RESUMO

A method is proposed to measure the photon polarisation parameter λ γ in b → s γ transitions using an amplitude analysis of B → K π π γ decays. Simplified models of the K π π system are used to simulate B + → K + π - π + γ and B 0 → K + π - π 0 γ decays, validate the amplitude analysis method, and demonstrate the feasibility of a measurement of the λ γ parameter irrespective of the model parameters. Similar sensitivities to λ γ are obtained with both the charged and neutral hadronic systems. In the absence of any background and distortion due to experimental effects, the statistical uncertainty expected from an analysis of B + → K + π - π + γ decays in an LHCb data set corresponding to an integrated luminosity of 9  fb - 1 is estimated to be 0.009. A similar measurement using B 0 → K + π - π 0 γ decays in a Belle II data sample corresponding to an integrated luminosity of 5  ab - 1 would lead to a statistical uncertainty of 0.018.

BACKGROUND: Rheumatoid arthritis (RA) has an increased number of comorbidities compared with the general population. OBJECTIVE: Study aim was to collect epidemiological data on prevalence, incidence and comorbidities of RA as well as utilization of outpatient and inpatient care services. MATERIAL AND METHODS: In an age and gender-adjusted case control study, a total of 3.4 million patients insured by the AOK Baden-Württemberg were analysed with respect to visits to physicians, prevalence, incidence and comorbidities of RA. The study was based on out- and inpatient diagnoses from 2013. RESULTS: The RA prevalence was 0.64% (n = 26,919), the incidence was 0.04%. Patients with RA have significant more comorbidities in almost all diagnosis groups, especially in musculoskeletal and cardiovascular diseases, compared to a control group (n = 181,209). 22.8% of RA patients had not contacted an internist rheumatologist, orthopedist or orthopedic surgeon. Biological disease-modifying anti-rheumatic drugs (DMARDs) were almost exclusively prescribed by internist rheumatologists, while conventional DMARDs were equally prescribed by general practitioners and rheumatologists. Of the RA patients 32.6% were hospitalized at least once a year and were nearly twice as frequently inpatient as the control group. CONCLUSION: RA patients need more in- and outpatient healthcare services and suffer significantly more often from comorbidities. The general practitioner is the most frequently visited physician. Other consulted physicians are rheumatologists, ophthalmologists, orthopedists/orthopedic surgeons and internists not specialized in rheumatology. The study highlights the need to create consensus treatment algorithms and maintain a close interdisciplinary and intersectoral cooperation and communication.

BACKGROUND: Musculoskeletal illnesses and injuries are among the most common ailments in the Federal Republic of Germany. In 2008 they generated costs of nearly 29 billion euros. Figures about their incidence and prevalence are necessary for a demand-oriented planning of future patient-centred care. METHOD: Pseudonymised data of 3.8 million people insured by AOK Baden-Württemberg between 2008 and 2013 were evaluated. The diagnoses were assigned to nine injury groups. For outpatient care confirmed diagnoses were considered, and for inpatient care both primary and secondary diagnoses were considered. For all patients with structural knee injuries, it was evaluated whether they made use of one of five eligible treatment paradigms either in the quarter in which they were injured or in the following quarter. RESULTS: 418,257 patients were treated in 2013 for at least one new-onset injury (10.9 % of all insurees); 86,783 insurees (2.3 % of all insurees) had a newly occurring knee injury. The vast majority of the patients were treated by specialist doctors. While magnetic resonance imaging clearly increased during the observation period, the incidence of surgical therapy did not change. Striking are the different age distributions regarding the types of injuries, with a high injury incidence amongst young men and a significant increase in injuries between 2008 and 2013, especially amongst women. CONCLUSION: For the first time, the data quantify the knee injury incidences of a large cohort in Germany. They show which inpatient and outpatient health care services have been claimed and that an age- and gender-adapted prevention and an increased awareness are needed.

