Wild fruits play a vital role in the diets and economic well-being of tribal communities of Uttarakhand, India. Despite their significance, there is limited information on some of the wild Ficus species such as Ficus drupacea, Ficus rumphii, Ficus semicordata, Ficus subincisa and Ficus hispida. This research aimed to analyse the nutritional potential, anti-nutritional compositions, mineral composition, antioxidant and finally the phytochemical properties of Ficus fruits to ascertain their medicinal and nutritional significance. The proximate analysis results of all the five wild Ficus species displayed varying percentages of protein, moisture, fat, ash, crude fibre, carbohydrates. Ficus species exhibited superior nutritional and mineral parameters along with a significant amount of vitamin C. Additionally, these fruits significantly showed lower levels of anti-nutritional and good amount of antioxidant parameters. Thus, adequate consumption of these wild Ficus fruits could potentially contribute to human nutritional needs and may serve as sources for dietary enhancements.

Ficus drupacea is a medicinal tree found in temperate regions. Various parts of this plant had been used traditionally for the treatment of various ailments such as root powder applied externally for skin infections. Analysis was carried out on the bioactive lipids extracted from Ficus drupacea fruit using both petroleum-based solvent (Hexane) and an environmentally friendly solvent Dimethyl carbonate (DMC). The results showed that DMC extraction yielded a high oil content in Ficus drupacea fruit (6.51 %). When examining the fatty acid composition using GC-FID analysis, Ficus drupacea oil extracted with DMC contained significant proportions of essential fatty acids such as linoleic acid (32.317 %), oleic acid (20.946 %), palmitic acid (25.841 %), etc. Additionally, DMC extraction resulted in higher levels of total phenolics in Ficus drupacea fruit oil compared to hexane. Moreover, DMC extracted oil exhibited stronger antioxidant properties, such as radical scavenging, anti- arthritic, photoprotective activity while displayed similar anti-inflammatory and anti-microbial activity as hexane-extracted oil. In summary, these findings demonstrate that DMC is an efficient and safer alternative to conventional solvent hexane for extracting oils from Ficus drupacea fruit. It is rich in bioactive compounds essential for human nutrition, including polyunsaturated fatty acids, flavonoids, and phenolic compounds, with enhanced biological activities.

Water is crucial for life. Being the world's third-largest industry, the textile industry pollutes 93 billion cubic meters of water each year. Only 28% of textile wastewater is treated by lower- to middle-income countries due to the costly treatment methods. The present work demonstrates the utilization of surface oxygen defects and nanopores in Mg0.8Li0.2Fe2O4 (Li-MgF) to treat textile effluents by a highly economical, scalable, and eco-friendly process. Nanoporous, oxygen-deficient Li-MgF splits water by a nonphotocatalytic process at room temperature to produce green electricity as hydroelectric cell. The adsorbent Li-MgF can be easily regenerated by heat treatment. A 70-90% reduction in the UV absorption intensity of adsorbent-treated textile effluents was observed by UV-visible spectroscopy. The oxygen defects on Li-MgF surface and nanopores were confirmed by X-ray photoelectron spectroscopy and Brunauer-Emmett-Teller (BET) measurements, respectively. To analyze the adsorption mechanism, three known organic water-soluble dyes, brilliant green, crystal violet, and congo red, were treated with nanoporous Li-MgF. The dye decolorization efficiency of Li-MgF was recorded to be 99.84, 99.27, and 99.31% at 250 µM concentrations of brilliant green, congo red, and crystal violet, respectively. The results of Fourier transform infrared (FTIR) spectroscopy confirmed the presence of dyes on the material surface attached through hydroxyl groups generated by water splitting on the surface of the material. Total organic carbon analysis confirmed the removal of organic carbon from the dye solutions by 82.8, 77.0, and 46.5% for brilliant green, Congo red, and crystal violet, respectively. Based on the kinetic and isotherm models, the presence of a large number of surface hydroxyl groups on the surface of the material and OH- ions in solutions generated by water splitting was found to be responsible for the complete decolorization of all of the dyes. Adsorption of chemically diverse dyes by the nanoporous, eco-friendly, ferromagnetic, economic, and reusable Li-MgF provides a sustainable and easy way to treat textile industry effluents in large amounts.

