RESUMO

The objective of this study was to determine the effects of different lipid sources, with or without a probiotic, on the gastrointestinal tract, immune system and blood parameters of Ross 308 male chickens. In this study, 360 one-day-old chickens were randomly allotted to six treatments with six replicates. Experimental diets were: (1) control (CTL); (2) a diet containing 30 g/kg lipid from tallow (CTL+TLW); (3) a diet containing 30 g/kg lipid from soybean oil (CTL+SO); (4) the basal diet plus a probiotic (CTL+PRO), (5) a diet containing 30 g/kg tallow plus probiotic (TLW+PRO); and (6) a diet containing 30 g/kg soybean oil plus probiotic (SO+PRO). The percentage of liver and jejunum in the treatments that used tallow alone or tallow with probiotics had a significant increase as compared to the control. The villus height and crypt depth of the ileum and villus height/crypt depth in the treatments that used soybean oil and probiotic alone had a significant increase compared to the control. The weight of the spleen, bursa of Fabricius, and thymus in the treatments that used probiotics had a significant increase compared to the control. The amount of alkaline phosphatase and alanine aminotransferase as well as triacylglycerol in the treatment containing probiotic and its mixture with soybean oil had the least significant difference with the control. The results showed that the use of soybean oil, probiotics, and their mixture can improve intestinal morphology, strengthen the immune system, and reduce liver enzymes in chickens.

RESUMO

In the present investigation, the mechanical properties of mouse normal and carcinomatous (LL/2) lung tissue cells were investigated using atomic force microscopy (AFM). The normal lung cells have been derived directly from C57BL mice. Initially, the elastic modulus of LL/2 cells was measured following chemotherapy with the anti-cancer drug Cisplatin and plasma treatment. MTT evaluation was used to determine the optimal dosages for 24- and 48-h incubations based on the IC50 cell viability concentration during chemotherapy treatment. After 24 and 48 h, the results demonstrated that Cisplatin-based chemotherapy increases the elastic modulus of LL/2 cells by 1.599 and 2.308 times compared to untreated cells. LL/2 cells were subsequently treated with plasma for 30 and 60 s for 24 and 48-h incubation. The plasma treatment decreased the LL/2 cell's elastic modulus, and the time duration of plasma treatment increased the reduction amount of elastic modulus. During the second section of the study, theoretical (finite element analysis [FEM]) and experimental techniques were used to examine the resonant frequencies and magnitude of the frequency response function (FRF) of the AFM cantilever's movements when applying normal and cancerous cells before and after chemo and plasma treatments as specimens. The results indicated that increasing the samples' elastic modulus raises the resonant frequency, so the resonant frequency of treated cells as a sample is greater than untreated cells. In conclusion, the FEM and experimental results were compared and found to be in good agreement.

RESUMO

In the present investigation, the mechanical properties of normal and carcinomatous cells of kidney tissue (HEK-293, ACHN, respectively) were investigated using atomic force microscopy (AFM). Initially, the elastic modulus of ACHN cells was measured following chemotherapy with the anti-cancer drug Cisplatin and plasma treatment. The MTT assay was employed to ascertain the most effective dosages for incubation periods of 12, 24, 48, 72, and 96 h, guided by the IC50 concentration for cell viability during chemotherapy treatment. Analysis at these specified time points revealed a progressive increase in the elastic modulus of ACHN cells when subjected to Cisplatin-based chemotherapy. Specifically, the elastic modulus increased by 1.847, 4.416, 6.035, 8.029, and 9.727 times in comparison to untreated cells at 12, 24, 48, 72, and 96 h, respectively. ACHN cells were subsequently treated with plasma for 30 and 60 s for 24 and 48-h incubation periods. The plasma treatment increased the ACHN cell's elastic modulus. In the subsequent phase of the research, a combination of theoretical (finite element method [FEM]) and experimental methodologies was employed to investigate the resonant frequencies and magnitude of the frequency response function (FRF) concerning the movement of the AFM cantilever. This examination was conducted using ACHN cells as specimens, both before and after exposure to chemotherapy and plasma treatments. The results showed that higher sample elastic modulus increased the resonant frequency, indicating that treated cells had a higher resonant frequency than untreated cells. In conclusion, the FEM and experimental results were compared and found to be in good agreement. HIGHLIGHTS: Using Cisplatin anti-cancer drug increases the elastic modulus of ACHN cell. Applying plasma treatment increases the elastic modulus of ACHN cell. For both of the chemo and plasma therapies, increasing the incubation time increases the influence of therapies oh the cell mechanics. Using finite element modeling (FEM) the real dynamic behavior of atomic force microscope cantilever by considering human kidney cells as the soft samples is possible.