Bone marrow mesenchymal stem cells' (BM-MSCs) role in multiple myeloma (MM) pathogenesis is recognized. Recently, we have published that co-culture of MM cell lines with BM-MSCs results in mutual modulation of phenotype and proteome (via translation initiation (TI) factors eIF4E/eIF4GI) and that there are differences between normal donor BM-MSCs (ND-MSCs) and MM BM-MSCs (MM-MSCs) in this crosstalk. Here, we aimed to assess the involvement of soluble BM-MSCs' (ND, MM) components, more easily targeted, in manipulation of MM cell lines phenotype and TI with specific focus on microvesicles (MVs) capable of transferring critical biological material. We applied ND and MM-MSCs 72h secretomes to MM cell lines (U266 and ARP-1) for 12-72h and then assayed the cells' (viability, cell count, cell death, proliferation, cell cycle, autophagy) and TI (factors: eIF4E, teIF4GI; regulators: mTOR, MNK1/2, 4EBP; targets: cyclin D1, NFκB, SMAD5, cMyc, HIF1α). Furthermore, we dissected the secretome into >100kDa and <100kDa fractions and repeated the experiments. Finally, MVs were isolated from the ND and MM-MSCs secretomes and applied to MM cell lines. Phenotype and TI were assessed. Secretomes of BM-MSCs (ND, MM) significantly stimulated MM cell lines' TI, autophagy and proliferation. The dissected secretome yielded different effects on MM cell lines phenotype and TI according to fraction (>100kDa- repressed; <100kDa- stimulated) but with no association to source (ND, MM). Finally, in analyses of MVs extracted from BM-MSCs (ND, MM) we witnessed differences in accordance with source: ND-MSCs MVs inhibited proliferation, autophagy and TI whereas MM-MSCs MVs stimulated them. These observations highlight the very complex communication between MM and BM-MSCs and underscore its significance to major processes in the malignant cells. Studies into the influential MVs cargo are underway and expected to uncover targetable signals in the regulation of the TI/proliferation/autophagy cascade.

Neutron reflectometry is used to study in situ the intercalation of lithium into amorphous silicon electrodes. The experiments are done using a closed three-electrode electrochemical cell setup. As a working electrode, an about 40 nm thick amorphous silicon layer is used that is deposited on a 1 cm thick quartz substrate coated with palladium as a current collector. The counter electrode and the reference electrode are made of lithium metal. Propylene carbonate with 1 M LiClO4 is used as an electrolyte. The utility of the cell is demonstrated during neutron reflectometry measurements where Li is intercalated at a constant current of 100 µA (7.8 µA cm(-2)) for different time steps. The results show (a) that the change in Li content in amorphous silicon and the corresponding volume expansion can be monitored, (b) that the formation of the solid electrolyte interphase becomes visible and (c) that an irreversible capacity loss is present.

First observations of the B(s)(0) → D(s)(*-) π+, B(s)(0) → D(s)(-) ρ+ and B(s)(0) → D(s)(*-) ρ+ decays are reported together with measurements of their branching fractions: B(B(s)(0) → D(s)(*-) π+) = [2.4(-0.4)(+0.5)(stat) ± 0.3(syst) ± 0.4(f(s))]×10(-3), B(B(s)(0) → D(s)(-) ρ+) = [8.5(-1.2)(+1.3)(stat) ± 1.1(syst) ± 1.3(f(s))]×10(-3) and B(B(s)(0) → D(s)(*-) ρ+) = [11.9(-2.0)(+2.2)(stat) ± 1.7(syst) ± 1.8(f(s))]×10(-3) (f(s) = N(B(s)(*) B(s)(*))/N(bb)). From helicity-angle distributions, we measured the longitudinal polarization fraction in B(s)(0) → D(s)(*-) ρ+ decays to be f(L)(B(s)(0) → D(s)(*-) ρ+) = 1.05(-0.10)(+0.08)(stat)(-0.04)(+0.03)(syst). These results are based on a 23.6 fb(-1) data sample collected at the Υ(5S) resonance with the Belle detector at the KEKB e+ e- collider.

