A series of experiments was conducted to examine the effects of chemical pretreatment on biodegradation of 14C-labeled PCB congeners in aqueous systems. Fenton's reagent was used to generate hydroxyl radicals (OH) which were successful in partially oxidizing/transforming otherwise recalcitrant molecules of tetrachlorinated PCB, but had little or no impact on the biodegradation of a monochlorinated congener. Application of Fenton's reagent (1% H2O2, 1 mM FeSO4) followed by inoculation with pure cultures Pseudomonas sp, strain LB 400 and Alcaligenes eutrophus, strain H850 resulted in the removal of approximately 38% of 2-chlorobiphenyl and 51% of 2,2',4,4'-tetrachlorobiphenyl in the form of 14CO2. Comparison of the rate and extent of biodegradation of 2,2',4,4'-tetrachlorobiphenyl after the application of Fenton's reagent with the dynamic and final level of radioactivity in the aqueous phase of experimental system suggests two possible means of microbial utilization of tetrachlorinated PCB congener altered by chemical oxidation: (a) consumption of the partially oxidized chemical dissolved in the aqueous phase, and (b) direct microbial attack on the transformed compound, which may still be adhered to the solid surface.

In-situ bioremediation of benzene, toluene, ethylbenzene and the xylenes (BTEX) was carried out in an O2-poor (approx. 1 mg O2/l) fuel-contaminated aquifer. Extracted groundwater, enriched with ammonium polyphosphate (nutrients) and KNO3 (electron acceptor), was piped to an infiltration gallery over the contaminated site. Before, during and after infiltration, BTEX, nitrate and different populations of culturable bacteria were measured. BTEX declined by 78% in water from the monitoring well which was most contaminated initially and by nearly 99% in water from one of the extraction wells. These declines persisted after cessation of nutrient and nitrate addition. During the second half of the nutrient and nitrate addition period (weeks 107 to 160.5), nitrate appeared in the monitoring well, denitrifying bacteria increased about 50-fold and bacteria degrading benzene, toluene and xylenes (BTX) and phenanthrene (enumerated aerobically) increased 16- and 121-fold, respectively. At one of the extraction wells, down-gradient of the monitoring well, nitrate appeared in significant concentrations after week 124; this appearance coincided with a marked decline (> 90%) in BTEX concentration and 21- and 10-fold increases, respectively, in BTX- and phenanthrene-degrading bacteria. Low concentrations of BTEX and nitrate in down-gradient, off-site wells showed that water washing did not mobilize BTEX from the aquifer. The data indicate that the BTEX in this nitrate-enriched aquifer was biodegraded in-situ under denitrifying conditions.

Published reports of posttransplant myeloma are extremely uncommon (three cases); to the best of our knowledge there have been no reported cases in cardiac transplant recipients. We are also unaware of any report of Epstein-Barr virus (EBV) genome studies in posttransplant myeloma. We report here the case of a 48-year-old man who developed multiple myeloma 1.5 years after cardiac transplantation. The results of a serum analysis were consistent with past EBV infection. Biopsy of a skull lesion showed a monomorphous population of malignant immature plasma cells that showed monotypic cytoplasmic staining with antibodies to lambda light chains. A monoclonal immunoglobulin heavy chain gene rearrangement was detected by polymerase chain reaction (PCR). Both EBER-1 in situ hybridization and EBNA-1 PCR were negative for the EBV genome. Cyclosporin withdrawal was followed by transient clinical and biological improvement, but the tumor later progressed and eventually stabilized in response to treatment with dexamethasone alone. This case illustrates that posttransplant lymphoproliferative disorders (PTLPDs) encompass not only EBV-positive but also EBV-negative cases and not only lymphomas but also myelomas.

RESUMO

Dystocia is one of the leading causes of calf mortality between birth and weaning in beef cattle. An understanding of the effects of parturition and dystocia on perinatal calf viability is paramount for the development of breeding and calving programs that will minimize calf losses. This article discusses the factors affecting birth weight and replacement heifer development and redefines dystocia as any birth that reduces calf viability, causes maternal injury, or reduces maternal reproduction.

