INTRODUCTION: Memory CD8+ T cells are essential for long-term immune protection in viral infections, including COVID-19. METHODS: This study examined the responses of CD8+ TEM, TEMRA, and TCM subsets from unvaccinated individuals who had recovered from mild and severe COVID-19 by flow cytometry. RESULTS AND DISCUSSION: The peptides triggered a higher frequency of CD8+ TCM cells in the recovered mild group. CD8+ TCM and TEM cells showed heterogeneity in CD137 expression between evaluated groups. In addition, a predominance of CD137 expression in naïve CD8+ T cells, TCM, and TEM was observed in the mild recovered group when stimulated with peptides. Furthermore, CD8+ TCM and TEM cell subsets from mild recovered volunteers had higher TNF-α expression. In contrast, the expression partner of IFN-γ, IL-10, and IL-17 indicated an antiviral signature by CD8+ TEMRA cells. These findings underscore the distinct functional capabilities of each memory T cell subset in individuals who have recovered from COVID-19 upon re-exposure to SARS-CoV-2 antigens.

A novel modification technique employing a layer-by-layer (LbL) self-assembly method, integrated with a pressure-assisted filtration system, was developed for enhancing a commercial polyethersulfone (PES) microfiltration (MF) membrane. This modification involved the incorporation of tannic acid (TA) in conjunction with graphene oxide (GO) nanosheets. The effectiveness of the LbL method was confirmed through comprehensive characterization analyses, including ATR-FTIR, SEM, water contact angle (WCA), and mean pore size measurements, comparing the modified membrane with the original commercial one. Sixteen variations of PES MF membranes were superficially modified using a three-factorial design, with the deposited amount of TA and GO as key factors. The influence of these factors on the morphology and performance of the membranes was systematically investigated, focusing on parameters such as pure water permeability (PWP), blue corazol (BC) dye removal efficiency, and flux recovery rate (FRR). The membranes produced with the maximum amount of GO (0.1 mg, 0.55 wt%) and TA as the inner and outer layers demonstrated remarkable FRR and significant BC removal, exceeding 80%. Notably, there was no significant difference observed when using either 0.2 (1.11 wt%) or 0.4 mg (2.22 wt%) in the first layer, as indicated by the Tukey mean test. Furthermore, the modified membrane designated as MF/TA0.4GO0.1TA0.4 was evaluated in the filtration of a simulated dye bath wastewater, exhibiting a BC removal efficiency of 49.20% and a salt removal efficiency of 27.74%. In conclusion, the novel PES MF membrane modification proposed in this study effectively enhances the key properties of pressure-driven separation processes.

Purpose: To investigate the potential relation between methylation of miR-9-3 and stages of diabetic retinopathy (DR). Additionally, we explored whether miR-9-3 methylation impacts the serum levels of Vascular Endothelial Growth Factor (VEGF). Methods: A cross-sectional study was conducted with 170 participants with type 2 diabetes, including a control group (n = 64) and a diabetes retinopathy group (n = 106), which was further divided into NPDR (n = 58) and PDR (n = 48) subgroups. Epidemiological, clinical, anthropometric, biochemical ELISA assay were analysed. DNA extracted from leukocytes was used to profile miR-9-3 methylation using PCR-MSP. Results: MiR-9-3 hypermethylated profile was higher in the DR group (p < 0.001) and PDR subgroup compared to DM2 control group (p < 0.001). The hypermethylated profile in the PDR subgroup was also higher compared to NPDR subgroup (p < 0.001). There was no difference between DM2 control and NPDR group (p = 0.234). Logistic regression showed that miR-9-3 hypermethylation increases the odds of presenting DR (OR: 2.826; p = 0.002) and PDR (OR: 5.472; p < 0.001). In addition, hypermethylation of miR-9-3 in the DR and NPDR subgroup was associated with higher serum VEGF-A levels (p = 0.012 and p = 0.025, respectively). Conclusion: The methylation profile of the miR-9-3 promoter increases the risk of developing PDR. Higher levels of VEGF-A are associated with miR-9-3 hypermethylated profile in patients in the DR and NPDR stages. Objetivou-se analisar o perfil epidemiológico das notificações de acidentes de trabalho com exposição a materiais biológicos entre profissionais de enfermagem no Paraná, por meio da série histórica determinada pelos anos de 2013 a 2022. Estudo epidemiológico observacional descritivo do tipo levantamento, fundamentado na epidemiologia descritiva, a partir de dados do Sistema de Informações de Agravos de Notificação e do Cadastro Nacional de Estabelecimentos de Saúde. Foram Identificados 48.405 casos, com prevalência de 98,54/1.000 profissionais. Destaca-se maior ocorrência no ano de 2019, na Regional de Saúde Metropolitana, entre técnicos de enfermagem, do sexo feminino, com faixa etária entre 31 e 40 anos. Entre enfermeiros, a faixa etária prevaleceu entre 18 e 30 anos, com evolução de alta sem conversão sorológica e fontes negativas para doenças. Destaca-se o alto número de tratamentos abandonados e subnotificações. Investir na formação profissional, na educação permanente, em equipamentos de proteção de qualidade e na efetivação dos protocolos para tratamento profilático é fundamental para prevenir o agravo.


