO
This article describes the environmental impacts of producing a single seedling in forest nurseries of selected districts (i.e., Haripur, Abbottabad, and Mansehra) of Hazara Division of Khyber Pakhtunkhwa, Pakistan using the life cycle assessment (LCA) approach. This study was based on the cradle-to-gate approach which begins with the pre-nursery stage and progresses toward the main nursery before transplanting seedlings into the plantation site. Data or life cycle inventory (LCI) of seedling production were collected through questionnaire surveys and personal meetings with forest nurseries managers and workers regarding consumption of different inputs such as electricity, diesel, fertilizers, herbicides, and polyethylene bags, organic manure, and water consumption. The SimaPro software version 8.5 and the CML2000 v2.05 environmental model was applied to perform life cycle impact assessment (LCIA) for a single seedling production in forest nurseries in the study area. In line with the objectives of the study, primary data regarding inputs and outputs of the nurseries were collected from 35 nurseries in the study area by using a random questionnaire method. In addition, secondary data were taken from online databases such as Eco-invent v.3.2 CORRIM and peer-reviewed published literature. For this study, a functional unit of a single seedling was considered. Production weighted average data were modeled in the latest environmental modeling software i.e., SimaPro v.8.5 for ten US-EPA most wanted environmental impacts, such as global warming potential (GWP), abiotic depletion (AD), eutrophication potential (EP), acidification potential (AP), freshwater aquatic eco-toxicity (FAE), marine water eco-toxicity (MWE), terrestrial eco-toxicity (TE), ozone layer depletion (OLD), photochemical oxidation (PO), and human toxicity (HT). The results showed that the highest environmental impact posed by a single seedling was marine aquatic eco-toxicity (11.31360 kg 1,4-DB eq), followed by global warming potential (0.02945 kg CO2 eq) and (0.01227 kg 1,4-DB eq) human toxicity. The primary reason for these environmental burdens was the use of synthetic fertilizers in forest nurseries and the consumption of fossil fuels in nursery mechanization and transportation activities. The total cumulative energy demand for a single seedling was (0.800 MJ) with more than 90% contribution from fossil fuel energy resources such as petrol and diesel. It is therefore highly recommended to use renewable energy resources and organic fertilizers instead of chemical fertilizers in forest nurseries to avoid and minimize greenhouse gas emissions (GHS) and other toxic emissions in the study area.

Aerobic vaginitis (AV) is a recently defined vaginal recurring infection, which is treated with antibiotics. However, excessive and prolonged use of antibiotics disrupts healthy vaginal microflora and leads to the emergence of antibiotic resistance among pathogens. This situation has directed researchers to explore alternative antimicrobials. The current study describes in vitro and in vivo antimicrobial efficacy and pharmaceutical interactions between plant essential oils (EOs) and five lactic acid bacteria (LABs), isolated from the healthy vagina, against E. faecalis, one of the major etiological agents of AV. In vitro experiments confirm good antimicrobial activity of both plant EOs and cell free supernatant (CFS) from LABs. Based on high antimicrobial efficacy, Moringa essential oil (MO) was selected to determine its nature of interaction with CFS of five LAB strains. Synergism was recorded between MO and CFS of L. reuteri (MT180537). To validate in vitro findings, prophylactic responses of individual and synergistic application of MO and L. reuteri (MT180537) were evaluated in an E. faecalis (MW051601) induced AV murine model. The prophylactic efficacy was evidenced by a reduction in intensity of clinical symptoms, E. faecalis (MW051601) count per vaginal tissue along with a reduction in AV associated changes in histological markers of infection in animals receiving Moringa essential oil and L. reuteri (MT180537) alone or in combination. However, significant synergism between Moringa essential oil and L. reuteri (MT180537) could not be observed. Our data confirms the importance of in vivo experiments in deducing pharmacological interactions.