The solubilisation of the functionalized silicone oil aminoethyl aminopropyl methylpolysiloxane WR 1300 has been investigated in aqueous solutions of the surfactant pentaethoxy-iso-tridecanol (iC(13)E(5)). The silicone oil consists of a linear backbone with an average of 200 dimethylsiloxane units and 2-3 functionalized side groups. In various applications the compound is considered a silicone oil. The surfactant iC(13)E(5) forms a L(alpha)-phase in water, and with decane a microemulsion can be obtained. The solubilisation experiments started with lamellar iC(13)E(5) phases. With proper amounts of WR 1300 transparent, optically isotropic but highly viscous single phases are formed. These phases cannot be considered true microemulsions with an oil core and a surrounding surfactant layer. Cryo-TEM micrographs show micelles with irregular shapes and about 25 nm diameter. Their size is independent of the oil/surfactant ratio. It can be explained on the basis of a model where the amino groups of the oil are all found at the surface of the micelles, limiting the radius of the aggregates to the largest length of the polydimethylsiloxane backbone between two such groups or between one end of the oil and one functionalized group. The micellar structures contain only a few siloxane molecules exposing their hydrophilic groups to the water. The functionalized silicone oil molecules can thus be considered as surfactant molecules with a few polar groups and a large hydrophobic chain from dimethyl siloxane.

We have studied B_(s);(0)-->D_(s);(-)pi;(+) and B_(s);(0)-->D_(s);(-/+)K;(+/-) decays using 23.6 fb;(-1) of data collected at the Upsilon(5S) resonance with the Belle detector at the KEKB e;(+)e;(-) collider. This highly pure B_(s);(0)-->D_(s);(-)pi;(+) sample is used to measure the branching fraction, B(B_(s);(0)-->D_(s);(-)pi;(+))=[3.67_(-0.33);(+0.35)(stat)-0.42+0.43(syst)+/-0.49(f_(s))]x10;(-3) (f_(s)=N_(B_(s);((*))B[over ]_(s);((*)))/N_(bb[over ])) and the fractions of B_(s);(0) event types at the Upsilon(5S) energy, in particular N_(B_(s);(*)B[over ]_(s);(*))/N_(B_(s);((*))B[over ]_(s);((*)))=(90.1_(-4.0);(+3.8)+/-0.2)%. We also determine the masses M(B_(s);(0))=(5364.4+/-1.3+/-0.7) MeV/c;(2) and M(B_(s);(*))=(5416.4+/-0.4+/-0.5) MeV/c;(2). In addition, we observe B_(s);(0)-->D_(s);(-/+)K;(+/-) decays with a significance of 3.5sigma and measure B(B_(s);(0)-->D_(s);(-/+)K;(+/-))=[2.4_(-1.0);(+1.2)(stat)+/-0.3(syst)+/-0.3(f_(s))]x10;(-4).

PURPOSE: To evaluate the advantages of multiplanar reconstruction and different axial slice thickness in diagnostic of rectal cancer recurrence after operation and radiotherapy. METHOD: We included 83 patients after operation and radiotherapy of rectal cancer in this study. All patients got a minimum of three CT-examinations in their follow-up program. A total of 294 CT-scans were evaluated. Each examination was reviewed by two experienced radiologists in respect to recurrence. Each examination was presented in axial reconstruction with a slice sickness of 8, 5, and 1.25 mm and in multiplanar reconstruction. The sensitivity, specificity, positive predictive value and accuracy were calculated. RESULTS: Multiplanar reconstructions showed better results for the detection of recurrence than axial reconstruction. A reduced slice thickness did not lead to better results in axial reconstruction. Multiplanar reconstruction showed a sensitivity of 0.88, a specificity of 0.98, an accuracy of 0.96 and a positive predictive value of 0,94, for axial reconstruction we calculated: 0.82, 0.97, 0.94 and 0.88, respectively. Sensitivity and accuracy showed a significant increase after the first and second examination. CONCLUSION: Multiplanar reconstructions allow a significant better detection of rectal cancer recurrence when compared to axial reconstructions. Thinner axial slice thickness shows no diagnostic advantage.

Snail hemocytes mobilise ROS-generating enzymes during oxidative burst similar to those of mammalian leukocytes. We report herein the identification of an inducible Cu/Zn superoxide dismutase, which converts O2- to H2O2, in hemocytes of the pond snail Lymnaea stagnalis. The deduced amino acid sequence with all characteristic residues (His44,46,61,69,78 and 118, Asp81, Cys55/144, Arg141 and the Greek Key loop region Glu119-Leu/Val142) includes an open reading frame of 155 AA. Changes in Cu/ZnSOD gene expression induced by stimulation with Zymosan or trematode larvae were examined in a time course. Activated hemocytes significantly up-regulate Cu/ZnSOD expression during 2-48 h upon stimulation with the maximal induction at 45 min during phagocytosis and at 12 h during encapsulations. This increase in Cu/ZnSOD expression paralleled the increasing production of hydrogen peroxide by hemocytes. Thus, intracellular or extracellular targets elicit an induced expression of Cu/ZnSOD and the generation of elevated amounts of hydrogen peroxide by L. stagnalis hemocytes, reflecting a significant activation of their host defense function.

