RESUMO
Hepatic and pulmonary lesions are common in cetaceans, despite their poorly understood viral etiology. Herpesviruses (HV), adenoviruses (AdV) and hepatitis E virus (HEV) are emerging agents in cetaceans, associated with liver and/or pulmonary damage in mammals. We isolated and molecularly tested DNA for HV and AdV (n = 218 individuals; 187 liver and 108 lung samples) and RNA for HEV (n = 147 animals; 147 liver samples) from six cetacean families. All animals stranded or were bycaught in Brazil between 2001 and 2021. Positive-animals were analyzed by histopathology. Statistical analyses assessed if the prevalence of viral infection could be associated with the variables: species, family, habitat, region, sex, and age group. All samples were negative for AdV and HEV. Overall, 8.7% (19/218) of the cetaceans were HV-positive (4.8% [9/187] liver and 11.1% [12/108] lung), without HV-associated lesions. HV-prevalence was statistically significant higher in Pontoporiidae (19.2%, 10/52) when compared to Delphinidae (4.1%, 5/121), and in southeastern (17.1%, 13/76)-the most industrialized Brazilian region-when compared to the northeastern region (2.4%, 3/126). This study broadens the herpesvirus host range in cetaceans, including its description in pygmy sperm whales (Kogia breviceps) and humpback whales (Megaptera novaeangliae). Further studies must elucidate herpesvirus drivers in cetaceans.

Pacas (Cuniculus paca) are highly hunted animals because of the flavor of their meat, and commercial breeding is recommended. However, this species has a relatively low reproductive rate. This study aimed to collect semen from pacas through electroejaculation and obtain the sperm parameters of this species for the first time. Seven male pacas were used, submitted to an anesthetic protocol before stimulation by an electroejaculator appropriate for the species. The stimulus protocol was performed in three series: series I, 10 stimuli with 1 and 2 V; series II, 10 stimuli with 3 and 4 V; and series III, 10 stimuli with 5 V and interval between series of 2 s. The collected material was evaluated for color, volume, motility, vigor, and concentration. The sperm parameters collected showed a mean volume of 0.43±0.33 mL, concentration of 45.5±42.44×106 sperm/mL, motility of 33.33±32.14%, and mean vigor of 2.6±1.15. In this study, the anesthetic protocol did not seem to favor semen collection by electroejaculation in the pacas. The electrical stimulation protocol was able to stimulate all animals in the study; however, there were few samples with sperm cells and a low rate of motility and vigor in most ejaculates.

Autophagy is a lysosomal degradation pathway that removes protein aggregates and damaged organelles maintaining cellular integrity. It seems to be essential for cell survival during stress, starvation, hypoxia, and consequently to the placenta implantation and development. Preeclampsia (PE) is a multisystemic disorder characterized by the onset of hypertension associated or not with proteinuria and other maternal complications. Considering that the placenta seems to play an important role in the pathogenesis of PE, the objective of the present study was to evaluate protein levels of light chain protein (LC3), beclin-1, and the mammalian target of rapamycin (mTOR) in the placenta of pregnant women with PE. Placental tissues collected from 20 women with PE and 20 normotensive (NT) pregnant women were evaluated for LC3, beclin-1, and mTOR expression by qPCR and immunohistochemistry. The mRNA for LC3 and beclin-1 were significantly lower, while mTOR gene expression was significantly higher in the placenta of pregnant women with PE than in the NT group. Placentas of PE women showed significantly decreased protein expression of LC3-II and beclin-1, whereas mTOR was significantly increased compared with the NT pregnant women. There was a negative correlation between protein expression of mTOR and LC3-II in the placental tissue of PE women. In conclusion, the results showed autophagy deficiency suggesting that failure in this degradation process may contribute to the pathogenesis of PE; however, new studies involving cross-talk between autophagy and inflammatory molecular mechanisms might help to better understand the autophagy process in this obstetric pathology.

