Androdioecy is a rare reproductive system. Fraxinus platypoda, a woody canopy species in Japan's mountainous riparian zones, is described as a morphologically androdioecious species. In this study, we tried to detect whether F. platypoda is also functionally androdioecious. We analyzed its sexual expression, seed development, pollen morphology and germination ability, pollination systems, and mast flowering behavior. We found that the hermaphrodite trees are andromonoecious, with inflorescences bearing male and hermaphroditic flowers, whereas male individuals had only male flowers. Pollen morphology was identical in male flowers, in hermaphrodite flowers of an andromonoecious individual, and in male flowers of male individuals. Pollen from both types of individuals was capable of germination both ex vivo (on nutrient medium) and in vivo in pollination experiments. However, compared with pollen from andromonoecious trees, pollen from male trees showed a higher germination rate. The self-pollination rate of bagged hermaphroditic flowers was almost zero. The fruit set rate following cross-pollination with male pollen from a male tree was higher than that following natural pollination, whereas the rate with hermaphroditic pollen was the same. The flowering and fruiting of F. platypoda have fluctuated over 17 years; the flowering of the two types of sexual individuals exhibited clear synchronization during this period. The frequency of male individuals within the populations is 50%. The maintenance of such a proportion of males in populations of the self-incompatible F. platypoda is either indicative of a true androdioecious species with a diallelic self-incompatibility system or a cryptic-dioecious species. Relict species play an important role in understanding the biogeography of intercontinental disjunctions. Pterocarya (a relict genus) is the valuable model taxon for studying the biogeography of East Asian versus southern European/West Asian disjunct patterns. This disjunction has not been as well studied as others (e.g., between Eastern Asia and North America). Several phylogenetic studies on Pterocarya have been conducted, but none have provided a satisfactory phylogenetic resolution. Here, we report the first well-resolved phylogeny of Pterocarya using restriction site-associated DNA sequencing data based on the sampling of all taxa across the entire distribution area of the genus. Taxonomic treatments were also clarified by combining morphological traits. Furthermore, fossil-calibrated phylogeny was used to explore the biogeography of Pterocarya. Our results support the existence of two sections in Pterocarya, which is in accordance with morphological taxonomy. Section Platyptera comprises three species: P. rhoifolia, P. macroptera, and P. delavayi. Section Pterocarya also comprises three species: P. fraxinifolia, P. hupehensis, and P. stenoptera. The divergence between the two sections took place during the early Miocene (20.5 Ma). The formation of the Gobi Desert and climate cooling of northern Siberia in the Middle Miocene (15.7 Ma) might have caused the split of the continuous distribution of this genus and the formation of the East Asian versus southern European/West Asian disjunct pattern. Lastly, the divergence between P. hupehensis and P. stenoptera as well as between P. rhoifolia and P. macroptera/P. delavayi (10.0 Ma) supports the late Miocene diversification hypothesis in East Asia.

Climate change is a major cause of changes in alpine and polar vegetation, particularly at the edges of distributions. In temperate regions, these changes are expected to occur at the timberline of alpine zones. On Mt. Fuji, the highest mountain in Japan, the timberline is located 2400-2500 m above sea level. Over a 40-year period (1978-2018), we researched changes in the timberline vegetation of Mt. Fuji. A permanent belt transect extending from the upper timberline to subalpine zones was set up in August 1978. Tree diameters and heights were recorded at the establishment of the transect and every 20 years afterwards. Over the 40 years of the study, the timberline advanced rapidly upwards, and the degree of vegetation cover above the timberline increased remarkably. Notably, the expansion of Salix reinii into the upper part of the timberline facilitated the subsequent spread of Larix kaempferi into this zone. Seedlings of L. kaempferi were particularly abundant at the upper timberline and became established on the uppermost part of the slope. The shape of L. kaempferi at the upper timberline changed from a prostrate form to an upright tree form. We conclude that the upward advance of the alpine timberline observed on Mt. Fuji is due to climate change.

How phenotypic or genetic diversity is maintained in a natural habitat is a fundamental question in evolutionary biology. Flower color polymorphism in plants is a common polymorphism. Hepatica nobilis var. japonica on the Sea of Japan (SJ) side of the Japanese mainland exhibits within population flower color polymorphism (e.g., white, pink, and purple), while only white flowers are observed on the Pacific Ocean (PO) side. To determine the relationships between flower color polymorphism, within and among populations, and the genetic structure of H. nobilis var. japonica, we estimated the genetic variation using simple sequence repeat (SSR) markers. First, we examined whether cryptic lineages corresponding to distinct flower colors contribute to the flower color polymorphisms in H. nobilis var. japonica. In our field observations, no bias in color frequency was observed among populations on Sado Island, a region with high variation in flower color. Simple sequence repeat (SSR) analyses revealed that 18% of the genetic variance was explained by differences among populations, whereas no genetic variation was explained by flower color hue or intensity (0% for both components). These results indicate that the flower color polymorphism is likely not explained by cryptic lineages that have different flower colors. In contrast, populations in the SJ and PO regions were genetically distinguishable. As with the other plant species in these regions, refugial isolation and subsequent migration history may have caused the genetic structure as well as the spatially heterogeneous patterns of flower color polymorphisms in H. nobilis var. japonica.

