Visible and near-infrared (NIR) upconversion luminescence (UCL) emissions originating from the BaY2F8: Yb3+, Tm3+ systems were investigated under a laser excitation at 980 nm. The BaY2F8:20 mol% Yb3+, x mol% Tm3+ and BaY2F8: y mol% Yb3+, 0.5 mol% Tm3+ phosphors showed prominent UCL at 800 and 810 nm. The optimized doping concentrations of Yb3+ and Tm3+ in the BaY2F8 host matrix were evaluated, their spectroscopic properties were determined, and studies on their temperature-dependent behaviour were carried out. The temperature-sensing properties were studied by generating the fluorescence intensity ratio (FIR) of the UCL peaks originating from the thermally-coupled energy levels of the Tm3+ ions. The Stark sublevels of 1G4 level of Tm3+ ions were utilized to estimate the temperature-sensing abilities of the phosphor.

RESUMO

Impending paradoxical embolism (IPDE) is a clinical emergency with adverse outcomes. Due to its rarity, larger research cannot be obtained to provide definitive therapy alternatives. We report a case of a tumor embolus from a renal cell carcinoma (RCC) that caused a right atrial mass, pulmonary embolus, and impending paradoxical embolus via a patent foramen ovale (PFO) and its management.

Reduced sulfur compounds are a nuisance in coastal industries causing heavy economical as well as ecological loss. One such compound, hydrogen sulfide, is proven toxic to aquatic animals as it interferes with their respiration and metabolism as well as overall development, thereby causing direct increase in mortality. Typically, 96-h LC50 values to freshwater and marine fishes are 0-25µM and 525-700µM, respectively. Management of sulfide and other reduced sulfur compounds from aquaculture water and sediment using bioremediating sulfur-oxidizing bacteria as probiotics has attracted attention in recent decades due to its efficiency and minimized environmental effects. In the present study, 201 native and indigenous probiotic candidates were isolated, from various coastal environments. The prospective candidates were screened based on pH reduction and 19 sulfur-oxidizing bacteria were selected and tested for salt tolerance. Further screening was done based on biosafety, ability to produce sulfate by oxidizing thiosulfate, and 16S rRNA-based identification to obtain nine probiotic candidates. Three strains (Enterobacter ludwigii HS1-SOB, Pseudomonas stutzeri B6-SOB, and Cytobacillus firmus C8-SOB) exerting highest sulfate-ion production were selected for formulating a probiotic consortium using mixture design matrix. The optimal composition was determined to be equal ratios of the three isolates that yielded 0.083 mM of sulfate from thiosulfate broth medium at room temperature in 7 days. This is a standalone report of sulfur-oxidizing probiotic consortium composed of the said bacteria. The consortium may be used as a strong tool for remediation of reduced sulfur in aquaculture and associated coastal environments.

BACKGROUND: Various suture materials are available for arthroscopic rotator cuff repair. More recently, suture tapes have become popular as they are perceived to be easier to use with less soft tissue irritation. However, little is known about their biomechanical and clinical properties compared with conventional sutures in rotator cuff repairs. PURPOSE: To perform a systematic review and meta-analysis on whether suture tapes are biomechanically superior to conventional sutures in arthroscopic rotator cuff repairs and whether this translates to superior functional outcomes and a lower incidence of retears. STUDY DESIGN: Meta-analysis. METHODS: The Cochrane Controlled Register of Trials, PubMed, Medline, and Embase were used to perform a systematic review and meta-analysis using the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) criteria with the following search terms: (rotator cuff repair OR arthroscopic rotator cuff repair) AND ("tape" OR "wire" OR "cord" OR "suture"). Data pertaining to certain biomechanical properties (contact area, contact pressure, gap formation, load to failure, and stiffness), retears, and patient-reported outcome measures (PROMs) were extracted. The pooled outcome data were analyzed by random- and fixed-effects models. RESULTS: After abstract and full-text screening, 7 biomechanical and 6 clinical studies were included. All biomechanical studies were on animals, with 91 suture tapes and 91 conventional sutures compared. Suture tapes had higher contact pressure (mean difference [MD], 0.04 MPa; 95% CI, 0.01-0.08; P = .02), higher load to failure (MD, 52.62 N; 95% CI, 27.34-77.90; P < .0001), greater stiffness (MD, 4.47 N/mm; 95% CI, 0.57-8.38; P = .02), and smaller gap formation (MD, -0.30 mm; 95% CI, -0.45 to -0.15; P < .0001) compared with conventional sutures. From the clinical analysis of the 681 rotator cuff repairs treated with a suture tape (n = 380) or conventional suture (n = 301), there were no differences in retear rates between the groups (16% vs 20% suture tape and wire, respectively; P = .26) at a mean of 11.2 months. Qualitatively, there were no differences in PROMs between the groups at a mean of 36.8 months. CONCLUSION: Although biomechanically superior, suture tapes showed similar retear rates and postoperative function to conventional sutures. However, higher-quality clinical studies are required to investigate whether there are no true differences.

