RESUMO

Barley scientifically known as Hordeum vulgare (HV) is a major grain crop. Over the course of time, great interest has been developed in the usage of barley, because of its various pharmacological activities. Current study is designed to determine the chemical constituents of Hordeum vulgare (HV) seed extract by GC-MS technique, and Invitro antioxidant assays i.e. 1,1-diphenyl-2-picryl-hydrazyl free radical (DPPH) and 2-azino-bis(3-ethyl benzthiazoline-6-sulfonic acid) (ABTS) methods. GC-MS identified 16 non-polar compounds in the hexane extract of HV plant, which includes carboxylic acid (6.25%), fatty acid (37.5%), carboxylic acid amide derivative of fatty acid (6.25%), triterpinoids (18.75%), fat soluble vitamin (6.25%), phytosterol (6.25%), stigmastanes (6.25%), beta diketones (6.25%), and cycloartenol (6.25%) respectively. The major compound includes Hexadecanoic acid, methyl ester (6.84%), n-Hexadecanoic acid (8.58%), 9,12-Octadecanoic acid (Z,Z)-, Methyl Ester (8.04%), 9,12-Octadecadienoic acid (Z,Z) (57.01%), Lup-20(29)-en-3-one (3.57%), γ-Sitosterol (3.31%). Some constituents such as Lup-20(29)-en-3-one, campesterol and squalene were observed and were not previously reported. Total phenolic and total flavonoid content were determined using spectrophotometric technique and calculated as gallic acid equivalents GAE/g dry weight and rutin equivalent RE/g of dry weight respectively.The highest phenolic content exhibited by the acetone extract of HV seedsi.e. 0.0597 mg GAE/g while the highest flavonoid content exhibited by dichloromethane extract i.e. 0.09 mg RE/g and 0.25 mg QE/g of dry weight respectively. All the extracts showed significant antioxidant activity in DPPH and ABTS cation decolorization assays. Methanol and dichloromethane extract showed the highest DPPH radical scavenging activity i.e. 52.41% and 42.07% at the concentration of 100 mg/ml respectively. Moreover, the IC50 has been determined by the acetone and methanol extract of HV seeds. The high antioxidant activity of its seed extracts has made this plant pharmacologically important. Conclusively, there is a vast scope to further explore the active principals of barley so that more of its pharmacological properties can be identified.

The objective of the present study was to determine the acute and subacute toxicity profile of a polyherbal formulation called "Goubion" in addition to the in vivo antihyperuricemic study using fructose-induced hyperuricemia. Goubion is a combination of Colchicum autumnale (tuber), Tribulus terresteris (fruit), Vitex negundo (leaves), Smilax chinensis (root), Glycyrrhiza glabra (root), and Curcuma amada (rhizome). The acute toxicity study revealed no signs of mortality and morbidity at a single dose of 2000 mg/kg. Similarly, the results of the subacute repeated dose toxicity study exhibited no signs of mortality at any of the doses. However, significant changes in hematological, biochemical, and renal parameters were recorded at the dose of 60 mg/kg. Antihyperuricemic activity was tested at the dose of 15 mg/kg and 20 mg/kg of Goubion, respectively against 5 mg/kg Allopurinol. Based on the antihyperuricemic study, we infer that the Goubion has a significant hypouricemic action, as it remarkably decreased the elevated uric acid levels. The results also suggest the potential inhibitory capability of Goubion on xanthine oxidase dehydrogenase might be the mechanism behind the hypouricemic effect.

Objectives: To determine risk factors of cervical cancer and role of healthcare providers regarding awareness and counseling of PAP-smear. Methods: It was case-control study conducted from Jan-2021 till may-2021 at two tertiary care hospitals of Karachi South. The intended sample size was 255 subjects as 105 cases and 150 controls. The inclusion criteria were diagnosed cases of CA Cervix and under the age of 50 years. Controls were recruited from the general population. All the data were entered into SPSS version 23. The odds ratio was calculated to compare the risks of occurring CA cervix among cases and controls. The relationship of risk factors was assessed by binary logistic regression. P-value < 0.05 was considered as statistically significant. Results: Generally, highly significant p-value (<0.000) was observed; depicted a positive association for a level of education, age at first intercourse, and number of parity in cases of CA cervix and controls (OR=4.3). The correct predicted rate was 68.8% for having CA cervix among controls due to family history, the knowledge of PAP smear screening, ever counseled for a PAP smear, ever tested for a PAP smear, never been tested for PAP smear due to cost. Conclusion: Educational level, age at first intercourse, number of parity was the risk factors of CA cervix. Family history of CA Cervix, knowledge of cervical cancer screening, ever counseled for PAP smear, ever tested for PAP smear, never been tested for PAP smear due to cost were significantly predicted for CA cervix among controls.