RESUMO

BACKGROUND: Although regional wall motion abnormality (RWMA) detection is foundational to transthoracic echocardiography, current methods are prone to interobserver variability. We aimed to develop a deep learning (DL) model for RWMA assessment and compare it to expert and novice readers. METHODS: We used 15,746 transthoracic echocardiography studies-including 25,529 apical videos-which were split into training, validation, and test datasets. A convolutional neural network was trained and validated using apical 2-, 3-, and 4-chamber videos to predict the presence of RWMA in 7 regions defined by coronary perfusion territories, using the ground truth derived from clinical transthoracic echocardiography reports. Within the test cohort, DL model accuracy was compared to 6 expert and 3 novice readers using F1 score evaluation, with the ground truth of RWMA defined by expert readers. Significance between the DL model and novices was assessed using the permutation test. RESULTS: Within the test cohort, the DL model accurately identified any RWMA with an area under the curve of 0.96 (0.92-0.98). The mean F1 scores of the experts and the DL model were numerically similar for 6 of 7 regions: anterior (86 vs 84), anterolateral (80 vs 74), inferolateral (83 vs 87), inferoseptal (86 vs 86), apical (88 vs 87), inferior (79 vs 81), and any RWMA (90 vs 94), respectively, while in the anteroseptal region, the F1 score of the DL model was lower than the experts (75 vs 89). Using F1 scores, the DL model outperformed both novices 1 (P = .002) and 2 (P = .02) for the detection of any RWMA. CONCLUSIONS: Deep learning provides accurate detection of RWMA, which was comparable to experts and outperformed a majority of novices. Deep learning may improve the efficiency of RWMA assessment and serve as a teaching tool for novices.

RESUMO

This study was conducted to evaluate the effects of different doses of selenium (Se) from Sel-Plex© (selenium-enriched Saccharomyces cerevisiae yeast) supplement on the antioxidant status, the antibody titers against the foot-and-mouth disease virus, and the expression of interleukin-2 (IL-2) and interferon-γ (IFN-γ) genes in ewes during the hot season. Six ewes were kept at 25 °C and received basal diet (the negative control group), and 24 ewes were kept at 38 °C for 5 h per day and received no supplement (the positive control), 0.15, 0.30, and 0.45 mg Se/kg. Ewes in the positive control had higher (P<0.001) liver enzyme activity, malondialdehyde (MDA), and cortisol levels, and lower antibody titer than the negative control. The liver enzymes' lowest (P<0.001) activities were observed in ewes receiving 0.30 and 0.45 mg Se/kg. Ewes receiving 0.30 and 0.45 mg Se/kg had lower MDA levels than other treatments. Ewes receiving 0.30 and 0.45 mg Se/kg had higher (P<0.001) total antioxidant capacity levels than those receiving 0.15 mg Se/kg and the positive control. Se-supplemented groups had lower (P<0.001) relative expression of IL-2 and higher (P<0.04) expression of IFN-γ than the positive control. The antibody titer was the same in the positive control and the group receiving 0.15 mg Se/kg. Ewes fed a diet with 0.30 and 0.45 mg Se/kg had higher (P<0.011) antibody titer than the positive control. The Se supplementation can reverse the decrease of antioxidant capacity and immune function caused by heat stress, and 0.3 mg Se/kg from Sel-Plex©is the best dose.