The objective was to analyze the epidemiological profile of notifications of work accidents with exposure to biological material among nursing professionals in Parana, through the historical series determined for the years 2013 to 2022. Descriptive observational epidemiological study of the survey type, based on descriptive epidemiology, based on data from the System Information on Notifiable Diseases and the National Registry of Health Establishments. 48,405 cases were identified, with a prevalence of 98.54/1,000. More occurrences among female nursing technicians aged between 31 and 40 years old, from the Metropolitan Health Regional Facilities, in year 2019, stand out. Among nurses, the age range was between 18 and 30 years old, with discharge progressing without serological conversion and negative sources of disease. The high number of abandoned treatments and underreporting stands out. Investing in professional training, continuing education, quality protective equipment and implementing protocols for prophylactic treatment are essential to prevent the disease.


El objetivo fue analizar el perfil epidemiológico de las notificaciones entre profesionales de enfermería de Paraná, a través de la serie histórica determinada para los años 2013 a 2022. Estudio epidemiológico observacional descriptivo, tipo encuesta, con base en epidemiología descriptiva, a partir de datos del Sistema de Información de Enfermedades de Notificación Obligatoria. Se identificaron 48.405 casos, con una prevalencia de 98,54/1.000. Sobresalen más ocurrencias en 2019 entre técnicos de enfermería, del sexo femenino, edades entre 31 y 40 años, en la Regional de Salud Metropolitana. Entre los enfermeros, el rango de edad estuvo comprendido entre 18 y 30 años, progresando el alta sin conversión serológica y focos negativos de enfermedad. Destaca elevado número de tratamientos abandonados y subregistro. Invertir en formación profesional, educación continua, equipos de protección de calidad e implementar protocolos de tratamiento profiláctico son fundamentales para prevenir la enfermedad.

A tuberculose é uma das doenças mais antigas do mundo. Os fatores de vulnerabilidade social, individual, programática, permitem que a tuberculose fique em latência por vários anos no indivíduo. Objetivo: Analisar, por meio de dados secundários, incidência e protocolo de infecção latente de tuberculose no município de São Paulo. Método: Trata-se de revisão sistemática da literatura. Em 29 de agosto de 2023, com a busca no Descritores em ciências da saúde: Tuberculose AND Infecção Latente, foram encontrados 4.527 artigos, após aplicação dos filtros: Base de dados LILACS e MEDLINE, texto completo, em Português, 2018 a 2023, restaram 38 documentos que foram submetidos ao checklist PRISMA. Resultados: Restaram 16 artigos que embasaram os resultados e a discussão. Conclusão: Iniciativas, como a busca ativa de comunicantes de tuberculose, devem ser construídas a partir de estratégias propostas em conjunto com pesquisadores e gestores alinhados ao Ministério da Saúde.(AU)


Tuberculosis is one of the oldest diseases in the world. Social, individual and programmatic vulnerability factors allow tuberculosis to remain latent for several years in the individual. Objective: To analyze, using secondary data, the incidence and protocol of latent tuberculosis infection in the city of São Paulo. Method: This is a systematic literature review. On August 29, 2023, 4,527 articles were found after applying the filters in the Health Sciences Descriptors: Tuberculosis AND Latent Infection: LILACS and MEDLINE database, full text, in Portuguese, 2018 to 2023, 38 documents remained that were submitted to the PRISMA checklist. Results: 16 articles were found to support the results and discussion. Conclusion: Initiatives, such as the active search for tuberculosis communicants, should be built on strategies proposed jointly with researchers and managers in line with the Ministry of Health.(AU)


