RESUMO

The sequence of 2630 3'-terminal nucleotides has been determined for the genomic RNA of potato virus M (PVM), a type member of the carlavirus group. Analysis of this nucleotide sequence revealed five open reading frames coding for proteins of mol. wt. 25, 12, 7, 34 and 11 kDa (in 5'----3' direction). The PVM genome organization has been shown to be basically analogous to that of potexviruses, except that the latter at their 3' end lack the gene encoding 11K protein. Amino acid sequence comparison between the PVM 34K protein and the coat proteins of potexviruses has shown a high extent of homology in the C-terminal amino acids. Homology has also been found between the 25, 12 and 7K proteins encoded by PVM RNA and corresponding potexvirus proteins. All this gave grounds for suggestion that carlaviruses and potexviruses belong to the same subgroup of phytoviruses.

RESUMO

The complete nucleotide sequence was determined for three variants of the third genomic component of BSMV strain Argentina mild. The common variant, RNA 3 (2797 nucleotide), contains two open reading frames (ORFs) coding for two proteins with Mr of 74,229 (putative BSMV RNA polymerase) and Mr of 16,994. The second ORF is expressed from a subgenomic RNA. The extended variant RNA 3 differs from the common one only by the presence of a direct tandem repeat 351-363 nucleotides in length (with some variability) encompassing part of the leader sequence and the beginning of the first ORF. The resulting protein has a Mr of about 86,000. The defective variant, RNA 4, carries a deletion of 185 nucleotides in the 3'-end proximal part of the first ORF, which shortens the product to a Mr of 60,344.

RESUMO

The nucleotide sequence of the 3'-proximal 2630 nucleotides of potato virus M (PVM) genomic RNA was determined. The sequenced region contained five long open reading frames (ORFs). The ORF nearest to the 3'-terminal pol(A) tail corresponds to a polypeptide of Mr 10,848. This ORF is preceded by one which encodes a protein of Mr 33,906 (34K) which has an amino acid sequence that is very similar in its carboxyterminal part to that of the coat proteins of some potexviruses. Three other ORFs encoding polypeptides of Mr 24,615, 11,983 and 6739 are present in the region 5' to the 34K protein gene. There is extensive similarity between these proteins and the corresponding proteins encoded by the conserved triple gene block in the RNA of potexviruses.

RESUMO

The data given testify that picornavirus RNA-dependent RNA-polymerase, RNA-polymerase encoded by the genome of MS2 phage and the certain polypeptides involved in the replication of RNA genomes of alphaviruses, tobamoviruses and tricornaviruses include the homologous stretches of the amino acids. The common sequences are located in the COOH-terminal regions of the viral proteins. These sequences have been found to be conserved also in RNA-replicase MS2 phage. The similarity of the primary structure between the RNA-polymerase phages and proteins of eucaryotic plus-RNA-containing viruses testifies in favour of the hypothesis on possible ancestral relationship of virus RNA-polymerases genes. These data point out that it is possible to localize an indispensable functional domain conserved upon evolutionary divergence of an ancestral RNA-polymerase gene. Such conservative region is recently found in the composition of RNA-dependent DNA-polymerases animals and plants virus. An attention is drawn to the region of protein similarity between conservative domains of viral RNA-dependent DNA-polymerases and RNA-polymerases.

RESUMO

This paper describes the sequence of 257 nucleotides from the 3' end of RNA 2 of barley stripe mosaic virus ( BSMV , strain Argentina Mild) including an internal oligo (A) tract localized at a distance of 236 nucleotides from the 3' end, and the 3' terminal tRNA-like structure accepting tyrosine. This sequence is shown to be the same with RNAs 1,2 and 3 of another BSMV strain, Norwich , for at least the first 106 nucleotides from the 3' end. The 3' extremity of BSMV RNA bears some resemblance to tRNATyr from yeast in its primary structure. The possible secondary structures of the tRNA-like sequence in BSMV genome are discussed.

RESUMO

Barley stripe mosaic virus (BSMV) is a representative of the hordeiviruses which has a functionally fragmented RNA genom. 3'-terminal nucleotide sequence of the noncoding region of BSMV RNA 2 has been determined. This region contains the internal olygo(A) sequence (n = 21) and the tyrosine accepting tRNA-like structure at 3'-extreme end. tRNA-like sequence can fold into the two distinct secondary structure which strikingly similar to such a structures which has been proposed recently for cucumo- and bromoviruses.

We have previously detected in TMV-infected cells virus-specific informosome-like ribonucleoproteins (vRNP) that differed in the CsCl buoyant density from mature TMV particles. It is shown in the present work that [3H]uridine-labelled TMV-specific structures, when fractionated in Cs2SO4, produce three types of structures, i. e. with a buoyant density of 1.23 g/cm3 (the so-called 1.23 material), 1.29 g/gm3 (mature virus) and 1.34--1.49 g/cm3 (vRNP). The 1.23 material has been investigated. The incorporation of [3H]palmitic acid and the sensitivity of this material to 0.1% Na dodecyl sulphate was interpreted to mean the presence of membrane components. Treatment of the 1.23 material Na dodecyl sulphate induces the release of the mature virus, vRNP and free viral RNA. vRNP was shown to contain genome TMV RNA (mol. weight, 2.0 x 10(6)) and a considerable amount of subgenomic TMV RNA (mol. weight, 1.1--1.3 x 10(6) and 0.6--0.8 x 10(6)). It is demonstrated that RNA isolated from vRNP codes for TMV-specific proteins and is able to hybridize with recombinant plasmid containing DNA-copy of 3'-end RNA TMV fragment (about one half of genome).

RESUMO

The dissociation of histones H1, H2B and H2A from DNA was shown for the first time in the medium of physiological ionic strength (0.15M NaCl + 0.7 mM Na-phosphate buffer, pH 7.0) by treating the whole histone--DNA complex with high concentrations of whole histone from calf thymus chromatin. The histones can be separated into three groups according to the strength of their binding to DNA: H1 < H2B + H2A < H3 + H4. It is suggested that histone octamer is not a conservative formation and the histone content of nucleosomes may change under physiological conditions being affected by competition of chromatin components for DNA.

RESUMO

New methods of radiometry and fractionation of cell lysates on hydroxyapatite were applied to determine the crosslinkage between DNA and protein induced by embiquin in cell culture of normal human skin fibroblasts. Such crosslinks were found to be formed; they were eliminated in long-term cultivation of the cells after mutagen treatment.
