RESUMO

Narrow gap welding is a prevalent technique used to decrease the volume of molten metal and heat required to fill a joint. Consequently, deleterious effects such as distortion and residual stresses may be reduced. One of the fields where narrow groove welding is most employed is pipeline welding where misalignment, productivity and mechanical properties are critical to a successful final assemblage of pipes. This work reports the feasibility of joining pipe sections with 4 mm-wide narrow gaps machined from API X80 linepipe using cold wire gas metal arc welding. Joints were manufactured using the standard gas metal arc welding and the cold wire gas metal arc welding processes, where high speed imaging, and voltage and current monitoring were used to study the arc dynamic features. Standard metallographic procedures were used to study sidewall penetration, and the evolution of the heat affected zone during welding. It was found that cold wire injection stabilizes the arc wandering, decreasing sidewall penetration while almost doubling deposition. However, this also decreases penetration, and incomplete penetration was found in the cold wire specimens as a drawback. However, adjusting the groove geometry or changing the welding parameters would resolve this penetration issue.

Salmonella enterica infection is a major public health concern worldwide, particularly when associated with other medical conditions. The serovars Typhimurium and Enteritidis are frequently associated with an invasive illness that primarily affects immunocompromised adults and children with HIV, malaria, or malnutrition. These serovars can also cause infections in a variety of animal hosts, and they are the most common isolates in poultry materials. Here, we described S. Enteritidis mutants, where hupA and hupB genes were deleted, and evaluated their potential use as live-attenuated vaccine candidates. In vitro, the mutants behaved like S. Typhimurium described previously, but there were some particularities in macrophage invasion and survival experiments. The virulence and immunogenicity of the mutant lacking both hupA and hupB (PT4ΔhupAB) were evaluated in a BALB/c mice model. This mutant was highly attenuated and could, therefore, be administrated at doses higher than 109 CFU/treatment, which was sufficient to protect all treated mice challenged with the wild-type parental strain with a single dose. Additionally, the PT4ΔhupAB strain induced production of specific IgG and IgA antibodies against Salmonella and TH1-related cytokines (IFN-γ and TNF-α), indicating that this strain can induce systemic and mucosal protection in the murine model. Additional studies are needed to better understand the mechanisms that lead to attenuation of the double-mutant PT4ΔhupAB and to elucidate the immune response induced by immunization using this strain. However, our data allow us to state that hupAB mutants could be potential candidates to be explore as live-attenuated vaccines.

Pyrolysis of industrial fatty wastes (soybean soapstock, beef tallow, and poultry industry waste) was carried out in the absence of catalysts. In all cases, organic mixtures of hydrocarbons and oxygenated compounds were obtained. These mixtures were distilled and diesel-like fractions were isolated and characterized by GC-FID, GC-MS and FT-IR, showing the formation of olefins, paraffins, and some oxygenated compounds such as carboxylic acids and esters. The main physical-chemical properties of those isolated diesel-like fuels (density, viscosity, distillation curve, carbon residue, copper corrosion test, cetane index, cold finger plugging point, acid index and heating value) were determined using ASTM standard methods and matched the Brazilian specification for diesel fuel.