A photometrically-measured indirect fluorescent antibody (IFA) test was developed to measure antibody levels in three heifers vaccinated intramuscularly with two injections (two-week interval) of a vaccine containing formalin-killed Tritrichomonas foetus in oil adjuvant; a separate animal served as a nonvaccinated control. Sera were collected weekly and tested for specific IgG against T. foetus. Antibodies were detectable within two weeks of the initial injection and reached reciprocal titers as high as 7700 as estimated from previously tested reference sera. Titers peaked at six to eight weeks and remained at relatively high levels for the eleven-week study period. The fluorometric assay was easily developed and economical to perform; in addition, it more accurately estimated IgG levels than standard slide IFA tests.

During the fall of 1982, increased incidence of left displaced abomasum (LDA) was seen in a group of cows fed a complete pelleted ration (3/16 inch) at the university's dairy center during the early postpartum period. Of these, 17.4% developed LDA when fed this experimental ration, whereas only 1.6% of the remaining cows in the herd developed LDA. The latter cows (as a control) were fed loose alfalfa hay, sorghum silage, and an 18% crude protein mix. Most of the fiber in the experimental diet or pelleted diet was alfalfa hay (30% as-fed basis) that was ground through a quarter-inch screen for incorporation into the pellet. Another 10% of the dietary fiber was contributed by cottonseed hulls. Thus, increased incidence of LDA may be associated with short length of the dietary fiber in the pelleted form.

RESUMO

Pyloric adenocarcinoma was diagnosed in a ferret with clinical signs of gastric distention. The development of this tumor may be related to Helicobacter mustelae infection in the gastric mucosa of ferrets. Pyloric adenocarcinoma has only recently been reported in ferrets and should be considered as a differential diagnosis for gastric distention in this species.

The concept of feeding for optimal reproductive performance is not new. The effects of prepartum and postpartum nutritional levels on return to postpartum estrus and conception rates was reported in 1962. The effect of body condition regardless of nutrition regimen was first reported in 1975. Body condition scoring has become a popular topic for Extension and practitioner sponsored producer meetings. To effectively promote a management change that requires increased cash flow and does not have an immediate return requires demonstration of cost-effectiveness. This requires knowledge of the biologic information and transformation of that knowledge (through partial budget analysis) into additional profit for the producer. Controlling reproduction through proper nutrition at critical times of the reproductive cycle requires: 1. Calving and breeding seasons of 60 days. 2. Cows to be in moderate to good body condition at calving (BCS 5-7). 3. Cows to be slightly gaining at the beginning of the breeding season. 4. Cows to be bred to fertile bulls. 5. Evaluation and correction of body condition well in advance of the actual impact of BCS on reproductive function. 6. Producer to be prepared to initiate "alternate" plans (i.e., early weaning or 48 hour calf removal).

Does the system work? After having been on a controlled heifer replacement program for 3 to 5 years, the entire herd will develop a short calving season resulting in more weaned calf weight per cow exposed to breeding. Wiltbank has reported on the advantage of feeding heifers for a selected target weight at breeding. Two groups of Brahman-cross heifers were fed to weigh 600 or 700 lbs at the beginning of the breeding season. They were pastured and fed similarly from breeding through weaning their first calves. The heifers that weighted 700 lb at breeding weaned 86 lb more calf per heifer exposed. They bred earlier and more became pregnant. The return above feed costs after 2 years breeding was $52 greater for the heavier heifers. Obviously, the benefits continue through subsequent calvings, emphasizing the importance of managing the replacement heifers for 3 years, through their second breeding and calving.

