RESUMO

Advances in genomics, molecular biology, and statistical genetics have created a paradigm shift in the way livestock producers pursue genetic improvement in their herds. The nexus of these technologies has resulted in combining genotypic and phenotypic information to compute genomically enhanced measures of genetic merit of individual animals. However, large numbers of genotyped and phenotyped animals are required to produce robust estimates of the effects of SNP that are summed together to generate direct genomic breeding values (DGV). Data on 11,756 Angus animals genotyped with the Illumina BovineSNP50 Beadchip were used to develop genomic predictions for 17 traits reported by the American Angus Association through Angus Genetics Inc. in their National Cattle Evaluation program. Marker effects were computed using a 5-fold cross-validation approach and a Bayesian model averaging algorithm. The accuracies were examined with EBV and deregressed EBV (DEBV) response variables and with K-means and identical by state (IBS)-based cross-validation methodologies. The cross-validation accuracies obtained using EBV response variables were consistently greater than those obtained using DEBV (average correlations were 0.64 vs. 0.57). The accuracies obtained using K-means cross-validation were consistently smaller than accuracies obtained with the IBS-based cross-validation approach (average correlations were 0.58 vs. 0.64 with EBV used as a response variable). Comparing the results from the current study with the results from a similar study consisting of only 2,253 records indicated that larger training population size resulted in higher accuracies in validation animals and explained on average 18% (69% improvement) additional genetic variance across all traits.

The objective of this study was to estimate genetic parameters required for genetic evaluation of retail product percentage (RPP) in Simmental cattle. Carcass weight (HCW), subcutaneous fat thickness (FAT), longissimus muscle area (REA) and kidney, pelvic, and heart fat (KPH) records were available to compute RPP on steers (n = 5171) and heifers (n = 1400) from the American Simmental Association database; animals were sired by 561 Simmental bulls and out of 5886 crossbred dams. Genetic parameters were estimated using residual maximal likelihood and a four trait animal model for the components of RPP including fixed harvest contemporary group effects, random animal genetic effects, and a linear covariate for age at harvest. Heritability estimates were 0.51 +/- 0.05, 0.36 +/- 0.05, 0.46 +/- 0.05, and 0.18 +/- 0.05 for HCW, FAT, REA and KPH respectively. Non-zero genetic correlations were estimated between HCW and REA (rg = 0.51 +/- 0.06) and between REA and FAT (rg = -0.43 +/- 0.08), but other genetic correlation estimates among the component traits were low. As a linear function of its components, heritability and genetic correlations involving RPP were estimated using index methods. The heritability estimate for RPP was 0.41, and genetic correlations were -0.17, -0.83, 0.67, and 0.01 with HCW, FAT, REA and KPH respectively. Therefore, RPP was strongly associated with muscle and fat deposition, but essentially independent of carcass weight and internal body cavity fat. Genetic evaluation of RPP would be straightforward using multiple trait index methods and genetic regression, although the inclusion of KPH would be of marginal value.

Carcass data from 6,795 Simmental-sired animals born from 1992 to 2001 were used to determine whether adjustment to a constant age, back-fat, HCW, or marbling score would result in differences in heritability of the carcass traits and, correspondingly, if EPD calculated using those variance components and adjustments would result in sire reranking. The endpoints were age (EPA), backfat (EPF), HCW (EPC), or marbling (EPM). The traits analyzed were 12th-rib backfat (FAT), HCW, marbling (MRB), LM area (LMA), and percentage retail cuts (PRC). The data were analyzed using an animal model, where contemporary group was included as a fixed effect and was composed of slaughter date, sex, and herd. Random effects included in the model were direct genetic and residual. Estimates of heritability ranged from 0.12 to 0.14, 0.32 to 0.34, and 0.26 to 0.27 for FAT, HCW, and LMA, respectively, for the corresponding endpoints. Heritability for MRB was estimated to be 0.27 at all endpoints. For PRC, estimates of heritability were more variable, with estimates of 0.23 +/- 0.05, 0.32 +/- 0.05, 0.21 +/- 0.05, and 0.20 +/- 0.04 for EPA, EPF, EPC, and EPM, respectively. However, because the EPF and EPC adjustments adjust for a component trait of PRC (FAT and HCW, respectively), they may be altering the trait to one different from PRC. Spearman rank correlations between EPD within a trait using EPA compared with the other endpoints were >0.90 (P < 0.01) for FAT, HCW, MRB, and LMA. For PRC, Spearman rank correlations with EPA EPD were 0.73 (P < 0.01), 0.93 (P < 0.01), and 0.95 (P < 0.01) for EPF, EPC, and EPM, respectively. For most traits and endpoints, there was little reranking among sires when alternative endpoints were used. However, adjusting PRC to EPF appears to result in a greater heritability and substantial re-ranking of sires, potentially due to the adjustment changing the trait to one other than PRC.

