RESUMO

Modern drug development increasingly requires comprehensive models that can be utilized in the earliest stages of compound and target discovery. Here we report a phenotypic screening exercise in a high-throughput Organ-on-a-Chip setup. We assessed the inhibitory effect of 1537 protein kinase inhibitors in an angiogenesis assay. Over 4000 micro-vessels were grown under perfusion flow in microfluidic chips, exposed to a cocktail of pro-angiogenic factors and subsequently exposed to the respective kinase inhibitors. Efficacy of compounds was evaluated by reduced angiogenic sprouting, whereas reduced integrity of the main micro-vessel was taken as a measure for toxicity. The screen yielded 53 hits with high anti-angiogenicity and low toxicity, of which 44 were previously unassociated with angiogenic pathways. This study demonstrates that Organ-on-a-Chip models can be screened in high numbers to identify novel compounds and targets. This will ultimately reduce bias in early-stage drug development and increases probability to identify first in class compounds and targets for today's intractable diseases.

Here, we present the first bone histological and microanatomical study of thalattosaurians, an enigmatic group among Triassic marine reptiles. Two taxa of thalattosaurians, the askeptosauroid Askeptosaurus italicus and one as yet undescribed thalattosauroid, are examined. Both taxa have a rather different microanatomy, tissue type, and growth pattern. Askeptosaurus italicus from the late Anisian middle Besano Formation of the southern Alpine Triassic shows very compact tissue in vertebrae, rib, a gastralium, and femora, and all bones are without medullary cavities. The tissue shows moderate to low vascularization, dominated by highly organized and very coarse parallel-fibred bone, resembling interwoven tissue. Vascularization is dominated by simple longitudinal vascular canals, except for the larger femur of Askeptosaurus, where simple vascular canals dominate in a radial arrangement. Growth marks stratify the cortex of femora. The vertebrae and humeri from the undescribed thalattosauroid from the late Carnian of Oregon have primary and secondary cancellous bone, resulting in an overall low bone compactness. Two dorsal vertebral centra show dominantly secondary trabeculae, whereas a caudal vertebral centrum shows much primary trabecular bone, globuli ossei, and cartilage, indicating an earlier ontogenetic stage of the specimens or paedomorphosis. The humeri of the thalattosauroid show large, simple vascular canals that are dominantly radially oriented in a scaffold of woven and loosely organized parallel-fibred tissue. Few of the simple vascular canals are thinly but only incompletely lined by parallel-fibered tissue. In the Oregon material, changes in growth rate are only indicated by changes in vascular organization but no distinct growth marks were identified. The compact bone of Askeptosaurus is best comparable to some pachypleurosaurs, whereas its combination of tissue and vascularity is similar to eosauropterygians in general, except for the coarse nature of its parallel-fibred tissue. The cancellous bone of the Oregon thalattosauroid resembles what is documented in ichthyosaurs and plesiosaurs. However, in contrast to these its tissue does not consist of fibro-lamellar bone type. Tissue types of both thalattosaurian taxa indicate rather different growth rates and growth patterns, associated with different life history strategies. The microanatomy reflects different life styles that fit to the different environments in which they had been found (intraplatform basin vs. open marine). Both thalattosaurian taxa differ from each other but in sum also from all other marine reptile taxa studied so far. Thalattosaurian bone histology documents once more that bone histology provides for certain groups (i.e., Triassic Diapsida) only a poor phylogenetic signal and is more influenced by exogenous factors. Differences in lifestyle, life history traits, and growth rate and pattern enabled all these Triassic marine reptiles to live contemporaneously in the same habitat managing to avoid substantial competition.

The aim of the study was to investigate whether jerk cost (JC) can discriminate between swimming levels. Nine elite and nine non-elite swimmers swam a 50-m front-crawl sprint wearing a 3D accelerometer on their back between the inferior angles of the scapulae. Lap times and JC were calculated from the acceleration signal and compared between groups and between swimmers within a group. The elite swimmers swam significantly faster lap times than the non-elite swimmers (p < 0.001). They did so with significantly lower levels of JC compared to the non-elite swimmers (p = 0.005). Furthermore, a stepwise multiple linear regression showed JC accounted for 32.9% of the variation in lap time of the elite swimmers. These results indicate that it is possible to discriminate elite from non-elite swimmers using JC: elite swimmers swim with lower JCs than non-elite swimmers. Additionally, swimming at higher speed is associated with more accelerations and decelerations in both elite and non-elite swimmers, which is reflected by higher JCs and lower smoothness. In sum, JC provides an index of swimming technique that is easy to use in training practice.