East Asia's temperate deciduous forests served as sanctuary for Tertiary relict trees, but their ages and response to past climate change remain largely unknown. To address this issue, we elucidated the evolutionary and population demographic history of Cercdiphyllum, comprising species in China/Japan (Cercdiphyllum japonicum) and central Japan (Cercdiphyllum magnificum). Fifty-three populations were genotyped using chloroplast and ribosomal DNA sequences and microsatellite loci to assess molecular structure and diversity in relation to past (Last Glacial Maximum) and present distributions based on ecological niche modelling. Late Tertiary climate cooling was reflected in a relatively recent speciation event, dated at the Mio-/Pliocene boundary. During glacials, the warm-temperate C. japonicum experienced massive habitat losses in some areas (north-central China/north Japan) but increases in others (southwest/-east China, East China Sea landbridge, south Japan). In China, the Sichuan Basin and/or the middle-Yangtze were source areas of postglacial northward recolonization; in Japan, this may have been facilitated through introgressive hybridization with the cool-temperate C. magnificum. Our findings challenge the notion of relative evolutionary and demographic stability of Tertiary relict trees, and may serve as a guideline for assessing the impact of Neogene climate change on the evolution and distribution of East Asian temperate plants.

PREMISE OF THE STUDY: Microsatellite markers were developed and characterized in a typically coastal, widespread, and dominant tree species of the evergreen broadleaf forests, Machilus thunbergii, for comparison of the genetic diversity and structure of inland populations surrounding the ancient Lake Biwa and coastal populations in Japan. METHODS AND RESULTS: Eighteen polymorphic microsatellites of this species were isolated using an improved technique for isolating codominant compound microsatellite markers. These isolated loci provided compound simple sequence repeat (SSR) markers with polymorphisms of three to 19 alleles per locus, with an average of 10.9. The expected and observed within-population heterozygosities ranged from 0.16 to 0.86 and from 0.13 to 0.72, respectively. CONCLUSIONS: These markers may be useful tools for further investigation of the population genetic structure and biogeographic history of M. thunbergii in the warm-temperate zone of East Asia.

The alpine timberline on Mt. Fuji (central Japan) is at 2,400-2,500 m above sea level. Over a 21-year period (1978-1999), we tracked changes in this vegetation boundary on a transect at a site impacted by the 1707 volcanic eruption. The timberline advanced rapidly upwards during this time period. Dominant tree species at the timberline (Alnus maximowiczii, Salix reinii, and Larix kaempferi) colonized sites that were initially largely free of vegetation at higher altitudes. Seedlings of L. kaempferi were particularly abundant at the border of advancing vegetation. According to tree age, we found that this was the first canopy species in the colonized areas. L. kaempferi is drought resistant, and this probably contributes to its establishment capability in the high-altitude climate. Most seedlings of Abies veitchii invaded patches of herbs and shrubs. These vegetation patches in the upper kampfzone provide important shelter for seedlings of invading tree species. We predict that the upward advance of the alpine timberline is a recovery process following the volcanic eruption, and that climate change may accelerate this advance.

Japanese horse chestnut (Aesculus turbinata: Hippocastanaceae) is one of the typical woody plants that grow in temperate riparian forests in the Japanese Archipelago. To analyze the phylogeography of this plant in the Japanese Archipelago, we determined cpDNA haplotypes for 337 samples from 55 populations covering the entire distribution range. Based on 1,313 bp of two spacers, we determined ten haplotypes that are distinguished from adjacent haplotypes by one or two steps. Most of the populations had a single haplotype, suggesting low diversity. Spatial analysis of molecular variance suggested three obvious phylogeographic structures in western Japan, where Japanese horse chestnut is scattered and isolated in mountainous areas. Conversely, no clear phylogeographic structure was observed from the northern to the southern limit of this species, including eastern Japan, where this plant is more common. Rare and private haplotypes were also found in southwestern Japan, where Japanese horse chestnuts are distributed sparsely. These findings imply that western Japan might have maintained a relatively large habitat for A. turbinata during the Quaternary climatic oscillations, while northerly regions could not.
