RESUMO

A meta-analysis was conducted to evaluate the effect of biochar-amended compost (BAC) on plant productivity (PP) and soil quality. The analysis was based on observations from 47 peer-reviewed publications. The results showed that BAC application significantly increased PP by 74.9 %, the total nitrogen content of soil by 37.6 %, and the organic matter content of soil by 98.6 %. Additionally, BAC application significantly decreased the bioavailability of cadmium (-58.3 %), lead (-50.1 %), and zinc (-87.3 %). However, the bioavailability of copper increased by 30.1 %. The study explored the key factors regulating the response of PP to BAC through subgroup analysis. It was found that the increase in the organic matter content of the soil was the key mechanism for PP improvement. The recommended rate of BAC application for improving PP was found to be between 10 and 20 t ha-1. Overall, the findings of this study are significant in providing data support and technical guidance for the application of BAC in agricultural production. However, the high heterogeneity of BAC application conditions, soil properties, and plant types suggests that site-specific factors should be considered when applying BAC to soils.

The ordered structure of partially melted isotactic polypropylene (iPP) was investigated using polarized optical microscopy (POM) and small-/wide-angle X-ray scattering (SAXS/WAXS) measurements. The crystalline morphology was first examined by means of pulling a glass fiber through the iPP melt, which was generated by partially melting a preformed spherulite. The results from the POM experiments indicated that, even at a minimal pulling rate, the surviving ordered structure could also relocate along the direction of fiber pulling and grow into cylindrites eventually. In addition, during the quiescent crystallization from the partially melted sample, which had the same thermal history of fiber-pulling experiments, the obvious memory effect of melt was also observed from the results of X-ray experiments. Moreover, the SAXS profile derived from the partially melted iPP at 170 °C was fitted by the theory of scattering amplitude with the cylindrical form factor. The fit result implied that the surviving ordered structure is of cylindrical nanocrystals with a diameter D ≈ 30 ± 3 nm and height h ≈ 45 ± 3 nm, which can significantly influence the crystallization morphology and kinetics during the subsequent crystallization process.

BACKGROUND: Little is known about whether UVB can directly influence epigenetic regulatory pathways to induce cutaneous squamous cell carcinoma (CSCC). This study aimed to identify epigenetic-regulated signalling pathways through global methylation and gene expression profiling and to elucidate their function in CSCC development. METHODS: Global DNA methylation profiling by reduced representation bisulfite sequencing (RRBS) and genome-wide gene expression analysis by RNA sequencing (RNA-seq) in eight pairs of matched CSCC and adjacent normal skin tissues were used to investigate the potential candidate gene(s). Clinical samples, animal models, cell lines, and UVB irradiation were applied to validate the mechanism and function of the genes of interest. FINDINGS: We identified the downregulation of the TGF-ß/BMP-SMAD-ID4 signalling pathway in CSCC and increased methylation of inhibitor of DNA binding/differentiation 4 (ID4). In normal human and mouse skin tissues and cutaneous cell lines, UVB exposure induced ID4 DNA methylation, upregulated DNMT1 and downregulated ten-eleven translocation (TETs). Similarly, we detected the upregulation of DNMT1 and downregulation of TETs accompanying ID4 DNA methylation in CSCC tissues. Silencing of DNMT1 and overexpression of TET1 and TET2 in A431 and Colo16 cells led to increased ID4 expression. Finally, we showed that overexpression of ID4 reduced cell proliferation, migration, and invasion, and increased apoptosis in CSCC cell lines and reduced tumourigenesis in mouse models. INTERPRETATION: The results indicate that ID4 is downregulated by UVB irradiation via DNA methylation. ID4 acts as a tumour suppressor gene in CSCC development. Understanding community assembly mechanisms is helpful to predict community dynamics. To explore which community assembly mechanism(s) drive(s) the grassland restoration in semi-arid region, we investigated the relationships between plant trait and species relative abundance (SRA), and estimated community functional diversity indices for each community under different treatments (enclosure, grazing and mowing treatment) in a restoration region of Stipa grandis - Leymus chinensis communities in the northern China from 2010 to 2012. There was a high fraction of significant relationships between trait value and SRA, suggesting that niche theory structured the grassland restoration in this region. The functional richness was higher and the functional divergence was lower in the enclosure community than that in the grazing or mowing community, and significantly positive plant height - SRA relationship was found in the enclosure community. These findings demonstrated that limiting similarity based on niche theory was more important in structuring the enclosure community and that environmental filtering based on niche theory played a more important role in driving the grazing or mowing community. Only the factor of year significantly affected the functional evenness (FEve), and the lowest FEve in 2011 implied that the relatively lower precipitation could enhance the effect of limiting similarity on community assembly in the semi-arid grassland.