The lossy mode resonance (LMR) phenomenon is almost exclusively limited to fiber optics, since the thin film configuration yields a relatively shallow resonance dip compared to its fiber counterpart. In this Letter, we bridge this frustrating intensity gap between these basic configurations by choosing vacuum-deposited metallic indium-rich indium tin oxide as the coating material. LMR attenuation as high as -14.3dB for transverse electric and -6.4dB for transverse magnetic polarization is achieved experimentally via a commonly used Kretschmann-Raether geometry and that too for a film thickness of ≈70nm. Such a high degree of LMR response is attributed to the metallic indium generated interbands, leading to a high extinction coefficient in the visible range. A modified transfer matrix method, which takes into account the surface roughness of the films through application of the anisotropic Bruggeman effective medium approximation, is developed to realize the experimental LMR spectra numerically.

Enzymatic pretreatment is emerging as an efficient tool for the extraction of biofuel precursors from algal biomass. However, yardsticks for end-use directed selection of optimal pretreatment conditions are not yet identified. The present study, for the first time, reveals different optimal conditions for algal biomass solubilization and sugar release. Algal biomass pretreatment optimization was carried out using the Taguchi method. Crude enzyme from Aspergillus fischeri was found effective for pretreatment of Chlorella pyrenoidosa. Maximum sugar yield (190 mg g-1 biomass) from algal biomass was observed at a substrate concentration of 4 g L-1, with a 5% enzyme load at temperature 60°C, pH 5.5, and shaking speed of 80 rpm. In contrast, maximum sCOD (1350 mg g-1 biomass) was obtained at 2 g L-1 substrate concentration with enzyme load of 20% v/v, at 60°C, pH 4, and shaking speed of 100 rpm. Hence, the first set of conditions would be more beneficial for bioethanol production. Whereas another set of conditions would improve the biofuel production that requires maximum solubilization of algal biomass, such as fermentative methane production. Overall, the present observations established that process conditions required for enzymatic pretreatment of algal biomass should be selected according to the desired biofuel type.

COVID-19 outbreak is the biggest threat to human health in recent history. Currently, there are over 1.5 million related deaths and 75 million people infected around the world (as of 22/12/2020). The identification of virulence factors which determine disease susceptibility and severity in different cell types remains an essential challenge. The serine protease TMPRSS2 has been shown to be important for S protein priming and viral entry, however, little is known about its regulation. SPINT2 is a member of the family of Kunitz type serine protease inhibitors and has been shown to inhibit TMPRSS2. Here, we explored the existence of a co-regulation between SPINT2/TMPRSS2 and found a tightly regulated protease/inhibitor expression balance across tissues. We found that SPINT2 negatively correlates with SARS-CoV-2 expression in Calu-3 and Caco-2 cell lines and was down-regulated in secretory cells from COVID-19 patients. We validated our findings using Calu-3 cell lines and observed a strong increase in viral load after SPINT2 knockdown, while overexpression lead to a drastic reduction of the viral load. Additionally, we evaluated the expression of SPINT2 in datasets from comorbid diseases using bulk and scRNA-seq data. We observed its down-regulation in colon, kidney and liver tumors as well as in alpha pancreatic islets cells from diabetes Type 2 patients, which could have implications for the observed comorbidities in COVID-19 patients suffering from chronic diseases.

Exacerbated pro-inflammatory immune response contributes to COVID-19 pathology. However, despite the mounting evidence about SARS-CoV-2 infecting the human gut, little is known about the antiviral programs triggered in this organ. To address this gap, we performed single-cell transcriptomics of SARS-CoV-2-infected intestinal organoids. We identified a subpopulation of enterocytes as the prime target of SARS-CoV-2 and, interestingly, found the lack of positive correlation between susceptibility to infection and the expression of ACE2. Infected cells activated strong pro-inflammatory programs and produced interferon, while expression of interferon-stimulated genes was limited to bystander cells due to SARS-CoV-2 suppressing the autocrine action of interferon. These findings reveal that SARS-CoV-2 curtails the immune response and highlights the gut as a pro-inflammatory reservoir that should be considered to fully understand SARS-CoV-2 pathogenesis.