La tuberculosis es una de las enfermedades más antiguas del mundo. Factores sociales, individuales y programáticos de vulnerabilidad permiten que la tuberculosis permanezca latente por varios años en el individuo. Objetivo: Analizar, a partir de datos secundarios, la incidencia y el protocolo de infección tuberculosa latente en el municipio de São Paulo. Método: Se trata de una revisión sistemática de la literatura. El 29 de agosto de 2023, fueron encontrados 4.527 artículos después de la aplicación de los filtros en los Descriptores de Ciencias de la Salud: Tuberculosis E Infección Latente: Base de datos LILACS y MEDLINE, texto completo, en portugués, 2018 a 2023, permanecieron 38 documentos que fueron sometidos a la lista de verificación PRISMA. Resultados: Quedaron 16 artículos, que constituyeron la base de los resultados y la discusión. Conclusión: Iniciativas como la búsqueda activa de comunicantes de tuberculosis deben construirse a partir de estrategias propuestas conjuntamente por investigadores y gestores en consonancia con el Ministerio de Salud.(AU)

Objective: To report on the experience and impressions of the Brazilian orthopedic trauma surgeons attending the Leadership Development Program (LDP) hosted by the Sociedade Brasileira do Trauma Ortopédico (SBTO) in Sao Paulo, Brazil on November 4, 2022. Methods: Forty-eight orthopedic trauma surgeons from five different regions throughout Brazil were provided a link to complete The Big Five Test, a validated online personality assessment. The questionnaire was available in Portuguese and was intended to provide a background on individual personality traits and their influence on interpersonal interactions. The LDP integrated content from literature reviews specific to Latin America, established leadership programs from leading business schools, and various subject matter experts. Prior to the start of the LDP, participants received a pre-course survey evaluating demographic information, a needs assessment, and the prioritization of leadership topics utilizing a 5-point Likert-scale. Attendees participated in the one-day, interactive LDP focusing on the fundamental principles of leadership development, communication, personal development, emotional intelligence and negotiation. Following the LDP, a post-course evaluation was administered to determine the participants' overall experience, and suggestions for LDP improvement. Results: Forty-one of the forty-eight course participants completed the pre-course evaluation, whereas forty-six of the forty-eight participants completed the post-course evaluations. Overwhelmingly, the lack of opportunity was most prevalently reported as the main obstacle to attending a leadership course, as cited by 56% of respondents. Conclusion: Expanding the accessibility, diversity, and customizability of leadership programs can facilitate the development of personal tools needed to move healthcare forward. Critical topics include emotional intelligence and other differentiating leadership qualities that distinguish true transformational and servant leaders. Advancing leadership skills can stimulate networking, expose learners to experiential learning styles, inspire others to create positive change, and engender creative solutions for systematic improvements and health outcomes. Objetivo: Relatar a experiência e as impressões de cirurgiões de trauma ortopédico brasileiros participantes do Programa de Desenvolvimento de Liderança (PDL), organizado pela Sociedade Brasileira do Trauma Ortopédico (SBTO), em São Paulo, Brasil, em 4 de novembro de 2022. Métodos: Quarenta e oito cirurgiões de trauma ortopédico de cinco regiões diferentes do Brasil receberam um link para preencher o The Big Five Test, uma avaliação de personalidade on-line validada. O questionário estava disponível em português e pretendia fornecer informações básicas sobre traços de personalidade individuais e sua influência nas interações interpessoais. O PDL integrou conteúdo de análises de literatura específicas da América Latina, e programas de liderança estabelecidos pelas principais escolas de negócios e por vários especialistas no assunto. Antes do início do PDL, os participantes receberam uma pesquisa pré-curso solicitando informações demográficas, uma avaliação de necessidades e a priorização de tópicos de liderança utilizando uma escala Likert de 5 pontos. Os participantes participaram do PDL interativo de um dia com foco nos princípios fundamentais de desenvolvimento de liderança, comunicação, desenvolvimento pessoal, inteligência emocional e negociação. Após o PDL, foi realizada uma avaliação pós-curso para determinar a experiência geral dos participantes e sugestões para melhoria do PDL. Resultados: Quarenta e um dos quarenta e oito participantes do curso concluíram a avaliação pré-curso, enquanto quarenta e seis dos quarenta e oito participantes concluíram a avaliação pós-curso. A falta de oportunidade foi relatada com maior prevalência como o principal obstáculo para frequentar um curso de liderança, conforme citado por 56% dos entrevistados. Conclusão: Expandir a acessibilidade, a diversidade e a personalização dos programas de liderança podem facilitar o desenvolvimento de ferramentas pessoais necessárias para fazer avançar os cuidados de saúde. Os tópicos críticos incluem inteligência emocional e outras qualidades de liderança diferenciadas, que distinguem verdadeiros líderes transformacionais e servidores O avanço das competências de liderança pode estimular o networking, expor os alunos a estilos de aprendizagem experiencial, inspirar outros a criar mudanças positivas e gerar soluções criativas para melhorias sistemáticas dos resultados na saúde. BACKGROUND: Understanding the causes of intracerebral hemorrhage (ICH) is crucial for effective treatment and preventing recurrences. The SMASH-U scale is a suggested method for classifying and predicting the outcomes of ICH. OBJECTIVE: To describe the SMASH-U classification and outcomes by etiology in patients admitted to a comprehensive stroke center in São Paulo, Brazil. METHODS: A retrospective analysis was conducted on patients admitted to the hospital or outpatient clinic between April 2015 and January 2018. Two stroke neurologists evaluated the SMASH-U classification, and patients with incomplete medical records were excluded. RESULTS: Out of the 2000 patients with a stroke diagnosis evaluated, 140 were included in the final analysis. The mean age was 57.9 (± 15.5) years, and 54.3% were male. Hypertension was the most frequent etiology, accounting for 41.4% of cases, followed by amyloid angiopathy (18.5%) and structural lesions (14.1%). Structural lesions were more common among women and patients under 45 years old. Favorable outcomes were observed in 61% of patients with structural lesions, compared to 10% of patients with medication-related etiologies. CONCLUSION: This study provides important evidence regarding the etiological classification of Brazilian patients with ICH. Hypertension and amyloid angiopathy were the most frequent causes, while structural lesions and systemic diseases were more common in younger patients.

