RESUMO

The allergenic action of influenza vaccines was evaluated by the determination of the capacity of sera, taken before and after immunization, for inducing the degranulation of rat mast cells in the presence of the vaccine preparation. In this test sera from three groups of volunteers immunized with experimental and commercial batches of influenza inactivated vaccines, purified and concentrated by gradient centrifugation, was studied. The absence of immediate-type allergization after a single administration of the vaccines under study was shown.

RESUMO

Experimental batches of adsorbed subunit influenza vaccine were prepared from the envelope of glycoprotein antigens separated from the influenza virion by treatment with a cationic detergent (cetyltrimethylammonium bromide). Purified and concentrated influenza virus strains A(H1N1) and A(H3N2) were obtained by gradient centrifugation; additional purification, with a view to removing ovalbumin and structural components of the chorioallantoic membrane, was achieved by gelfiltration. The composition and biological properties of the vaccine are described.

RESUMO

The materials substantiating the possibility of using the method for the determination of the lethal effect of endotoxin on dactinomycin-treated mice are presented. This determination is made with a view to detecting the admixtures of endotoxins in whole-virion and subvirion inactivated influenza vaccines at different stages of their manufacture, as well as in the final product. The proposed test is highly sensitive, rather simple in its practical realization and can be used for evaluating the degree of the purification of influenza vaccines from endotoxins.

RESUMO

The capacity of S. paratyphi B endotoxin and its high molecular (HMF) and low molecular (LMF) fractions to stimulate the resistance of mice to shigella infection in intraperitoneal and intranasal challenge was investigated. The HMF developed high activity after the intraperitoneal injection (efficiency index--EI = 2.8). All the preparations were less active on the lung model (EI = 1.6--1.7). Materials of this work lead to the conclusion that in studying the phenomenon of nonspecific resistance induction conditions providing antitoxic and antiinfectious resistance should be considered separately.

RESUMO

Chemical (detergent) antigens administered intraintestinally or intravenously to rabbits were applied to reproduce postvaccinal immunity to experimental shigellosis. There was noted an increase of the antibody level in the serum, an intensification of its preventive proerties, and the absence of dilatation of the isolated loops of the small intestine after the administration of the corresponding dysentery culture. Oral immunization of rabbits with the detergent antigen sorbed on ionexchange resin also led to immunity.

The authors evaluated the immunological results of enteral and subcutaneous injection of detergent Sonne dysentery vaccine to volunteers (changes in homo- and heterologous antibodies in the blood and saliva recorded by means of common indirect hemagglutination test and that modified according to Coombs' test, and IgG, IgM, and IgA concentration recording. Both immunization methods induced specific antibody formation: enteral vaccination caused chiefly the accumulation of secretory antibodies of IgA class, and subcutaneous -- of serum antibodies belonging to IgG class. The supposed mechanisms determining the capacity of enteral administration of the antigen to provoke an immune response different from that caused by its subcutaneous injection are discussed.

RESUMO

The authors elaborated a method of isolation under industrial conditions of Shigella sonnei surface antigens by the treatment of the culture with a detergent (sodium laurylsulfate). The antigens obtained possessed a much lesser toxicity in comparison with the tryptic ones. A method of preparation of a detergent Sonne monovaccine with an increased antigen content (1.3 mg/ml) was described. Experimental series of the preparation were tested on 38 volunteers. A lower reactogenicity of the detergent vaccine (in comparison with the tryptic) and its marked immunological efficacy was demonstrated.
