RESUMO

Combined vaccine "Bubo-Kok" is characterized by safety and high immunological activity. The number of postvaccinal reactions in children aged 1 and 2 years, immunized with vaccine "Bubo-Kok", was not statistically different from those in groups of children immunized with adsorbed DPT vaccine, as well with such vaccine in combination with vaccine against hepatitis B. After the completion of the primary course of immunization 100% of children had protective antibody titers against diphtheria, tetanus and hepatitis B. Antibody titers against pertussis, equal to or exceeding protective titers, were found in more than 70% of immunized children. The immunogenic potency of vaccine "Bubo-Kok" with respect to all its components was not inferior to that of adsorbed DPT vaccine and vaccine against hepatitis B, when introduced simultaneously in different areas of the body. Vaccine "Bubo-Kok" successfully passed state trials and was recommended for registration.

The dynamics of ultrafiltration was found to depend on the composition of the nutrient medium in which C.perfringens toxin accumulated. A new composition of casein-pancreatic medium with the reduced content of high-molecular peptides ensuring a high effect of membrane separation was developed. The influence of the main factors (pressure, concentration, the number of diafiltration cycles) on the characteristics of the membrane separation of type A C.perfringens culture. The optimum conditions for the purification of C.perfringens toxin by ultrafiltration were established with due regard to the data obtained in this investigation. Toxin concentrates were found to contain a fraction inhibiting their sorption capacity, and the method of the elimination of the inhibiting component was developed. C.perfringens toxoids, considerably exceeding commercial preparations in specific activity and immunogenic potency, were obtained.

The reactor technology for obtaining suspensions of Staphylococcus aureus (strain Cowan 1) with active protein A was developed. On the basis of the sensitization of bacterial suspensions with antitoxic rabbit sera staphylococcal reagents to diphtherial, gas gangrene (perfringens, oedematiens), botulinic toxins-toxoids were obtained and for the first time the coagglutination test with toxoids was carried out. More sensitive "specifically directed" reagents for the coagglutination test were obtained; for this purpose bacterial suspensions were sensitized with pure antibodies isolated from rabbit immune sera by immunosorption. The possibility of using the coagglutination test with specifically directed staphylococcal reagents for the rapid evaluation of the dynamics of toxin formation in reactors for the titration of purified toxoids, for the control of the purification of toxoids by ultrafiltration, as well as for the evaluation of the sorption-desorption of toxoids in the production of the corresponding preparations, was shown.

A gene for imidazole glycerophosphate dehydratase (HIS) has been selected from the library of Hansenula polymorpha genes by complementation of Escherichia coli hisB mutations or his3 mutation of Saccharomyces cerevisiae. Inactivation of the gene in Hansenula polymorpha strain 48V, as well as Leu(-)-Leu+ conversion of phenotype and arising of antibiotic G418 resistance resulted from transformation of the strain by the linear DNA molecules of the cloned HIS-gene with the in vitro inserted LEU2 gene from Saccharomices cerevisiae and KmR gene. The Southern hybridization analysis of the Leuf+His- G418R clones representing 1-2% of transformants population together with the DNA analysis of the monospore clones from the hybrid Leu+His-G418R transformant tetrad and tester strain have revealed the locus specific nature of the clones.

RESUMO

The ploidy of a mutant of Saccharomyces cerevisiae defective in recombination (Rec-) has been determined using tetrad analysis and flowing fluorometry. Evidence is obtained that the effect of the Rec- phenotype, i.e. the increase of the stability of plasmids with 2 mkm DNA ori replication in the yeast cirO cells is not the result of the diploidy in cells of the Rec- mutant developed in the process of transformation.