Mature, pregnant Hereford cows (n = 17) were used to determine the effect of nutrition and body energy reserves on fetal development, concentrations of nutrients and estrogens in placental fluids, and on progesterone, estrogens and placental lactogen in maternal plasma. On d 145 of gestation, cows were assigned by breeding date to two groups and fed to achieve either a thin (TH; n = 8) or a moderate (M; n = 9) body condition score (BCS) by d 195 of gestation. Body weights, BCS, estrogens, placental lactogen and progesterone in plasma were determined weekly between d 200 and 256 of gestation. Cows were slaughtered on d 259 +/- 1 of gestation, and amnionic and allantoic fluids were sampled and analyzed for concentrations of protein, fructose and estrogens. Body weights and BCS were less (P less than .01) for TH (419 kg; 3.7) than for M (511 kg; 5.7) cows at slaughter. Uterine weights were less (P less than .07), but chorioallantoic weights were greater (P less than .07) in TH than in M cows. Cotyledonary weights were greater (P less than .05) for TH than for M cows, and total fructose in amnionic fluid was reduced (P less than .01) in TH compared with M cows. Concentrations of estradiol, estrone and placental lactogen were greater between d 240 and 256 of gestation for TH than for M cows. We conclude that nutrient intake and(or) BCS of beef cows during late gestation influence placental weight, fructose in amnionic fluid, and placental lactogen, estrone and estradiol in plasma.

Ten of 100 mature ewes were afflicted with acute oxalate toxicosis within 40 hours after being temporarily penned in a lot that contained considerable growing Rumex crispus (curly dock). Clinical signs of toxicosis included excess salivation, tremors, ataxia, and recumbency. Affected ewes were markedly hypocalcemic and azotemic. Oxalate crystals were not observed in urine. Gross postmortem lesions were minimal and nondiagnostic in 2 ewes that died peracutely, but perirenal edema and renal tubular degeneration were clearly observable in ewes euthanatized on the third day of toxicosis. Diagnosis of oxalate toxicosis was confirmed by histopathologic findings. Samples of Rumex spp contained 6.6 to 11.1% oxalic acid on a dry-weight basis, a concentration comparable with that in other oxalate-containing plants that have caused acute oxalate toxicosis.

The fetal animal undergoes a tremendous transition from intrauterine to extrauterine life at parturition. In this article, the maternal-fetal interactions of parturition are discussed with the aim of examining the normal stress reactions of parturition. Dystocia is discussed from the standpoint of additional distress of the newborn, with an aim toward the development of rational therapeutic support.

One hundred seventy-two bulls with preputial injuries were treated from 1980 through 1985. Most of the injuries developed during the breeding season (April through August), with a peak during June. The number (111) of bulls represented by 3 beef breeds (Brangus, Brahman, Beefmaster) was greater than the number (61) of the other 9 breeds included in the survey. Preputial prolapse (PP) and posthitis without obvious laceration was diagnosed in 82 of 172 bulls. Preputial laceration, abscessation, stricture with phimosis, and PP with penile hematoma were diagnosed in 46, 22, 15, and 9 bulls, respectively. These abnormalities were treated medically and/or surgically. Questionnaires (n = 145) were mailed, and 83 (57.4%) replies were received. Owners (80/83) of affected bulls responded to questions regarding the future use of their bulls for breeding. Of the bulls with PP and posthitis (n = 34), laceration (n = 17), abscessation (n = 15), stricture with phimosis (n = 8) and PP with penile hematoma (n = 6) and treated medically and/or surgically, 30, 11, 7, 7, and 4 bulls, respectively, (74% overall) were returned to breeding use successfully.

Estrous response, pregnancy rate, and the relationship between palpable corpora lutea (CL) and plasma progesterone concentrations were studied during the spring (n = 149) and fall (n = 146) breeding seasons using postpartum, crossbred beef cows consisting of 0-Brahman, 1 4 - Brahman , or 1 2 Brahman breeding. At the start of each breeding season, a jugular blood sample was collected for progesterone analysis; each cow was palpated for the presence of a CL and randomly alloted within breed to a non-treated control or Synchro-Mate-B (SMB) treatment. Fewer (P<0.01) fall than spring bred cows exhibited estrus within the first 21 d of breeding. Within 48 h of implant removal, 84 and 64% (spring and fall, respectively) of synchronized cows exhibited estrus. In both seasons, more SMB than control cows became pregnant by 7 d of breeding. Percentage of Brahman breeding did not influence estrous response or pregnancy rate. On the basis of 292 evaluations, palpation per rectum correctly assessed ovarian status 74% of the time. These data indicate that although season can influence response. SMB can be used effectively to synchronize Brahman crossbred cows. In addition, it was found that there is a close relationship between palpation per rectum and plasma concentrations of progesterone.

