O Brasil vive uma transição demográfica que favorece o aparecimento e desenvolvimento de doenças crônicas não transmissíveis (DCNT). Em um cenário em que as DCNT estão entre as principais causas de mortes, reforçar a importância de uma alimentação saudável é imprescindível, e medidas preventivas tomam lugar de destaque. Pensando nisso, foram realizadas 11 intervenções de educação alimentar e nutricional em um grupo de atividade física intergeracional, com o objetivo de promover autonomia alimentar. O estudo contou com 25 participantes, na faixa etária de 37 a 83 anos, sendo a maior incidência de 61 a 69 anos (n=12) e predominância de pessoas do gênero feminino (92%). Os parâmetros para avaliar o grupo foram os feedbacks das(os) participantes e o grau de adesão, no qual houve aumento significativo ao longo do período interventivo. Destaca-se a importância de ampliar a abordagem dos hábitos alimentares saudáveis em diversos ambientes, sendo os grupos de atividades físicas um campo fértil, que requer a adaptação da linguagem e formato das atividades, considerando o nível de escolaridade das(os) participantes. Assim, a educação alimentar e nutricional pode ser usada como ferramenta efetiva para a promoção da saúde, a prevenção e o tratamento de DCNT.


Brazil is undergoing a demographic transition that favors the onset and development of chronic non-communicable diseases (NCDs). In a scenario where NCDs feature among the main causes of death, reinforcing the importance of healthy eating is essential and preventive measures should take place. Consequently, eleven Food and Nutritional Education interventions were conducted with an intergenerational physical activity group to promote food autonomy. A total of 25 individuals aged 37 to 83 years participated in the study, with the highest prevalence of 61 to 69 years (n=12) and women (92%). Evaluation parameters were participant feedback and the degree of adherence, which increased significantly over the interventional period. Results highlight the importance of broadening the approach to healthy eating habits in various environments, with physical activity groups being a fertile setting which requires adapting language and the format of activities according to participants' schooling level. Nutritional Food Education can be used as an effective tool for health promotion, prevention and treatment of NCDs.


Brasil vive una transición demográfica que favorece la aparición y desarrollo de enfermedades crónicas no transmisibles (ECNT). En un escenario en que entre las principales causas de muerte están las ECNT, se hace imprescindible reforzar la importancia de una alimentación saludable; para ello se destacan las medidas preventivas. Así, se realizaron 11 intervenciones de educación alimentaria y nutricional en un grupo de actividad física intergeneracional, con el objetivo de promover la autonomía alimentaria. Este estudio contó con una muestra de 25 participantes, en la franja etaria de 37 a 83 años, siendo la mayor prevalencia de los de entre 61 y 69 años (n=12), y predominancia de personas del género femenino (92%). Los parámetros que evalúan al grupo fueron las respuestas de las(los) participantes y el grado de adhesión, en el cual hubo un aumento significativo a lo largo de la intervención. Se destaca la importancia de ampliar el foco en los hábitos alimentarios saludables en diversos ambientes, y los grupos de actividades físicas son un campo fértil que requiere la adaptación del lenguaje y el formato de las actividades, considerando el nivel de escolaridad de las(los) participantes. Así, la educación alimentaria nutricional puede ser utilizada como herramienta efectiva para la promoción de la salud, la prevención y el tratamiento de las ECNT.

This study evaluated the effects of probiotics (PRO) as an adjuvant to the treatment of experimental periodontitis (EP) in rats immunosuppressed with 5FU.108 rats were randomly allocated to six different groups: EP; SS - systemic treatment with saline solution (SS); 5FU - systemic treatment with 5FU; 5FU+PRO - systemic treatment with 5FU, followed by the local administration of Saccharomyces cerevisiae ; 5FU+SRP - systemic treatment with 5-FU, followed by scaling and root planing (SRP); and 5FU+SRP+PRO - systemic treatment with 5FU followed by local treatments with SRP and PRO. Immunosuppression was obtained at two points: at the time of ligature installation and after 48 h. Six animals from each group were euthanized at seven, 15, and 30 d and hemimandibles were collected and processed for histopathological, histometric, and immunohistochemical analysis. Data were subjected to statistical analysis (α=5%). At 7 d, the 5FU+PRO group showed less bone resorption and better structured connective tissue compared with the EP, SS, 5FU+SRP, and 5FU+SRP+PRO groups. At 15 d, the 5FU+SRP group showed a greater intensity of the inflammatory response (p<0.05). At 30 d, the 5FU+SRP+PRO group showed better structured bone tissue and a higher percentage of bone tissue (PBT) than the EP, SS, 5FU, and 5FU+PRO groups (p<0.05). The use of Saccharomyces cerevisiae as monotherapy or as an adjuvant to periodontal therapy may have a positive effect on bone repair in immunosuppressed conditions.

