Microplastic pollution in aquatic environments has been a ubiquitous concern in recent years, owing to their rapid production combined with poor waste management practices. However, information on freshwater microplastics in the Global South is still scarce, despite growing research on freshwater microplastics in recent years, particularly within the Global North. To help address this knowledge gap, we studied water and sediment microplastic dynamics along a subtropical river system, i.e. Crocodile River around the Nelspruit City area (South Africa), across three different seasons (i.e. cool-dry, hot-dry, hot-wet) using a combination of diversity indices and multivariate analyses. Microplastics were more abundant during the cool-dry season in the surface water samples (mean 1058 particles m-3) and high during the hot-dry season (mean 568 particles kg-1 dwt) in the sediment samples. The hot-wet season had a low particle density in both surface water (mean 625 particles m-3) and sediments (mean 86 particles kg-1 dwt) samples. Microplastic shapes were dominated by fibres and fragments, with the colour scheme dominated by transparent, blue, and black. The abundance of microplastics was positively correlated with pH and resistivity, and negatively with river flow. Wastewater was attributed as a primary source of microplastics, particularly because of the observed dominant fibre microplastics, usually released during laundry. Our results suggest that Crocodile River and its tributaries are temporary sinks of microplastics during periods of low rainfall. Implications of this pollution are far-reaching, including effects on residents who are dependent on the Crocodile River as a source of drinking water and aquatic biota which may be exposed to these pollutants.

Pan-wetland systems are one of the world's essential and productive ecosystems and are considered important, unique and complex ecosystems. Anthropogenic activities around the temporary pans in the Khakhea Bray Transboundary Aquifer region are increasingly becoming a big issue of concern as this may affect pan biodiversity. The study specifically aimed to investigate spatial and temporal distributions of metal and nutrient concentrations within the pans in relation to land use, identify potential pollution sources in this water-scarce region, and assess macroinvertebrate diversity and distribution in relation to pan limnochemistry using a combination of multivariate analyses from 10 pans across three seasons. Environmental and anthropogenic variables influence water quality and the distribution of metals concentration in Khakhea-Bray pan systems. Anthropogenic activities such as animal grazing, infrastructure degradation, water withdrawal and littering have resulted in poor water quality within temporary pans, which may strongly influence macroinvertebrate diversity and distribution. Forty-one macroinvertebrate species from 5 insect orders (i.e., Coleoptera, Hemiptera, Odonata, Ephemeroptera, Diptera), Crustacea and Mollusca were identified. Significant differences were observed across the seasons for macroinvertebrate taxa, with high and low species richness being observed in autumn and winter, respectively. Water (i.e., temperature, dissolved oxygen, pH, salinity, conductivity), physical (i.e., stone composition) and sediment (i.e., sulphur, sodium) parameters were found to have a significant impact on the macroinvertebrate communities. Therefore, understanding the relationships between macroinvertebrates and their environment is crucial in understanding how the ecosystem taxa are structured and is vital for informing conservation managers on how to properly manage and protect these systems from further degradation.

Feline immunodeficiency virus (FIV) is a pathogenic lentivirus related to human and simian immunodeficiency viruses that has been associated with AIDS-like pathologies in domestic and wild cats, as well as in hyenas. Despite known pathologies, progressive immunosuppression and ill health effects driven by these lentiviruses in association with other secondary infections remain understudied in free-ranging species. Here, the role of coinfections by gastrointestinal parasites and tick-borne hemoparasites for FIV disease progression was explored in 195 free-ranging African lions (Panthera leo) living in Kruger National Park (KNP), South Africa. Using statistical methodology, we evaluated the effects of FIV on a range of health indicators to explore how direct and indirect effects of FIV and associated coinfections align to determine lion health outcomes. Findings show direct negative effects of FIV on host immunity and nutritional status, and exacerbation of aggressive behaviors, conditions which may increase exposure/susceptibility to other secondary infections. When taken together, the contribution of coinfecting parasites to morbidity in lions is of similar magnitude as direct effects of FIV infection alone, suggesting that the particular coinfection assemblage may play a role in mediating disease progression within natural lion populations.

The International Species Information System has set forth an extensive database of reference intervals for zoologic species, allowing veterinarians and game park officials to distinguish normal health parameters from underlying disease processes in captive wildlife. However, several recent studies comparing reference values from captive and free-ranging animals have found significant variation between populations, necessitating the development of separate reference intervals in free-ranging wildlife to aid in the interpretation of health data. Thus, this study characterizes reference intervals for six biochemical analytes, eleven hematologic or immune parameters, and three hormones using samples from 219 free-ranging African lions ( Panthera leo ) captured in Kruger National Park, South Africa. Using the original sample population, exclusion criteria based on physical examination were applied to yield a final reference population of 52 clinically normal lions. Reference intervals were then generated via 90% confidence intervals on log-transformed data using parametric bootstrapping techniques. In addition to the generation of reference intervals, linear mixed-effect models and generalized linear mixed-effect models were used to model associations of each focal parameter with the following independent variables: age, sex, and body condition score. Age and sex were statistically significant drivers for changes in hepatic enzymes, renal values, hematologic parameters, and leptin, a hormone related to body fat stores. Body condition was positively correlated with changes in monocyte counts. Given the large variation in reference values taken from captive versus free-ranging lions, it is our hope that this study will serve as a baseline for future clinical evaluations and biomedical research targeting free-ranging African lions.

OBJECTIVE: To evaluate the effects of the combination butorphanol, medetomidine and midazolam (BMM) and its reversibility in lions. STUDY DESIGN: Prospective clinical trial. ANIMALS: Thirty free-ranging lions, 10 male and 20 female, weighing 81-210 kg. METHODS: Lions were immobilised with butorphanol mean 0.31 ± SD 0.034 mg kg(-1), medetomidine 0.052 ± 0.006 mg kg(-1), midazolam 0.21 ± 0.024 mg kg(-1) and hyaluronidase 1250 IU administered intramuscularly with a dart gun. Upon recumbency, physiological parameters and anaesthetic depth were monitored 10-15 minutes after darting (T1) and repeated every 10 minutes for a further 30 minutes (T2, T3, T4). Arterial blood gas analyses were performed at T1 and T4. At the end of the procedure, 45-60 minutes after initial darting, immobilisation was reversed with naltrexone 0.68 ± 0.082 mg kg(-1), atipamezole 0.26 ± 0.031 mg kg(-1), and flumazenil 0.0032 ± 0.0007 mg kg(-1) administered intravenously and subcutaneously. RESULTS: The BMM combination rapidly induced immobilisation and lateral recumbency was reached within 7.25 ± 2.3 minutes. Median induction score [scored 1 (excellent) to 4 (poor)] was 1.4 (range 1-2). Cardio-respiratory parameters were stable. Heart rate varied from 32 to 72 beats per minute, respiratory rate from 14 to 32 breaths minute(-1) and rectal temperature from 36.6 to 40.3 °C. No sudden arousals were observed. Arterial blood gas analyses revealed a mean pH of 7.33, PaCO(2) of 33 mmHg and PaO(2) of 87 mmHg. Mild to moderate hypoxemia was seen in four lions. Recovery was smooth and lions were walking within 4.4 ± 4.25 minutes. Median recovery score [scored 1 (excellent) to 4 (poor)] was 1.3 (range 1-2). CONCLUSION AND CLINICAL RELEVANCE: The drug combination proved to be effective in immobilising free-ranging healthy lions of both sexes with minimal cardio-respiratory changes.