RESUMO

In cis and trans geometrical configurations of the polyacetylene molecule, one-dimensional chain is constructed by attaching a number of identical -HC=CH- units one-by-one. We attach as many units as required to obtain the chain of the desired length. In case of a very long polyacetylene chain, which is practically considered infinite in length, a periodic unit is defined, so that its band structure would be calculable. Then, the electronic properties and topological properties of the chain can be predicted. Since experimental synthesis of single-layer polyacetylene chain has lots of limitations, in an alternative approach, emulation of a tight-binding model is used to describe the electron transfer in polyacetylene polymer chain. In case of either synthesis or testing the polyacetylene molecule, it is necessary to improvise a one-to-one correspondence between polyacetylene polymer and topological circuit, which is introduced for the first time in the present study. To this aim, the outputs of density functional theory calculations alongside with the calculations based on the physical chemistry formalisms are used. Here, we observed that the electronic response of the circuit is topologically sustained at frequencies where the coupling was pre-determined via high precision quantum system equivalent topolectrical circuit, as an alternative classical system, to study electron transfer of trans-polyacetylene polymer quantum chain by the precision of one-electron.

RESUMO

Each anti-cancer drug has special effects on the target cells. One of the most important reasons to recommend an anti-cancer drug is related to the influences of it on the mechanical properties of the target cells. In this study, the effects of cetuximab and cisplatin anti-cancer drugs on the mechanical properties of A-549 and Calu-6 cells as the cancerous lung cells have been investigated. For both of the cells and anti-cancer drugs, MTT assessment has been used to define the convenient dosages for 24 and 48 h incubations based on IC50 concentration for the cell line viability. The mechanical specifications of the cells before and after treatment were obtained using nanoindentation by the JPK Instruments' NanoWizard3 atomic force microscope. The results show that cetuximab increases the stiffness of A-549 cell from 1225 to 3403 and 12 690 Pa for 24 and 48 h incubations. The influence of cetuximab on the Calu-6 shows that the elastic modulus after 24 and 48 h culture times increases about cisplatin anti-cancer drug, for A-549 cell indicates that the elastic modulus rises from 1225 to 1506 and 2375 Pa for 24 and 48 h, respectively. For Calu-6 cell, cisplatin has an important role to increase the stiffness of the cell. Applying cisplatin increases the elastic modulus from 33 to 682.8 Pa for 24 h and 1105 Pa after 48 h incubations.

RESUMO

This study was aimed to evaluate the effects of vegetable oils as calcium salt on immune responses and the expression of immune-related genes in vaccinated lambs. Twenty-four lambs (35 kg body weight, 6 months old) were assigned to four treatments with six replicates in a completely randomized design for 40 days. Four concentrates were formulated in which the calcium salts of palm oil, canola oil, corn oil, and flaxseed oil were used. On day 30 of the experiment, lambs were vaccinated by a dose of foot-and-mouth disease virus. The blood samples were collected from jugular vein 10 days after vaccination. The level of malondialdehyde and the activity of liver enzymes were the highest in lambs receiving corn oil and the lowest in lambs receiving flaxseed oil. The highest lymphocytes and the lowest neutrophil percentages were observed in lambs receiving flaxseed oil. There was a significant difference among treatments for the relative genes expression. Flaxseed oil significantly upregulated interferon-γ and corn oil upregulated interleukin-1ß. The highest titer against foot-and-mouth disease virus was related to lambs receiving flaxseed oil, and the lowest titer was related to lambs that received corn oil. Flaxseed oil had more beneficial effects on immune response than other oils.

RESUMO

This study was implemented to evaluate the effects of different zinc doses as Zinc-Met supplement (Zinpro®) on the antioxidant status, blood immune cells, antibody titers, and the expression of IL-4 and IL-6 genes of ewes in the hot season. In a completely randomized design, 24 ewes were assigned to treatments as follow: 0, 15, 30 and 45 mg/kg zinc as Zinc-Met supplementation for 40 days in region with 40 °C and vaccinated against food-and-mouth disease as an immune challenge at day 30, and then blood samples were collected on day 40. Ewes were fed a basal diet containing 29.9 mg zinc/kg. The highest activity of the antioxidant enzyme and the lowest lipid peroxidation values were found in ewes receiving 30 and 45 mg/kg zinc following a linear trend. The highest lymphocytes count and antibody titers were found in ewes received 30 mg zinc/kg. There were no significant differences among treatments for the relative expression of genes. In overall, zinc supplementation non-significantly up-regulate interleukin-4 and down-regulate interleukin-6. It was concluded that zinc supplementation as Zinc-Met could enhance the antioxidant status and immune response of ewes under heat stress; supplementation of diet with 30 mg zinc/kg (300 mg/kg Zinpro®) appeared to be the most effective dose.

