RESUMO

INTRODUCTION: Despite advancements in the methods for prevention and early diagnosis of gastric cancer (GC), GC continues to be the fifth in incidence among major cancers and the third most common cause of cancer-related death. The therapeutic effects of surgery and drug treatment are still unsatisfied and show notable differences according to the tumor microenvironment (TME) of GC. METHODS: Through screening Pubmed, Embase, and Web of Science, we identified and summarized the content of recent studies that focus on the investigation of Helicobacter pylori (Hp) infection, regulatory T cells (Tregs), and tumor-associated macrophages (TAMs) in the TME of GC. Furthermore, we searched and outlined the clinical research progress of various targeted drugs in GC treatment including CTLA-4, PD-1\PD-L1, and VEGF/VEGFR. RESULTS: In this review, the findings indicate that Hp infection causes local inflammation and leads to immunosuppressive environment. High Tregs infiltration in the TME of GC is associated with increased induction and recruitment; the exact function of infiltrated Tregs in GC was also affected by phenotypes and immunosuppressive molecules. TAMs promote the development and metastasis of tumors, the induction, recruitment, and function of TAMs in the TME of gastric cancer are also regulated by various factors. CONCLUSION: Discussing the distinct tumor immune microenvironment (TIME) of GC can deepen our understanding on the mechanism of cancer immune evasion, invasion, and metastasis, help us to reduce the incidence of GC, and guide the innovation of new therapeutic targets for GC eventually.

Objective:Using different machine learning methods to construct and screen the best prediction model for predicting the risk of death within 30 days after surgery in patients with acute type A aortic dissection.Methods:Five hundred and twenty-one patients with acute type A aortic dissection who underwent surgery between 2015 and 2022 were included, after collecting their perioperative date and screening them, 329 patients were retained. two different groups of predictor variables were generated by using Lasso regression and principal component analysis, after that, logistic regression, support vector machine algorithm, random forest algorithm, gradient boosting algorithm, and super learning algorithm were used to develop prediction models for the risk of death within 30 days after surgery. Finally, we compare the models and select the best one. Results:The AUC values for all models rangrd from 0.791-0.959. The model using Lasso regression to determine the predictor variables and built by the super learning algorithm had the best prediction with an AUC value of 0.959. Conclusion:The super learning algorithm better than other algorithms in predicting death within 30 days after acute type A aortic dissection.

@#Objective To investigate the efficacy of low-dose inhaled nitric oxide (iNO) in the treatment of severe hypoxemia after Sun’s operation. Methods The clinical data of patients undergoing Sun’s operation for acute Type A aortic dissection in our hospital from January 2020 to June 2022 were retrospectively analyzed. Patients who received conventional treatment before November 2021 were enrolled as a control group. After November 2021, iNO was used in our hospital, and the patients who received iNO as an iNO group. The preoperative clinical baseline data, perioperative clinical data and oxygenation index were compared between the two groups. Results A total of 54 patients were included in the control group, including 45 males and 9 females, with an average age of 53.0±10.9 years. A total of 27 patients were included in the iNO group, including 21 males and 6 females, with an average age of 52.0±10.6 years. The preoperative body mass index of the two groups was greater than 25 kg/m2, white blood cell count, C-reactive protein were significantly higher than normal level, but there was no statistical difference between the groups (P>0.05). There were no statistical differences in intraoperative data between the two groups (P>0.05). The iNO group had significantly shorter duration of mechanical ventilation, postoperative ICU stay, and postoperative hospital stay than the control group (P<0.001). After 12 h of iNO treatment, hypoxic condition improved obviously, oxygenation indices in 12 h, 24 h, 36 h,48 h, 60 h and 72 h in the iNO group were significantly higher than those in the control group (P<0.05). Conclusion The treatment of severe hypoxemia after Sun’s surgery with low-dose of iNO is safe and effective, can significantly improve oxygenation function, and has significant advantages in shortening ventilator use time, postoperative ICU stay and postoperative hospital stay, but it is not significant in changing postoperative mortality.

ObjectiveTo clone coumarate-3-hydroxylase gene (C3H) from Angelica sinensis, and analyze the correlation between its bioinformatics, expression patterns and content of ferulic acid, and to explore the functions of ASC3H. MethodReal-time polymerase chain reaction (Real-time PCR) was used to clone the full-length cDNA of ASC3H based on the transcriptome dataset of A. sinensis, and the bioinformatics analysis of the gene sequence was carried out. Real-time PCR and high performance liquid chromatography (HPLC) were used to determine relative expression of ASC3H and content of ferulic acid in different root tissues of A. sinensis (periderm, cortex and stele). ResultThe open reading frame (ORF) of ASC3H (GenBank accession number: MN2550298) was 1 530 bp, encoding 509 amino acids, with a theoretical molecular weight of 57.86 kDa and an isoelectric point of 8.36. It was a hydrophilic protein that was located in the chloroplast with multiple phosphorylation sites and a transmembrane region, and contained a conserved domain CGYDWPKGYGPIINVW_P450 (383-399 aa) in cytochrome P450. Multiple amino acid sequence alignment analysis showed that ASC3H had high similarity with C3H from other plants, especially Ammi majus in Umbelliferae. The Real-time PCR revealed that ASC3H had different expressions in periderm, cortex and stele tissues of A. sinensis roots. It was found from HPLC that the cortex tissues had the highest content of ferulic acid, and the stele tissues had the lowest. ConclusionASC3H was successfully cloned from A. sinensis, and its sequence characteristics were understood more clearly, suggesting that ASC3H might be involved in the ferulic acid biosynthesis pathway of A. sinensis. This paper provided a basis for further studying the functions of the gene and exploring the biosynthesis and regulation mechanism of ferulic acid in A. sinensis, while laying the foundation for the genetic improvement of A. sinensis.