Neuroblastoma is a pediatric tumor of the developing sympathetic nervous system. However, the cellular origin of neuroblastoma has yet to be defined. Here we studied the single-cell transcriptomes of neuroblastomas and normal human developing adrenal glands at various stages of embryonic and fetal development. We defined normal differentiation trajectories from Schwann cell precursors over intermediate states to neuroblasts or chromaffin cells and showed that neuroblastomas transcriptionally resemble normal fetal adrenal neuroblasts. Importantly, neuroblastomas with varying clinical phenotypes matched different temporal states along normal neuroblast differentiation trajectories, with the degree of differentiation corresponding to clinical prognosis. Our work highlights the roles of oncogenic MYCN and loss of TFAP2B in blocking differentiation and may provide the basis for designing therapeutic interventions to overcome differentiation blocks.

The migrational propensity of neuroblastoma is affected by cell identity, but the mechanisms behind the divergence remain unknown. Using RNAi and time-lapse imaging, we show that ADRN-type NB cells exhibit RAC1- and kalirin-dependent nucleokinetic (NUC) migration that relies on several integral components of neuronal migration. Inhibition of NUC migration by RAC1 and kalirin-GEF1 inhibitors occurs without hampering cell proliferation and ADRN identity. Using three clinically relevant expression dichotomies, we reveal that most of up-regulated mRNAs in RAC1- and kalirin-GEF1-suppressed ADRN-type NB cells are associated with low-risk characteristics. The computational analysis shows that, in a context of overall gene set poverty, the upregulomes in RAC1- and kalirin-GEF1-suppressed ADRN-type cells are a batch of AU-rich element-containing mRNAs, which suggests a link between NUC migration and mRNA stability. Gene set enrichment analysis-based search for vulnerabilities reveals prospective weak points in RAC1- and kalirin-GEF1-suppressed ADRN-type NB cells, including activities of H3K27- and DNA methyltransferases. Altogether, these data support the introduction of NUC inhibitors into cancer treatment research.

Half of the children diagnosed with neuroblastoma (NB) have high-risk disease, disproportionately contributing to overall childhood cancer-related deaths. In addition to recurrent gene mutations, there is increasing evidence supporting the role of epigenetic deregulation in disease pathogenesis. Yet, comprehensive cis-regulatory network descriptions from NB are lacking. Here, using genome-wide H3K27ac profiles across 60 NBs, covering the different clinical and molecular subtypes, we identified four major super-enhancer-driven epigenetic subtypes and their underlying master regulatory networks. Three of these subtypes recapitulated known clinical groups; namely, MYCN-amplified, MYCN non-amplified high-risk and MYCN non-amplified low-risk NBs. The fourth subtype, exhibiting mesenchymal characteristics, shared cellular identity with multipotent Schwann cell precursors, was induced by RAS activation and was enriched in relapsed disease. Notably, CCND1, an essential gene in NB, was regulated by both mesenchymal and adrenergic regulatory networks converging on distinct super-enhancer modules. Overall, this study reveals subtype-specific super-enhancer regulation in NBs.

A BaY2 F8 :Yb3+ ,Ho3+ phosphor was synthesized using a simple precipitation method. The temperature-sensing properties of BaY2 F8 :Yb3+ ,Ho3+ phosphor were investigated using the fluorescence intensity ratio (FIR) of the thermally coupled energy levels (5 F4 and 5 S2 ) of Ho3+ . Energy transitions 5 F4 →5 I8 and 5 S2 →5 I8 gave rise to emission peaks at 538 nm and 549 nm, respectively. The areas under these two emission peaks were used to calculate the FIR. Temperature-dependent upconversion luminescence was recorded in the temperature range 303-623 K using a 980 nm laser excitation source. The results suggested that the BaY2 F8 :Yb3+ , Ho3+ phosphor has the potential to be used as a non-contact optical temperature sensor.