Cancer remains a leading cause of death worldwide, partly owing to late detection which entails limited and often ineffective therapeutic options. Most cancers lack validated screening procedures, and the ones available disclose several drawbacks, leading to low patient compliance and unnecessary workups, adding up the costs to healthcare systems. Hence, there is a great need for innovative, accurate, and minimally invasive tools for early cancer detection. In recent years, multi-cancer early detection (MCED) tests emerged as a promising screening tool, combining molecular analysis of tumor-related markers present in body fluids with artificial intelligence to simultaneously detect a variety of cancers and further discriminate the underlying cancer type. Herein, we aim to provide a highlight of the variety of strategies currently under development concerning MCED, as well as the major factors which are preventing clinical implementation. Although MCED tests depict great potential for clinical application, large-scale clinical validation studies are still lacking.

Objective: The aim of this study was to evaluate the effects of probiotic (PRO) Lactobacillus reuteri (DSM17938) as an adjuvant to the treatment of experimental periodontitis (EP). Material and methods: Fifty-four male adult Wistar rats were included. EP was induced and maintained for 7 days. Subsequently, the ligature was removed and the animals were allocated into three different experimental groups (n = 18/group): EP - no local treatment, the animals received four systemic saline solution (SS) administrations; SRP+SS, the animals underwent SRP treatment, followed by SS administration; and SRP+PRO, the animals received SRP treatment, followed by the systemic administration of PROs (Lactobacillus reuteri ; 0.16 ml/day). Six animals from each group were euthanised at 7, 15 and 30 days. Histological and histometric analyses of alveolar bone loss (BL) and immunohistochemical analyses for TRAP, RANKL, OPG, OCN, and PCNA were performed. Shapiro-Wilk, ANOVA, post-hoc Tukey, Kruskal-Wallis, Student-Newman Keuls were performed. Results: The SRP+PRO group presented a reduction in inflammation. At 15 days, a lower BL was observed in the SRP+SS and SRP+PRO groups. Greater immunolabeling was noticed for PCNA at 15 days in the SRP+PRO group than in the SRP +SS group. The SRP+PRO group demonstrated a higher OCN immunolabeling pattern than the EP group at 15 and 30 days. Conclusion: The use of Lactobacillus reuteri as an adjuvant to SRP for the treatment of EP showed promising results in the control of local inflammatory responses, and enhanced the periodontal tissue repair process according to the employed concentration.

Testicular germ cell tumors (TGCTs) are the most common cancers in young-adult male patients aged between 15 and 39 years. Hsa-miR-371a-3p is currently the most reliable biomarker for diagnosis and monitoring of these patients non-invasively in liquid biopsies, and it is destined to be introduced in the clinic due to improved performance compared to the classical serum tumor markers available. Current studies have focused on real-time quantitative PCR (RT-qPCR) protocols for its determination; still, some challenges remain, since these protocols often require preamplification steps (costly and time-consuming), and report relative levels normalized to a housekeeping microRNA, not always performed the same way. Droplet digital PCR (ddPCR) shows the promise to overcome these challenges, skipping normalization and preamplifications, but has hardly been explored in the field of TGCTs. In this work, we provide a report of a ddPCR-based pipeline for the quantification of hsa-miR-371a-3p (the DigiMir pipeline) and compare it with two RT-qPCR protocols. A total of 107 plasma samples were investigated in the validation setting. The DigiMir pipeline detected TGCTs in a manner representative of tumor burden, with a sensitivity and specificity of 94% and 100%, respectively, outperforming the combined sensitivity of all three classical serum tumor markers (61.5%). Therefore, in this proof-of-concept investigation, we have shown that the DigiMir pipeline constitutes a new promising methodology to accurately report hsa-miR-371a-3p in the clinical setting.

BACKGROUND: The repair of bone defects has been the subject of many studies that have shown inconclusive results as to what is the best bone substitute. MATERIAL AND METHODS: Bone defects (Ø 2 mm) were induced on the tibia of seventy-two rats, which were distributed into the following four groups/treatments (n=18 each): Control: no treatment; EMD: enamel matrix derived protein; PBM: photobiomodulation therapy (660 nm, 0,035 W, 60 s); EMD + PBM: EMD and immediate treatment with PBM (660 nm, 0,035 W, 60 s). Six animals from each group were euthanized after 10, 30 and 60 days. Histological and immunohistochemistry analyses (osteocalcin - OCN and tartrate-resistant acid phosphatase - TRAP) were performed with scores for each of the biological events. RESULTS: All performed treatments resulted in an increased filling and maturation of bone tissue, being greater in the EMD and EMD + PBM groups in the 30 day period, compared to the Control group. The immunostaining of OCN was greater at 60 days in all treated groups than in the Control over the same period. TRAP immunostaining was higher at 30 days in all treated groups, and lower in groups EMD and PBM after 60 days, compared to the Control over the same period. There was greater immunostaining in the EMD + PBM group after 10 days than in the Control and EMD groups in the same period. CONCLUSIONS: These results lead to the conclusion that treatments with EMD and PBM, both separate and in association were effective in filling and maturing bone tissue in tibial bone cavities, with greater effectiveness in the period of 30 days in the EMD and EMD + PBM groups. O presente relato de experiência apresenta uma vivência ocorrida durante um estágio básico em um Centro de Atenção Psicossocial (CAPS), realizado de forma online por três graduandas do curso de Psicologia de uma Universidade do interior de Minas Gerais. Buscou-se observar e descrever como ocorreram as práticas realizadas no Grupo Operativo do CAPS em meio à pandemia da Covid-19, dialogando com a técnica do Grupo Operativo postulada por Pichon-Revière. Para tanto, as estagiárias participaram de dez encontros via Google Meet, com duração média de uma hora, com o intuito de observar e coordenar alguns encontros nos dias combinados com a equipe técnica. Esse relato tem como objetivo compartilhar e discutir as vivências realizadas com os usuários do CAPS de modo remoto. Foram apresentadas características observadas nos encontros online, algumas peculiaridades do manejo do atendimento frente às demandas, bem como os alcances e limites advindos desse novo modelo. Através desse relato, percebe-se que o contato virtual permitiu a conquista de novas aprendizagens e mudanças pelos participantes, além de reflexões sobre as dinâmicas do funcionamento grupal vivenciadas.


