RESUMO

We compared the content of adenylyl cyclase-associated protein 1 (CAP1) in the blood and tissues of patients with head and neck squamous cell carcinomas (with and without regional metastases), patients with chronic inflammatory diseases aggravated by laryngeal and laryngopharyngeal dysplasia, and healthy individuals. The data suggest that serum CAP1 concentration correlated with the depth of primary tumor invasion and the presence of regional metastases. In cancer patients, the serum level of CAP1 was lower than in patients with laryngeal and laryngopharyngeal dysplasia, which can be of importance for differential and timely diagnostics of malignant tumors.

RESUMO

The article is devoted to conservative and reconstructive-plastic surgical interventions using biocompatible implants accompanied by various types of radiotherapy and modern anticancer drugs that have allowed improving basic survival with maintaining a high level of quality of life. Our method of combined treatment for larynx, hypopharynx, oral cavity and oropharynx cancer of stage II-III by chemoradotherapy shows high immediate effectiveness and allows achieving statistically significant increase of overall and disease-free survival in the remote period of observation. The use of NiTi implants provides reliable recovery of deleted structures, pressurization of the cranial cavity, reduces the duration of surgery, shortens the time of wound healing.

A comparative study of four series of pyrrhotite-type chalcogenide compounds Fe(7-y)M(y)X(8) (X = S, Se) with substitution of Ti or Co for iron has been performed by means of x-ray and neutron powder diffraction, and by magnetization measurements. In Fe(7-y)M(y)X(8) compounds having a ferrimagnetic order at y = 0, the substitution of either Ti or Co for iron is observed to result in a monotonous decrease of the magnetic ordering temperature, while the resultant magnetization shows a non-monotonous behavior with a minimum around y = 1.0-1.5 in all the Fe(7-y)M(y)X(8) families except Fe(7-y)Co(y)Se(8). Suppression of a magnetically ordered state with substitutions in Fe(7-y)M(y)X(8) is ascribed to nearly zero values of Ti and Co magnetic moments, while the non-monotonous changes of the resultant magnetization are explained by the compensation of the sublattice magnetizations due to the non-random substitutions in alternating metallic layers. The difference in the cation partitioning observed in Fe(7-y)Ti(y)X(8) and Fe(7-y)Co(y)X(8) is attributed to the difference in the spatial extension of Ti and Co 3d orbitals. High coercive field values (20-24 kOe) observed at low temperatures in the Ti-containing compounds Fe(7-y)Ti(y)X(8) with y ⩾ 3 are suggested to result from the enhancement of Fe orbital moment due to the Ti for Fe substitution.

RESUMO

There was studied an expression of transcription factors NF-kappaB p65, NF-kappaB p50 and HIF-1 in the tissue of squamous cell carcinomas of the head and neck. An association of development of regional metastasis of the disease with increased expression of the transcription factor NF-kappaB p50 was revealed. After treatment there were found relapses within 3 to 47 months in 10 patients, regional lymph node metastases in 3 patients, 12 patients died. Metastasis-free survival of patients with squamous cell carcinomas of the head and neck depended on the threshold level of an expression of the transcription factors NF-kappaB p65, p50, HIF-1 equal to 9.6, 7.8 and 9 CU/mg protein respectively.

RESUMO

The development of malignant neoplasm of the head and neck is related to the state of intracellular proteasome system. Elevation of total activity of proteasomes and specific activity of 20S proteasomal pool are accompanied by changes in proteasomal composition. Tumor size correlated with the content of regulatory proteasomal complex PA28. In the presence of regional metastases, 26S proteasome activity decreases and the content of proteasome immune subunit LMP7 in the tumor increases.

The paper compares the levels of TIMP-1 and TIMP-2 in blood serum from patients with head and neck squamous cell carcinoma (HNSCC) to establish correlations with clinico-morphological parameters of the disease and prognosis. Our study involved 140 patients with HNSCC (T1-3N0-3M0) and 38 healthy donors. The level of serum TIMP-1 in patients was significantly higher (p < or = 0.02) while that of TIMP-2 was in inverse correlation with histological grade of tumor (p < or = 0.04) and age (p < or = 0.05). High level of TIMP-1 appeared an unfavorable prognosticator for 2-year relapse-free survival in HNSCC patients (p < or = 0.05). Those of both TIMP-1 and TIMP-2 identified in blood serum prior to therapy involved high risk of metastasis development (p < or = 0.05) and (p < or = 0.05), respectively.

The levels of metalloproteinases (MMP-2,-9), their tissue inhibitors (TIMP-1,-2) and extracellular matrix metalloproteinase inducer (EMMPRIN) were studied in tumor tissue and blood serum from patients with head and neck squamous sell carcinoma. Immunohistochemical investigation showed much higher expression of MMP-9 and TIMP-1 in tumor tissue than MMP-2 and TIMP-2. Different expression of the studied parameters (except TIMP-1) in cancer cells and stroma was observed. Enhanced level of MMP-2,-9 and TIMP-2 expression was revealed mainly in fibrocytes, leukocytes and stromal extracellular matrix. Expression of EMMPRIN in tumor cells was higher than in stromal cells. It is possible that carcinoma cells express EMMPRIN to increase MMP production by surrounding cells. Significant decrease of inhibitor expression was observed in cells of carcinomas with N1 grade of metastasis than in tumors without metastasis. Also the decrease in TIMP-1 level was determined in blood serum of patients with metastasis to regional lymph nodes compared with serum of patients without metastasis. Thus, MMP-9 and TIMP-1 play an important role in the development of head and neck squamous sell carcinoma and TIMP-1 level in blood serum and cancer tissues is linked to first grade of regional lymph node metastasis.

Levels of matrix metalloproteinase (MMP-2) and endogenous inhibitors (TIMP-1,-2) in tumor and blood serum from patients with squamous-cell laryngeal carcinoma were studied. Immunohistochemical investigation showed much higher MMP-9 expression (76.9%) and TIMP-1 (100%) in tumor tussue than MMP-2 (23.8%) and TIMP-2 (33.3%). Therefore, MMP-9 and TIMP-1 levels in blood serum alone were assayed. TIMP-1 levels in serum from patients were significantly higher than those in healthy subjects (p=0.023). High TIMP-1 levels in blood serum were recorded in patients with stage III laryngeal carcinoma and differed significantly from control (p=0.009). However, no correlation was found between them and MMP concentration in blood serum, on the one hand, and tumor size, on the other. High TIMP-1 levels in blood serum were recorded in patients free from metastasis to regional lymph nodes (124.5+39.5 ng/ml); however, they were lower than in those with such metastases (102.7+10.9 ng/ml) and significantly lower than in healthy subjects (p=0.023). There was a correlation between MMP-9 expression in tumor tissue, on the one hand, and involvement of regional lymph nodes, on the other, (p=0.054). In blood serum from patients showing tumor regression by 75-100% after radiotherapy, TIMP-1 levels were higher than in those with relatively lower response to the same treatment (p=0.072). Hence, assay of TIMP-1 levels in blood serum and MMP-9 concentration in tumor tissue may be used for assessment of tumor processes alongside standard procedures. Moreover, the former may be instrumental in predicting laryngeal response to radiotherapy.