Objectives of the study were to investigate influence of artificial insemination (AI) in caged and floored flock in comparison to natural mating (NM) on broiler breeders welfare, productive performance and economic appraisal. To execute this experiment, a total of 1440 pullets of Ross-308 Parent Stock along with 168 males were picked from a commercial flock and divided into three groups i.e., AIC=AI in cages, AIF=AI on floor and third was NM=NM on floor. Each group carried 480 females (HH) while 168 males were divided into 41, 48 and 58 for AIC, AIF and NM, respectively. During both phases all procedures and studied parameters were the same except the sperm dose rates which were changed during post peak. According to the results, significantly (p0.05) higher levels of serum corticosterone, glucose and cholesterol were recorded in birds of AIC and respectively as compared to birds being allowed to mate naturally. Whereas, the experimental males and females of AIF and AIC kept for AI exhibited (p0.05) higher body weight, depletion % and feed consumption particularly in post peak phase. However, among the birds subjected to AI, the birds kept in the cages had better performance (p0.05) than the birds kept in the floor. Moreover, frequency rate of insemination in females and milking of males found (p0.05) reciprocal to the depletion, feed intake and body weight during peak but particularly in post peak. Perhaps, various sperm doses remained inert to implicit any impact on studied parameters. Similarly, egg weight was neither affected by housing systems nor by mating strategies during both phases. However, various insemination and milking frequencies noticeably swayed the productive traits under this study. AI in floored flock was found ineffective even in comparison to natural mating. In conclusion, AI in cages brought forth the better productive performance and lesser male depletion, hence, can be recommended.

A positive crossmatch has been associated with increased risk in liver transplantation. To study the clinical significance of preformed donor-specific human leukocyte antigen antibodies (DSAs) in liver transplantation, we reviewed patients who underwent liver transplantation with a strongly positive flow cytometry crossmatch. DSAs were evaluated with a Luminex solid phase assay. The complement-fixing ability of DSAs was tested with a complement component 1q (C1q) assay. Using an assay correlation between complement-dependent cytotoxicity crossmatch, flow cytometry crossmatch, and DSA results, we reviewed the effects of DSAs on the outcomes of our patients as well as reported cases in the literature. Five of 69 liver recipients had a strongly positive crossmatch: 4 had a positive T cell crossmatch [median channel shift (MCS) = 383.5 ± 38.9], and 5 had a positive B cell crossmatch (MCS = 408.8 ± 52.3). The DSAs were class I only in 1 patient, class I and II in 3 patients, and class II only in 1 patient. Cholestasis, acute rejection, or both were observed in 3 of the 4 patients with a positive T cell crossmatch with an MCS approximately greater than 300. The C1q assay was positive for 3 patients. Two had either persistent cholestasis or early acute rejection. One patient who was treated with preemptive intravenous immunoglobulin had an unremarkable outcome despite a positive C1q result. One of the 2 patients with a negative C1q assay experienced persistent cholestasis and early and recurrent acute rejection; the other had an unremarkable outcome. None of the patients died or lost a graft within the first year of transplantation. Our study suggests that human leukocyte antigen antibody screening, flow cytometry crossmatch MCS levels, DSA mean fluorescent intensity levels, and C1q assays may be useful in assessing the risk of antibody-mediated rejection and timely interventions in liver transplantation.

The concentrations of serum lipids were measured in patients with lepromatous (LL/BL) leprosy and erythema nodosum leprosum (ENL). The relationships between serum lipid levels and serum amyloid A (SAA) and C-reactive protein (CRP) were also examined in these patients. LL/BL patients had significantly higher serum triglyceride and lower HDL-cholesterol concentrations compared to the endemic controls. ENL patients had significantly lower total, HDL- and LDL-cholesterol levels compared to the endemic controls. The levels of all lipid metabolites also were significantly lower in ENL patients compared to LL/BL patients. The concentrations of SAA and CRP were markedly elevated in ENL patients but were not statistically different in LL/BL patients compared to control subjects. There was a significant negative correlation between SAA and HDL-cholesterol levels in both stable lepromatous and reactional (ENL) patients; there was no statistically significant correlation between CRP and HDL-cholesterol levels. SAA levels also had a significant negative correlation with total and LDL-cholesterol levels. Our results indicate that serum lipids are significantly altered in patients with lepromatous disease and ENL reaction. Our results also suggest that an increase in SAA levels may divert the metabolism of lipoproteins from hepatocytes toward macrophages, resulting in a decrease in serum lipoprotein levels.