The present experience report presents an experience that took place during a basic internship in a Psychosocial Care Center (CAPS), carried out online by three undergraduate students of the Psychology of a University in the interior of Minas Gerais. We sought to observe and describe how the practices carried out in the CAPS Operative Group occurred in the midst of the Covid-19 pandemic, dialoguing with the Operative Group technique postulated by Pichon-Revière. To this end, the interns participated in ten meetings via Google Meet, with an average duration of one hour, in order to observe and coordinate some meetings on the days agreed with the technical team. This report aims to share and discuss the experiences carried out with CAPS users within the remote model. Characteristics observed in online meetings were presented, as well as some peculiarities of the handling of care in the face of demands, as well as the scope and limits arising from this new model. Through this report, it is clear that the virtual contact allowed the conquest of new learning and changes by the participants, as well as reflections on the dynamics of group functioning experienced.


El presente relato de experiencia presenta una experiencia que tuvo lugar durante una pasantía básica en un Centro de Atención Psicosocial (CAPS), realizada en línea por tres estudiantes de pregrado de la Psicología de una Universidad del interior de Minas Gerais. Buscamos observar y describir cómo ocurrieron las prácticas realizadas en el Grupo Operativo CAPS en medio de la pandemia de la Covid-19, dialogando con la técnica del Grupo Operativo postulada por Pichon-Revière. Para ello, los becarios participaron en diez reuniones vía Google Meet, con una duración promedio de una hora, con el fin de observar y coordinar algunas reuniones en los días acordados con el equipo técnico. Este informe tiene como objetivo compartir y discutir las experiencias realizadas con los usuarios de CAPS dentro del modelo remoto. Se presentaron las características observadas en los encuentros en línea, así como algunas peculiaridades del manejo del cuidado frente a las demandas, así como los alcances y límites derivados de este nuevo modelo. A través de este relato, queda claro que el contacto virtual permitió la conquista de nuevos aprendizajes y cambios por parte de los participantes, así como reflexiones sobre la dinámica de funcionamiento grupal vivida.

Este estudo tem por objetivo investigar quais os desafios da elaboração de uma demanda subjetiva para a clínica de pacientes com diagnóstico de fibromialgia, e parte da observação de que esses sujeitos frequentemente chegam à clínica psicanalítica por encaminhamento, marcados pelo discurso hegemônico biomédico e por queixas somáticas. Para tanto, seis mulheres com diagnóstico de fibromialgia foram entrevistadas. Os dados foram analisados segundo as diretrizes da análise de conteúdo temática, a partir do referencial teórico da psicanálise. A discussão foi dividida em três eixos temáticos: (I) dor e demanda; (II) fibromialgia e fenômeno psicossomático; e (III) o sujeito entre o discurso científico e a dor. Como resultado, notou-se a existência de uma dificuldade na elaboração de uma demanda para o acompanhamento psicológico no caso da fibromialgia. Essa dificuldade pode ser abordada pelo rechaço simbólico apresentado nos casos de dor crônica, que parece não articular um sintoma analítico ou elaboração psíquica. Ademais, ficou evidente a alienação das participantes ao discurso científico, o que as priva de um saber acerca do próprio sofrimento.


This study aims to investigate the challenges of developing a subjective demand for the clinic of patients diagnosed with fibromyalgia, starting with the observation that these subjects frequently arrive at the psychoanalytic clinic by referral, marked by biomedical hegemonic discourse and somatic complaints. To do so, six women diagnosed fibromyalgia were interviewed. Data were analyzed according to thematic content analysis guidelines, based on the psychoanalysis theoretical framework. The discussion was divided into three thematic axes: (I) pain and demand; (II) fibromyalgia and psychosomatic phenomenon; and (III) the subject between scientific discourse and pain. As a result, it was noticed that there is a difficulty in elaborating a demand for psychological care in the case of fibromyalgia. This difficulty can be addressed by the symbolic rejection that is noted in cases of chronic pain, which does not seem to articulate an analytic symptom or psychic elaboration. In addition, the alienation of the participants from the scientific discourse became evident, which deprives them of knowledge about their own suffering.


