RESUMO

Objective To observe the clinical efficacy of acupuncture combined with rehabilitation training under the guidance of Chinese medicine-enhanced recovery after surgery(CMERAS)in postoperative rehabilitation of tibial plateau fracture.Methods Sixty patients with tibial plateau fracture in postoperative rehabilitation period were randomly divided into observation group and control group,30 cases in each group.The control group was given routine postoperative rehabilitation training,and the observation group was treated with combined acupuncture treatment on the basis of the rehabilitation training of the control group.Both groups were treated for 3 consecutive months.After 3 months of treatment,the clinical efficacy of the two groups was evaluated,and the changes in the Visual Analogue Scale(VAS)scores of pain were observed before and after treatment,and the changes in the knee scores of the Hospital for Special Surgery knee score(HSS)of the United States were compared before and after treatment between the two groups.As well as the time of fracture healing on the operative side of the two groups of patients,and the fracture healing rate within 3 months of the two groups of patients were compared.Results(1)After 1 week,1 month and 3 months of postoperative treatment,the VAS scores of patients in the two groups were significantly improved compared with the pre-treatment period(P<0.05),and the observation group was significantly superior to the control group in improving the VAS scores,with a statistically significant difference(P<0.05).(2)After treatment,the HSS scores of patients in the two groups were significantly improved compared with those before treatment(P<0.05),and the observation group was significantly superior to the control group in improving the HSS scores,and the difference was statistically significant(P<0.05).(3)After 3 months of treatment,the fracture healing rate was 56.67%(17/30)in the observation group and 30.00%(9/30)in the control group.The fracture healing rate of the observation group was superior to that of the control group,and the difference was statistically significant(P<0.05).(4)After 3 months of acupuncture treatment,the total effective rate was 96.67%(29/30)in the observation group and 73.33%(22/30)in the control group.The efficacy of the observation group was superior to that of the control group,and the difference was statistically significant(P<0.05).Conclusion The clinical efficacy of acupuncture in postoperative rehabilitation of tibial plateau fractures under the guidance of CMERAS is significant,which reduces the clinical symptom of postoperative pain of the patients,shortens the healing time of fracture breaks,and significantly improves the patients'knee joint function.

