RESUMO

The key role played by host-microbiota interactions on human health, disease onset and progression, and on host response to treatments has increasingly emerged in the latest decades. Indeed, dysbiosis has been associated to several human diseases such as obesity, diabetes, cancer and also neurodegenerative disease, such as Parkinson, Huntington and Alzheimer's disease (AD), although whether causative, consequence or merely an epiphenomenon is still under investigation. In the present study, we performed a metabologenomic analysis of stool samples from a mouse model of AD, the 3xTgAD. We found a significant change in the microbiota of AD mice compared to WT, with a longitudinal divergence of the F/B ratio, a parameter suggesting a gut dysbiosis. Moreover, AD mice showed a significant decrease of some amino acids, while data integration revealed a dysregulated production of desaminotyrosine (DAT) and dihydro-3-coumaric acid. Collectively, our data show a dysregulated gut microbiota associated to the onset and progression of AD, also indicating that a dysbiosis can occur prior to significant clinical signs, evidenced by early SCFA alterations, compatible with gut inflammation.

Fluids are known to trigger a broad range of slip events, from slow, creeping transients to dynamic earthquake ruptures. Yet, the detailed mechanics underlying these processes and the conditions leading to different rupture behaviors are not well understood. Here, we use a laboratory earthquake setup, capable of injecting pressurized fluids, to compare the rupture behavior for different rates of fluid injection, slow (megapascals per hour) versus fast (megapascals per second). We find that for the fast injection rates, dynamic ruptures are triggered at lower pressure levels and over spatial scales much smaller than the quasistatic theoretical estimates of nucleation sizes, suggesting that such fast injection rates constitute dynamic loading. In contrast, the relatively slow injection rates result in gradual nucleation processes, with the fluid spreading along the interface and causing stress changes consistent with gradually accelerating slow slip. The resulting dynamic ruptures propagating over wetted interfaces exhibit dynamic stress drops almost twice as large as those over the dry interfaces. These results suggest the need to take into account the rate of the pore-pressure increase when considering nucleation processes and motivate further investigation on how friction properties depend on the presence of fluids.

Large, destructive earthquakes often propagate along thrust faults including megathrusts. The asymmetric interaction of thrust earthquake ruptures with the free surface leads to sudden variations in fault-normal stress, which affect fault friction. Here, we present full-field experimental measurements of displacements, particle velocities, and stresses that characterize the rupture interaction with the free surface, including the large normal stress reductions. We take advantage of these measurements to investigate the dependence of dynamic friction on transient changes in normal stress, demonstrate that the shear frictional resistance exhibits a significant lag in response to such normal stress variations, and identify a predictive frictional formulation that captures this effect. Properly accounting for this delay is important for simulations of fault slip, ground motion, and associated tsunami excitation.

Chronic elevation of pulmonary microvascular pressure (Pmv) consistently leads to alveolocapillary barrier thickening and reduction in the filtration coefficient. In animal models of chronic heart failure (CHF) the lung remains dry despite hydrostatic forces. As fluid flux is bi-directional, it has been postulated that an increase in alveolar fluid clearance may facilitate the dry lung when Pmv is chronically elevated. In this study we aimed to examine alveolar fluid clearance in ambulatory patients with CHF secondary to left ventricular (LV) systolic dysfunction compared against non-CHF controls. Lung clearance following aerosol delivery of 99mtechnetium (Tc)-diethyl triaminepentaacetic acid (DTPA) was measured non-invasively by scintigraphy and half time of 99mTc-DTPA clearance (T (1/2)) was calculated by mono-exponential curve fit. Alveolar fluid clearance measured as half time DTPA clearance was significantly faster in CHF patients than controls (P=0.001). This was further defined by NYHA classification. No correlation was found between DTPA clearance and plasma epinephrine, norepinephrine or aldosterone hormone (P>0.05). Our results support an association between increasing alveolar fluid clearance and disease severity in CHF, and the concept of controlled bi-directional fluid flux in CHF associated with increasing Pmv, and represents another defence mechanism of the lung against pulmonary oedema.