Plants are the prime source of phytoconstituents that can act as potent agents for the prevention and treatment of various diseases. Heterospathe elata is a plant belonging to the Arecaceae family having numerous medicinal properties. The present study was undertaken to prepare crude extracts of Heterospathe elata leaves with solvents of different polarity dimethyl carbonate (DMC), isopropyl alcohol (IPA), hydro alcohol (HYA) and water (WTR) by using successive Soxhlet extraction method. Further, the antioxidant, antidiabetic, and anti-inflammatory activities were assessed by the spectrophotometric method and possible bioactive phytoconstituents from the hydro alcohol extract of Heterospathe elata leaves using GC/MS. In our study, it was found that the GC/MS analysis revealed the presence of nineteen bioactive phytoconstituents. The highest antioxidant activity was found in the water extract. In antidiabetic and anti-inflammatory activity highest potential was shown by hydro alcohol extract and the lowest was in the dimethyl carbonate extract. These findings support the Heterospathe elata leaves showed the high biological potential attributed to a high amount of bioactive phytoconstituents and could be utilized as value-added functional food and medicine.

OBJECTIVES: Mechanical ocular trauma can result in wide spectrum of injuries with varying levels of severity. A complete ophthalmic examination is important to achieve a thorough evaluation and provide optimal management. The ocular adnexa play a significant role in the protection of the globe via various mechanisms. Open and closed globe injuries directly affect the visual outcome. Although adnexal injuries may not affect the vision directly, they can lead to severe ocular morbidity in terms of a cosmetic blemish, watering, a long healing period, severe complications, or permanent disfigurement. Currently, there is no appropriate classification of adnexal injuries. This study examined the causes, and effects of adnexal injuries to prepare a new classification system for adnexal injuries. METHODS: A retrospective study of ocular injuries was conducted at a tertiary care center in Gwalior, India. The data of 600 mechanical ocular trauma patients were analyzed and the demographic details, mode of injury, type and sub-type of ocular injury were recorded. The injuries were divided into open and closed globe injuries using the Birmingham Eye Trauma Terminology classification system. Adnexal injuries were categorized by type and sub-type using a proposed classification system. RESULTS: Among the 600 cases, there were 34 closed globe injuries, 26 open globe injuries, and 540 adnexal injuries. Only 10% of the cases could be classified using the conventional classification system. CONCLUSION: The existing classification system is not optimally comprehensive; the majority of the cases could not be categorized. A more detailed framework of classification of adnexal injuries will facilitate better assessment and management.

With the rising incidences of cancer cases, the quest for new metal based anticancer drugs has led to extensive research in cancer biology. Zinc complexes of amino acid residue side chains are well recognized for hydrolysis of phosphodiester bond in DNA at faster rate. In the presented work, a Zn(II) complex of cyclen substituted with two l-tryptophan units, Zn(II)-Cyclen-(Trp)2 has been synthesized and evaluated for antiproliferative activity. Zn(II)-Cyclen-(Trp)2 was synthesized in ∼70% yield and its DNA binding potential was evaluated through QM/MM study which suggested good binding (G=-9.426) with B-DNA. The decrease in intensity of the positive and negative bands of CT-DNA at 278nm and 240nm, respectively demonstrated an effective unwinding of the DNA helix with loss of helicity. The complex was identified as an antiproliferative agent against U-87MG cells with 5 fold increase in apoptosis with respect to control (2h post incubation, IC50 25µM). Electrophoresis and comet assay studies exhibited an increase in DNA breakage after treatment with complex while caspase-3/ß-actin cleavage established a caspase-3 dependent apoptosis pathway in U-87 MG cells after triggering DNA damage. In vivo tumor specificity of the developed ligand was validated after radiocomplexation with 99mTc (>98% radiochemical yield and specific activity of 2.56GBq/µmol). Avid tumor/muscle ratio of >6 was depicted in biodistribution and SPECT imaging studies in U-87 MG xenograft model nude mice.