The concern regarding the harm caused by biocides to human health has been increasing over the years, making the natural products an alternative to less toxic and more efficient biocides. Therefore, this paper reports the investigation of the disinfectant potential of extracts and isolated compounds from Baccharis dracunculifolia. For this purpose, extracts of aerial parts (BD-C), tricomial wash (BD-L) and roots (BD-R) of B. dracunculifolia were obtained by maceration. The extracts were submitted to different chromatographic techniques, including high-speedy countercurrent chromatography (HSCCC) furnishing nine isolated compounds. The extracts and isolated compounds were evaluated regarding their antimicrobial activity by the broth microdilution method, according to the Clinical and Laboratory Standards Institute, and regarding their sanitizing activity according to Standard Operating Procedure No. 65·3210·007 (INCQS, 2011), developed by the National Institute for Quality Control in Health (INCQS) - Oswaldo Cruz Foundation (FIOCRUZ). In the antimicrobial evaluation the BD-C extract showed minimum inhibitory concentration (MIC) values of 200 and 100 µg/ml against Staphylococcus aureus and Tricophyton mentagrophytes, respectively. BD-L extract showed MIC value of 200 µg/ml against S. aureus. The isolated compounds caffeic acid (MBC 2·22 µmol l-1 ), ferulic acid (MBC 2·06 µmol l-1 ) and baccharin (MBC 0·27 µmol l-1 ) showed significant inhibitory activity against S. aureus. All B. dracunculifolia isolated compounds were active with exception of aromadrendin-4´-O-methyl-ether for T. mentagrophytes. Additionally, isosakuranetin was active against Salmonella choleraesuis (MIC 1·4 µmol l-1 ). Regarding the sanitizing activity, the hydroalcoholic solution containing 0·2% of B. dracunculifolia extract in 40°GL ethanol was effective in eliminating the microbial contamination on all carrier cylinders and against all microorganisms evaluated in the recommended exposure time of 10 min. Therefore, B. dracunculifolia has potential for the development of sanitizing agents to be used in hospitals, food manufactures and homes.

The objective of this study was to investigate the effect of whole body vibration (WBV) exercise on oxidative stress markers in a group of women with fibromyalgia (FM) compared to a group of healthy women (CT). Twenty-one women diagnosed with FM and 21 age- and weight-matched healthy women were enrolled the study. Plasma oxidative stress markers (primary outcomes) were evaluated at rest and after WBV, and included thiobarbituric acid reactive substances (TBARS), iron reduction capacity (FRAP), superoxide dismutase antioxidant enzymes activity (SOD), and catalase (CAT). At rest, the FM group had higher TBARS (P<0.001) and FRAP (P<0.001), and lower CAT (P=0.005) compared to the CT. In the CT group, the WBV had no effect on TBARS (P=0.559) and FRAP (P=0.926), whereas it increased both SOD (P<0.001) and CAT (P<0.001). In the FM group, the WBV reduced TBARS (p <0.001), FRAP (P<0.001), and CAT (P=0.005), while it increased SOD (P=0.019). There was an interaction effect (moments vs groups) in the TBARS (effect size=1.34), FRAP (effect size=0.93), CAT (effect size=1.45), and SOD (effect size=1.44) (P<0.001). A single trial of WBV exercise improved all oxidant and antioxidant parameters towards a greater adaptation to the stress response in FM women.

This study aimed to verify if random regression models using linear splines (RRMLS) are suitable for identifying genetic parameters in multiple-breed populations and also to investigate whether an interaction exists between the breeding value (BV) of sires and their progeny breed group. Ten populations were simulated by crossing 2 breeds with distinct genetic variance and nonzero segregation variance. To obtain the genetic parameters, 2 models were used: a multiple-trait model (MULT), in which the trait was considered distinct when evaluated in each group (1/2P1 + 1/2P2, 5/8P1 + 3/8P2, and 3/4P1 + 1/4P2), and a RRMLS with the spline polynomial knots adjusted to these same groups. The genetic parameters estimated through MULT and RRMLS did not differ from the simulated values. The correlations between BV (simulated and estimated) of animals were high and varied from 0.74 to 0.76, which indicates the efficiency of using MULT and RRMLS for predicting BV. Using field data, the traits age at first calving (AFC), first lactation length (LL), and 305-d milk yield (MY-305) from a multiple-breed population of Holstein-Gyr cattle were analyzed. The BV of animals were modeled through RRMLS with 3, 5, and 7 knots, distributed in accordance with the fraction of Holstein breed in each progeny breed group. It was verified that RRMLS with 7 knots for adjusting mean trajectories and genetic effects, with homogeneous residual variance, best fit AFC and LL. For MY-305, the best fit for mean trajectory and genetic effects was the RRMLS with 5 knots and with homogeneous residual variance. The posterior means of heritability varied from 0.21 to 0.48, 0.21 to 0.38, and 0.10 to 0.33 for AFC, LL, and MY-305, respectively. Estimates from genetic parameters obtained by using RRMLS with field data showed that this model is a useful tool for genetic evaluations of populations formed by a great number of breed groups. An interaction occurred between the BV of sires and their progeny breed group, and the genetic parameters for AFC, LL, and MY-305 traits from a multiple-breed population depend on breed composition of the progeny from which the evaluations are based.

