RESUMO

The neuropeptide pigment-dispersing factor (PDF) is important for the generation and entrainment of circadian rhythms in the fruitfly Drosophila melanogaster. Recently two pdf homologs, pdf-1 and pdf-2, and a PDF receptor, pdfr-1, have been found in Caenorhabditis elegans and have been implicated in locomotor activity. In this work, we have studied the role of the PDF neuropeptide in the circadian system of C. elegans and found that both pdf-1 and pdf-2 mutants affect the normal locomotor activity outputs. In particular, loss of pdf-1 induced circadian arrhythmicity under both light-dark (LD) and constant dark (DD) conditions. These defects can be rescued by a genomic copy of the pdf-1 locus. Our results indicate that PDF-1 is involved in rhythm generation and in the synchronization to LD cycles, as rhythmic patterns of activity rapidly disappear when pdf-1 mutants are recorded under both entrained and free-running conditions. The role of PDF-2 and the PDF receptors is probably more complex and involves the interaction between the two pdf paralogues found in the nematode.

Impulsiveness is a pivotal personality trait representing a core domain in all major personality inventories. Recently, impulsiveness has been identified as an important modulator of cognitive processing, particularly in tasks that require the processing of large amounts of information. Although brain imaging studies have implicated the prefrontal cortex to be a common underlying representation of impulsiveness and related cognitive functioning, to date a fine-grain and detailed morphometric analysis has not been carried out. On the basis of ahigh-resolution magnetic resonance scans acquired in 1620 healthy adolescents (IMAGEN), the individual cortical thickness (CT) was estimated. Correlations between Cloninger's impulsiveness and CT were studied in an entire cortex analysis. The cluster identified was tested for associations with performance in perceptual reasoning tasks of the Wechsler Intelligence Scale for Children (WISC IV). We observed a significant inverse correlation between trait impulsiveness and CT of the left superior frontal cortex (SFC; Monte Carlo Simulation P<0.01). CT within this cluster correlated with perceptual reasoning scores (Bonferroni corrected) of the WISC IV. On the basis of a large sample of adolescents, we identified an extended area in the SFC as a correlate of impulsiveness, which appears to be in line with the trait character of this prominent personality facet. The association of SFC thickness with perceptual reasoning argues for a common neurobiological basis of personality and specific cognitive domains comprising attention, spatial reasoning and response selection. The results may facilitate the understanding of the role of impulsiveness in several psychiatric disorders associated with prefrontal dysfunctions and cognitive deficits.

Video game playing is a frequent recreational activity. Previous studies have reported an involvement of dopamine-related ventral striatum. However, structural brain correlates of video game playing have not been investigated. On magnetic resonance imaging scans of 154 14-year-olds, we computed voxel-based morphometry to explore differences between frequent and infrequent video game players. Moreover, we assessed the Monetary Incentive Delay (MID) task during functional magnetic resonance imaging and the Cambridge Gambling Task (CGT). We found higher left striatal grey matter volume when comparing frequent against infrequent video game players that was negatively correlated with deliberation time in CGT. Within the same region, we found an activity difference in MID task: frequent compared with infrequent video game players showed enhanced activity during feedback of loss compared with no loss. This activity was likewise negatively correlated with deliberation time. The association of video game playing with higher left ventral striatum volume could reflect altered reward processing and represent adaptive neural plasticity.

The authors have analysed the frequency and structure of congenital anomalies in children born in the Pomeranian district in the period from 01.07.1997 to 31.12.1998. Among a total of 28.361 births in that area, 748 (2.64%) were affected by congenital anomalies. Among 28.361 births, 620 (2.18%) were from multiple pregnancies. 23 (3.71%) among births from multiple pregnancies were affected by congenital malformations. The prevalence rate of inborn anomalies in births from multiple pregnancy in our area were higher (3.71%) in comparison to births from singleton pregnancy (2.61%). It implies that children born from multiple pregnancy are at higher risk of developing congenital anomalies.

Metabolites of sphingomyelin, ceramide and sphingosine, have previously been implicated in cell growth regulation. Here we show that cell-permeable ceramide analogs and treatment with sphingomyelinase, which hydrolyzes sphingomyelin located on the outer leaflet of the bilayer, increase the progression of quiescent Swiss 3T3 fibroblasts through the S phase of the cell cycle leading to an increase in cell division. Although both potentiate the mitogenic effects of several growth factors [14], sphingomyelinase treatment antagonized the mitogenic effect of the tumor promoter, 12-O-tetradecanoylphorbol 13-acetate (TPA), while ceramide analogs had no effect, and sphingosine, a further metabolite of ceramide, potentiated the mitogenic effect of TPA. Concomitantly, sphingomyelinase, but not ceramide analogs, blunted the rapid increase in membrane-associated protein kinase C (PKC) activity induced by TPA without affecting the translocation of PKC alpha, delta, epsilon or zeta isoforms. Moreover, in contrast to sphingosine which activates phospholipase D (PLD) leading to an increase in phosphatidic acid levels, sphingomyelinase, but not ceramide analogs, reduced TPA-stimulated PLD activity. Our results suggest that the signaling pathways utilized by sphingomyelinase differ from those of cell-permeable ceramide analogs, and both act differently than sphingosine. The differential effects of exogenous short-chain ceramide analogs and sphingomyelinase call for caution in using these analogs as tools to study the role of ceramide in diverse cellular functions.

We describe the characterization of a mutation of the locus GLR1. This mutation allowed for (i) the glucose repression-insensitive synthesis ot the enzymes maltase, galactokinase, alpha-galactosidase, reduced nicotinamide adenine dinucleotide-cytochrome c reductase, and cytochrome c oxidase and (ii) growth on maltose in the presence of the gratuitous glucose repressor D-glucosamine. The glucosamine resistance cosegregated with the glucose-insensitive synthesis of the enzymes listed above. In addition, crosses between the glucosamine-resistant mutant and isogenic sensitive strains gave only tetrads containing two resistant and two sensitive spores. Thus, a single pleiotropic mutation is responsible for both phenotypes. We call the locus GLR1, for glucose regulation, and the glucose repression-insensitive mutation glr1-1.