RESUMO

Nowadays, the utilization and production of polymeric nanofibers are expanding rapidly due to their unique and exclusive properties. The current study examined silk fibroin nanofibers, poly-L lactic acid (PLLA), and polycaprolactone (PCL) as three widely-used biopolymers. In the first section, the mechanical properties of nanobiofibers, including elastic modulus as the first research variable, were investigated using atomic force microscopy (AFM)-based nanoindentation and tensile testing (through a universal testing machine). The results indicated that increasing the concentration of the biopolymer solution increased its elastic modulus. The total amount of elastic modulus for silk fibroin (SF), PCL, and PLLA nanofibers with random and aligned scaffold formats ranged from 1.207 MPa to 3.71 MPa. Based on the molecular weight and concentrations of the solutions, PLLA, PCL, and silk fibroin nanofibers yielded the highest elastic modulus, respectively. In addition, the elastic modulus was greater in an aligned scaffold as opposed to a random format and in retraction as opposed to an extended stroke. During the second section of the study, resonant frequencies as the second research variable and the amplitude of the FRF of the AFM beam's vertical movements using nanobiopolymers with various scaffolds as the samples were investigated theoretically (using the Finite Element Method [FEM]) and experimentally (using AFM). For the first to third modes, the resonant frequency of the AFM beam considering biopolymer nanofibers as samples varied from 12658.43 Hz to 241208.4 Hz. The results revealed that increasing the elastic modulus of the samples increases the resonant frequency. The mechanical and vibrational properties of nanofibers calculated by various methods were subsequently compared separately. Overall, the comparison indicated good agreement between the properties.

RESUMO

Today, in developing countries, women's need to be present in urban public spaces and the establishment of everyday social interactions seem to be ignored more than other groups. Therefore, the present study aims to achieve the environmental components related to women's presence in urban spaces and prioritize them. In this study, by scrutinizing the criteria obtained from the interviews, a questionnaire was prepared and randomly distributed among 256 women in the population. IBM SPSS has been used to analyze the data and explain the priorities. The results of the T-test show that security (T-value = 6.508 in T1 test), compatibility with behavioral patterns (T-value = 4.975 in T2 test), eventuality (T-value = 11.064 in T3 test), permeability (T-value = 10.220 in T4 test), attention to climate (T-value = 5.692 in T5 test), liberty (T-value = 11.184 in T6 test), collective memory (T-value = 7.367 in T7 test), variety (T-value = 1.816 in T8 test), complexity (T-value = 13.228 in T9 test), and identity (T-value of 18.905 in T10 test) are the most important criteria in motivating presence in urban public spaces, respectively. According to the results, among the individual characteristics of the respondents, the components of collective memory (r = 0.805), identity (r = 0.784), liberty (r = 0.703), and safety and security (r = 0.644) have had a positive correlation with the age of the respondents. The results of this study indicated that improving security (individual, social, and psychological) is the main environmental priority for women over 18 to be present in urban public spaces.

RESUMO

This study was done to investigate the effects of thymol, fumagillin, oxalic acid (Api-Bioxal) and hops extract (Nose-Go) on Nosema sp. spore load, the expression of vitellogenin (vg) and superoxide-dismutase-1 (sod-1) genes and mortality of bees infected with N. ceranae. Five healthy colonies were assigned as the negative control, and 25 Nosema sp. infected colonies were assigned to five treatment groups including: the positive control: no additive to sirup; fumagillin 26.4 mg/L, thymol 0.1 g/L, Api-Bioxal 0.64 g/L and Nose-Go 5.0 g/L sirup. The reduction in the number of Nosema sp. spores in fumagillin, thymol, Api-Bioxal and Nose-Go compared to the positive control was 54, 25, 30 and 58%, respectively. Nosema sp. infection in all infected groups increased (p < .05) Escherichia coli population compared to the negative control. Nose-Go had a negative effect on lactobacillus population compared to other substances. Nosema sp. infection decreased vg and sod-1 genes expression in all infected groups compared to the negative control. Fumagillin and Nose-Go increased the expression of vg gene, and Nose-Go and thymol increased the expression of sod-1 gene than the positive control. Nose-Go has the potential to treat nosemosis if the necessary lactobacillus population is provided in the gut.