Age-of-dam adjustment factors are used to preadjust birth and weaning weight data for national beef cattle genetic evaluations. Adjustments are used in order to make the means of the different age-of-dam subclasses similar so that a fair comparison of animals can be performed. A review was made of various research studies that have estimated age-of-dam adjustment factors for birth weight and weaning weight of beef cattle. In general, birth weight age-of-dam adjustment factors are the same across the sexes, but weaning weight age-of-dam adjustment factors differ across the sexes, with heifer calves receiving smaller adjustments than their male counterparts. Additionally, adjustment factors vary greatly across breeds. Preadjustment of records is difficult to do because a perfect estimate of adjustments is not possible. A more appropriate method for adjusting for age-of-dam is to simultaneously adjust during national genetic evaluations

Genetic parameters of mature weight are needed for effective selection and genetic evaluation. Data for estimating these parameters were collected from 1963 to 1985 and consisted of 32,018 mature weight records of 4,175 Hereford cows that were in one control and three selection lines that had been selected for weaning weight, for yearling weight, or for an index combining yearling weight and muscle score for 22 yr. Several models and subsets of the data were considered. The mature weight records consisted of a maximum of three seasonal weights taken each year, at brand clipping (February and March), before breeding (May and June), and at palpation (August and September). Heritability estimates were high (0.49 to 0.86) for all models considered, which suggests that selection to change mature weight could be effective. The model that best fit the data included maternal genetic and maternal permanent environmental effects in addition to direct genetic and direct permanent environmental effects. Estimates of direct heritability with this model ranged from 0.53 to 0.79, estimates of maternal heritability ranged from 0.09 to 0.21, and estimates of the genetic correlation between direct and maternal effects ranged from -0.16 to -0.67 for subsets of the data based on time of year that mature weight was measured. For the same subsets, estimates of the proportions of variance due to direct permanent environment and maternal permanent environment ranged from 0.00 to 0.09 and 0.00 to 0.06, respectively. Using a similar model that combined all records and included an added fixed effect of season of measurement of mature weight, direct heritability, maternal heritability, genetic correlation between direct and maternal effects, proportion of variance due to direct permanent environmental effects, and proportion of variance due to maternal permanent environmental effects were estimated to be 0.69, 0.13, -0.65, 0.00, and 0.04, respectively. Mature weight is a highly heritable trait that could be included in selection programs and maternal effects should not be ignored when analyzing mature weight data.

Records of 59 U.S. Army service members identified as at-risk for coronary heart disease were reviewed. Soldiers were divided into groups that completed and did not complete cholesterol nutrition education classes. Differences in the means of repeated cholesterol measures were compared within each group. Results showed statistically significant declines in total cholesterol, with t(35) = 2.94, p < 0.01 for the class completion group and t(22) = 2.18, p < 0.05 for the dropout group. Similar results were noted for low-density lipoprotein measures. However, cholesterol differences in the means between the two groups were not statistically significant. Results indicate that decreases in serum cholesterol may occur due to at-risk reactive behavior, but that full compliance with the current nutrition education program does not appear to further reduce cholesterol levels to any appreciable degree.

RESUMO

An adsorption high-performance liquid chromatographic method suitable for the quantitative determination of the purity of alprostadil (prostaglandin E1) bulk drug and of the concentration of alprostadil in Prostin V.R. Pediatric Sterile Solution is described. A variety of isomers and related compounds including 8-iso-PGE1, 11-epi-PGE1, 15-epi-PGE1, PGE2, 5,6-trans-PGE2, and 8-iso-PGE2 are separated from alprostadil and are quantifiable. The 2-naphthacyl ester derivatives are employed and derivatization conditions providing maximum response are described.

RESUMO

An absorption high-performance liquid chromatography assay for the quantitative determination of prostaglandins A1 and B1 in alprostadil (prostaglandin E1) and in Prostin VR Pediatric Sterile Solution was developed. Prostaglandins A1 and B1 have been shown to be the major degradation products of prostaglandin E1. The adsorption system provided baseline resolution of the 2-naphthacyl esters of prostaglandin A1 from prostaglandin B1. Derivatization conditions providing maximal response for prostaglandins A1 and B1 while minimizing the conversion of prostaglandin E1 to prostaglandins A1 and B1 were established.