The direct electropolymerization of benzene dissolved in the ionic liquid 1-hexyl-3-methylimidazolium tris(pentafluoroethyl)trifluorophosphate was studied at room temperature applying the electrochemical quartz-crystal microbalance technique. Analysis of the damping changes showed that the Sauerbrey equation could be applied for data evaluation. In the polymer, every third to fourth benzene ring carried a positive charge in the oxidized state. During electropolymerization, some ionic liquid was absorbed in the growing polymer. The redox behavior was characterized by wide peaks typical for conducting polymers. Charge neutrality of the polymer during redox cycling was maintained by anion and cation exchange with the ionic liquid. With increasing scan rate, cation exchange became more and more important.

The Cerec method offers experienced users the possibility of accomplishing veneers chairside which are capable of meeting the highest functional and esthetic requirements. "Esthetic shaping" directly on the patient requires a good eye for color and form, and this requires training. The present report outlines the clinical procedure and presents three clinical cases step by step, specifically discussing the external staining technique with Pro-CAD blocks.

Hemocytes from schistosome-resistant and schistosome-susceptible Biomphalaria glabrata differ fundamentally in their behavior towards an invading parasite. When the schistosome infects a resistant snail host it is quickly surrounded by hemocytes, encapsulated and destroyed. Hemocytes from susceptible hosts fail to kill the parasite. To detect the differences between these two host phenotypes, we used differential-display reverse-transcription PCR (DDRT-PCR), based on RNA extracted from isolated hemocytes. A number of differentially expressed fragments from resistant and susceptible snails were detected by DDRT-PCR and confirmed using single-strand conformation polymorphism. These methods proved to be sensitive enough to allow comparison and verification of differential gene expression in our system, where only small numbers of cells are available. The most interesting phenotype-specific fragments detected so far show sequence homologies to an adhesion molecule, defensin, serine/ threonine kinases, peroxidases and glycosidases.

The purpose of Computer-Assisted Surgery (CAS) is to help physicians and surgeons plan and execute optimal strategies from multimodal image data. The execution of such planned strategies may be assisted by guidance systems. Some of these systems, called synergistic systems, are based on the cooperation of a robotic device with a human operator. We have developed such a synergistic device: PADyC (Passive Arm with Dynamic Constraints). The basic principle of PADyC is to have a manually actuated arm that dynamically constrains the authorized motions of the surgical tool held by the human operator during a planned task. Dynamic constraints are computed from the task definition, and are implemented by a patented mechanical system. In this paper, we first introduce synergistic systems and then focus on modeling and algorithmic issues related to the dynamic constraints. Finally, we describe a 6-degree-of-freedom prototype robot designed for a clinical application (cardiac surgery) and report on preliminary experiments to date. The experimental results are then discussed, and future work is proposed.

OBJECTIVE: Follistatin (FS) is the specific binding protein of activin and expression of both factors is regulated by inflammatory agents. Therefore, FS concentrations were determined in cerebrospinal fluid (CSF) of patients with bacterial and viral meningitis or multiple sclerosis (MS), as well as in the CSF of patients without meningial inflammation or autoimmune diseases. Furthermore, a mouse pneumococcal meningitis model was used to localise the cellular sources of FS in brains of normal and meningitic mice. METHODS: FS concentrations in CSF were determined by ELISA; FS in mice was localised by in situ hybridisation and immunohistochemistry. RESULTS: FS concentrations were > or =0.4 microg/l in 22 of 66 CSF samples of meningitis patients versus 2 of 27 CSF samples from patients with multiple sclerosis (P<0.05) and 2 of 41 CSF specimen from patients without neuroinflammatory diseases (P<0.01). In the CSF of patients with meningitis, the concentration of FS was correlated with total protein (P<0.005) and lactate concentrations (P<0.05), but not with leukocyte counts, interval between onset of disease and CSF analysis, or clinical outcome. The CSF-to-serum ratios of FS and albumin also correlated significantly (P<0.0005). In some patients with meningitis the CSF-to-serum ratios suggested that the elevated FS in CSF did not originate from serum alone. FS was localised in mice brains to neurones of the hippocampus, dentate gyrus, neocortex, and to the choroid plexus. Analyses of brains and other organs from uninfected and infected animals sacrificed 6-36 h after infection did not reveal any obvious differences in the distribution and intensity of FS mRNA and protein expression. CONCLUSIONS: The concentration of FS in humans is elevated during meningitis. In some patients the increase is caused by a release of FS from brain into CSF. Data from the mouse meningitis model suggest that increased CSF concentrations of FS in meningitis appear not to be accompanied by an elevated number of cells containing FS mRNA or protein in the brain.