A escala SMASH-U é um método sugerido para classificar e prever os resultados da HIC. OBJETIVO: Descrever a classificação SMASH-U e os resultados por etiologia em pacientes admitidos em um centro de acidente vascular cerebral (AVC) em São Paulo, Brasil. MéTODOS: Foi realizada uma análise retrospectiva de pacientes admitidos no hospital ou ambulatório entre abril de 2015 e janeiro de 2018. Dois neurologistas especializados em doenças cerebrovasculares avaliaram a classificação SMASH-U e pacientes com prontuários incompletos foram excluídos. RESULTADOS: Dos 2000 pacientes com diagnóstico de AVC avaliados, 140 foram incluídos na análise final. A idade média foi de 57,9 (±15,5) anos e 54,3% eram do sexo masculino. A hipertensão foi a etiologia mais frequente, correspondendo a 41,4% dos casos, seguida pela angiopatia amiloide (18,5%) e lesões estruturais (14,1%). As lesões estruturais foram mais comuns em mulheres e pacientes com menos de 45 anos. Resultados favoráveis foram observados em 61% dos pacientes com lesões estruturais, em comparação com 10% dos pacientes com etiologias relacionadas a medicamentos. CONCLUSãO: Este estudo fornece evidências importantes sobre a classificação etiológica de pacientes brasileiros com HIC. A hipertensão e a angiopatia amiloide foram as causas mais frequentes, enquanto lesões estruturais e doenças sistêmicas foram mais comuns em pacientes mais jovens.

The objective of this research was to evaluate how pregnancy and feeding regimens affect the feed intake, digestibility, and efficiency of microbial nitrogen (N) synthesis in beef cows. Forty-four multiparous Nellore cows, comprising 32 gestating and 12 non-gestating cows, with an average weight of 451 ± 10 kg, were assigned to either a HIGH (ad libitum) or LOW (limited feeding at 1.2 times maintenance based on the NRC) feeding regimen during the gestational period. The dry matter intake (DMI) in kg/d was significantly greater (P < 0.01) in HIGH-fed cows. The DMI reduced (P < 0.05) in proportion to the shrunk body weight (SBW) as days of pregnancy (DOP) increased. The interaction between feeding level and DOP was significant (P < 0.05) for the digestibility of dry matter (DM), organic matter (OM), N compounds, ether extract (EE), ash- and protein-free neutral detergent fiber (NDFap), gross energy (GE), and total digestible nutrients (TDN). Except for DM and TDN digestibility, there was a reduced nutrient digestibility as gestation progressed in HIGH-fed cows. In contrast, digestibility increased as a function of DOP in LOW-fed cows. Microbial N synthesis (g/day) was significantly higher in HIGH-fed cows (P < 0.001) compared to LOW-fed cows. The efficiency of microbial N production per g of N intake and kg of digestible OM intake was (P = 0.021) and tended (P = 0.051) to be greater in LOW-fed cows compared to HIGH-fed cows. In summary, HIGH-fed Nellore cows reduce feed intake and digestibility with advancing gestation, affecting feed utilization. In addition, LOW-fed cows, showed higher microbial protein synthesis efficiency, potentially making them more nutrient-efficient under challenging nutritional conditions.