Lisfranc injuries are relatively uncommon but carry devastating consequences if left untreated. Although many surgical techniques have been proposed for best operative management, there is an ongoing debate over which procedure is superior. We performed a systematic review and meta-analysis comparing the outcomes of transarticular screw fixation and dorsal bridge plating in management of Lisfranc injuries. Ovid MEDLINE, Ovid Embase and Cochrane Central Register of Controlled Trials (CENTRAL) were searched to identify randomised controlled trials (RCTs) and cohort studies comparing the outcomes between screw and dorsal plate fixation. The pooled outcome data were calculated by random and fixed effect models. One prospective cohort and three retrospective studies were identified with a total of 210 patients with mean follow up of 40.6 months. All papers were analysed for quality using the modified Newcastle Ottawa score. The results show that dorsal bridge plating is associated with better American Orthopaedic Foot and Ankle Society score (AOFAS) compared with transarticular screw fixation (OR - 0.71, 95% CI -1.31 to -0.10, p = 0.02). Dorsal plating may also be associated with fewer cases of arthritis, although this was not significant (OR 2.46, 95% CI 0.89 to 6.80, p = 0.08). We found no significant differences between the groups in terms of Foot Function Index (FFI), post traumatic arthritis and failure of hardware material. Although our results suggest dorsal bridge plating may provide superior functional outcomes, there is a scarcity of literature with little robustness to make definitive conclusions. High quality randomised trials are required.

In the present work, nanoparticles of copper and silver synthesized via pulsed laser ablation of the respective targets in distilled water are applied to cellulose filter paper to check their effectiveness in the annihilation of bacteria from contaminated water. The treatment of the filter paper with the nanoparticles is found to be an excellent way to get rid of two common bacteria, Staphylococcus aureus and Escherichia coli, from contaminated water. The spread plate method on agar, employed to test the antibacterial efficacy of the nanoparticle-treated papers, clearly shows the absence of bacterial growth upon coming into contact with the nanoparticles in the filter paper. These results were further substantiated by the growth kinetic study of the bacteria that exhibited slow growth of the bacteria that were exposed to the nanoparticles. The morphology of the bacteria that came into contact with the nanoparticles is found to be adversely affected by the nanoparticles. Both copper and silver nanoparticles show a similar extent of antibacterial activity.

PURPOSE: To determine whether the anthocyanin, pelargonidin 3-glucoside (P3G), attenuates symptoms of inflammatory bowel disease (IBD) and metabolic syndrome in rats. METHODS: We tested P3G-enriched strawberry in two models of chronic inflammation in rats, chronic IBD induced by 0.5% dextran sodium sulphate in the drinking water for 12 weeks (D) and metabolic syndrome induced by a high-carbohydrate, high-fat diet (H) for 16 weeks. P3G-enriched strawberry was added to the diet for the final 6 weeks in IBD rats (DP) or 8 weeks in H rats (HP) to provide a dose of 8 mg P3G/kg/day. RESULTS: D rats had marked diarrhoea, bloody stools, erosion of mucosal epithelium, crypt atrophy, loss of villi and goblet cells, and inflammatory cell infiltration. These symptoms were reversed by P3G with healthy stools and mucosal lining of ileum and colon including increased villi, crypts and goblet cells and reduced inflammation. H rats developed hypertension, dyslipidaemia, central obesity, increased ventricular stiffness, cardiac and liver inflammation, and steatosis. P3G treatment in H rats improved systolic blood pressure, ventricular stiffness, and cardiac and liver structure, and reduced abdominal fat, abdominal circumference and body weight gain. CONCLUSIONS: Our study indicates that dietary P3G decreased inflammation to decrease the symptoms of IBD, and to improve cardiovascular, liver and metabolic functions in metabolic syndrome.

Background: In 2007, a field observation from India reported 11% misclassification among 'new' patients registered under the revised national tuberculosis (TB) control programme. Ten years down the line, it is important to know what proportion of newly registered patients has a past history of TB treatment for at least one month (henceforth called 'misclassification'). Methods: A study was conducted among new smear-positive pulmonary TB patients registered between March 2016 and February 2017 in 18 randomly selected districts to determine the effectiveness of an active case-finding strategy in marginalised and vulnerable populations. We included all patients detected through active case-finding. An equal number of randomly selected patients registered through passive case-finding from marginalised and vulnerable populations in the same districts were included. Before enrolment, we enquired about any history of previous TB treatment through interviews. Results: Of 629 patients, we interviewed 521, of whom, 11% (n=56) had past history of TB treatment (public or private) for at least a month: 13% (34/268) among the active case-finding group and 9% (22/253) among the passive case-finding group (p=0.18). No factors were found to be significantly associated with misclassification. Conclusion: Around one in every ten patients registered as 'new' had previous history of TB treatment. Corrective measures need to be implemented, followed by monitoring of any change in the proportion of 'previously treated' patients among all registered patients treated under the programme at national level.