Herein, olivine LiFePO4 covered with graphene and carbon layers is prepared via a sol-gel method, followed by calcination, and the resultant composite is used as a cathode material in aqueous rechargeable lithium-ion batteries (ARLBs). The phase structure and morphology of the composite are characterized via X-ray diffraction (XRD), transmission electron microscopy (TEM), high-resolution transmission electron microscopy (HRTEM) and specific surface area analysis (BET). The ARLB system is fabricated using LiFePO4/C/graphene as the cathode and a zinc anode in 1 mol L-1 ZnSO4·7H2O and saturated LiNO3 aqueous solution without dissolved oxygen, which delivers a capacity of 153 mA h g-1 at 0.5C rate. Even at a 50C rate, it maintains a capacity of 95 mA h g-1 after 200 cycles. The excellent rate capabilities show that this cathode material exhibits good electrochemical performance and this novel ARLB has great potential in the fields of energy storage and high power sources.

The transformation of prostate cancer from an androgen-dependent state to an androgen-independent state is a lethal progression. Alterations in transcriptional programs are the basis of prostate cancer deterioration. The androgen receptor (AR), a member of the nuclear hormone receptor superfamily, mediates prostate cancer progression by functioning primarily through the ligand-activated transcription of target genes. Therefore, a detailed map of AR-regulated genes and AR genomic binding sites is required for hormone-naive and castration-resistant prostate cancer. Through the use of chromatin immunoprecipitation in combination with direct sequencing, 4,143 AR binding sites were defined in the LNCaP androgen-sensitive prostate cancer cell line. Using the same method, 2,380 AR binding regions were identified in the LNCaP-AI long-term androgen-deprived cell line. Approximately 8.5% (354/4,143) of the binding regions were mapped to within 2 kb of the transcription start site (TSS) in the LNCaP cells, while ∼12.6% (299/2,380) were mapped to within 2 kb of the TSS in the LNCaP-AI cells. In total, the study mapped 2,796 genes in LNCaP cells and 1,854 genes in LNCaP-AI cells. The cell lines shared 789 mutual genes. In addition, gene ontology (GO) analysis of the genes revealed that there was a notable overlap between the GO terms in the LNCaP cells and LNCaP-AI cells. However, GO terms within the biological process domain that were only observed in the LNCaP-AI cells included the reproduction process, death, immune system process, multi-organism process, pigmentation and viral reproduction. The major genes in the different GO terms were TNFAIP8, RTN4, APP and SYNE1. Through analyzing the AR binding sites in the two cell types, the present study aimed to map potential AR-regulated genes, identify their associated transcription factors and provide a new perspective on the biological processes in the development of prostate cancer. The results provided a valuable data set that furthered the understanding of the genome-wide analysis of AR binding sites in prostate cancer cells, which may be exploited for the development of novel prostate cancer therapeutic strategies.

BACKGROUND: Age-related physiological, biochemical and functional changes in mammalian skeletal muscle have been shown to begin at the mid-point of the lifespan. However, the underlying changes in DNA methylation that occur during this turning point of the muscle aging process have not been clarified. To explore age-related genomic methylation changes in skeletal muscle, we employed young (0.5 years old) and middle-aged (7 years old) pigs as models to survey genome-wide DNA methylation in the longissimus dorsi muscle using a methylated DNA immunoprecipitation sequencing approach. RESULTS: We observed a tendency toward a global loss of DNA methylation in the gene-body region of the skeletal muscle of the middle-aged pigs compared with the young group. We determined the genome-wide gene expression pattern in the longissimus dorsi muscle using microarray analysis and performed a correlation analysis using DMR (differentially methylated region)-mRNA pairs, and we found a significant negative correlation between the changes in methylation levels within gene bodies and gene expression. Furthermore, we identified numerous genes that show age-related methylation changes that are potentially involved in the aging process. The methylation status of these genes was confirmed using bisulfite sequencing PCR. The genes that exhibited a hypomethylated gene body in middle-aged pigs were over-represented in various proteolysis and protein catabolic processes, suggesting an important role for these genes in age-related muscle atrophy. In addition, genes associated with tumorigenesis exhibited aged-related differences in methylation and expression levels, suggesting an increased risk of disease associated with increased age. CONCLUSIONS: This study provides a comprehensive analysis of genome-wide DNA methylation patterns in aging pig skeletal muscle. Our findings will serve as a valuable resource in aging studies, promoting the pig as a model organism for human aging research and accelerating the development of comparative animal models in aging research.
