RESUMO

We present a comprehensive phylogeny derived from 5 genes, nucSSU, nucLSU rDNA, TEF1, RPB1 and RPB2, for 356 isolates and 41 families (six newly described in this volume) in Dothideomycetes. All currently accepted orders in the class are represented for the first time in addition to numerous previously unplaced lineages. Subclass Pleosporomycetidae is expanded to include the aquatic order Jahnulales. An ancestral reconstruction of basic nutritional modes supports numerous transitions from saprobic life histories to plant associated and lichenised modes and a transition from terrestrial to aquatic habitats are confirmed. Finally, a genomic comparison of 6 dothideomycete genomes with other fungi finds a high level of unique protein associated with the class, supporting its delineation as a separate taxon.

The class Dothideomycetes (along with Eurotiomycetes) includes numerous rock-inhabiting fungi (RIF), a group of ascomycetes that tolerates surprisingly well harsh conditions prevailing on rock surfaces. Despite their convergent morphology and physiology, RIF are phylogenetically highly diverse in Dothideomycetes. However, the positions of main groups of RIF in this class remain unclear due to the lack of a strong phylogenetic framework. Moreover, connections between rock-dwelling habit and other lifestyles found in Dothideomycetes such as plant pathogens, saprobes and lichen-forming fungi are still unexplored. Based on multigene phylogenetic analyses, we report that RIF belong to Capnodiales (particularly to the family Teratosphaeriaceae s.l.), Dothideales, Pleosporales, and Myriangiales, as well as some uncharacterised groups with affinities to Dothideomycetes. Moreover, one lineage consisting exclusively of RIF proved to be closely related to Arthoniomycetes, the sister class of Dothideomycetes. The broad phylogenetic amplitude of RIF in Dothideomycetes suggests that total species richness in this class remains underestimated. Composition of some RIF-rich lineages suggests that rock surfaces are reservoirs for plant-associated fungi or saprobes, although other data also agree with rocks as a primary substrate for ancient fungal lineages. According to the current sampling, long distance dispersal seems to be common for RIF. Dothideomycetes lineages comprising lichens also include RIF, suggesting a possible link between rock-dwelling habit and lichenisation.

Melanised fungi were isolated from rock surfaces in the Central Mountain System of Spain. Two hundred sixty six isolates were recovered from four geologically and topographically distinct sites. Microsatellite-primed PCR techniques were used to group isolates into genotypes assumed to represent species. One hundred and sixty three genotypes were characterised from the four sites. Only five genotypes were common to two or more sites. Morphological and molecular data were used to characterise and identify representative strains, but morphology rarely provided a definitive identification due to the scarce differentiation of the fungal structures or the apparent novelty of the isolates. Vegetative states of fungi prevailed in culture and in many cases could not be reliably distinguished without sequence data. Morphological characters that were widespread among the isolates included scarce micronematous conidial states, endoconidia, mycelia with dark olive-green or black hyphae, and mycelia with torulose, isodiametric or moniliform hyphae whose cells develop one or more transverse and/or oblique septa. In many of the strains, mature hyphae disarticulated, suggesting asexual reproduction by a thallic micronematous conidiogenesis or by simple fragmentation. Sequencing of the internal transcribed spacers (ITS1, ITS2) and 5.8S rDNA gene were employed to investigate the phylogenetic affinities of the isolates. According to ITS sequence alignments, the majority of the isolates could be grouped among four main orders of Pezizomycotina: Pleosporales, Dothideales, Capnodiales, and Chaetothyriales. Ubiquitous known soil and epiphytic fungi species were generally absent from the rock surfaces. In part, the mycota of the rock surfaces shared similar elements with melanised fungi from plant surfaces and fungi described from rock formations in Europe and Antarctica. The possibility that some of the fungi were lichen mycobionts or lichen parasites could not be ruled out.