Este estudio tiene como objetivo investigar los desafíos del desarrollo de una demanda subjetiva para la clínica de pacientes diagnosticados de fibromialgia, y parte de la observación de que estos sujetos frecuentemente llegan a la clínica psicoanalítica por derivación, marcados pelo discurso hegemónico biomédico y quejas somáticas. Por lo tanto, se entrevistó a seis mujeres diagnosticadas con fibromialgia. Los datos se analizaron de acuerdo con las directrices de análisis del contenido temático, basadas en el referencial teórico del psicoanálisis. La discusión se dividió en tres ejes temáticos: (I) dolor y demanda; (II) fibromialgia y fenómeno psicosomático; y (III) el sujeto entre el discurso científico y el dolor. Como resultado, se notó que hay una dificultad en el desarrollo de una demanda de asistencia psicológica en el caso de la fibromialgia. Esta dificultad puede abordarse mediante el rechazo simbólico que aparece en los casos de dolor crónico, que no parece articular un síntoma analítico o elaboración psíquica. Además de eso, era evidente que los participantes estaban alienados del discurso científico, lo que les priva del conocimiento de su propio sufrimiento.

Após a cirurgia para câncer de mama, algumas complicações podem levar a restrições da amplitude de movimento (ADM), dor e impacto negativo na realização das tarefas de vida diária. Neste contexto, a fisioterapia é importante na recuperação da funcionalidade. Objetivo: Avaliar o efeito da fisioterapia na amplitude de movimento, na intensidade de dor e no desempenho funcional do membro superior após a cirurgia para câncer de mama de mulheres no menacme. Métodos: Ensaio clínico não controlado, considerando mulheres que realizaram cirurgia para câncer de mama associada a linfonodectomia axilar. Foram avaliados a ADM pela fleximetria, intensidade de dor pela escala visual analógica (EVA) e o desempenho funcional pelo questionário "deficiência do ombro braço e mão" (DASH). Resultados: Após as 10 sessões de fisioterapia, foi observado aumento significativo da ADM de todos os movimentos, diminuição da intensidade de dor de 4,09 ± 2,51) para 2,54 ± 2,18, (p = 0,04), e dos escores do DASH de 35,31 ± 17,23 para 17,75 ± 13,09, (p = 0,001), indicando significativa melhora do desempenho funcional. Conclusão: A abordagem fisioterapêutica foi satisfatória em melhorar a ADM, intensidade de dor e o desempenho funcional em mulheres na pré-menopausa. Contudo, acompanhamento por mais longo prazo e incrementar os exercícios podem trazer melhora adicional. (AU)

With the fast increase in the demand for location-based services and the proliferation of smartphones, the topic of indoor localization is attracting great interest. In indoor environments, users' performed activities carry useful semantic information. These activities can then be used by indoor localization systems to confirm users' current relative locations in a building. In this paper, we propose a deep-learning model based on a Convolutional Long Short-Term Memory (ConvLSTM) network to classify human activities within the indoor localization scenario using smartphone inertial sensor data. Results show that the proposed human activity recognition (HAR) model accurately identifies nine types of activities: not moving, walking, running, going up in an elevator, going down in an elevator, walking upstairs, walking downstairs, or going up and down a ramp. Moreover, predicted human activities were integrated within an existing indoor positioning system and evaluated in a multi-story building across several testing routes, with an average positioning error of 2.4 m. The results show that the inclusion of human activity information can reduce the overall localization error of the system and actively contribute to the better identification of floor transitions within a building. The conducted experiments demonstrated promising results and verified the effectiveness of using human activity-related information for indoor localization.

OBJECTIVE: To analyze whether maternal age at first pregnancy and parity are mediators of the association between early menarche and metabolic syndrome in a sample of middle-aged and older women. METHODS: Cross-sectional study of 428 women (40 to 80 y), who had experienced a pregnancy in their lifetime, was performed between 2014 and 2016. Age at first pregnancy, parity, and early menarche were self-reported. Metabolic syndrome was assessed using the criteria described by the National Cholesterol Education Program's Adult Treatment Panel III. The association between metabolic syndrome and early menarche was assessed by logistic regression analysis. The mediating role of age at first pregnancy and multiparity in the relationship between early menarche and metabolic syndrome was assessed through mediation analysis, adjusted for covariates. RESULTS: According to adjusted logistic regression models, early menarche was associated with higher odds of prevalent metabolic syndrome (OR: 2.26; 95% CI: 1.15-4.46). Mediation analysis showed a significant direct effect of early menarche on metabolic syndrome (ß: 0.808; 95% CI: 0.107-1.508). Of the two mediators tested, age at first pregnancy was significant (ß: 0.065; 95% CI: 0.004-0.221), ie, participants with and without early menarche differ, on average, by 0.879 SDs in the log odds of MetS (total effect), of which 0.065 SDs (8%), on average, would be attributable to the effect of early menarche on age at first pregnancy (indirect effect), which, in turn, affects MetS. CONCLUSIONS: Age at first pregnancy may partially contribute to the association between early menarche and metabolic syndrome among middle-aged and older women who had experienced a pregnancy over their lifetime.