Prior to the start of the LDP, participants received a pre-course survey evaluating demographic information, a needs assessment, and the prioritization of leadership topics utilizing a 5-point Likert-scale. Attendees participated in the one-day, interactive LDP focusing on the fundamental principles of leadership development, communication, personal development, emotional intelligence and negotiation. Following the LDP, a post-course evaluation was administered to determine the participants' overall experience, and suggestions for LDP improvement. Results: Forty-one of the forty-eight course participants completed the pre-course evaluation, whereas forty-six of the forty-eight participants completed the post-course evaluations. Overwhelmingly, the lack of opportunity was most prevalently reported as the main obstacle to attending a leadership course, as cited by 56% of respondents. Conclusion: Expanding the accessibility, diversity, and customizability of leadership programs can facilitate the development of personal tools needed to move healthcare forward. Critical topics include emotional intelligence and other differentiating leadership qualities that distinguish true transformational and servant leaders. Advancing leadership skills can stimulate networking, expose learners to experiential learning styles, inspire others to create positive change, and engender creative solutions for systematic improvements and health outcomes. RESUMO Objetivo: Relatar a experiência e as impressões de cirurgiões de trauma ortopédico brasileiros participantes do Programa de Desenvolvimento de Liderança (PDL), organizado pela Sociedade Brasileira do Trauma Ortopédico (SBTO), em São Paulo, Brasil, em 4 de novembro de 2022. Métodos: Quarenta e oito cirurgiões de trauma ortopédico de cinco regiões diferentes do Brasil receberam um link para preencher o The Big Five Test, uma avaliação de personalidade on-line validada. O questionário estava disponível em português e pretendia fornecer informações básicas sobre traços de personalidade individuais e sua influência nas interações interpessoais. O PDL integrou conteúdo de análises de literatura específicas da América Latina, e programas de liderança estabelecidos pelas principais escolas de negócios e por vários especialistas no assunto. Antes do início do PDL, os participantes receberam uma pesquisa pré-curso solicitando informações demográficas, uma avaliação de necessidades e a priorização de tópicos de liderança utilizando uma escala Likert de 5 pontos. Os participantes participaram do PDL interativo de um dia com foco nos princípios fundamentais de desenvolvimento de liderança, comunicação, desenvolvimento pessoal, inteligência emocional e negociação. Após o PDL, foi realizada uma avaliação pós-curso para determinar a experiência geral dos participantes e sugestões para melhoria do PDL. Resultados: Quarenta e um dos quarenta e oito participantes do curso concluíram a avaliação pré-curso, enquanto quarenta e seis dos quarenta e oito participantes concluíram a avaliação pós-curso. A falta de oportunidade foi relatada com maior prevalência como o principal obstáculo para frequentar um curso de liderança, conforme citado por 56% dos entrevistados. Conclusão: Expandir a acessibilidade, a diversidade e a personalização dos programas de liderança podem facilitar o desenvolvimento de ferramentas pessoais necessárias para fazer avançar os cuidados de saúde. Os tópicos críticos incluem inteligência emocional e outras qualidades de liderança diferenciadas, que distinguem verdadeiros líderes transformacionais e servidores O avanço das competências de liderança pode estimular o networking, expor os alunos a estilos de aprendizagem experiencial, inspirar outros a criar mudanças positivas e gerar soluções criativas para melhorias sistemáticas dos resultados na saúde. Asthma is a disease characterized by chronic inflammation and hyper responsiveness of airways. We aimed to assess the relaxant potential of phosphodiesterase-4 (PDE4) inhibitors N-sulfonilhidrazonic derivatives on non-asthmatic and asthmatic guinea pig trachea. Firstly, guinea pigs were sensitized and challenged with ovalbumin, and then morphological, and contractile changes were evaluated resulting from asthma, followed by evaluation of relaxant effect of derivatives on guinea pig trachea and the cAMP levels measurement by ELISA. It has been evidenced hypertrophy of airway smooth muscle, inflammatory infiltrate, and vascular abnormalities. Moreover, only sensitized tracheal rings were responsive to OVA. Contractile response to histamine, but not to carbachol, was greater in sensitized animals, however the relaxant response to aminophylline and isoprenaline were the same in non-asthmatics and asthmatics. N-sulfonilhidrazonic derivatives presented equipotent relaxant action independent of epithelium, with exception of LASSBio-1850 that presented a low efficacy (< 50%) and LASSBio-1847 with a 4-fold higher potency on asthmatics. LASSBio-1847 relaxant curve was impaired in the presence of propranolol and potentiated by isoprenaline in both groups. Furthermore, relaxation was potentiated 54- and 4-fold by forskolin in non-asthmatics and asthmatics, respectively. Likewise, LASSBio-1847 potentiated relaxant curve of aminophylline 147- and 4-fold in both groups. The PKA inhibitor H-89 impaired the relaxant potency of the derivative. Finally, LASSBio-1847 increased tracheal intracellular cAMP levels similarly to rolipram, selective PDE4 inhibitor, in both animals. LASSBio-1847 showed to be promising to relax guinea pig trachea from non-sensitized and sensitized guinea pigs by activation of ß2-adrenergic receptors/AC/cAMP pathway.

