RESUMO

Modern livestock production systems are characterized by a greater focus on intensification, involving managing larger numbers of animals to achieve higher productive efficiency and animal health and welfare within herds. Therefore, animal breeding programs need to be strategically designed to select animals that can effectively enhance production performance and animal welfare across a range of environmental conditions. Thus, this review summarizes the main methodologies used for assessing the levels of genotype-by-environment interaction (G × E) in cattle populations. In addition, we explored the importance of integrating genomic and phenotypic information to quantify and account for G × E in breeding programs. An overview of the structure of cattle breeding programs is provided to give insights into the potential outcomes and challenges faced when considering G × E to optimize genetic gains in breeding programs. The role of nutrigenomics and its impact on gene expression related to metabolism in cattle are also discussed, along with an examination of current research findings and their potential implications for future research and practical applications. Out of the 116 studies examined, 60 and 56 focused on beef and dairy cattle, respectively. A total of 83.62% of these studies reported genetic correlations across environmental gradients below 0.80, indicating the presence of G × E. For beef cattle, 69.33%, 24%, 2.67%, 2.67%, and 1.33% of the studies evaluated growth, reproduction, carcass and meat quality, survival, and feed efficiency traits, respectively. By contrast, G × E research in dairy cattle populations predominantly focused on milk yield and milk composition (79.36% of the studies), followed by reproduction and fertility (19.05%), and survival (1.59%) traits. The importance of G × E becomes particularly evident when considering complex traits such as heat tolerance, disease resistance, reproductive performance, and feed efficiency, as highlighted in this review. Genomic models provide a valuable avenue for studying these traits in greater depth, allowing for the identification of candidate genes and metabolic pathways associated with animal fitness, adaptation, and environmental efficiency. Nutrigenetics and nutrigenomics are emerging fields that require extensive investigation to maximize our understanding of gene-nutrient interactions. By studying various transcription factors, we can potentially improve animal metabolism, improving performance, health, and quality of products such as meat and milk.

The wide use of genomic information has enabled the identification of lethal recessive alleles that are the major genetic causes of reduced conception rates, longer calving intervals, or lower survival for live-born animals. This study was carried out to screen the Nellore cattle genome for lethal recessive haplotypes based on deviation from the expected population homozygosity, and to test SNP markers surrounding the lethal haplotypes region for association with heifer rebreeding (HR), post-natal mortality (PNM) and stayability (STAY). This approach requires genotypes only from apparently normal individuals and not from affected embryos. A total of 62,022 animals were genotyped and imputed to a high-density panel (777,962 SNP markers). Expected numbers of homozygous individuals were calculated, and the probabilities of observing 0 homozygotes was obtained. Deregressed genomic breeding values [(G)EBVs] were used in a GWAS to identify candidate genes and biological mechanisms affecting HR, STAY and PNM. In the functional analyses, genes within 100 kb down and upstream of each significant SNP marker, were researched. Thirty haplotypes had high expected frequency, while no homozygotes were observed. Most of the alleles present in these haplotypes had a negative mean effect for PNM, HR and STAY. The GWAS revealed significant SNP markers involved in different physiological mechanisms, leading to harmful effect on the three traits. The functional analysis revealed 26 genes enriched for 19 GO terms. Most of the GO terms found for biological processes, molecular functions and pathways were related to tissue development and the immune system. More phenotypes underlying these putative regions in this population could be the subject of future investigation. Tests to find putative lethal haplotype carriers could help breeders to eliminate them from the population or manage matings in order to avoid homozygous.

Covariance components were estimated for growth traits (BW, birth weight; WW, weaning weight; YW, yearling weight), visual scores (BQ, breed quality; CS, conformation; MS, muscling; NS, navel; PS, finishing precocity), hip height (HH), and carcass traits (BF, backfat thickness; LMA, longissimus muscle area) measured at yearling. Genetic gains were obtained and validation models on direct and maternal effects for BW and WW were fitted. Genetic correlations of growth traits with CS, PS, MS, and HH ranged from 0.20 ± 0.01 to 0.94 ± 0.01 and were positive and low with NS (0.11 ± 0.01 to 0.20 ± 0.01) and favorable with BQ (0.14 ± 0.02 to 0.37 ± 0.02). Null to moderate genetic correlations were obtained between growth and carcass traits. Genetic gains were positive and significant, except for BW. An increase of 0.76 and 0.72 kg is expected for BW and WW, respectively, per unit increase in estimated breeding value (EBV) for direct effect and an additional 0.74 and 1.43, respectively, kg per unit increase in EBV for the maternal effect. Monitoring genetic gains for HH and NS is relevant to maintain an adequate body size and a navel morphological correction, if necessary. Simultaneous selection for growth, morphological, and carcass traits in line with improve maternal performance is a feasible strategy to increase herd productivity.