【Objective】 To explore the molecular mechanism of Gasdermin B (GSDMB) regulating the fate of intestinal epithelial cells. 【Methods】 The human GSDMB plasmid was overexpressed into two human intestinal epithelial cell lines (NCM460 and HT-29 cells) and human colon-derived organoids. Western blotting was used to confirm the efficiency of electroporation. Cell counting kit (CCK8), cell apoptosis, and cell cycle by flow cytometry were performed to analyze the effect of GSDMB overexpression on cell function. Transcriptome sequencing was used to analyze the downstream effector molecules of GSDMB. T test was used to compare the data between the two groups. 【Results】 The overexpression of GSDMB protein in the two intestinal epithelial cell lines was successfully reconstructed. The absorbance value (A) of human intestinal epithelial cells overexpressing GSDMB protein [NCM460 cells: (1.17±0.01), HT-29 cells: (0.96±0.06)] was significantly lower than that of blank control cells [NCM460 cells: (1.67±0.12), HT-29 cells: (1.24±0.07)] (t=7.24 and 5.46, P<0.05). The number of apoptotic cells in the GSDMB overexpression group [NCM460 cells: (12.03±1.55), HT-29 cells: (29.30±4.48)] was significantly higher than that in the blank group [NCM460 cells: (4.96±1.74), HT-29 cells: (6.95±3.42)] (t=5.26 and 6.97, P<0.05). Cell cycle analysis showed that the ratio of cells at G0/G1 phase in the GSDMB overexpression group [NCM460 cells: (47.98±5.28)%, HT-29 cells: (38.04±3.45)%] was significantly lower than that in the control group [NCM460 cells: (59.54±3.90) %, HT-29 cells: (63.81±1.76) %] (t=3.05 and 11.53, P<0.05). Transcriptome sequencing results showed that the dual specificity phosphatase 4 and 6 (DUSP4 and DUSP6) genes were significantly upregulated after GSDMB protein expression. Fluorescence quantitative PCR results confirmed that the relative expression levels of DUSP4 (2.45±0.15) and DUSP6 (4.34±0.22) in intestinal epithelial cells transfected with GSDMB were significantly higher than those in the control group (1.06±0.05 and 1.01±0.02) (t=15.08 and 26.52, P<0.05). After GSDMB-expressing NCM460 cells were treated with the DUSP inhibitor BCI, the BCI treatment group had a significantly increased expression level of p-ERK compared to the control group [(1.14±0.17) vs. (0.58±0.12)] (t=5.42, P=0.002); the A value (1.84±0.07) and G0/G1 phase ratio (59.83±2.17)% in the BCI treatment group were significantly higher than those in the non-treatment group [(1.52±0.10) and (52.10±2.23)%] , and the number of apoptosis in the BCI treated group (7.60±0.56) was significantly lower than that in the untreated group (12.57±1.00) (t=4.71, 4.31, 7.52, P<0.05). TUNEL staining in human colon organoids showed a significant increase in apoptotic cells, and the relative expression level of DUSP6 protein (0.85±0.09) was significantly higher than that of the control group (0.21±0.04), accompanied by a decrease in p-ERK levels [(0.83±0.18) vs. (0.19±0.06)] , with statistical significance (t=11.95, P<0.001; t=6.56, P<0.001). 【Conclusion】 GSDMB may inhibit cell proliferation, induce cell cycle arrest, and promote apoptosis by upregulating dual specificity phosphatase DUSP6-mediated ERK phosphorylation, thus affecting the fate of intestinal epithelial cells.

Objective:To observe effect of Jingulian capsule on the proliferation of human breast cancer MDA-MB-231 cells and investigate its action mechanism against triple negative breast cancer (TNBC). Method:The ingredients of Jingulian capsule were identified by ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS). The inhibitory effect of Jingulian capsule at different doses (0.125,0.25,0.5,1,and 2 g·L<sup>-1</sup>) against the proliferation of MDA-MB-231 cells were detected by methyl thiazolyl tetrazolium (MTT) assay. After treatment for 24 h, the morphological changes in nuclear apoptosis of MDA-MB-231 cells were detected by Hoechst 33258 staining. The effect of different concentrations of Jingulian capsule on the apoptosis and cycle of MDA-MB-231 cells after different treatment time were determined by flow cytometry. The protein expression levels of Poly-ADP-ribose polymeras (PARP), proto-oncogene c-Myc, cyclin B<sub>1</sub>, and phosphorylated extracellular signal-regulated kinase (p-ERK) in each group were assayed by Western blot. Result:A total of 113 compounds in Jingulian capsule were identified by UPLC-MS/MS. As revealed by MTT assay,compared with blank group,Jingulian capsule (0.125,0.25,0.5,1,2 g·L<sup>-1</sup>) significantly inhibited viability of MDA-MB-231 cells (<italic>P</italic><0.01), with the half maximal inhibitory concentration ( IC<sub>50</sub>) of(0.13±0.02)g·L<sup>-1</sup>. According to flow cytometry,compared with the blank group,Jingulian capsule at 1 g·L<sup>-1</sup> significantly induced the apoptosis of MDA-MB-231 cells (<italic>P</italic><0.05)and Jingulian capsule at 0.5, 1 g·L<sup>-1</sup> obviously increased the number of MDA-MB-231 cells in S phase (<italic>P</italic><0.05,<italic>P</italic><0.01). The results of Western blotting demonstrated that the protein expression levels of PARP,c-Myc,and cyclin B<sub>1</sub> in 0.5, 1 g·L<sup>-1 </sup>Jingulian capsule groups were remarkably down-regulated as compared with those in the blank group(<italic>P</italic><0.01), and the protein expression level of p-ERK in 1 g·L<sup>-1 </sup>Jingulian capsule group was also down-regulated (<italic>P</italic><0.01). Conclusion:Jingulian capsule is able to inhibit the proliferation of MDA-MB-231 cells,induce S phase cell cycle arrest, and promote their apoptosis, which may be related to the inactivation of the MAPK signaling pathway.