(1) Background: Stretching is known to improve range of motion (ROM), and evidence has suggested that strength training (ST) is effective too. However, it is unclear whether its efficacy is comparable to stretching. The goal was to systematically review and meta-analyze randomized controlled trials (RCTs) assessing the effects of ST and stretching on ROM (INPLASY 10.37766/inplasy2020.9.0098). (2) Methods: Cochrane Library, EBSCO, PubMed, Scielo, Scopus, and Web of Science were consulted in October 2020 and updated in March 2021, followed by search within reference lists and expert suggestions (no constraints on language or year). Eligibility criteria: (P) Humans of any condition; (I) ST interventions; (C) stretching (O) ROM; (S) supervised RCTs. (3) Results: Eleven articles (n = 452 participants) were included. Pooled data showed no differences between ST and stretching on ROM (ES = -0.22; 95% CI = -0.55 to 0.12; p = 0.206). Sub-group analyses based on risk of bias, active vs. passive ROM, and movement-per-joint analyses showed no between-protocol differences in ROM gains. (4) Conclusions: ST and stretching were not different in their effects on ROM, but the studies were highly heterogeneous in terms of design, protocols and populations, and so further research is warranted. However, the qualitative effects of all the studies were quite homogeneous.

RESUMO A maioria das atuais demandas em saúde mental são atravessadas por questões provenientes do movimento da Reforma Psiquiátrica, que teve como objetivo o rompimento de modelos manicomiais no tratamento de sujeitos em sofrimento psíquico. Sendo assim, o presente estudo buscou investigar sujeitos em tratamento em um Centro de Atenção Psicossocial (CAPS), seus modos de relacionamento familiar e a importância das mesmas durante o acompanhamento nestes dispositivos de assistência diante os desafios impostos pelo processo de desinstitucionalização. Para tanto foram realizadas sete visitas semanais em um CAPS situado em uma cidade do interior de Minas Gerais, no período entre março e junho de 2018. A partir das observações realizadas, foram levantadas questões como: a importância da família no acompanhamento terapêutico, as mudanças na dinâmica familiar diante do adoecimento, o desenvolvimento de autonomia e situações de violência. Dentre as diversas temáticas abordadas nessa experiência, destaca-se a importância dada ao CAPS e aos vínculos familiares pelos usuários na reconstrução de sua autonomia, assim como os encargos subjetivos gerados pelas demandas de cuidado.


ABSTRACT Most of the current demands on mental health are issues that once arose from the Psychiatric Reform movement, which aimed to discontinue the mental asylum model in the treatment of patients suffering psychiatrically. Thus, the present research has sought to investigate patients treated at a CAPS (Centros de Atenção Psicossocial), their family relationships and the importance of them during the follow-up in these care centers faced with the challenges caused by the deinstitutionalization. Therefore, weekly visits were made at a CAPS located in a small town of Minas Gerais. From the observations made, questions were raised such as the importance of the family in the therapeutic support, the changes in family dynamics in the face of illness, development of selfreliance, violence occurrences with confinement suffered inside psychiatric wards, and power relations. Among the different themes addressed in this experiment, the importance given to the CAPS and family bonds by the users in the restoration of their autonomy stands out, as well the subjective burdens generated by the demands of care.


RESUMEN La mayoría de las actuales demandas en salud mental son atravesadas por cuestiones provenientes del movimiento de la Reforma Psiquiátrica, que tuvo como objetivo el ruptura de los modelos manicomiales en el tratamiento de sujetos con sufrimiento psíquico. Por esta razón, el presente estudio busca investigar cómo los sujetos en tratamiento en un Centro de Atención Psicosocial (CAPS), sus modos de relaciones familiares y la importancia de las mismas durante el seguimiento en estos dispositivos de asistencia ante los desafíos impuestos causadas por la desinstitucionalización. Por lo tanto, se realizaron visitas semanales en un CAPS situado en una ciudad en el interior de Minas Gerais. A partir de las observaciones realizadas, se plantearon cuestiones como la importancia de la familia en el acompañamiento terapéutico, desarrollo de autonomía, los cambios en la dinámica familiar ante la enfermedad, situaciones de violencia y reclusión sufridas dentro de hospitales psiquiátricos y relaciones de poder. Entre las diversas temáticas abordadas en esta experiencia se destaca la importancia dada al CAPS y los vínculos familiares por los usuarios en la reconstrucción de su autonomia, así como las cargas subjetivas generadas por las demandas de cuidado.