Plesiosauria is an extinct clade of diapsid marine reptiles that evolved in the Late Triassic and radiated globally for the remainder of the Mesozoic. The recent description of a pregnant specimen of Polycotylus latipinnis demonstrates that some plesiosaurs were viviparous. To establish a baseline of histological data on plesiosaur ontogeny, we sampled the mother and fetus of the gravid plesiosaur specimen. To widen the base of data concerning ontogeny and life history of plesiosaurs, we gathered additional morphologic and histologic data from a securely identified growth series of polycotylids from the Pierre Shale of South Dakota. Paleohistological thin sections were prepared from the three humeri. Both adults show a dense, heavily remodeled cortex consisting entirely of longitudinally oriented secondary osteons, except for a thin rind of superficial primary bone. The mother exhibits an external fundamental system, indicating it was fully mature; the other adult does not. In both adults the cortex grades into a spongy medulla, comprising large vascular canals and erosion rooms surrounded by secondary lamellar trabecular bone, and lacking a marrow cavity. The fetal humerus possesses a medullary region similar to that of the Dolichorhynchops bonneri adult, although its lamellar bone is primary and deposited around calcified cartilage. The medulla is demarcated from the cortex by a prominent Kastschenko's line. The cortex of the fetus is a relatively thin layer of periosteal woven bone, longitudinally to radially vascularized, and interfingered with columns of osteoblasts surrounded by rapidly-deposited extracellular matrix. The neonate humerus resembles the fetus, with its trabeculae identical in both size and histology, although it lacks calcified cartilage. The cortex is also similar but much thicker, consisting entirely of rapidly deposited, radially vascularized, woven to fibrolamellar bone. The cortex carries a line near its surface. This feature is not a line of arrested growth, but a sudden change in vascular angle and increase in bone density. We argue this feature is a birth line indicating a change in growth regime, possibly in response to increased hydrodynamic forces after birth. The birth line indicates that the neonate was about 40% of maternal length when born. Our histological data demonstrate that polycotylids had very high fetal growth rates, and that birth size was large. Comparison with the geologically oldest plesiosaur confirms that rapid growth evolved in the Triassic, although histological details differ, and the degree to which the polycotylid ontogenetic pattern is generalizable to other plesiosaurs is currently unknown. Further histological research utilizing full growth series is needed, particularly for Jurassic taxa.

For training to be optimal, daily training load has to be adapted to the momentary status of the individual athlete, which is often difficult to establish. Therefore, the current study investigated the predictive value of heart-rate recovery (HRR) during a standardized warm-up for training load. Training load was quantified by the variation in heart rate during standardized training in competitive swimmers. Eight female and 5 male Dutch national-level swimmers participated in the study. They all performed 3 sessions consisting of a 300-m warm-up test and a 10 × 100-m training protocol. Both protocols were swum in front crawl at individually standardized velocities derived from an incremental step test. Velocity was related to 75% and 85% heart-rate reserve (% HRres) for the warm-up and training, respectively. Relative HRR during the first 60 s after the warm-up (HRRw-up) and differences between the actual and intended heart rate for the warm-up and the training (ΔHRw-up and ΔHRtr) were determined. No significant relationship between HRRw-up and ΔHRtr was found (F1,37 = 2.96, P = .09, R2 = .07, SEE = 4.65). There was considerable daily variation in ΔHRtr at a given swimming velocity (73-93% HRres). ΔHRw-up and ΔHRtr were clearly related (F1,37 = 74.31, P < .001, R2 = .67, SEE = 2.78). HRR after a standardized warm-up does not predict heart rate during a directly subsequent and standardized training session. Instead, heart rate during the warm-up protocol seems a promising alternative for coaches to make daily individual-specific adjustments to training programs.

The extinct parareptilian clade of pareiasaurs was in the past often presented to constitute a morphocline from larger, less armoured forms to smaller, well armoured forms, indicating that the osteoderm cover became an increasingly prominent aspect in the post-cranial skeleton of these animals. Here, we describe microanatomical and microstructural aspects of osteoderms of the three pareiasaur taxa Bradysaurus, Pareiasaurus and Anthodon from the Permian of South Africa. A generalized mode of osteoderm formation, consistent with intramembraneous skeletogenesis, is hypothesized to be present in all pareiasaurs. Few characters are shared between pareiasaur dermal armour and turtle shell bones and osteoderms. Otherwise, there is strong evidence from microanatomy and histology (i.e. absence of structures that formed via metaplasia of dermal tissue) that indicates nonhomology between pareiasaur dermal armour and the armour of living eureptiles. Analysis with bone profiler revealed no clear connection between bone compactness and lifestyle in the amniote osteoderm sample.