It is not fully clear why there is a higher contribution of pluripotent stem cells (PSCs) to the chimera produced by injection of PSCs into 4-cell or 8-cell stage embryos compared with blastocyst injection. Here, we show that not only embryonic stem cells (ESCs) but also induced pluripotent stem cells (iPSCs) can generate F0 nearly 100% donor cell-derived mice by 4-cell stage embryo injection, and the approach has a "dose effect". Through an analysis of the PSC-secreted proteins, Activin A was found to impede epiblast (EPI) lineage development while promoting trophectoderm (TE) differentiation, resulting in replacement of the EPI lineage of host embryos with PSCs. Interestingly, the injection of ESCs into blastocysts cultured with Activin A (cultured from 4-cell stage to early blastocyst at E3.5) could increase the contribution of ESCs to the chimera. The results indicated that PSCs secrete protein Activin A to improve their EPI competency after injection into recipient embryos through influencing the development of mouse early embryos. This result is useful for optimizing the chimera production system and for a deep understanding of PSCs effects on early embryo development.

A pH- and temperature-responsive magnetic adsorbent [poly(N-isopropylacrylamide) grafted chitosan/Fe3O4 composite particles, CN-MCP], was synthesized for the removal of the endocrine-disrupting chemical nonylphenol. According to the structural characteristics (changeable surface-charge and hydrophilic/hydrophobic properties) of the targeted contaminant, CN-MCP was designed owning special structure (pH- and temperature-responsiveness for the changeable surface-charge and adjustable hydrophilic/hydrophobic properties, respectively). Compared to chitosan magnetic composite particles without grafting modification (CS-MCP) and several other reported adsorbents, CN-MCP exhibited relatively high adsorption capacity for nonylphenol under corresponding optimal conditions (123 mg/g at pH 9 and 20 °C; 116 mg/g at pH 5 and 40 °C). Meanwhile, high selectivity of the novel adsorbent in selective adsorption of nonylphenol from bisolute solution of nonylphenol and phenol was found. Effects of grafting ratio of the grafted polymer branches and coexisting inorganic salts on the adsorption were systematically investigated. Moreover, CN-MCP demonstrated desired reusability during 20 times of adsorption-desorption recycling. The high adsorption capacity, high selectivity, and desired reusability aforementioned revealed the significant application potential of CN-MCP in the removal of NP. On the basis of the adsorption behaviors, isotherms equilibrium, thermodynamics and kinetics studies, and instrumental analyses including X-ray photoelectron spectroscopy, BET specific surface area, zeta potential, and static water contact angle measurements, distinct adsorption mechanisms were found under various conditions: charge attraction between CN-MCP and the contaminant, as well as binding between polymeric branches of CN-MCP and nonyls, contributed to the adsorption at pH 9 and 20 °C; whereas hydrophobic interaction between CN-MCP and nonylphenol played a dominant role at pH 5 and 40 °C. The current study provided a strategy for the structural design of adsorbents according to the features of targeted emerging contaminants, and the continuity of the work was discussed and proposed.

A novel hydrogen peroxide sensor was fabricated by the seed-mediated growth method. First, polyethyleneimine(PEI) functionalized multiwalled carbon nanotubes(MWNTs) were used as growth scaffold on the glass carbon electrode ( GCE). Then, Au nanoparticles were electrodeposited uniformly as seeds. Finally, Pt nanoparticles ( PtNPs ) grew on Au nanoparticles to form Pt @ Au core-shell structure nanocomposite. A new type of electrochemical sensor based on Pt @ Au / PEI-MWNTs nanocomposites for detection of hydrogen peroxide was developed, and the designed Pt@ Au / PEI-MWNTs/ GCE was characterized by electrochemical methods and field emission scanning electron microscopy (FESEM). The Differential pulse experimental results showed that the modified electrode exhibited excellent electrocatalytic activity towards the reduction of H2 O2 with the wide linear range from 9. 2 ×10-8 mol/ L to 1. 3 ×10-3 mol/ L. The correlation coefficient was 0. 9994 and the low detection limit was 3. 1×10-8 mol/ L at the signal-to-noise of 3.