AIM: Curcumin (CUR) has been used clinically in several studies as a subgingival irrigant or as a photoantimicrobial in combination with a blue light-emitting diode (LED) in antimicrobial photodynamic therapy (aPDT) adjuvant to scaling and root planing (SRP). The aim of this study was to assess the effectiveness of CUR as an irrigant or as a photoantimicrobial in conjunction with the blue LED in aPDT adjuvant to SRP, compared to SRP as conventional mechanical treatment. MATERIALS AND METHODS: Fifteen randomized controlled trials (RCT) were included in a qualitative analysis after researching the databases: PubMed / MEDLINE, SCOPUS, EMBASE, Cochrane Central, Web of Science and Scielo. Manual searches were also performed. Five studies were submitted to quantitative analysis, evaluating periodontal clinical parameters such as probing depth (PD) and clinical attachment level (CAL). RESULTS: The obtained results have shown clinical benefits in PD reduction and CAL gains at 3 months with the use of CUR as adjuvant therapy to SRP, both as an irrigant or photoantimicrobial, in comparison with SRP monotherapy. CONCLUSION: Currently, there is evidence that treatment with CUR applied as irrigant or in conjunction with the blue LED as aPDT presents superior clinical results in the short term, for clinical periodontics parameters like as PD reduction and CAL gain, when compared to SRP monotherapy in the non-surgical treatment of periodontitis. However, these results cannot be proven in the long term.

AIM: The aim of the present study was to compare the effects of the phenothiazine photosensitizers methylene blue (MB), toluidine blue-O (TBO) and butyl toluidine blue (BuTB) in antimicrobial photodynamic therapy (aPDT), as adjuvant therapy to scaling and root planing (SRP) in the treatment of experimental periodontitis (EP) in rats. MATERIAL AND METHODS: 120 Wistar rats underwent ligation around the lower left molar. After seven days, the ligature was removed. The animals were separated into the following groups (n = 15): EP, no treatment; SRP, SRP and irrigation with saline solution; MB, SRP and deposition of MB; TBO, SRP and deposition of TBO; BuTB, SRP and deposition of BuTB; MB-aPDT, SRP and aPDT with MB; TBO-aPDT, SRP and aPDT with TBO and; BuTB-aPDT, SRP and aPDT with BuTB. The aPDT session was performed after SRP, with deposition of the photosensitizer and irradiation with a diode laser (DL; InGaAlP, 660 nm, 40 mW, 60 s, 2.4 J). Histological and histometric analysis was performed. RESULTS: BuTB-aPDT group had a lesser extent of the inflammatory process compared to the EP, SRP, MB and TBO at all experimental periods (p < 0.05). At 15 days, the aPDT treated groups had a greater bone tissue structure than groups EP and SRP (p < 0.05) The BuTB showed lower Alveolar Bone Loss (ABL) compared to the TBO-aPDT group at 30 days (p < 0.05). CONCLUSION: aPDT using the photosensitizer BuTB proved to be the adjuvant therapy that most favored the reduction of inflammatory infiltrate in the furcation area and ABL.

IMPORTANCE: Menopause at younger ages is associated with a greater risk of adverse health outcomes such as osteoporosis, chronic diseases, and death. However, the association with physical function has not been well established. OBJECTIVE: Assess the association between timing of menopause and different measures of physical function. EVIDENCE REVIEW: Searches on the PubMed, Cochrane Library, SciELO, LILACS, and Web of Science databases were conducted. Observational studies on the association between age at menopause and measures of physical function were included, with no restriction for publication date or language. Methodological quality was assessed by the "Quality Assessment Tool for Observational Cohort and Cross-Sectional Studies." FINDINGS: Four cross-sectional studies were included, totaling 13,846 participants. These investigated five measures of physical function: gait speed, grip strength, standing balance, chair stand, and self-reported functional limitations. Poor physical function was associated with premature (<40 y) or early menopause (<45 y) in all the studies, with significant results only for grip strength, gait speed, and functional limitation. Premature and early menopause were associated with weaker grip strength [between 2.58 kg (95% CI = 0.74 to 4.43) and 5.21 kg (2.18 to 8.25)], and lower gait speed [between 0.03 m/s (0.01 to 0.06) and 0.06 m/s (0.02 to 0.09)]. Menopause after the age of 50 is associated with less likelihood of functional limitation [OR between 0.52 (95% CI = 0.29 to 0.95) and 0.61 (0.40 to 0.95)] compared with premature and early menopause. Two measures of physical function (chair stand test and standing balance) were not significantly associated with age at menopause. CONCLUSION: Only four cross-sectional studies showed that earlier ages at menopause are associated with poor physical function (grip strength, gait speed, and self-reported functional limitations), but given the high heterogeneity of the studies, no consensus is possible. Longitudinal studies are needed to explore the association between age at menopause and different measures of physical function as well as the influence of different socioeconomic conditions between countries on functioning.

