RESUMO

The United Nations (UN) Decade of Ocean Science highlights a need to improve the way in which scientific results effectively inform action and policies regarding the ocean. Our research contributes to achieving this goal by identifying practical actions, barriers, stakeholder contributions and resources required to increase the sustainability of activities carried out in the context of artisanal fisheries to meet UN Sustainable Development Goals (SDG) and International Year of Artisanal Fisheries and Aquaculture (IYAFA) Global Action Plan (GAP) Pillar targets. We conducted a novel 'social value chain analysis' via a participatory workshop to elicit perspectives of value chain actors and fisheries stakeholders associated with two Spanish artisanal common octopus (Octopus vulgaris) fisheries (western Asturias-Marine Stewardship Council [MSC] certified, and Galicia-non-MSC certified) about their priorities regarding sustainable octopus production and commercialization. Our adapted Rapfish sustainability framework emphasised the importance of economic, environmental, ethical, institutional, social, and technological indicators to different actors across the value chain. We mapped participants' shared sustainability priorities (e.g. integrated fisheries management, knowledge-based management, product traceability) to six Rapfish indicators, seven IYAFA Pillars and twelve SDGs to reveal how our results can inform ocean policy and actions. This identified how certification incentives and other cooperative approaches can facilitate environmental, economic and social sustainability (e.g. value-added products, price premiums for producers, gender inclusive organisations); support IYAFA priority outcomes (raised awareness, strengthened science-policy interface, empowered stakeholders, partnerships); and help to achieve UN SDG targets (e.g. SDG 14.b, SDG 17.17). The results can inform actors, stakeholders and policymakers about how different actors contribute to efforts to achieve the SDGs and how to manage priorities for sustainable actions within artisanal fisheries and their value chains. We recommend inclusive and equitable participatory knowledge transfer and governance platforms as part of the UN Decade of Ocean Science and beyond where participants can create theories of change towards sustainability involving the development of multi-sectoral ocean policies framed at the level of the value chain and supported by appropriate governance structures. Here we list species-specific recommendations for housing, care and management of cephalopod molluscs employed for research purposes with the aim of contributing to the standardization of minimum requirements for establishments, care and accommodation of these animals in compliance with the principles stated in Directive 2010/63/EU. Maximizing their psychophysical welfare was our priority. General recommendations on water surface area, water depth and tank shape here reported represent the outcome of the combined action of the analysis of the available literature and an expertise-based consensus reached - under the aegis of the COST Action FA1301 - among researchers working with the most commonly used cephalopod species in Europe. Information on water supply and quality, environmental conditions, stocking density, feeding and handling are also provided. Through this work we wish to set the stage for a more fertile ground of evidence-based approaches on cephalopod laboratory maintenance, thus facilitating standardization and replicability of research outcomes across laboratories, at the same time maximizing the welfare of these animals.

This study aimed to explore different substances (or cold sea water) as potential anesthetic agents to facilitate short-term handling in Octopus maya juveniles. We investigated oxygen consumption before (baseline), during (first 600 s of exposure) and after anesthesia (recovery) of octopuses (n = 98; 1.67 ± 0.5 g) exposed to cold sea water (SW; 11 and 13°C), ethanol (EtOH; 0.5; 1.5 and 3.0%), magnesium chloride (MgCl2; 0.75; 1.5 and 3.75%), ethanol combined with magnesium chloride (Mix; 1.5:0.75%; 0.75:1.13%; and 2.25:0.37%) and clove oil (0.15 mL L-1). After exposure, the animals were handled for 180 s (exposed to air) and weighted. Two experimental groups not exposed to anesthetics (with or without handling) were also evaluated. The criteria for general anesthesia were analysed. Times of induction and recovery, incidence of attack response after recovery and possible longer-term effects of repeated general anesthesia on growth and mortality of the octopuses were evaluated. During anesthesia, O. maya juveniles exposed to SW (11 and 13°C), EtOH (0.5; 1.5 and 3.0%), Mix (0.75:1.13%), and clove oil, presented a significant decrease on oxygen consumption. In animals exposed to different concentrations of EtOH and Mix 0.75:1.13%, this decrease was registered after an increase on oxygen consumption. Animals exposed to MgCl2 did not show significant changes on oxygen consumption, except for animals exposed MgCl2 3.75%, which showed a significant increase on oxygen consumption. At the end of recovery, except for octopuses exposed to clove oil and MgCl2 0.75%, the values of oxygen consumption observed were comparable to the ones registered during baseline. Animals exposed to SW 11°C, EtOH 3.0%, Mix 1.5:0.75% and MgCl2 3.75% fulfilled the criteria defined for general anesthesia. Exposure to MgCl2 (all concentrations), SW 13°C and clove oil reduced or inhibited the incidence of attack response after recovery. Except for animals exposed to clove oil, growth of the juveniles was not affected by the exposure to the different substances. Short-term handling (180 s) of O. maya juveniles can eventually be carried out without anesthesia. However, to facilitated handling, we suggest the use of EtOH 3.0% or cold sea water 11°C.