The TORC2 gene is responsible for nutrient metabolism, gluconeogenesis, myogenesis and adipogenesis through the PI3K-Akt, AMPK, glucagon and insulin resistance signaling pathways. Sequencing of PCR amplicons explored three novel SNPs at loci g.16534694G>A, g.16535011C>T, and g.16535044A>T in the promoter region of the TORC2 gene in the Qinchuan breed of cattle. Allelic and genotypic frequencies of these SNPs deviated from Hardy-Weinberg equilibrium (HWE) (P < 0.05). SNP1 genotype GG, SNP2 genotype CT and SNP3 genotype AT showed significantly (P <0.05) larger body measurement and improved carcass quality traits. Haplotype H1 (GCA) showed significantly (p<0.01) higher transcriptional activity (51.44%) followed by H4 (ATT) (34.13%) in bovine preadipocytes. The diplotypes HI-H3 (GG-CC-AT), H1-H2 (GG-CT-AT) and H3-H4 (GA-CT-TT) showed significant (P<0.01) associations with body measurement and improved carcass quality traits. Analysis of the relative mRNA expression level of the TORC2 gene in different tissues within two different age groups revealed a significant increase (P<0.01) in liver, small intestine, muscle and fat tissues with growth from calf stage to adult stage. We can conclude that variants mapped within TORC2 can be used in marker-assisted selection for carcass quality and body measurement traits in breed improvement programs of Qinchuan cattle.

OBJECTIVE@#To investigate the down-regulation effect of let-7b-5p on the expression of FTO in acute myeloid leukemia cell line THP-1 and inhibitory effect on THP-1 proliferation via m@*METHODS@#The acute myeloid leukemia cell line THP-1 and the normal human peripheral blood mononuclear cells (PBMNC) were selected as subjects. The expression of let-7b-5p and FTO mRNA in those cells was detected by qPCR, further the expression of FTO protein in those cells was detected by Western blot. And, the luciferase reporter gene assay was used to verify the targeting effect of let-7b-5p on FTO. Finally, THP-1 cells were transfected respectively with let-7b-5p mimic, and PBMNC with let-7b-5p inhibitor, there after the C-MYC mRNA m@*RESULTS@#Compared with PBMNC, the expression of let-7b-5p in THP-1 significantly decreased, while the expression of FTO was significantly increased (P<0.05). After transfection with let-7b-5p mimic combined with FTO 3'-UTR, the luciferase activity of transfected THP-1 cells significantly decreased, but the luciferase activity significantly increased after transfection with mutant 3'-UTR, which was significantly different from the negative control group(blank vector) (P<0.05). Let-7b-5p inhibitor down-regulated c-MYC mRNA m@*CONCLUSION@#Human acute myeloid leukemia cell line THP-1 low expresses the let-7b-5p, which regulates c-MYC expression through let-7b-5p-/FTO-/m

The propagation characteristics of annular cos-Gaussian beams and Bessel-Gaussian beams in turbulence have been widely investigated. However, the expression of the average intensity of Bessel-Gaussian beams involves a complex double integral, which requires large computational resources and does not easily ensure accuracy. Under this background, through the general cos-Gaussian beam rotation, a new annular cos-Gaussian beam is proposed which not only has some properties of the general cos-Gaussian beam, but also properties similar to those of the Bessel-Gaussian beam. Moreover, the annular cos-Gaussian beam is simpler in form than the general one and much more time-saving in calculation than the Bessel-Gaussian beams. Based on the extended Huygens-Fresnel principle, the average intensity on a receiver plane of the annular cos-Gaussian beams can be simplified to a summation of several polynomials and it is in good agreement with that of low-order Bessel Gaussian beams. The average radius and the power in the bucket are also derived. In summary, the annular cos-Gaussian beam has promising applications for its quick calculation and simple form among annular beams and hollow beams.

