RESUMO

Inspired by the pharmacological interest generated by 6-substituted purine roscovitine for cancer treatment, 5-aminoimidazole-4-carboxamidine precursors containing a cyanamide unit were prepared by condensation of 5-amino-N-cyanoimidazole-4-carbimidoyl cyanides with a wide range of primary amines. When these amidine precursors were combined with acids, a fast cascade cyclization occurred at room temperature, affording new 6,8-diaminopurines with the N-3 and N-6 substituents changed relatively to the original positions they occupied in the amidine and imidazole moieties of precursors. The efficacy and wide scope of this method was well demonstrated by an easy and affordable synthesis of 22 6,8-diaminopurines decorated with a wide diversity of substituents at the N-3 and N-6 positions of the purine ring. Preliminary in silico and in vitro assessments of these 22 compounds were carried out and the results showed that 13 of these tested compounds not only exhibited IC50 values between 1.4 and 7.5 µM against the colorectal cancer cell line HCT116 but also showed better binding energies than known inhibitors in docking studies with different cancer-related target proteins. In addition, good harmonization observed between in silico and in vitro results strengthens and validates this preliminary evaluation, suggesting that these novel entities are good candidates for further studies as new anticancer agents.

The objective is to verify the effects of pesticides on the health of farmers in the southern region of Brazil, as well as the possible symptoms and cases of intoxication. This study has a quantitative, descriptive and exploratory approach, carried out in 12 agricultural locations in the municipality of Serra Catarinense. This research was approved by the research ethics committee and was applied through a structured questionnaire to 79 farmers who grow grains in the conventional cultivation system. The pesticide most used by farmers was glyphosate, followed by acephate. Of the individuals, 21.5% reported that they had suffered poisoning by pesticides by the respiratory route (58.8%). In addition, 28% had three symptoms of intoxication, demonstrating possible probable cases of intoxication. And 41% of workers reported headaches as a predominant symptom in applications with the use of pesticides, followed by dizziness/vertigo (16%). Farmers who use PPE during the preparation of the pesticide spray are neither chronic diseases (p<0.003) nor psychological problems (p<0.000). All four individuals who had cancer, all also eat while applying pesticides (p<0.049). The exposure to pesticides causes changes in the organism of those exposed, using them more vulnerable to health problems.

Autonomous control of gene expression through engineered quorum-sensing processes is broadly applicable to biosynthetic pathways, including simultaneous control of different genes. It is also a powerful tool for balancing growth and production. We had previously engineered a modular autoinduction device for the control of gene expression in B. subtilis. Now, we expand its functionality to repress gene expression autonomously. The engineered R8 promoter responds to AHL accumulation in the culture medium. In a riboflavin-producing strain, the AHL-Lux complex exerts 5-fold repression on the R8-driven expression of the flavokinase/FAD synthetase gene ribC, resulting in a higher titer of the vitamin. We engineered a strain able to autonomously induce and repress different genes simultaneously, demonstrating the potential of the device for use in metabolic engineering.

Pathogenic spirochetes from genus Leptospira are etiologic agents of leptospirosis. Cellular vaccines against Leptospira infection often elicit mainly response against the LPS antigen of the serovars present in the formulation. There is no suitable protein candidate capable of replacing whole-cell vaccines, thus requiring new approaches on vaccine development to improve leptospirosis prevention. Our goal was to develop a whole-cell vaccine sorovar-independent based on LPS removal and conservation of protein antigens exposure, to evaluate the protective capacity of monovalent or bivalent vaccines against homologous and heterologous virulent Leptospira in hamster. Leptospire were subjected to heat inactivation, or to LPS extraction with butanol and in some cases further inactivation with formaldehyde. Hamsters were immunized and challenged with homologous or heterologous virulent serovars, blood and organs were collected from the survivors for bacterial quantification, chemokine evaluation, and analysis of sera antibody reactivity and cross-reactivity by Western blot. Immunization with either heated or low LPS vaccines with serovar Copenhageni or Canicola resulted in 100% protection of the animals challenged with homologous virulent bacteria. Notably, different from the whole-cell vaccine, the low LPS vaccines produced with serovar Canicola provided only partial protection in heterologous challenge with the virulent Copenhageni serovar. Immunization with bivalent formulation results in 100% protection of immunized animals challenged with virulent serovar Canicola. All vaccines produced were able to eliminate bacteria from the kidney of challenged animals. All the vaccines raised antibodies capable to recognize antigens of serovars not present in the vaccine formulation. Transcripts of IFNγ, CXCL16, CCL5, CXCL10, CXCR6, and CCR5, increased in all immunized animals. Conclusion: Our results showed that bivalent vaccines with reduced LPS may be an interesting strategy for protection against heterologous virulent serovars. Besides the desirable multivalent protection, the low LPS vaccines are specially promising due to the expected lower reatogenicity.