Although interest in several areas of cephalopod research has emerged over the last decades (e.g., neurobiology, aquaculture, genetics, and welfare), especially following their 2010 inclusion in the EU Directive on the use of animals for experimental purposes, knowledge regarding the parasites of cephalopods is lacking. Cephalopods can be intermediate, paratenic, or definitive hosts to a range of parasites with a wide variety of life cycle strategies. Here, we briefly review the current knowledge in cephalopod parasitological research, summarizing the main parasite groups that affect these animals. We also emphasize some topics that, in our view, should be addressed in future research, including: (i) better understanding of life cycles and transmission pathways of common cephalopod parasites; (ii) improve knowledge of all phases of the life cycle (i.e., paralarvae, juveniles, adults and senescent animals) and on species from polar deep sea regions; (iii) exploration of the potential of using cephalopod-parasite specificity to assess population boundaries of both, hosts and parasites; (iv) risk evaluation of the potential of standard aquacultural practices to result in parasite outbreaks; (v) evaluation and description of the physiological and behavioral effects of parasites on their cephalopod hosts; (vi) standardization of the methods for accurate parasite sampling and identification; (vii) implementation of the latest molecular methods to facilitate and enable research in above mentioned areas; (viii) sharing of information and samples among researchers and aquaculturists. In our view, addressing these topics would allow us to better understand complex host-parasite interactions, yield insights into cephalopod life history, and help improve the rearing and welfare of these animals in captivity.

RESUMO

Here, three researchers who have recently embarked on careers in cephalopod biology discuss the current state of the field and offer their hopes for the future. Seven major topics are explored: genetics, aquaculture, climate change, welfare, behavior, cognition, and neurobiology. Recent developments in each of these fields are reviewed and the potential of emerging technologies to address specific gaps in knowledge about cephalopods are discussed. Throughout, the authors highlight specific challenges that merit particular focus in the near-term. This review and prospectus is also intended to suggest some concrete near-term goals to cephalopod researchers and inspire those working outside the field to consider the revelatory potential of these remarkable creatures.

Cephalopods are the sole invertebrates included in the list of regulated species following the Directive 2010/63/EU. According to the Directive, achieving competence through adequate training is a requisite for people having a role in the different functions (article 23) as such carrying out procedures on animals, designing procedures and projects, taking care of animals, killing animals. Cephalopod Biology and Care Training Program is specifically designed to comply with the requirements of the "working document on the development of a common education and training framework to fulfil the requirements under the Directive 2010/63/EU". The training event occurred at the ICM-CSIC in Barcelona (Spain) where people coming from Europe, America and Asia were instructed on how to cope with regulations for the use of cephalopod molluscs for scientific purposes. The training encompasses discussion on the guidelines for the use and care of animals and their welfare with particular reference to procedures that may be of interest for neuroscience. Intensive discussion has been carried out during the training sessions with focus on behavioural studies and paradigms, welfare assessment, levels of severity of scientific procedures, animal care, handling, transport, individual identification and marking, substance administration, anaesthesia, analgesia and humane killing.

This study evaluated for two years the haematological parameters and the occurrence of gill parasites in silver catfish Rhamdia quelen, farmed in the state of Santa Catarina, southern Brazil. Between March 2010 and January 2012, a total of 137 fish were caught in a fish farm to compare the haematological and parasitological analysis, during hot and cold seasons. Simultaneously, water quality parameters were measured in the pond to analyze the relationship between environmental conditions and haematology and parasitism. During the hot season, there was an increase in water temperature, electrical conductivity and ammonia followed by a decrease in dissolved oxygen, pH, transparency and alkalinity. Two species of Monogenea were observed in the gills, Aphanoblastella mastigatus (Dactylogyridae) and Scleroductus sp. (Gyrodactylidae). Additionally, during this season, there was a significant increase (p<0.05) in the mean intensity of infection by Monogenea, as well as total leukocytes number, thrombocytes and lymphocytes, and a reduction in monocytes. The changes might be caused by parasitism and/or environmental variations between seasons.