In the winter of 1983, practitioners reported extensive photosensitization in 7 herds of cattle. All herds had a history of having been fed water-damaged alfalfa hay. A cow from one herd was referred to the veterinary teaching hospital at Oklahoma State University. In this herd of approximately 40 adult Polled Herefords, all cattle had had some degree of clinical involvement over the past 4 to 6 weeks. Clinical signs included scaling and erythema of sparsely haired skin, muzzle, and teats, as well as icterus, anorexia, and weight loss. One cow died, and the remaining cattle recovered over an 8- to 10-week period after removal of the hay from the ration. In the referred cow, values for total and conjugated bilirubin, BUN, creatinine, sorbitol dehydrogenase, serum alkaline phosphatase, serum aspartate transaminase, and serum gamma-glutamyl transferase were higher than normal. In the herd of origin, extremely high serum gamma-glutamyl transferase values (180 to 1,400 IU/L) persisted (normal, 2 to 35 IU/L). Feeding the same alfalfa hay to 2 clinically normal cows reproduced the syndrome. The characteristic hepatic lesion was bile duct necrosis, with secondary bile duct hyperplasia.

Clenbuterol is a highly specific, long-acting (four to eight hours) beta-two sympathomimetic which causes bronchodilation and tocolysis (myometrial paralysis). The tocolytic effect was explored as a means to control parturition and reduce dystocia. Forty-six heifers were injected i.m. with either Clenbuterol or saline placebo in a randomly controlled experiment. Animals were treated when a cervical dilation of five centimeters or more was detected by vaginal examination. Length of first, second and third stages of parturition, ease of parturition, maternal pelvic area and calf viability were compared between treatment groups. Treatment with Clenbuterol increased (P<0.025; 119 vs 468 mins) the time heifers were in Stage I. However, the lengths of Stages II and III, pelvic area at birth and calf viability were not influenced by treatment. Diameter of the cervix at treatment was negatively related to the length of Stage I delay. Pelvic area also significantly affected the length of Stage II. Clenbuterol effectively delays Stage I of parturition with no adverse effects on the fetus or dam.

Complement levels were determined in 12 normal human volunteers while receiving intravenous infusions of inulin for determination of standard inulin clearances. Significant decreases in beta IC globulin and total hemolytic complement activity were observed when inulin was infused, but not following infusion of saline as a control. These effects were noted only when steady-state inulin levels were greater than 26 mg%, suggesting a dose-dependent response. No evidence of increased coagulation was noted as measured by fibrinogen and fibrin degradation products. Since C4 levels remained largely unchanged following inulin infusion, it was concluded that the results most likely occurred through activation of the alternate complement pathway.

The effects of 4-pentenoic acid (4-PA) on renal excretion of phosphate and calcium were studied in anesthetized mongrel dogs. The major metabolic action of 4-PA is inhibition of long-chain fatty acid oxidation. In intact dogs undergoing modest saline diuresis, 4-PA caused significant decrease in the percentage of filtered phosphate reabsorbed. No statistically significant calciuria was observed. Similar results were observed in intact dogs undergoing more brisk saline diuresis as well as in acutely and chronically thyroparathyroidectomized dogs. However, when results were pooled, both phosphate and calcium excretions were significantly increased. The results are interpreted as demonstrating the importance of fatty acid oxidation in providing energy for the work of tubular phosphate reabsorption. Either the energy source for calcium reabsorption differs from fatty acid oxodation, or calcium reabsorption is less sensitive to changes in energy sources. The action of 4-PA is independent of the forces involved in saline diuresis and of parathyroid hormone.

Renovascular hypertension developed in an anephric 37-year-old patient after he received a cadaveric renal transplant from a 2-year-old donor. Despite adequate homograft function, a transplant nephrectomy was perfomed because of intractable, lifethreatening hypertension. There was relative stenosis throughout the course of the transplanted renal artery. Pathologic examination of the kidney did not demonstrate evidence of technical failure or immunological or hypertensive damage. Atrophic changes in the media of the renal artery may have resulted from radiation damage. The hypertension appears to have been caused by disproportionate growth between the parenchyma in the hypertrophying pediatric homograft and its renal artery.