Objective To investigate the correlation between dynamic contrast enhancement magnetic resonance imaging (DCE-MRI) quantitative parameters and pathological grading in esophageal squamous cell carcinoma(SCC). Methods Prospective analysis of esophageal squamous cell carcinoma confirmed by electronic gastrointestinal endoscopy was performed. Thirty nine patients who underwent radical resection of esophageal carcinoma with MRI examination one weeks before operation were included. All patients underwent routine chest MRI and DCE-MRI scans, and DCE-MRI quantitative parameters including volume transfer constant (Ktrans), exchange rate constant (Kep) and extravascular extracellular volume fraction(Ve)were measured.Pathological analysis of postoperative specimens,including pathological grading(highly differentiated,moderately differentiated,poorly differentiated,undifferentiated),gross tumor pathological type(ulcerative type,medullary type,fungating type,sclerotic type)and local infiltration degree (T staging) were performed. Kruskal-Wallis H test was used to compare the differences of quantitative parameters between different pathological T staging,gross tumor pathological types and pathological grades of DCE-MRI,and a Dunn-Bonferroni test for post hoc comparisons.Spearman rank correlation analysis was used to evaluate the correlation between DCE-MRI parameters and pathological grading of esophageal squamous cell carcinoma.The ROC curves was used to evaluate the efficiency of different parameters in the diagnosis of poorly differentiated esophageal squamous cell carcinoma. Result Among the thirty nine patients, they were divided into three group according to pathological findings: well differentiated (12 patients),moderately differentiated(15 patients)and poorly differentiated group(12 patients);ulcerative type (19 patients), fungating type(10 patients), medullary type(10 patients);T1, 2 stage(16 patients), T3 stage(14 patients), and T4 stage(9 patients). There was no significant difference in the value of Ktrans, Kepand Ve between different T staging groups and different tumor pathological types groups(all P>0.05).The differences of Ktrans, Kepand Vebetween different pathological grading groups were statistically significant (all P<0.05). There were positive correlation between Ktrans, Kep, Veand the pathological grading, rs value were 0.874, 0.672, 0.578 respectively, all P<0.01. The ROC curve area of Ktrans, Kepand Vein the diagnosis of poorly differentiated esophageal squamous cell carcinoma was 0.941,0.809 and 0.773 respectively.The diagnostic efficiency of Ktranswas the best.Conclusions The quantitative parameters of DCE-MRI are correlated with the pathological grading of esophageal squamous cell carcinoma. Ktrans, Kepand Vecan reflect the perfusion characteristics of esophageal squamous cell carcinoma.

AIMS: To investigate the association between triglyceride-glucose(TyG) index and the risk of hypertension. METHOD: A cross-sectional study was conducted in Bengbu, China. The participants received relevant questionnaire survey, anthropometric tests, and laboratory examination. Multivariate logistic regression analysis was performed to estimate the possible association between TyG index and hypertension risk. The additive interaction evaluated by the relative excess risk due to interaction (RERI), attributable proportion due to interaction (AP), and synergy index(SI) was calculated. RESULTS: A total of 1777 participants (748 men and 1029 women) were investigated. There was a significant increase in the risk of hypertension and isolated systolic hypertension (ISH) when comparing the highest TyG index (the fourth quartile) to the lowest TyG index (the first quartile) and corresponding ORs were 2.446 (95% CI: 1.746-3.426) and 2.621(95%CI: 1.627-4.224), respectively. However, no significant relationship was observed between TyG index and isolated diastolic hypertension (IDH). In males, significant interactions between TyG index and WHtR (RERI:1.978, 95%CI: 0.162-3.792; AP: 0.359, 0.113-0.605; SI:1.782, 1.017-3.122), smoking (AP: 0.437, 95%CI: 0.048-0.825), family history of hypertension (AP:0.433, 95%CI: 0.203-0.662; SI:2.248, 95%CI: 1.333-3.791) were observed. As for females, there were also significant interactions between TyG index and WHtR (RERI:1.415, 95%CI: 0.693-2.136; AP: 0.198, 95%CI: 0.104-0.291; SI:1.298, 95%CI:1.101-1.530), family history of hypertension (RERI:1.744, 95%CI: 0.221-3.267; AP:0.405, 95%CI: 0.113-0.697) on risk of hypertension. CONCLUSIONS: Increased TyG index was significantly associated with higher risk of hypertension and ISH, but not for IDH in middle-aged and elderly adults. Our results also demonstrated interactions of TyG index and abdominal obesity and family history of hypertension on hypertension risk.