Fungal strains isolated from rocks and lichens collected in the Antarctic ice-free area of the Victoria Land, one of the coldest and driest habitats on earth, were found in two phylogenetically isolated positions within the subclass Dothideomycetidae. They are here reported as new genera and species, Recurvomyces mirabilisgen. nov., sp. nov. and Elasticomyces elasticusgen. nov., sp. nov. The nearest neighbours within the clades were other rock-inhabiting fungi from dry environments, either cold or hot. Plant-associated Mycosphaerella-like species, known as invaders of leathery leaves in semi-arid climates, are also phylogenetically related with the new taxa. The clusters are also related to the halophilic species Hortaea werneckii, as well as to acidophilic fungi. One of the latter, able to grow at pH 0, is Scytalidium acidophilum, which is ascribed here to the newly validated genus Acidomyces. The ecological implications of this finding are discussed.

The levels of 5-hydroxytryptamine and its main metabolite 5-hydroxyindoleacetic acid were assessed in two brain regions, hypothalamus and telencephalon, of rainbow trout (Oncorhynchus mykiss) submitted to increases or decreases in plasma glucose levels through different experimental approaches. Thus, intraperitoneal glucose treatment (500 mg kg(-1)) increased 5-hydroxytryptamine telencephalic levels. Long-term food deprivation up to 3 weeks significantly increased hypothalamic (2 weeks and 3 weeks) and telencephalic (1 week, 2 weeks, and 3 weeks) levels of 5-hydroxyindoleacetic acid, whereas the ratio 5-hydroxyindoleacetic acid/5-hydroxytryptamine significantly increased throughout the food-deprivation period assessed. Intraperitoneal treatment with bovine insulin (4 mg kg(-1)) decreased the 5-hydroxyindoleacetic acid/5-hydroxytryptamine ratio in hypothalamus after 1 h. Intraperitoneal administration of fenfluramine (3 mg kg(-1)) caused a depression in food intake coincident with a significant decrease of the hypothalamic 5-hydroxyindoleacetic acid/5-hydroxytryptamine ratio. These data are discussed in the context of the involvement of serotonergic system in the control of food intake in rainbow trout.

AIMS: The diversity within a collection of worldwide isolates of Epicoccum nigrum has been studied using several phenotypic approaches. In addition, the abilities of phenotypic and genotypic techniques for the differentiation of a set of isolates are compared. METHODS AND RESULTS: The methodology used include the study of isozymes (acetyl esterase and alkaline phosphatase), HPLC profile of metabolites and antibiotic activities against a panel of bacteria, yeasts and filamentous fungi, and cytotoxicity against three mammalian cell lines. Two procedures for assessing the relationships within a collection of isolates, using a combination of the techniques, were evaluated, comparing the advantages and disadvantages of each method. CONCLUSIONS: The results showed that each individual technique allows differentiation of the isolates studied to some degree and that the information provided by each technique could be considered as complementary. Genotypic techniques were more powerful than the phenotypic ones to discriminate among the strains. SIGNIFICANCE AND IMPACT OF THE STUDY: This work evaluates the predictive value of several phenotypic techniques on a collection of fungal isolates, and compares the results obtained with genotypic techniques performed on the same strains.

The present paper discusses the effect of a single melatonin treatment (0.5 mg/kg, i.p.) on the dopaminergic metabolism in the hypothalamus and pituitary of the rainbow trout. The effects of exogenous melatonin on dopamine (DA) and 3,4-dihydroxyphenylacetic acid (DOPAC) contents were compared with the variations in the content of these catecholamines associated to the natural increase in the endogenous melatonin from daytime (3 hr before lights off) to nighttime (3 hr after lights off). Animals treated with melatonin showed a rapid (maximal values at 30 min post-injection) and relatively sustained rise in plasma melatonin levels, which reached supraphysiological ranges. The increase in circulating melatonin was accompanied by a reduction in the amount of DOPAC in both the hypothalamus (30, 60, and 120 min after i.p. melatonin) and the pituitary (120 min after i.p. melatonin) as well as in the pituitary DOPAC/DA ratio (60 and 120 min after i.p. melatonin). Similarly, the increase in circulating melatonin levels from the daytime to nighttime was associated with decreases in the contents of DOPAC in both the hypothalamus and pituitary and in the DOPAC/DA ratio in the pituitary. These data suggest that the inhibition of the hypothalamic-pituitary dopaminergic metabolism may be a specific mechanism of melatonin action in the trout brain that might operate following changes in the secretion of the hormone from the pineal gland.