Vertically unstable pelvic injuries associated with sacroiliac disruption are challenging. Although percutaneous iliosacral fixation using two screws at S1 vertebral body has been shown beneficial, the use of two transiliac screws at S1 has been proposed to increase the fixation strength of the construct. In the herein study, the finite element method (FEM) was performed to analyse the biomechanical behaviour of five different constructions using iliosacral screws only, transiliac screws only, and combining an iliosacral and a transiliac screw. A vertically unstable AO 61C1.2 type pelvic injury was produced for the evaluation of the posterior pelvic displacement and implant stress, and the anterior implant stress using FEM. The symphysis pubis was fixed with a 3.5-mm reconstruction plate in all cases. The model was axially loaded with 800N applied in the centre of S1 body, perpendicular to the ground (Y-axis), simulating the bipodal stance moment. There was a statistically significant reduction in both posterior displacement and implant stresses in the groups fixed with at least one transiliac screw compared to the groups fixed with iliosacral screws. In our FEM study, the construct using two transiliac screws in S1 is biomechanically superior for stabilizing the sacroiliac joint in vertically unstable pelvic ring injuries compared to the other configurations. Lateral displacement, posterior displacement, and von Mises stress were reduced with this construct. A good option can be the use of one iliosacral screw and one transiliac screw in S1.

Many studies have focused on SARS-CoV-2 and Mycobacterium tuberculosis (Mtb) co-infection consequences. However, after a vaccination plan against COVID-19, the cases of severe disease and death are consistently controlled, although cases of asymptomatic and mild COVID-19 still happen together with tuberculosis (TB) cases. Thus, in this context, we sought to compare the T cell response of COVID-19-non-vaccinated and -vaccinated patients with active tuberculosis exposed to SARS-CoV-2 antigens. Flow cytometry was used to analyze activation markers (i.e., CD69 and CD137) and cytokines (IFN-γ, TNFα, IL-17, and IL-10) levels in CD4+ and CD8+ T cells upon exposure to SARS-CoV-2 peptides. The data obtained showed that CD8+ T cells from non-vaccinated TB patients present a high frequency of CD69 and TNF-α after viral challenge compared to vaccinated TB donors. Conversely, CD4+ T cells from vaccinated TB patients show a high frequency of IL-10 after spike peptide stimulus compared to non-vaccinated patients. No differences were observed in the other parameters analyzed. The results suggest that this reduced immune balance in coinfected individuals may have consequences for pathogen control, necessitating further research to understand its impact on clinical outcomes after COVID-19 vaccination in those with concurrent SARS-CoV-2 and Mtb infections.

This study evaluated collagen solubility and gene expression of biomarkers for intramuscular collagen (IMCT) deposition and remodeling in the Longissimus muscle of bulls and steers through the finishing phase. Thirty-six Nellore calves were used (18 bulls and 18 steers), and six of each sexual condition were randomly assigned to be harvested at 0, 100, or 200 days on feed (DOF) to evaluate collagen characteristics in different time points throughout the finishing phase. Bulls showed a greater collagen solubility than steers (P = 0.03). The gene expression of fibrogenic markers (TGFß1, COL1A1, and COL3A1) and IMCT remodeling mediators (MMPII, TIMPII, and LOX) were not affected by sexual condition or DOF (P > 0.05). Our data indicate that young Nellore bulls have a higher percentage of soluble intramuscular collagen, possibly due to higher collagen remodeling associated with a faster growth rate and muscle hypertrophy. Moreover, castration and DOF did not modify mRNA levels of fibrogenic and collagen remodeling markers.