OBJECTIVE: The relation of activins (dimers of the beta-subunits of inhibin) and follistatin (FS) (their binding protein) affect the growth and differentiation of many cell types. Activin- and FS-mRNAs show a widespread co-expression throughout the organism, indicating an essential role for the FS/activin system in diverse physiological processes. The present study was performed to investigate FS-, activin betaA-, and activin beta B-mRNA expression in porcine tissues and to compare the relative mRNA tissue distribution by a newly developed multiplex S1 nuclease protection assay. METHODS: Twenty micrograms total RNA from different porcine tissues were subjected to multiplex S1 analysis. Specific mRNA expression was determined by measurements of optical densities on autoradiographs. RESULTS: Activin beta A-mRNA expression was abundant in the ovary, adrenal gland, fat, vein, artery and uterus, activin beta B-mRNA was highly expressed in the ovary, pituitary, uterus, placenta, aorta and cerebellum. FS-mRNA showed a widespread expression with high levels in ovary, uterus, cerebellum, placenta and fat. The comparison of relative activin beta A-, activin beta B- and FS-mRNA expression within a certain tissue showed a predominance of activin beta A-mRNA in the adrenal gland, fat, artery, spinal cord, cerebrum and colon and of activin beta B-m RNA in pituitary, testis and placenta, while FS-mRNA levels exceeded those of activin subunits in epididymis, liver, lymphoid tissue, muscle, intestine, cerebellum, ovary and uterus. CONCLUSIONS: The presented data provide an overview of FS-, activin beta A-, and activin beta B-mRNA steady state levels in porcine tissues.

Lipoteichoic and teichoic acids are components of the cell wall of Streptococcus pneumoniae. A recently developed enzyme immunoassay was used in patients with pneumococcal meningitis to investigate lipoteichoic and teichoic acid concentrations in CSF at the first lumbar puncture in relation to the clinical outcome determined by the Glasgow Outcome Scale. Lipoteichoic and teichoic acid concentrations in CSF were significantly associated with neurologic sequelae and mortality in S. pneumoniae meningitis.

1. Chemical risk assessment integrates the identification of hazards and the human exposure levels which can be established from external and/or internal exposure data. 2. The availability of biomonitoring and metabolism animal data, the skin penetration ability, and the existence of atmospheric threshold limit values were examined for twelve substances of the European first list of priority existing substances. This investigation was focused on workplace exposures and on urinary biomarkers of exposure. Appropriate biomonitoring data appeared to be available for two substances: styrene and trichloroethylene. Some biomonitoring research has been conducted on acrylonitrile, buta-1,3-diene, cyclohexane, 1,4-dichlorobenzene, hydrogen fluoride, 2-(2-methoxyethoxy)ethanol, however additional studies could be usefully carried out. No biomonitoring data are available for alkanes, C10-13, chloro; benzene, C10-13-alkyl derivatives; bis(pentabromophenyl)ether; diphenylether, octabromo-derivative. 3. It was concluded that in some cases, biomonitoring data are either lacking or scarce. This is rather surprising since the selection of the substances of the priority list was based on high tonnage, widespread use, extent of human exposure, and toxicological concern. The development of biomonitoring information could be helpful in assessing individual or population chemical exposure whatever the source and route, and would result in both more realistic and more accurate risk assessments.