RESUMO

This study aimed to investigate the effects of grains, and protein sources in the starter diet on daily gain, pro- and anti-inflammatory genes expression, ruminal volatile fatty acid (VFA) concentration, and blood metabolites in the dairy calves. Forty Holstein calves were randomly assigned to treatments in a 2 × 2 factorial arrangement with the factors of grain source (steam-flaked corn, CG vs. steam-rolled barley, BG) and pelleted protein source (soybean meal, SBM vs. canola meal, CM). The daily gain of calves who fed with SBM was higher than those fed with CM. Total VFA concentration in the rumen of calves fed with CG was greater than BG at day 35 (p < 0.05), and calves receiving SBM had higher total VFA concentration than calves fed with CM. The relative expression of tumor necrosis factor-α and interleukin-1ß genes were significantly decreased in the calves fed with BG and CM compared to calves fed with CG and SBM. Calves receiving the starter diet based on BG had the highest (p < 0.05) gene expression of interferon-γ. Feeding calves with SBM is recommended because it resulted in a greater daily gain than CM. Complete replacement of SBM with CM, and CG with BG is not recommended in dairy calves.

RESUMO

The ability to adapt and conform to angular and uneven surfaces improves the suction cup's performance in grasping and manipulation. However, in most cases, the adaptation costs lack of required stiffness for manipulation after surface attachment; thus, the ideal scenario is to have compliance during adaptation and stiffness after attachment to the surface. Inspired by the capability of stiffness regulation in octopus suction cup, this article presents a suction cup that adapts to steep angular surfaces due to compliance and has high stiffness after attachment. In this design, the stiffness after attachment is provided by using granular jamming as vacuum driven stiffness modulation. Thus, the design is composed of a conventional active suction pad connected to a granular stalk, emulating a hinge behavior during adaptation and creating high stiffness by jamming granular particles driven by the same vacuum as the suction pad. During the experiment, the suction cup can adapt to angles up to 85° with a force lower than 0.5 N. We also investigated the effect of granular stalk's length on the adaptation and how this design performs compared to passive adaptation without stiffness modulation.

RESUMO

Terahertz (THz) technology is developing a non-invasive imaging system for biosensing and clinical diagnosis. THz medical imaging mainly benefits from great sensitivity in detecting changes in water content and structural variations in diseased cells versus normal tissues. Compared to healthy tissues, cancerous tumors contain a higher level of water molecules and show structural changes, resulting in different THz absorption. Here we described the principle of THz imaging and advancement in the field of translational biomedicine and early detection of pathologic tissue, with a particular focus on oncology. In addition, although the main forte of THz imaging relies on detecting differences in water content to distinguish the exact margin of tumor, THz displays limited contrast in living tissue for in-vivo clinical imaging. In the last few years, nanotechnology has attracted attention to aid THz medical imaging and various nanoparticles have been investigated as contrast enhancements to improve the accuracy, sensitivity, and specificity of THz images. Most of these multimodal contrast agents take advantage of the temperature-dependent of THz spectrum to the conformational variation of the water molecule. We discuss advances in developing THz contrast agents to accelerate the advancement of non-invasive THz imaging with improved sensitivity and specificity for translational clinical oncology.

RESUMO

This study was conducted to compare nutrient digestibility, performance and immune response of dairy cows received live and autolyzed yeast during the transition period in high ambient temperature. Cows (n = 25) were randomly divided and received a basal diet with or without live yeast or autolyzed yeast as on top three weeks pre-parturition until three weeks post-parturition. The Control group received a basal diet without yeast products; other groups received 0.5 g live yeast; 1.0 g live yeast; 10 g autolyzed yeast and 20 g/d/head autolyzed yeast. Live yeast resulted in higher nutrient digestibility compared with autolyzed yeast and the control. Methane production was the highest in autolyzed yeast and the lowest in live yeast. Average milk production was the highest in cows that received live yeast. The highest IgG level was for cows that received autolyzed yeast at a dose of 20 g/d/head. Live yeast had no significant effect, but autolyzed yeast increased the relative expression of γ-Interferon and interleukin-2 as compared with the control group. It was concluded that live yeast at a dose of 1.0 g/d/head could influence ruminal fermentation and milk production, but autolyzed yeast at a dose of 20 g/d/head could influence the immune response of dairy cows during the transition period and heat stress.