We constructed a UC-VEGF-SMC three dimensional (3D) scaffold to explore its effect on blood vessel regeneration and bladder repair function in a rabbit model with bladder injury. Rabbit adipose tissue-derived stem cells (ADSCs) were cultured to construct pluripotent stem cell systems that can be induced to differentiate into urothelial cells (UCs) and smooth muscle cells (SMCs). Reverse transcriptase-polymerase chain reaction (RT-PCR), enzyme-linked immunosorbent assay (ELISA), immunofluorescence, MTT assay and HE staining were used in our study. Rabbit models were divided into an experimental group, control group and sham group. The bladder histology, urodynamics, smooth muscle function, stent degradation rate, urothelial permeability and biomechanical determination of rabbits were detected after grafting the scaffold. Correct stem cells based on the ADSC surface marker and found that CD90 and CD105 were positive and that CD34 and CD45 were negative. RT-PCR showed that ADSC-iPS cells expressed the marker gene of embryonic stem cells (ESCs), which indicated that Sox2, Klf4, Oct4 and c-Myc were inserted into the iPS nucleus and that the ADSC-iPS system was constructed successfully. Immunofluorescence and MTT assays indicated that iPS differentiated into mature SMCs and UCs. ELISA and HEMC culturing methods revealed that vascular endothelia growth factor (VEGF) could promote the growth of HMECs. Rabbit bladder repair function (urodynamics, smooth muscle function, urothelial permeability and biomechanical determination) was stronger in the experimental group than in the control group. UC-VEGF-SMC 3D nano scaffold exhibits good repair function for bladder damage, which may helpful for treatment of damaged bladders.

Objective To investigate the effects of monosodium tetrahexose ganglioside sodium (GM-1) combined with edaravone in the treatment of acute cerebral infarction (ACI) in patients with serum homocysteine (Hcy), high sensitivity C-reactive protein (Hs-CRP). Methods 66 patients with ACI were randomly divided into observation group and control group, 33 cases were treated with conventional neurology. The control group was treated with edaravone. The observation group was treated with edaravone and GM-1. (NIHSS) score, serum hs-CRP and Hcy levels were compared between the two groups. Results The total effective rate of the observation group was 90.91%, which was significantly higher than that of the control group (P<0.05). After 21 days of treatment, the NIHSS score, serum Hcy and hs-CRP in the observation group were significantly lower than those in the control group (P<0.05 ). Conclusion GM-1 combined with edaravone is effective in the treatment of ACI, which can promote the rehabilitation of neurological function and living activity. Its mechanism may be related to its anti-inflammatory response, reduced Hcy level and protection of brain cell injury.