Este trabalho foi desenvolvido com o objetivo de avaliar a resistência aos ectoparasitas em bovinos jovens da raça Crioula Lageana e meio-sangue Angus em condições naturais. Foram utilizados 10 machos castrados em cada grupo, recém desmamados (6-8 meses) e mantidos sob as mesmas condições de manejo, em pastagens cultivadas de inverno e verão, no município de Monte Castelo - SC, sofrendo infestação natural por ectoparasitas. A cada 28 dias, fêmeas de carrapatos (Rhipicephalus (Boophilus) microplus) com tamanho acima de 4 mm foram quantificadas nos dois lados do corpo de cada animal, bem como os nódulos com larvas de Dermatobia hominis. A espessura da capa do pelame foi também avaliada a cada coleta e os animais foram classificados quanto à coloração do pelame. Os animais da raça Crioula Lageana foram mais resistentes às infestações por D. hominis e R. microplus. Não houve diferença entre a disposição de carrapatos e bernes conforme os lados direito e esquerdo dos animais. Os animais de pelagem escura albergaram a maior porcentagem de ectoparasitas nos grupos avaliados.


This study was carried out to evaluate the breed resistance against ectoparasites infestations in Crioula Lageana and crossbred Angus male calves under natural condition. Ten weaned and neutered male calves (6-8 months) of each group were kept together in winter and summer pastures in Monte Castelo, Santa Catarina State. Every 28 days, female ticks (Rhipicephalus (Boophilus) microplus) more than 4 mm long were counted on the right and left side of the body of each animal, as well as the D. hominis nodules. Coat thickness also was measured at each sampling and all animals were classified according coat color. Crioula Lageana group was less infested by D. hominis than crossbred Angus in five times and by ticks in the infestation peak (P<0.05). There were no differences in the distribution of the ticks and grubs between animal body sides. Dark-haired animal in both groups were the most susceptible.

Gastrointestinal nematode (GIN) infection is a major cause of production losses in cattle. This study was carried out to evaluate the natural resistance against nematode infection in Crioulo Lageano and crossbred Angus male calves. Crioulo Lageano is a local cattle breed in the state of Santa Catarina, in southern Brazil. Ten weaned calves of each breed were grazed together on pasture and naturally infected with nematodes between July 2009 and December 2010. Once every 28 days, we collected fecal and blood samples for parasitological and immunological tests, as well as recording body weights. After 19 samplings, all animals were slaughtered for quantification and identification of GINs. We found that the animals had been infected with the following nematode species, in decreasing order by the mean number of specimens: Trichostrongylus axei, Cooperia punctata, Ostertagia ostertagi, Haemonchus placei, Oesophagostomum radiatum, and Trichuris spp. There were no significant differences between the Crioulo Lageano and crossbred Angus groups in terms of worm burden or nematode fecal egg count, nor in terms of the mean levels of immunoglobulin (G and A) against C. punctata and H. placei antigens, except in IgA mean level in abomasal mucus against H. placei adult worms that was significantly higher in crossbred Angus cattle (p<0.05). At the end of the study, the crossbred Angus cattle were heavier than were the Crioulo Lageano cattle (mean live weight, 507.35 and 390.3 kg, respectively). Comparative parasitological and immunological evaluation revealed no difference between two breeds in terms of their natural resistance against GINs.