T-cell immunity plays a critical role in controlling viral infections, making it essential to identify specific viral targets to develop effective vaccines. In this study, we focused on identifying and understanding the potential effects of different SARS-CoV-2-derived peptides, including spike, nucleocapsid, and ORFs, that have the potential to serve as T-cell epitopes. Assessing T cell response through flow cytometry, we demonstrated that PBMC collected from vaccinated individuals had a significantly higher expression of important biomarkers in controlling viral infection and proper regulation of immune response mediated by T CD4+ and T CD8+ cells stimulated with immunodominant peptides. These data highlight how cellular immune responses to some of these peptides could contribute to SARS-CoV-2 protection due to COVID-19 immunization.

Mycobacterium tuberculosis complex causes tuberculosis (TB), a disease that causes pulmonary inflammation but can also affect other tissues. Despite macrophages having a defined role in TB immunopathogenesis, other innate immune cells, such as neutrophils, are involved in this process. These cells have high phagocytic ability and a microbial-killing machine comprised of enzymes, antimicrobial peptides, and reactive oxygen species. In the last two decades, a new neutrophil immune response, the neutrophil extracellular traps (NETs), has been intensely researched. NETs comprise DNA associated with histones, enzymes, and antimicrobial peptides. These structures are related to antimicrobial immune response and some immuno-pathogenesis mechanisms. This mini review highlights the role of NETs in tuberculosis and how they can be helpful as a diagnostic tool and/or therapeutic target.

Hyperprolactinemia is a pathological condition resulting from increased prolactin that directly affects reproduction, as this condition inhibits the release of LH, FSH and gonadal steroidogenesis, bringing several negative clinical associations in reproduction. In contrast, melatonin (MEL) plays an important role in the regulation of steroidogenesis and modulates damages to the process of spermatogenesis. The objective was to analyze the protective effects of exogenous melatonin on the testis of hyperprolactinemic adult rats. Forty-eight male rats were used, divided into two treatment periods: 30 and 60 days, each treatment was subdivided into three groups: Control, Hyper (hyperprolactinemia), and Hyper+MEL (hyperprolactinemia and melatonin). Treatment with melatonin was 200 µg/100 g, subcutaneously. Induction of hyperprolactinemia was obtained with a dose of 4 mg/kg of domperidone, subcutaneously. The results of the histopathology demonstrated that the animals in the Hyper group presented degeneration of germ cells when compared to the control. In addition, the degenerations were presented in smaller quantities in the Hyper+MEL, in both treatment periods, evidencing the benefits of the melatonin in gonadal regeneration. The Hyper group of both treatment periods showed a decrease in tubular diameter, epithelium height, and tubular area, in addition to a decrease in Sertoli cells, when compared to the control and the Hyper+MEL group. In conclusion, the hyperprolactinemia can affect the germinal epithelium and testicular microstructure; the exogenous melatonin has a protective effect against hyperprolactinemia, reducing testicular damage.

We evaluated the effects of incremental amounts of ground flaxseed (GFX) on diversity and relative abundance of ruminal microbiota taxa, enteric methane (CH4) emissions, and urinary excretion of purine derivatives (PD) in lactating dairy cows in a replicated 4 × 4 Latin square design. Twenty mid-lactation Jersey cows were used in the study. Of these 20 cows, 12 were used for ruminal sampling, 16 for enteric CH4 measurements, and all for spot urine collection. Each period lasted 21 d with 14 d for diet adaptation and 7 d for data and sample collection. Diets were formulated by replacing corn meal and soybean meal with 0%, 5%, 10%, and 15% of GFX in the diet's dry matter. Ruminal fluid samples obtained via stomach tubing were used for DNA extraction. Enteric CH4 production was measured using the sulfur hexafluoride tracer technique. Diets had no effect on ruminal microbiota diversity. Similarly, the relative abundance of ruminal archaea genera was not affected by diets. In contrast, GFX decreased or increased linearly the relative abundance of Firmicutes (P < 0.01) and Bacteroidetes (P < 0.01), respectively. The relative abundance of the ruminal bacteria Ruminococcus (P < 0.01) and Clostridium (P < 0.01) decreased linearly, and that of Prevotella (P < 0.01) and Pseudobutyrivibrio (P < 0.01) increased linearly with feeding GFX. A tendency for a linear reduction (P = 0.055) in enteric CH4 production (from 304 to 256 g/d) was observed in cows fed increasing amounts of GFX. However, neither CH4 yield nor CH4 intensity was affected by treatments. Diets had no effect on the urinary excretion of uric acid, allantoin, and total PD. Overall, feeding GFX decreased linearly the relative abundance of the ruminal bacterial genera Ruminococcus and Clostridium and enteric CH4 production, but no change was seen for CH4 yield and CH4 intensity, or urinary excretion of total PD, suggesting no detrimental effect of GFX on microbial protein synthesis in the rumen.