Objective To evaluate the safety of Parecoxib Sodium for Injection.Methods Systemic active anaphylaxis test:Guinea pigs were injected respectively with Parecoxib Sodium for Injection (test sample,20 mg/mL),primary control,sodium chloride injection or human serum albumin,once every other day,continuously for three times.After 14 and 21 d from the end time of sensitization,to stimulation and to observe whether allergic reactions occurred within 30 mins.Passive cutaneous anaphylaxis test:Guinea pigs were received 0.1 mL antiserum injection for inducing passive sensitization,after 24 h of that we stimulated the guinea pigs,and the guinea pigs were sacrificed after 30 mins to examine the diameter of blue spots.Blood vessel irritation test:After continuous ear Ⅳ test sample for 5 d,HE staining was performed at the end of the administration and recovery period,and the stimulation of the blood vessel at the site of injection was observed.Hemolysis or agglutination of Parecoxib Sodium for Injection was examined by in vitro methods.Results Under the dosage of 20 mg/mL,guinea pigs showed no systemic allergy and passive skin allergy,and no hemolysis,agglutination,and irritation of vascular was observed.Conclusion Under the present experimental conditions,20 mg/mL Parecoxib Sodium for Injection shows no obvious allergic reactions,irritation and hemolysis,is safe.

Objective To prospectively determine the feasibility of high-resolution in vivo MR imaging in the evaluation of esophageal carcinoma invasion at 3.0 T.Methods One hundred and eighteen patients with esophageal carcinoma,proven by the gastroscopic biopsy,were prospectively studied using 3.0 T MR.The esophageal specimens were sectioned transversely to keep consistent in the orientation with the MR images,the histopathological stage was made and the thickness of the tumor on the largest diameter of the slice were measured.The MR images were reviewed in the transverse plane.According to the seventh American joint committee on cancer,the MR stage was made and the tumor's thickness was measured.The MR images and the histopathological slices were matched.The staging diagnostic efficacy of the MR imaging was evaluated with the histopathological results as the standard reference,Kappa test was used to compare the stage of MR imaging with that at the histopathological analysis.Bland-Altman scatterplots were used to compare the thickness of tumor measured on the MR images with that at the histopathological measurement.Results Ninety seven cases(82.2%,97/118) of MR stage were accurately made,including 7 T1a,15 T1b,18 T2,25 T3 and 32 T4a cases,furthermore,14 cases were over staged and 7 cased were underestimated.The MR stage was highly consistent with the histopathological stage (Kappa=0.772).The sensitivity for the staging of high-resolution MR imaging at 3.0 T was 58.3%(7/12) to 100.0%(32/32),the specificity was 95.3% (82/86) to 98.1% (104/106),and the accuracy was 91.5% (108/118) to 96.6% (114/118),respectively.Bland-Altman scatterplots demonstrated that the discrepancy of the mean thickness between the value obtained by three radiologists respectively and the histopathological analysis were 2.0,2.6 and 2.1 mm,which demonstrated a good consistency.Conclusion High-resolution MR images obtained at 3.0 T can be used to evaluate the depth of carcinoma invasion and provide excellent diagnostic accuracy for preoperative staging.

<p><b>OBJECTIVE</b>To establish an improved method for stereotactic location of the supraoptic nucleus in rats.</p><p><b>METHODS</b>Twenty-four SD rats were randomly divided into experimental group (12 rats) and control group (12 rats) for oblique (20° to the left) stereotactic puncture (OSP group) and vertical stereotactic puncture (VSP group), respectively, both targeting the supraoptic nucleus (SON). The surgical data and postoperative (within 24) mortality of the rats were compared between the two groups.</p><p><b>RESULTS</b>The nucleus locating time was longer in OSP group than in VSP group (59.55∓3.64s vs 27.44∓2.18 s, P=0.000), and the postoperative mortality rate of the rats did not differ significantly between the groups (0 vs 44.4%, P=0.082). In OSP group, compared with VSP group, the procedure was associated with a lowered rupture rate of the superior sagittal sinus (11.1% vs 88.9%, P=0.003), a shortened hemostatic time after craniotomy (52.89∓24.05 s vs 157.445 ime a s, P=0.000) and after puncture (24.33 reas 45 s vs 133.89∓28.81 s, P=0.000), and also a shortened operation time (178.89 on tims vs 362.44 timees, P=0.000).</p><p><b>CONCLUSION</b>The improved method for locating supraoptic nucleus in rats is convenient, stable and reproducible, and helps to avoid important blood vessels and specific nuclei according to the needs of different experiments and allows the operators to choose different surgical paths.</p>