O presente estudo teve a finalidade de avaliar os efeitos do uso do probiótico (PRO) Saccharomyces ceveriseae coadjuvante à raspagem e alisamento radicular (RAR) no tratamento da periodontite experimental (PE) induzida em animais imunossuprimidos com o quimioterápico 5-Fluorouracil (5FU). Cento e oito animais foram submetidos à indução da PE por meio da instalação de um fio de algodão ao redor do primeiro molar inferior esquerdo ou direito. A imunossupressão foi obtida pela injeção intraperitoneal do 5FU aplicada em dois momentos: no ato da instalação da ligadura e após 48 horas. Sete dias após instalação a ligadura foi removida e os animais separados aleatoriamente em seis grupos experimentais que receberam os seguintes tratamentos: PE (n=18) - animais que não sofreram nenhum tratamento sistêmico ou local; SS (n=18) - animais que não sofreram tratamento local, apenas tratamento sistêmico com solução salina (SS); 5FU (n=18) - animais tratados sistemicamente com 5FU que não sofreram nenhum tratamento local; 5FU-PRO (n=18) - animais tratados sistemicamente com 5FU que receberam irrigação local com 0,6 ml de PRO; 5FURAR (n=18) - animais tratados sistemicamente com 5FU que receberam RAR e; 5FURAR-PRO (n=18) - animais tratados sistemicamente com 5FU, que receberam tratamento com RAR e irrigação local com 0,6 ml de PRO. Seis animais de cada período foram submetidos à eutanásia aos 7, 15 e 30 dias após os tratamentos. Os espécimes contendo a área interessada foram obtidos e processados para a análise histopatológica, histométrica da perda óssea alveolar (POA), porcentagem de tecido ósseo (PTO), porcentagem de osso vital (POV) e porcentagem de osso não vital (PONV), e análise imunoistoquímica (TRAP, RANKL e OPG). Os dados obtidos foram submetidos à análise estatística (α=5%). Os resultados histológicos demonstraram que nos espécimes tratados com 5FU houve agravamento da perda óssea na região de furca comparado com os animais sistemicamente saudáveis (grupos PE e SS); que o tratamento com PRO como monoterapia (grupo 5FU-PRO) promoveu redução do processo inflamatório, denotando estabilização da doença periodontal; que o tratamento com RAR (grupo 5FU-RAR) reduziu o volume do infiltrado inflamatório mostrando sinais de reparação e a associação PRO-RAR (grupo 5FU-RAR-PRO) evidenciou nítida reestruturação tecidual aos 15 dias e menor área de reabsorção óssea aos 30 dias. Observou-se na análise histométrica que a POA foi maior no grupo 5FU-PRO em relação ao grupo SS aos 30 dias (p<0,05); a PTO foi maior no grupo 5FU-RAR-PRO aos 30 dias em comparação aos grupos EP, SS, 5FU e 5FU-PRO (p<0,05) e a POV foi maior no grupo 5FU-RAR-PRO em comparação ao grupo 5FU aos 7 dias, e maior aos 30 dias em comparação a todos os grupos experimentais (p<0,05). Não houve diferença entre os grupos e períodos na imunomarcação das células TRAP positivas. A RANKL mostrou alto padrão de imunomarcação nos grupos PE, SS e 5FU em todos períodos, e aos 7 dias no grupo 5FU-PRO, 5FU-RAR e 5FURAR-PRO, moderado padrão de imunomarcação nos grupos 5FU, 5FU-PRO, 5FURAR, e 5FU-RAR-PRO, mantendo-se baixo padrão neste último grupo, no período de 30 dias. Houve baixo padrão de imunomarcação de OPG em todos os grupos e períodos, exceto no grupo 5FU-RAR-PRO que mostrou moderado padrão. Diante dos resultados obtidos pode ser concluído que o uso do quimioterápico 5FU agravou a PE; que o uso do Saccharomyces ceveriseae associado à RAR, minimizou os efeitos do quimioterápico nos tecidos periodontais reduzindo o processo inflamatório, e reduzindo a porcentagem de destruição de tecido ósseo na área de furca em animais imunossuprimidos(AU)