The aims of this study were 1) to characterize the intensity of the vibration stimulation in women diagnosed with fibromyalgia (FM) compared to a control group of healthy women (HW) matched by age and anthropometric parameters, and 2) to investigate the effect of a single session of whole body vibration (WBV) on inflammatory responses. Levels of adipokines, soluble tumor necrosis factor receptors (sTNFr1, sTNFr2), and brain-derived neurotrophic factor (BDNF) were determined by enzyme-linked immunosorbent assay. Oxygen consumption (VO2) was estimated by a portable gas analysis system, heart rate (HR) was measured using a HR monitor, and perceived exertion (RPE) was evaluated using the Borg scale of perceived exertion. Acutely mild WBV increased VO2 and HR similarly in both groups. There was an interaction (disease vs vibration) in RPE (P=0.0078), showing a higher RPE in FM compared to HW at rest, which further increased in FM after acute WBV, whereas it remained unchanged in HW. In addition, there was an interaction (disease vs vibration) in plasma levels of adiponectin (P=0.0001), sTNFR1 (P=0.000001), sTNFR2 (P=0.0052), leptin (P=0.0007), resistin (P=0.0166), and BDNF (P=0.0179). In conclusion, a single acute session of mild and short WBV can improve the inflammatory status in patients with FM, reaching values close to those of matched HW at their basal status. The neuroendocrine mechanism seems to be an exercise-induced modulation towards greater adaptation to stress response in these patients.

Aimed to verify if the autoregressive model (MAR) is adequate to obtain genetic parameters for Gyr dairy cattle milk yield on the test day in the three first lactations. Analysis was performed on 125,191 records of daily production of 9,242 cows using repeatability model (MREP) and MAR. On MREP, a long-term environment was considered, on MAR, the short-term environment was also taken into consideration. The models were compared by logarithm of the maximum likelihood function (−2logL) . The heritability estimated using the MREP model was 0.18, while the heritability estimated by MAR for first, second, and third lactations were 0.32, 0.28 and 0.26, respectively. The autocorrelation estimates of the components of long-term variance were close to zero, and those of the short-term were of high magnitude for first (0.79), second (0.79) and third (0.81) lactations. Therefore, the influence of the short-term environment within each lactation is not the same. The value of −2logL closer to zero was obtained for MAR (-294,884.7778) in relation to MREP (-329,266.4810). Thus, MAR is suitable for obtaining genetic parameters estimates for PLDC in the first three lactations of dairy cattle.(AU)

This study describes the osteology and radiology of the pelvic limb in maned wolves. Ten (five live and five dead) maned wolves (Chrysocyon brachyurus), five males and five females, aged from 2 to 7 years old were used. Digital radiographs were taken and recorded for both pelvic limbs in all animals. Osteology was correlated with the radiographic images. The pelvis had a rectangular shape, and the obturator foramen (foramen obturatum) was oval. The femoral neck (collum femoris) was short and thick. The greater trochanter (trochanter major) extended proximally to near the dorsum of the femoral head (caput ossis femoris). The lateral femoral condyle (condylus lateralis) was larger than the medial condyle (condylus medialis), and the intercondylar fossa (fossa intercondylaris) had a slightly oblique orientation. The proximal tibia displayed medial and lateral condyles with the medial larger. The femur was slightly shorter than the tibia. Seven tarsal bones (ossa tarsi) were present, four long metatarsal bones (ossa metatarsalia II - V) and a short first metatarsal bone (os metatarsal I).