Objetivou-se analisar a associação entre características sociodemográficas e clínicas com o desenvolvimento dos graus de incapacidades físicas 1 ou 2 em pessoas com diagnóstico de hanseníase na Paraíba, Brasil. Estudo ecológico, de base populacional, que teve como unidades de análises os 223 municípios do estado. Os dados foram coletados no Núcleo de Doenças Crônicas e Negligenciadas/Hanseníase, pertencente à Gerência Executiva de Vigilância em Saúde da Secretaria de Saúde do estado da Paraíba em junho de 2021, após extração do Sistema de Informação Nacional de Agravos de Notificação. O banco de dados reuniu 2.468 casos novos de hanseníase registrados no período de 2016 a 2020. A chance de uma pessoa diagnosticada com hanseníase desenvolver a incapacidade física 1 ou 2 é maior nas pessoas de sexo masculino, com 15 anos ou mais, estudo formal menor ou igual a nove anos, classificação operacional multibacilar, com mais de cinco lesões e mais de um nervo afetado, além de baciloscopia positiva. Políticas de educação em saúde são fortemente recomendadas no intuito de melhorar o conhecimento dos profissionais e da comunidade. Abordagens sobre a hanseníase, diagnóstico precoce, busca ativa, vigilância e acompanhamento dos casos e de seus contatos, além das incapacidades físicas, em especial para pessoas de maior vulnerabilidade a desenvolvê-las, são fundamentais.(AU)


The objective was to analyze the association between sociodemographic and clinical characteristics with the development of physical physical disabilities of grades 1 and 2 in people diagnosed with leprosy in Paraíba, Brazil. It's an ecological study, population-based, which had the 223 municipalities at the Center of Chronic and Neglected Diseases/Leprosy belonging to the Health Surveillance Executive Management of Health Department of Paraíba State in June 2021, after extraction from the Notifiable Diseases Information System. The database gathered 2,468 new cases of leprosy registered in the period from 2016 to 2020. The probability of a person diagnosed with leprosy developing physical disability 1 or 2 is greater in males, aged 15 or over, formal education less than or equal to nine years, operational classification multibacillary, with more than five lesions and more than one affected nerve, in addition to positive bacilloscopy. Health education policies are strongly recommended in order to improve the knowledge of professionals and the community. Approaches about leprosy, early diagnosis, active search, surveillance and follow-up of cases and their contacts, in addition to physical disabilities, especially for people who are more vulnerable to develop them, are fundamental.(AU)

BACKGROUND: Changes in Caveolin-1 (CAV-1) expression are related to tumorigenesis. The aim of this study was to evaluate the role of CAV-1 in tumor progression in oral squamous cell carcinoma (SCC) tissue samples and the effect of CAV-1 silencing on two oral tongue SCC (OTSCC) cell lines (SCC-25, from a primary tumor, and HSC-3 from lymph node metastases). METHODS: Mycroarray hybridization, mRNA expression, and immunohistochemistry were performed on OSCC tissue samples and corresponding non-tumoral margin tissues. The effects of CAV-1 silencing (siCAV-1) on cell viability, membrane fluidity, on the expression of epithelial to mesenchymal transition (EMT) markers and on cell migration and invasion capacity of OTSCC cell lines were evaluated. RESULTS: Microarray showed a greater CAV-1 expression (1.77-fold) in OSCC tumors than in non-tumoral tissues and 2.0-fold more in less aggressive OSCCs. However, significant differences in CAV-1 gene expression were not seen between tumors and non-tumoral margins nor CAV-1 with any clinicopathological parameters. CAV-1 protein was localized both in carcinoma and in spindle cells of the tumor microenvironment (TME), and CAV-1 positive TME cells were associated with smaller/more aggressive tumors, independent of the carcinoma cells' expression. Silencing of CAV-1 increased cell viability only in SCC-25 cells. It also stimulated the invasion of HSC-3 cells and increased ECAD and BCAT mRNA in these cells; however, the protein levels of the EMT markers were not affected. CONCLUSION: Decreased expression of CAV-1 by tumor cells in OSCC and an increase in the TME were associated with increased cell invasiveness and tumor aggressiveness.