The present study aimed to evaluate the effects of using probiotic (PRO) Saccharomyces ceveriseae as an adjuvant of scaling and root planing (SRP) in the treatment of experimental periodontitis (EP) induced in immunosuppressed animals with chemotherapeutic 5-Fluorouracil (5FU). One hundred and eight animals were submitted to EP induction by installing a cotton thread around the lower left or right first molar. Immunosuppression was obtained by intraperitoneal injection of 5FU applied in two moments: at the time of installing the ligature and after 48 hours. Seven days after installation, the ligature was removed and the animals were randomly separated into six experimental groups that received the following treatments: EP (n = 18) - animals that did not receive any systemic or local treatment; SS (n = 18) - animals that did not receive local treatment, only systemic treatment with saline solution (SS); 5FU (n = 18) - animals treated systemically with 5FU that did not receive any local treatment; 5FUPRO (n = 18) - animals treated systemically with 5FU that received local irrigation with 0.6 ml of PRO; 5FU-SRP (n = 18) - animals treated systemically with 5FU that received RAR and; 5FU-SRP-PRO (n = 18) - animals treated systemically with 5FU, which received treatment with SRP and local irrigation with 0.6 ml of PRO. Six animals from each period were euthanized at 7, 15 and 30 days after treatments. The specimens containing the interested area were obtained and processed for histopathological, histometric analysis of alveolar bone loss (ABL), percentage of bone tissue (PBT), percentage of vital bone (PVB) and percentage of non-vital bone (PNVB), and immunohistochemical analysis (TRAP, RANKL and OPG). The data obtained were subjected to statistical analysis (α = 5%). The histological results showed that in the specimens treated with 5FU there was worsening of bone loss in the furcation region compared to healthy animals (EP and SS groups); that treatment with PRO as monotherapy (group 5FU-PRO) reduced the inflammatory process, denoting stabilization of periodontal disease; that treatment with SRP (5FU-SRP group) reduced the volume of the inflammatory infiltrate showing signs of repair and the PRO-SRP association (5FU-SRP-PRO group) showed a clear tissue restructuring at 15 days and a smaller area of bone resorption at 30 days. It was observed in the histometric analysis that ABL was higher in the 5FU-PRO group compared to SS group at 30 days (p <0.05); PTO was higher in 5FU-SRP-PRO group at 30 days compared to EP, SS, 5FU and 5FU-PRO groups (p <0.05) and PVB was higher in the 5FU-SRP-PRO group compared to 5FU group at 7 days, and greater at 30 days compared to all experimental groups (p <0.05). There was no difference between groups and periods in the immunostaining of TRAP positive cells. RANKL showed a high immunostaining pattern in the EP, SS and 5FU groups in all periods, and at 7 days in 5FU-PRO, 5FU-SRP and 5FU-SRP-PRO groups, a moderate immunostaining pattern in the 5FU, 5FU-PRO, 5FU-SRP and 5FU-SRP-PRO groups, maintaining a low standard in 5FU-SRP-PRO group, in the period of 30 days. There was a low pattern of OPG immunostaining in all groups and periods, except in 5FU-SRP-PRO group, which showed a moderate pattern. In view of the results obtained, it can be concluded that the use of 5FU chemotherapy has worsened the evolution of EP; that the use of Saccharomyces ceveriseae associated with RAR, minimized the effects of chemotherapy on periodontal tissues, reducing the inflammatory process, and reducing the percentage of bone tissue destruction in the furcation area in immunosuppressed animals(AU)

Differences in hydroxyurea response in sickle cell anemia may arise due to a series of factors with genetic factors appearing to be predominant. This study aims to investigate the effects of single nucleotide polymorphisms in genes encoding drug-metabolizing enzymes and solute carriers on hydroxyurea response, in patients with sickle cell anemia. For that purpose, a total number of 90 patients with sickle cell anemia were recruited, 45 were undergoing hydroxyurea treatment, while 45 were not under the treatment. Association analyses were performed between CYP3A4 (rs2740574), CYP2D6 (rs3892097), CAT (rs7943316 and rs1001179), and SLC14A1 (rs2298720) variants and laboratory parameters. According to our findings, patients with hydroxyurea treatment demonstrated higher HbF levels and a significant improvement in hemolytic, hepatic, inflammatory, and lipid parameters in comparison to those without the treatment. We also found significant associations between the CYP2D6 (rs3892097), CAT (rs7943316 and rs1001179), and SLC14A1 (rs2298720) variants and an improvement of the therapeutic effects, specifically the hemolytic, hepatic, inflammatory, lipid, and renal parameters. In conclusion, our results highlight the importance of the investigated variants, and their strong association with hydroxyurea efficacy in patients with sickle cell anemia, which may be considered in the future as genetic markers.

OBJECTIVE: The aim of this study was to evaluate the effects of Lactobacillus reuteri applied locally or systemically with scaling and root planing (SRP) to the treatment of experimental periodontitis (EP) in rats treated with 5-fluorouracil. METHODS: A cotton ligature was installed on the molars of rats. The animals (n = 54) underwent chemotherapy and were divided into groups: SRP (n = 18), scaling and root planing only; LP (n = 18), SRP and 4 sessions of local probiotic (PRO); SP, SRP and 4 sessions of systemic PRO. The molar furcation area was submitted to histopathological, histometric of alveolar bone loss (ABL) and immunolabeling analysis after euthanasia at 7, 15 and 30 days. The data were submitted to statistical analysis (α = 5%). RESULTS: At 15 days ABL was higher in LP compared to SP. At 30 days, ABL was higher in LP compared to SRP and SP. Higher immunolabeling of TGF-ß1 was observed in LP and SP at 7 days compared to SRP (p < 0.05). Lower immunolabeling of OCN and higher immunolabeling of RANKL were observed at all periods in SRP compared to SP (p < 0.05). At 30 days, SRP showed lower immunolabeling of OPG compared to SP and LP (p < 0.05). In SP, lower immunolabeling was observed at 15 days compared at 30 days (p < 0.05). CONCLUSION: The ABL was similar among the groups treated with SRP associated or not to probiotic therapeutic, although the systemic use of Lactobacillus reuteri considerably reduced inflammation and favored periodontal tissues repair.