Wild animal breeders are always looking for effective ways to care for their livestock. Intestinal parasites affect animals in captivity, reducing their survival and reproduction. This study aimed to evaluate the effectiveness of quicklime for use in soil disinfection and prevention of reinfection of endoparasites in paca (Cuniculus paca) kept in captivity. Soil samples of pacas' stalls were analyzed using the modified Rugai method. Two experimental stages were evaluated; the first at 5, 45 and 105 minutes after treatment and the second at 7 days and 14 days after treatment. The pH values of the samples were checked after the application of four treatments. An application of 400g/m2 of quicklime spread out on ground or 500mL/m2 of three lime solutions (10%, 20% or 40%, w / v) applied on ground. In the first stage, the lime spread out on soil was ineffective in parasites control, while the 10% solution provided the best control of parasites. In the second stage, the solutions were effective in raising the pH, but there was no significant difference in larval findings. In conclusion, under the conditions of the study, 10% solution increased the soil pH and had good ability to inactivate the eggs of Strongyloides sp.; that is economically interesting. However, for effective control of Strongyloides sp. in pacas kept captive, both the frequency of application of solutions up to 20% of lime and the adoption of a good worming schedule should be taken into consideration.(AU)

In the present study, we demonstrated the in vitro activity of endophytic phosphate-solubilizing bacteria (PSB). Fifty-five endophytic PSB that were isolated from sap, leaves, and roots of maize were tested for their ability to solubilize tricalcium phosphate and produce organic acid. Partial sequencing of the 16S rRNA-encoding gene showed that the isolates were from the genus Bacillus and different species of Enterobacteriaceae. The phosphate solubilization index on solid medium and phosphate solubilization in liquid medium varied significantly among the isolates. There was a statistically significant difference (P ≤ 0.05) for both, the values of phosphate-solubilizing activity and pH of the growth medium, among the isolates. Pearson correlation was statistically significant (P ≤ 0.05) between P-solubilization and pH (R = -0.38), and between the gluconic acid production and the lowering of the pH of the liquid medium at 6 (R = 0.28) and 9 days (R = 0.39). Gluconic acid production was prevalent in all the PSB studied, and Bacillus species were most efficient in solubilizing phosphate. This is the first report on the characterization of bacterial endophytes from maize and their use as potential biofertilizers. In addition, this may provide an alternative strategy for improving the phosphorus acquisition efficiency of crop plants in tropical soils.

There is an increasing interest in improving neurocysticercosis (NCC) diagnosis through the search of new and alternative antigenic sources, as those obtained from heterologous antigens. The aim of this study was to obtain potential biomarkers for NCC diagnosis after gel filtration chromatography [gel filtration fraction (GFF)] from the total saline extract (SE) from Taenia saginata metacestodes, followed by protein identification and application in immunodiagnostic. SE and GFF proteic profiles were characterized in gel electrophoresis, and diagnostic performance was verified by testing 160 serum samples through enzyme-linked immunosorbent assay and immunoblotting. Sensitivity (Se), specificity (Sp) and other diagnostic parameters were calculated. Polypeptides of interest in the diagnosis of human NCC present at GFF were analysed by mass spectrometry (MS) and B-cell epitopes were predicted. GFF had the best diagnostic parameters: Se 93·3%; Sp 93%; AUC 0·990; LR+ = 13·42 and LR- = 0·07, and proved to be useful reacting with serum samples in immunoblotting. Proteic profile ranged from 64 to 68 kDa and enolase and calcium binding protein calreticulin precursor were identified after MS. The enolase and calcium-binding protein calreticulin precursor showed 18 and 10 predicted B-cell epitopes, respectively. In conclusion we identified important markers in the GFF with high efficiency to diagnose NCC.

The aim of this study was defined the biometric values and quantify hematological parameters such as hematocrit, concentration of total plasma proteins and total leukocytes for Podocnemis expansa, we collect one milliliter of blood from the femoral vein of 60 specimens apparently healthy. The animals were collected from a private farm, Estancia Terra, located in Rio Branco in the state of Acre, and were classified into six groups of different weights. During the collection of biometric data, weight measurements were taken, and length and width of the carapace and plastron, for the analysis of correlation between the size parameters and body mass between individuals of the same group. The values observed indicate that the group 06, which includes animals with mass above 26kg, showed the best correlation between the individual masses of each animal (R = 0.97), while the group 02 (weight 21 to 25kg) showed the worst correlation between the mass (r = 0.34). The result of the overall average hematocrit of all animals of this experiment was 10.56%. The concentration of total plasma proteins was 3,69g DL-1 and the average total number of leukocytes was 51466,66µL. These parameters are considered abnormal when compared to the values found in healthy animals of the same species. This work contributes with information about biometric data and hematological parameters of P. expansa captivity and demonstrates the importance of monitoring health, preventing diseases that may endanger the health of all animals in the farm.(AU)