Infection with the gastrointestinal nematode Haemonchus contortus causes considerable losses in the sheep industry. In this study, we evaluated the effect that climate has on third-stage larvae (L3) of H. contortus in terms of their migration from sheep feces to Brachiaria decumbens grass, as well as their distribution among the forage plants. Fecal samples containing H. contortus L3 was deposited on the soil among the herbage at an initial height of 30 cm. Sample collection began 24h after contamination and was performed on alternate days over 13 days. The L3 were recovered and quantified in three strata (heights) of grass (0-10 cm, 10-20 cm and >20 cm) as well as in the remaining feces and a superficial layer of soil, collected from beneath the feces. In order to obtain results under different environmental conditions, fecal samples containing H. contortus L3 were deposited on pasture in January (summer), in April (autumn), and July (winter). In all of the periods, the L3 were able to migrate from the feces to the herbage. However, rains, accompanied by high relative humidity and high temperatures, apparently favored migration. The highest L3 recovery rate in the pasture was in the summer observation period, which had the highest number of days with measurable precipitation, high relative humidity (>68.2%), and the highest temperatures at the soil level (minimum and maximum means of 19°C and 42°C, respectively). Under those conditions, larvae began to reach the upper stratum of the grass (>20 cm) by 24h after the deposition of fecal matter, the number of larvae having reached that stratum peaking at seven days after deposition. In the autumn observation period, there was no rainfall in the first five days post-contamination. During that period, high numbers of larvae were found in the fecal samples demonstrating that feces can act as a reservoir of larvae in the absence of rain. Except for two days in the summer observation period, when most of the L3 were recovered from the tops of blades of grass, L3 where located predominantly at the base of the herbage. In conclusion, rainfall favors the migration of L3 from feces to herbage. In addition, larval migration up and along blades of grass can occur relatively rapidly when the temperature is high.

The aim of this study was to evaluate the efficacy of formulations of sodium alginate matrix (pellets) of the nematode predatory fungi, Duddingtonia flagrans (AC001 isolate) and Arthrobotrys robusta (I-31 isolate), in the biological control of sheep gastrointestinal nematode infections. Thirty young Bergamacia ewes were allocated into three groups: In group 1 (control), the animals received 2 g/10 kg of live weight (l.w.) of pellets without fungus; in group 2, each animal received 2 g/10 kg of l.w. of pellets of D. flagrans (0.2 g of fungus/10 kg l.w.); and in group 3, each animal received 2 g/10 kg of l.w. of pellets of A. robusta (0.2 g of fungus/10 kg l.w.). The animals of each group were kept separately under rotational grazing. Pellets, with or without fungi, were mixed with 1 kg animal food and administered twice a week for 6 months. There was no significant difference in mean live weight and packed cell volume among groups (P > 0.05). Mean nematode fecal egg counts (FEC) did not significantly differ between the control and the remaining groups, except in one or two collections, when FEC was higher in the control group than in group 2 and group 3, respectively. The group that received A. robusta pellets needed less salvage anthelmintic treatments. Haemonchus contortus was the predominant species recovered from tracer lambs. The nematophagous fungi, D. flagrans and A. robusta, did not provide satisfactory results in the prophylaxis of parasitic gastroenteritis in sheep, under the conditions of the present study.

The influence of trichomes on vertical migration and survival of Haemonchus contortus infective larvae (L3) on different forages was investigated. Four different forages showing different distributions of trichomes (Brachiaria brizantha cv. Marandu, Brachiaria brizantha cv. Xaraes, Andropogon gayanus, and Stylosanthes spp.), and one forage species without trichomes (Panicum maximum cv. Tanzania), were used. Forages cut at the post-grazing height were contaminated with faeces containing L3. Samples of different grass strata (0-10, 10-20, >20 cm) and faeces were collected for L3 quantification once per week over four weeks. In all forages studied, the highest L3 recovery occurred seven days after contamination, with the lowest recovery on A. gayanus. In general, larvae were found on all forages' strata. However, most of the larvae were at the lower stratum. There was no influence of trichomes on migration and survival of H. contortus L3 on the forages.