RESUMO

Background: Clomiphene citrate (CC) has been suggested to increase the chance of sperm retrieval withmicrodissection testicular sperm extraction (micro-TESE). Objective: This study aimed to evaluate the effect of CC on micro-TESE results, due to the great controversy in this regard. Materials and Methods: 112 participants were included in this cross-sectional study and were divided into a case (n = 54) and a control group (n = 58) diagnosed with non-abstractive azoospermia. The case group received 25 mg of CC daily for 3 months, while the control group did not receive anything. All participants underwent micro-TESE by an andrologist, and at the end, the results were compared between groups. Hormone tests, including follicle-stimulating hormone, luteinizing hormone, testosterone, and prolactin were analyzed. Results: The mean age of participants was the same in the case and the control groups, and no significant relationship was observed between the 2 groups (p = 0.16). 25.9% of sperm and 31.0% of sperm were observed and extracted in the CC-treated and the control group, respectively. Conclusion: Our findings showed that after receiving CC, the number of sperm extraction did not increase but it rather decreased. However, the initial level of hormones such as testosterone, follicle-stimulating hormone, luteinizing hormone, and prolactin, and the men's age, testicle size, smoking, and opium addiction, underlying diseases had no significant relationship in the 2 groups and did not affect the results.

RESUMO

The neighborhood is one of the most fundamental urban elements and acts as the intermediary link between the city and citizens to enhance the quality of life. The present study examined the significance of the relationship between the subjective well-being of citizens and perceived neighborhood environment characteristics in urban historical fabrics for creating healthy neighborhoods. To this end, a survey research method was employed, and the data were collected via questionnaires. The population consisted of all the citizens of the historical neighborhood of Sange Siah in Shiraz, Iran, who lived or worked in the neighborhood and used the neighborhood spaces daily. A Nonparametric Spearman correlation coefficient was run to assess the correlation between the variables. The results showed that the component of social inclusion from among the six components of subjective well-being had a significant positive correlation with perceived neighborhood environment characteristics (r = 0.712). In the following, the components of satisfaction with life (0.614), mental well-being (0.569), positive and negative effect (0.526), and feeling of happiness (0.468) had a moderate positive correlation; and the component of physical and mental health also had a weak positive correlation with perceived neighborhood environment characteristics (0.230). In addition, the concept of subjective well-being with a correlation coefficient of 0.579 had a moderate positive correlation with perceived neighborhood environment characteristics, which indicates that the structural characteristics of the neighborhood have a significant relationship with the subjective well-being of the people living in the neighborhood.

RESUMO

This study was done to assess the effects of hydrolyzed yeast on blood cells counts, antioxidant status and gene expression of pro-inflammatory and anti-inflammatory cytokines of vaccinated dairy cows. Twenty Holstein lactating dairy cows were randomly divided into four groups received basal diet without (control) and with 10, 20 and 30 g/d/head of hydrolyzed yeast. Cows were vaccinated against foot and mouth disease virus at day 10 and blood samples were taken 10 days after. Cows received hydrolyzed yeast at doses of 20 and 30 g/d/head had the lowest and those in the control group had the highest malondialdehyde concentration. Cows received hydrolyzed yeast at dose of 20 g/d/head had the highest lymphocyte and the lowest neutrophil/lymphocyte ratio. IL-1ß and TNF-α downregulated and INF-γ up-regulated in cows received different doses of hydrolyzed yeast. The lowest expression of IL-1ß and TNF-α and the highest expression of INF-γ was for cows received hydrolyzed yeast at dose of 20 g/d/head. The highest titers against foot and mouth disease virus were for cows received hydrolyzed yeast at dose of 20 g/d/head and the lowest titer was for the control group. Supplementation of 20 g/d/head hydrolyzed yeast recommended in vaccinated cows.