A chromatographic method is described for the separation and quantification of polycyclic aromatic sulfur heterocycles (PASHs) using liquid chromatography coupled with diode array detection (DAD). The PASHs that were investigated in this study were chosen based on their similarity in molecular weight, volatility and polarity of their PAH analogues. The chromatographic separation of the compounds was optimized, and their analytical characteristics were evaluated. The limits of detection and quantification ranged from 0.05 for 2,3,4,7-tetramethylbenzothiophene to 2.16 µg L-1 for thieno[2,3-b]thiophene and from 0.16 for 2,3,4,7-tetramethylbenzothiophene to 6.53 µg L-1 for thieno[2,3-b]thiophene, respectively. Recoveries ranged from 84.9 for benzo[b]benzo[4,5]thieno[2,3-d]thiophene to 110% for dinaphtho[2,1-b:1',2'-d]thiophene. Intermediate precisions and repeatabilities lie between 1.4 and 3.0%, and 0.3 and 1.7%, respectively. The chromatographic method was applied for determination of PASHs directly in asphalt fumes, which were obtained by heating asphalt samples in a homemade closed system. The mutual interference of PAH analogues was also discussed. The method was successfully used for PASH determination in asphalt samples obtained from three different oil refineries in Brazil.

Objetivo: analisar os efeitos do método Pilates sobre a força muscular respiratória em crianças com sintomas de asma. Metodologia: nesse estudo clínico foram avaliadas 17 crianças em idade escolar (09 a 12 anos), de ambos os sexos. Inicialmente responderam ao questionário International Study of Asthma and Allergies in Childhood (ISAAC), que avalia a prevalência de asma e doenças alérgicas em crianças e adolescentes. Assim, aquelas que obtiveram uma pontuação no ISAAC, com nota de corte igual ou superior a 5, foram incluídas no estudo. Após as crianças realizarem uma avaliação através do manovacuômetro, a fim de mensurar as pressões respiratórias máximas (PImáx e PEmáx) foi realizada uma avaliação antropométrica para verificar a massa corporal e a estatura. Como intervenção, as crianças participaram de um programa de exercícios com base no método Pilates 2 vezes por semana, com duração aproximada de 50 minutos cada sessão, durante 8 meses, totalizando 32 semanas. Resultados: Em relação às pressões, tanto a PImáx quanto a PEmáx aumentaram pós-intervenção. Conclusão: o estudo encontrou contribuições significativas do método Pilates sobre a força muscular respiratória (PImáx e PEmáx) de crianças com sintomas de asma. Desta forma, sugere-se que o método representa uma ferramenta importante para melhorar a força muscular respiratória dessa população.


Objective: To analyze the effects of the Pilates method on respiratory muscle function in children with asthma symptoms. Methods: in this clinical study, a sample of school-age children (9 to 12 years old) of both sexes was included. Initially, the children answered the questionnaire International Study of Asthma and Allergies in Childhood (ISAAC). Thus, those who scored 5 or more were included in the study. Subsequent to this initial screening, the children performed an evaluation of respiratory muscle strength through the digital manovacuometer in order to measure the maximum respiratory pressures (MIP and MEP). An anthropometric assessment was also performed to verify body mass and height. The children participated in an exercise program based on the Pilates method, being performed twice a week, lasting about 50 minutes each session, for 8 months, totaling 32 weeks. Results: regarding the pressures, both inspiratory and expiratory pressure increased at the end of the intervention. Conclusion: the present study found significant contributions of the Pilates method in the inspiratory muscle strength of children with asthma symptoms. Thus, it is suggested that the method represents an important tool to improve the muscular strength of these children.

Our goal was to define a breeding objective for Brangus beef cattle in Brazil. Bioeconomic models were produced and used to estimate economic values (EVs). The scenarios simulated were typical full-cycle beef production systems that are used in tropical and subtropical regions. The breeding objective contained pregnancy rate (PR), warm carcass weight (WCW), mature cow weight (MCW), number of nematode eggs per gram of faeces (EPG) and tick count (TICK). Two models were used in series to estimate the EV. A deterministic model was used to simulate effects of PR, WCW and MCW on profitability with a constant parasite load. Subsequently, stochastic models were used to estimate economic values for TICK and EPG as consequences of their environmental effects on weight gains, mortality and health costs. The EV of PR, WCW, MCW, EPG and TICK, was US$1.59, US$2.11, -US$0.24, -US$5.35 and -US$20.88, respectively. Results indicate positive emphasis should be placed on PR (12.49%) and WCW (65.07%) with negative emphasis on MCW (13.92%), EPG (2.77%) and TICK (5.75%). In comparison with the indexes usually used, these results suggest a reformulation in the selection indexes of the beef production system in tropical and subtropical regions in order to obtain greater profitability.