Objective To investigate changes of the elastic modulus of astrocytes induced by injury. Methods The astrocytes were isolated and extracted from the 2-day old SD rats, and identified by immunofluorescence staining with glial fiber acidic protein (GFAP) antibody. Cells were divided into control group and injured group. The injured group was astrocytes 6 h after being injured by the cell damage instrument. The control group was astrocytes without any injury. The elastic modulus in liquid phase was tested by atomic force microscope in two groups. Results were compared and analyzed between two groups. Results The purification rate of rat astrocytes was more than 95%. Six hours after the injury, the astrocytes were in disorder, and some of cell bodies were swelling. The mechanical topographic maps and force indentation curves were obtained. The elastic modulus of astrocytes was significantly increased in injured group compared with that of control group[(1 689±693) Pa vs. (724±283) Pa, P<0.01]. Conclusion The injury stimulus increases the elastic modulus of astrocytes, which provides theoretical basis for understanding intracranial physical microenvironment after traumatic brain injury in animal experiments.

RESUMO

We reported a technical report of traumatic lower cervical spondylolisthesisca used by bilateral pedicle fracture, without neurological compression. The patient was treated with the minimally invasive technique of percutaneous pedicle screw fixation. Fracture healing and normal cervical motion were confirmed by plain films and physical examinations on the 18-monthpostoperatively. The technique of percutaneous pedicle screw fixation might be an alternative strategy for the treatment of traumatic lower cervical spondylolisthesis with pedicle fracture.

RESUMO

<p><b>OBJECTIVE</b>To observe the effects of different anesthesia ways on endorphin and hemodynamics of laparoscopic cholecystectomy patients in the perioperative phase.</p><p><b>METHODS</b>A total of 90 laparoscopic cholecystectomy patients, 29 to 80 years old, were randomly assigned to Group A (treated with electroacupuncture at acupoints combined general anesthesia), Group B (treated with electroacupuncture at non-acupoints combined general anesthesia), and Group C (treated with general anesthesia) according to American Society of Anesthesiologists (ASA) I-II, 30 cases in each group. All patients were induced by 3 microg/kg Fentanyl (Fen), 2 mg/kg Propofol (Pro), and 0.1 mg/kg Vecuronium (Vcr). Bispectral index (BIS), being 40 -65, indicated the state of general anesthesia. The anesthesia was maintained by intravenous injecting Pro, interruptedly intravenous injecting Fen and Vcr. Each patient received patient controlled intravenous analgesia (PCIA) after operation. On these bases, patients in Group A received electrical acupuncture at bilateral Hegu (LI4), Neiguan (PC6), Quchi (Ll11), Zusanli (ST36), and Yanglingquan (GB34). Patients in Group B received electrical acupuncture at the points beside acupoints. The electroacupuncture was lasted from 15 -30 min before anesthesia induction to the end of the operation in Group A and B. The heart rate (HR), mean arterial pressure (MAP), cardiac index (CI), cardiac output (CO), systemic vascular resistance index (SVRI), and acceleration index (ACI) were recorded before anesthesia induction, immediate before pneumoperitoneum, 5 min after pneumoperitoneum, excision of gallbladder, and at the end of operation. The time consumption from discontinuation to spontaneously breathing recovery, analeptic, and extubation were recorded. The blood samples (3 mL each time) were collected from the peripheral vein before anesthesia induction, 2 h after operation, the 1st day after operation, and the 3rd day after operation to detect the beta-endorphin (beta-EP) level. The visual analogue scale (VAS) were observed and recorded in the 3 groups at post-operative 4, 6, 8, 24, and 44 h, respectively.</p><p><b>RESULTS</b>(1) Compared with before anesthesia induction in the same group, the CI, CO, ACI of all patients decreased significantly at 5 min after pneumoperitoneum and at excision of gallbladder (P < 0.01, P < 0.05). The HR, MAP, SVRI obviously increased in Group B and Group C at each time point (P < 0.05, P < 0.01). Less change happened in Group A. Compared with Group C, the increment of MAP was less in Group A at 5 min after pneumoperitoneum, showing statistical difference (P < 0.05). (2) The time consumption from discontinuation to analeptic and extubation was obviously shorter in Group A than in Group B and Group C (P < 0.05, P < 0.01). (3) The level of beta-EP on the 1st day of operation was significantly lower in Group A than in Group B (P < 0.05) and Group C (P < 0.01). (4) The VAS score at post-operative 44 h was significantly lower in Group A than in Group B and Group C (P < 0.05).</p><p><b>CONCLUSIONS</b>Electroacupuncture at acupoints combined general anesthesia could maintain the stabilization of haemodynamics, and relieve the stress reaction after pneumoperitoneum and operation, and prolong it to early post-operative period, and strengthen the effects of post-operative analgesia. The post-operative recovery was fast, safe, and reliable.</p>