RESUMO

We detected Crimean-Congo hemorrhagic fever virus infections in 4 provinces of Pakistan during 2017-2018. Overall, seroprevalence was 2.7% in humans and 36.2% in domestic livestock. Antibody prevalence in humans was highest in rural areas, where increased contact with animals is likely.

Middle East Respiratory Syndrome Coronavirus (MERS-CoV) is a zoonotic pathogen capable of causing severe respiratory disease in humans. Although dromedary camels are considered as a major reservoir host, the MERS-CoV infection dynamics in camels are not fully understood. Through surveillance in Pakistan, nasal (n = 776) and serum (n = 1050) samples were collected from camels between November 2015 and February 2018. Samples were collected from animal markets, free-roaming herds and abattoirs. An in-house ELISA was developed to detect IgG against MERS-CoV. A total of 794 camels were found seropositive for MERS-CoV. Prevalence increased with the age and the highest seroprevalence was recorded in camels aged > 10 years (81.37%) followed by those aged 3.1-10 years (78.65%) and ≤ 3 years (58.19%). Higher prevalence was observed in female (78.13%) as compared to male (70.70%). Of the camel nasal swabs, 22 were found to be positive by RT-qPCR though with high Ct values. Moreover, 2,409 human serum samples were also collected from four provinces of Pakistan during 2016-2017. Among the sampled population, 840 humans were camel herders. Although we found a high rate of MERS-CoV antibody positive dromedaries (75.62%) in Pakistan, no neutralizing antibodies were detected in humans with and without contact to camels.

BACKGROUND: Reject analysis can be used as a quality indicator, and is an important tool in localizing areas where optimization is required. Reducing number of rejects is important yielding reduced patient exposure and increased cost-effectiveness. PURPOSE: To determine rejection rates and causes in direct digital radiography. MATERIAL AND METHODS: Data were collected during a three-month period in spring 2010 at two direct digital laboratories in Norway. All X-ray examinations, types, numbers, and reasons for rejections were obtained using automatic reject analysis software. Thirteen causes for rejection could be selected. RESULTS: Out of the 27,284 acquired images, 3206 were rejected, yielding an overall rejection rate of 12%. Highest rejection rates were found for examination of knees, shoulders, and wrist. In all, 77% of the rejected images arose from positioning errors. CONCLUSION: An overall rejection rate of 12% indicates a need for optimizing radiographic practice in the department.

Prunes are used by folks as a remedy of various diseases including hepatitis. A clinical trial was designed to see the effects of prunes (Prunus domestica) on liver function. 166 healthy volunteers were divided into three groups randomly. Either three (about 11.43g) or six (23g approx.) prunes were soaked in a glass of water (250ml) overnight. Each subject from two test groups was asked to drink prune juice & eat whole fruit(single or double dose of prunes) as well, early in the morning, daily for 8 weeks; whereas each subject from control group was given a glass of water to drink. Blood samples were taken at week 0 and week 8 for chemical analysis. There was significant reduction of serum alanine transaminase (p 0.048) and serum alkaline phosphatase (p 0.017) by the lower dose of prunes. There was no change in serum aspartate transaminase and bilirubin. Alteration in liver function by use of prunes may have clinical relevance in appropriate cases and prunes might prove beneficial in hepatic disease.

BACKGROUND: Fruits and vegetables are shown to reduce blood pressure. It is not merely the antioxidants contained in fruits and vegetables that have health benefits such as lowered systolic and diastolic blood pressures. This study was undertaken to see the cardiovascular protective effects of prunes. METHODS: A placebo controlled clinical trial study was designed to see the effects of Prunus domestica on blood pressure in 259 pre-hypertensive (Systolic BP = 120-139 mmHg, diastolic BP = 80-89 mmHg) volunteers. Treated groups drank prune juice and ate the whole fruit (dried plums) while either 3 (about 11.5 gm) or 6 prunes were soaked overnight in a glass of water whereas control group took only a glass of plain water early in the morning on empty stomach. Blood pressure was recorded fortnightly for 8 weeks, and blood samples were taken at 0 and 8 weeks. RESULTS: There was significant reduction of blood pressure by single dose of prunes daily group and the controls (p < 0.05). With the double dose of prunes, only systolic BP was reduced significantly (p < 0.05). Control group had significantly increased serum HDL whereas test groups had significantly reduced serum cholesterol and LDL (p < 0.05). Data was analysed by paired-sample t-test with 95% confidence interval. CONCLUSION: The data predicts cardiovascular protective effects of prunes.