In nautical literature, 'dead-water' refers to the obstructive effect encountered by ships moving in stratified water due to the ship generating waves on an interface that separates different water masses. To investigate the hypothesis that open water swimming may also be obstructed by an encounter of dead-water, possibly causing drowning, we performed two experiments that assess the impact of stratified water on swimming. In the first experiment, subjects made a single front-crawl stroke while lying on a carriage that was rolling just above the water surface. The gain in kinetic energy, as a result of the stroke, was far less in stratified than in homogeneous water. In the second experiment, four subjects swam a short distance (5 m) in homogeneous and in two different settings of stratified water. At the same stroke frequency, swimming in stratified conditions was slower by 15%, implying a loss in propulsive power by 40%. Although in nature stratification will be less strong, extrapolation of the results suggests that dead-water might indeed obstruct swimming in open water as well. This effect will be most pronounced during fair weather, when stratification of a shallow surface layer is most easily established. Our findings indicate that swimmers' anecdotal evidence on 'water behaving strangely' may have to be taken more seriously than previously thought.

The largest and most diverse collection of Pachypleurosauridae (Nothosauria, Reptilia) comes from Monte San Giorgio, Switzerland. Several hundred complete skeletons were collected from four distinct horizons of bituminous limestones and shales of Anisian-Ladinian boundary to early Ladinian age (Middle Triassic). Serpianosaurus mirigiolensis comes from the oldest strata, the Grenzbitumenzone Beds. The three younger strata, all in the Lower Meride Limestone, yield three species of Neusticosaurus. Neusticosaurus pusillus comes from the Cava Inferiore horizon, Neusticosaurus peyeri, new species, from the Cava Superiore horizon, and Neusticosaurus edwardsii, new combination, from the Alla Cascina horizon. Neusticosaurus pusillus is biostratigraphically important because it is one of the rare species reported from both the Germanic and the Alpine Triassic. Neusticosaurus pusillus and N. peyeri are small and very similar in their anatomy. Neusticosaurus species are easiest separated by their number of presacral vertebrae. Ornamentation of the bone surface is distinctive for all four pachypleurosaurids. Soft parts are rarely preserved, except for one partial squamation. The biological age of Neusticosaurus individuals can be determined by skeletochronology (aging by bone annuli). Small species of Neusticosaurus were sexually mature after three to four years and lived for six to nine years. Taphonomic analysis of the small species indicates attritional mortality and suggests weak bottom currents in the Monte San Giorgio basin during early Ladinian times. Morphometric comparison of all four pachypleurosaurids indicates that the changing vertebral numbers between species are largely due to a change in number of segments. All Monte San Giorgio pachypleurosaurids are sexually dimorphic in forelimb development. Sex x has poorly differentiated and relatively short humeri whereas sex y has well differentiated and relatively long humeri. The sexes are of about the same size and represented in roughly equal numbers. Identification of gender was not possible. Good growth series, especially of Neusticosaurus peyeri, from embryo to large adult permitted qualitative and quantitative study of ontogeny. The skull grows with negative allometry; the humerus grows isometrically or with positive allometry, depending on sex and species; the femur grows isometrically. The adult size range in N. peyeri is considerably larger than in modern reptiles. The Monte San Giorgio pachypleurosaurids are a monophyletic group. The phylogeny of this group is congruent with the stratigraphic distribution of its members.

The first nothosaur (Neusticosaurus sp.) embryo, one of the very few fossil embryos known, provides a rare glimpse at reproduction in extinct reptiles. The specimen from the southern Alpine Middle Triassic (about 230 million years ago) was recognized as an embryo in comparison with an exceptionally large and well-understood sample of juvenile and sexed adult Neusticosaurus sp. The skeleton shows many embryonic features and may well be the smallest fossil reptile known (body length 51 millimeters). It reached only 22% of mean adult length whereas modern reptiles of this size do not hatch before they reach about 30% of mean adult length. The question of ovipary versus vivipary in pachypleurosaurs is discussed in light of the embryo.