In the present study, the antioxidant activity of successive leaf extracts of Dracaena reflexa was investigated using the scavenging activity on 1,1-diphenyl-2-picrylhydrazyl and reducing power by ferric reducing antioxidant power assay. Methanol extract was found potent in both the assays. IC50 values of 1,1-diphenyl-2-picrylhydrazyl assay for methanol extract was 0.97 mg/ml and ferric reducing antioxidant power value for the same is 1.19. Phytochemical screening, proximate analysis and total phenolic content were also determined. Qualitative screening for phytochemical showed the presence of alkaloids, flavonoids, terpenoids, glycosides and saponins. Highest phenolic content was shown by methanol extract (49.69 mg gallic acid equivalent/g dry weight). Proximate analysis showed moisture content (3.31%), ash content (8.02%), crude fibre (1.31%), crude fat (0.97%), total protein (3.70%), total carbohydrate (86.01) and nutritive value (367.56 kcal/100 g), which would make it a potential nutraceutical. This study suggested that Dracaena reflexa, a potential natural free radical scavenger, which could find use as an antioxidative.

The ability of the human fungal pathogen Candida albicans to reversibly switch between different morphological forms and establish biofilms is crucial for establishing infection. Targeting phenotypic plasticity and biofilm formation in C. albicans represents a new concept for antifungal drug discovery. The present study evaluated the influence of cyclic lipopeptide biosurfactant produced by Bacillus amyloliquefaciens strain AR2 on C. albicans biofilms. The biosurfactant was characterized as a mixture of iturin and fengycin by MALDI-TOF and amino acid analysis. The biosurfactant exhibited concentration dependent growth inhibition and fungicidal activity. The biosurfactant at sub-minimum growth inhibition concentration decreased cell surface hydrophobicity, hindered germ tube formation and reduced the mRNA expression of hyphae-specific gene HWP1 and ALS3 without exhibiting significant growth inhibition. The biosurfactants inhibited biofilm formation in the range of 46-100 % depending upon the concentration and Candida strains. The biosurfactant treatment dislodged 25-100 % of preformed biofilm from polystyrene plates. The biosurfactant retained its antifungal and antibiofilm activity even after exposure to extreme temperature. By virtue of the ability to inhibit germ tube and biofilm formation, two important traits of C. albicans involved in establishing infection, lipopeptides from strain AR2 may represent a potential candidate for developing heat stable anti-Candida drugs.

An anthraquinone conjugated macrocyclic chelating agent, 2,2',2″-(10-(2-(9,10-dioxo-9,10-dihydroanthracen-1-ylamino)-2-oxoethyl)-1,4,7,10-tetraazacyclododecane-1,4,7-triyl)triacetic acid or DO3A-Act-AQ, was synthesized by reacting trisubstituted cyclen (DO3A) with 2-chloro-N-(9,10-dioxo-9,10-dihydro-anthracen-1-yl)-acetamide and radiolabeled with (68)GaCl3 in 84% efficiency and a specific activity of 4.62 MBq/nmol. The IC50 value for BMG-1 cells was 0.1 mM, while the same concentration of DO3A-Act-AQ rendered no significant toxicity in HEK cells. The exposure of BMG-1 cells with 0.1 mM DO3A-Act-AQ displayed a time-dependent increase in apoptosis (40.7% at 4 h and 53% at 24 h), and the effect was 2.8- and 3.6-fold % higher as seen in HEK cells. An increase in S-phase cell population suggested S-phase arrest concomitant with induction of apoptosis in BMG-1 cells reaching to 4.5 times after 24 h with respect to control cells. DNA binding studies on CT-DNA (calf thymus) revealed a quenching pattern in the presence of DO3A-Act-AQ (10-70 µM), and the Stern-Volmer quenching constant was 2.4157 × 10(6) L mol(-1), indicative of strong binding with ds-DNA. A decrease in the positive and negative bands of CT-DNA was seen at 278 nm and 240 nm, respectively, on addition of 0.05 mM of DO3A-Act-AQ in CD studies. (68)Ga-DO3A-Act-AQ was stable in vitro in both PBS and human serum for at least 2 h. The in vivo blood kinetics study performed on normal rabbits indicated fast clearance with t1/2(F) = 40 ± 0.3 min and t1/2(S) = 3 h 30 min ± 0.1 min. Ex vivo biodistribution analysis displayed a favorable tumor-to-muscle ratio of 8.4 after 2 h in athymic nude mice xenografted with BMG-1 cells, suggesting the specificity of (68)Ga-DO3A-Act-AQ toward tumors.