The objective of the present study was to evaluate the accuracy and bias of direct and blended genomic predictions using different methods and cross-validation techniques for growth traits (weight and weight gains) and visual scores (conformation, precocity, muscling, and size) obtained at weaning and at yearling in Hereford and Braford breeds. Phenotypic data contained 126,290 animals belonging to the Delta G Connection genetic improvement program, and a set of 3,545 animals genotyped with the 50K chip and 131 sires with the 777K. After quality control, 41,045 markers remained for all animals. An animal model was used to estimate (co)variance components and to predict breeding values, which were later used to calculate the deregressed estimated breeding values (DEBV). Animals with genotype and phenotype for the traits studied were divided into 4 or 5 groups by random and k-means clustering cross-validation strategies. The values of accuracy of the direct genomic values (DGV) were moderate to high magnitude for at weaning and at yearling traits, ranging from 0.19 to 0.45 for the k-means and 0.23 to 0.78 for random clustering among all traits. The greatest gain in relation to the pedigree BLUP (PBLUP) was 9.5% with the BayesB method with both the k-means and the random clustering. Blended genomic value accuracies ranged from 0.19 to 0.56 for k-means and from 0.21 to 0.82 for random clustering. The analyses using the historical pedigree and phenotypes contributed additional information to calculate the GEBV, and in general, the largest gains were for the single-step (ssGBLUP) method in bivariate analyses with a mean increase of 43.00% among all traits measured at weaning and of 46.27% for those evaluated at yearling. The accuracy values for the marker effects estimation methods were lower for k-means clustering, indicating that the training set relationship to the selection candidates is a major factor affecting accuracy of genomic predictions. The gains in accuracy obtained with genomic blending methods, mainly ssGBLUP in bivariate analyses, indicate that genomic predictions should be used as a tool to improve genetic gains in relation to the traditional PBLUP selection.

RESUMO

Este artigo pretende narrar sobre as potencialidades de uma oficina de hip hop, construída coletivamente com as(os) usuárias(os) do CAPS ad de Ourinhos, município localizado no interior do Estado de São Paulo. As oficinas acontecem em encontros quinzenais, no CAPS ad. Realizamos parcerias com diversos coletivos de hip hop do município, os quais têm, voluntariamente, ofertado sua envergadura cultural, estimulando as(os) usuárias(os) a desenvolverem tanto suas habilidades para a rima, a música, a dança e o desenho, quanto a capacidade de externalizar angústias, opiniões e alegrias. As oficinas têm proporcionado o fortalecimento do vínculo de muitas (os) usuárias (os) no serviço, proporcionando momentos de prazer, dando voz às suas demandas e promovendo cidadania. A expressão pelas canções aparece como uma maneira muito rica de elaboração e de ressignificação da vida, assim como de construção de novas formas de viver. Dessa maneira, espalhar a cultura do hip hop neste espaço é resistir às diversas exclusões sociais, dar voz àquelas (es) que são, diariamente, silenciados, e lutar por uma sociedade mais justa e menos excludente.

AIMS: Aluminum (Al) is an important environmental contaminant; however, there are not enough evidences of Al-induced cardiovascular dysfunction. We investigated the effects of acute exposure to aluminum chloride (AlCl3) on blood pressure, vascular reactivity and oxidative stress. METHODS AND RESULTS: Male Wistar rats were divided into two groups: Untreated: vehicle (ultrapure water, ip) and AlCl3: single dose of AlCl3 (100mg/kg,ip). Concentration-response curves to phenylephrine in the absence and presence of endothelium, the nitric oxide synthase inhibitor l-NAME, the potassium channel blocker tetraethylammonium, and the NADPH oxidase inhibitor apocynin were performed in segments from aortic and mesenteric resistance arteries. NO released was assessed in aorta and reactive oxygen species (ROS), malondialdehyde, non-protein thiol levels, antioxidant capacity and enzymatic antioxidant activities were investigated in plasma, aorta and/or mesenteric arteries. After one hour of AlCl3 exposure serum Al levels attained 147.7±25.0µg/L. Al treatment: 1) did not affect blood pressure, heart rate and vasodilator responses induced by acetylcholine or sodium nitroprusside; 2) decreased phenylephrine-induced vasoconstrictor responses; 3) increased endothelial modulation of contractile responses, NO release and vascular ROS production from NADPH oxidase; 4) increased plasmatic, aortic and mesenteric malondialdehyde and ROS production, and 5) decreased antioxidant capacity and affected the antioxidant biomarkers non-protein thiol levels, glutathione peroxidase, glutathione-S-transferase, superoxide dismutase and catalase enzymatic activities. CONCLUSION: AlCl3-acute exposure reduces vascular reactivity. This effect is associated with increased NO production, probably acting on K+ channels, which seems to occur as a compensatory mechanism against Al-induced oxidative stress. Our results suggest that Al exerts toxic effects to the vascular system.