A recent revival in using cephalopods as experimental animals has rekindled interest in their biology and life cycles, information with direct applications also in the rapidly growing ornamental aquarium species trade and in commercial aquaculture production for human consumption. Cephalopods have high rates of growth and food conversion, which for aquaculture translates into short culture cycles, high ratios of production to biomass and high cost-effectiveness. However, at present, only small-scale culture is possible and only for a few species: the cuttlefish Sepia officinalis, the loliginid squid Sepioteuthis lessoniana and the octopuses Octopus maya and O. vulgaris. These four species are the focus of this chapter, the aims of which are as follows: (1) to provide an overview of the culture requirements of cephalopods, (2) to highlight the physical and nutritional requirements at each phase of the life cycle regarded as essential for successful full-scale culture and (3) to identify current limitations and the topics on which further research is required. Knowledge of cephalopod culture methods is advanced, but commercialization is still constrained by the highly selective feeding habits of cephalopods and their requirement for large quantities of high-quality (preferably live) feed, particularly in the early stages of development. Future research should focus on problems related to the consistent production of viable numbers of juveniles, the resolution of which requires a better understanding of nutrition at all phases of the life cycle and better broodstock management, particularly regarding developments in genetic selection, control of reproduction and quality of eggs and offspring.

O presente estudo avaliou, durante dois anos, os parâmetros hematológicos e a ocorrência de parasitos branquiais em jundiá Rhamdia quelen, cultivado no Estado de Santa Catarina, Sul do Brasil. Entre março de 2010 e janeiro de 2012, um total de 137 jundiás foi coletado de uma piscicultura para comparar os parâmetros hematológicos e parasitológicos durante as estações quente e fria. Simultaneamente, foram medidos os parâmetros de qualidade da água do viveiro, para relacionar as condições ambientais com a hematologia e o parasitismo. Na estação quente, houve aumento da temperatura, condutividade elétrica e amônia, e redução do oxigênio dissolvido, pH, transparência e alcalinidade. Duas espécies de Monogenea foram observadas nas brânquias, Aphanoblastella mastigatus (Dactylogyridae) e Scleroductus sp. (Gyrodactylidae). Na estação quente, houve aumento significativo (p<0,05) da intensidade média de infecção por Monogenea, bem como dos valores de leucócitos totais, trombócitos e linfócitos, e redução significativa dos monócitos. As diferenças observadas podem ter sido causadas pelas variações ambientais entre as estações.

This study evaluated the influence of the clomazone herbicide (2-[(2-chlorophenyl)methyl]-4,4-dimethyl-3-isoxazolidinone) contamination on the hematological parameters and histological changes in gills and liver of silver catfish (Rhamdia quelen) from Madre River, Santa Catarina State, Southern Brazil. Fish were collected between March 2010 and January 2012 at two different sites of the Madre River, one site receiving residual water (contaminated site) from rice culture (n=49) and another that do not receive residual water (reference site) (n=48). The herbicide clomazone analysis detected 3.40±1.70 µg/L in the contaminated site and 1.1±0.33 µg/L in the reference site. Fish from contaminated site showed increased (P<0.05) number of monocytes suggesting the possible defense response as a result of chronic exposure to clomazone. On the other hand, no difference was found in the hematocrit percentage, red blood cell count, total thrombocyte number, white blood cell count, lymphocytes, and neutrophils number. Fish from both sites showed histopathological changes in gills and liver, possibly caused by chronic exposure to contamination. The influence of herbicide sub doses on fish health is also discussed.

RESUMO

A novel and easily constructed apparatus for collecting monogenean eggs released by the parasites into the water is described and illustrated. Use of this technique may lead to a better understanding of the parasite's biology, which, in turn, may lead to the improvement of parasite management strategies in fish farms where monogeneans are potentially harmful to their hosts. The technique is also useful for studies of eggs or free-living stages of other fish parasites.