<p><b>OBJECTIVE</b>To compare the efficacy and feasibility of neoadjuvant chemoradiotherapy with docetaxel plus cisplatin or with cisplatin plus fluorouracil in the treatment of local advanced esophageal squamous cell carcinoma.</p><p><b>METHODS</b>A total of 154 cases in the stage of cT3N0-1M0 were randomly assigned to two arms. The arm A received 2 cycles of doctaxel 75 mg/m(2) plus cisplatin 25 mg/m(2) d1-3 and 40 Gy of radiation therapy, and the arm B received 2 cycles of cisplatin 25 mg/m(2) d1-3 plus fluorouracil 600 mg/m(2) d1 ∼ 5 and 40 Gy of radiation therapy. The surgery was performed 3 - 4 weeks later.</p><p><b>RESULTS</b>Grade 3/4 toxicities occurred in 53.2% of the patients in arm A and in 36.4% of the patients in arm B (P = 0.035). Neutropenia occurred in 20.7% of the patients in arm A and 5.6% of the patients in arm B (P = 0.004). Nine patients aborted surgery due to tumor progression. 71 patients underwent resection in 73 cases of the arm A and 69 patients underwent complete resection, 70 patients underwent resection in 72 cases and 70 complete resection of the arm B, respectively (P > 0.05). No mortality was noted. The overall complication rate was similar in the two arms (21.9% vs. 23.6%). Pathological complete response was achieved in 27 patients (35.1%) in the arm A and 16 patients (20.8%) in the arm B (P = 0.048).</p><p><b>CONCLUSIONS</b>Neoadjuvant chemoradiotherapy with docetaxel plus cisplatin can be well tolerated and achieves a higher pathological complete response rate than with cisplatin plus fluorouracil.</p>

This study was aimed to investigate the expression of Hippo signaling pathway core element MST1 gene in acute leukemia (AL) patients, and explore its relation with AL pathogenesis and prognosis. 50 newly diagnosed patients with AL, 33 normal people, 23 patients with AL in complete remission, 12 refractory or relapsed patients were tested for the expression of MST1 gene by real-time quantitative PCR, Western blot was used to further validate the level of MST1 protein expression. And combined with clinical data, prognostic factors of the patients were analyzed. The results indicated that compared with the normal people, the expression level of MST1 in newly diagnosed patients with AL significantly decreased (P < 0.05), but significantly increased in AL patients with complete remission (CR), the difference of expression was statistically significant before CR and after CR (P < 0.05). Compared with refractory or relapsed patients, the expression level of MST1 gene in newly diagnosed patients was not significantly different (P > 0.05). Besides, the expression level of MST1 between the patients with CR and the normal people was not significantly different (P > 0.05). It is concluded that the low expression of MST1 may be related with the pathogenesis and prognosis of AL.
