RESUMO

The effects of helium cold plasma on some parameters of metabolism of rat erythrocytes were studied after a course exposure (10 daily procedures, 1 min each). The intensity of free-radical processes in erythrocyte membranes, malondialdehyde concentration, and aldehyde dehydrogenase activity were analyzed. The exposure to helium cold plasma led to a significant increase in malondialdehyde level mostly associated with inhibition of its utilization. These changes were not related to activation of malondialdehyde synthesis in LPO reactions.

RESUMO

We studied aldehyde dehydrogenase activity in erythrocytes from healthy rats and animals with thermal trauma after NO inhalation. NO had an activating effect on catalytic properties of aldehyde dehydrogenase in healthy rats and burned animals. The effect of NO was more pronounced during burn disease.

We have studied specific features of lipid peroxidation in whole human blood under the action of aqueous solutions of xymedone (19.6 microM), phytic acid (117.9 microM) and its complex (237.6 microM) synthesized in distilled water and isotonic (0.9%) solution of sodium chloride. The estimated parameters included lipid peroxidation (LPO) rate, total antioxidant potential, superoxide dismutase (SOD) level, and malonic dialdehyde (MDA) level in blood plasma and erythrocytes. It was established that the effect of phytic acid on blood samples includes moderate stimulation of total antioxidant activity and SOD activity with predominant prooxidant effect. The phytic acid--xymedone complex synthesized in distilled water exhibits an antioxidant action, while its synthesis in saline solution yields a prooxidant.

AIM: To make multiparametric analysis of blood metabolic profile in early period of burn disease. MATERIALS AND METHODS: We tested blood samples of 15 healthy adults (control group) and 60 patients with thermal trauma (main group--II-4IIIA, B degree of burn, more then 15 bsp). Parameters of lipid metabolism, level of glucose, lactate, malonic dialdehyde and some enzymes in blood plasma and erythrocytes were estimated. RESULTS: In early period of burn disease we fixed the clear metabolic disorders, including tissues hypoxia, activation of plasma transaminases and oxidoreductases, inhibition of detoxication system, induction of oxidative stress. Connection of metabolic changes, associated with burn disease, was registered. It supported by numerous correlations between studied parameters, formed from first day after trauma. CONCLUSIONS: Our data expand the knowledge about mating metabolic changes of catalytic activity of blood enzymes, forming in early period of burn disease (system metabolic disadaptation), and diagnostic value of some blood biochemical parameters in estimation of burned patient metabolism.

The effect of dinitrosyl iron complexes (DNIC) on the energy metabolism of erythrocytes under combined thermal trauma conditions has been studied on a group of 30 Wistar rats, which was divided into 3 groups: intact (n = 10), control (n = 10), and main (n = 10). Combined thermal trauma (skin burn + thermoinhalation damage) was modeled in animals of the control and main groups. Rats of control group received infusions of sodium chloride solution (n = 10) every day. Rats of the main group obtained infusions of DNIC solution in sodium chloride. Rat blood samples were characterized by the activity of lactate dehydrogenase in direct and reverse reaction, lactate level, and coefficients of the substrate provision and energy reactions balance. It was stated, that DNIC clearly normalized the energy metabolism of erythrocytes beginning with the third day after thermal trauma onset.

The aim of this work was to estimate the dynamics of blood physical and chemical parameters when blood specimens were processed by singlet oxygen in vitro. Our experiments were executed with whole blood specimens of healthy people (n=10). Each specimen was divided into five separate portions of 5 ml. The first portion was a control (without any exposures). The second one was processed by an oxygen-ozone mixture (at ozone concentration of 500 mcg/l, the third portion--by oxygen, and the fourth and fifth ones were processed by a gas mixture with singlet oxygen (50 and 100% of generator power). In blood samples after processing we studied the activity of lactate dehydrogenase, aldehyde dehydrogenase and superoxide dismutase, erythrocyte and plasma levels of glucose and lactate, acid-base balance and the partial pressure of gases in blood. It was found out, that blood processing by singlet oxygen leads to optimization of energy, detoxication and antioxidant enzymes functioning with changes in plasma and erythrocyte level of glucose and lactate, normalization of blood gases level and acid-base balance. Our results show, that the effect of singlet oxygen on enzyme activity is more pronounced than exposure to an oxygen-ozone gas mixture.

RESUMO

The dynamics in the oxidative and energy metabolism and enzyme systems of blood detoxification in animals with thermal trauma injected with dinitrosyl iron complexes was explored. The positive effect of dinitrosyl iron complexes on the state of blood pro- and antioxidant systems in animals with experimental thermal injury having profound oxidative stress is shown. This effect is observed as a considerable reduction of the intensity (normalization) of lipid peroxidation processes against significant elevation of antioxidant potential of blood plasma. This tendency was also fixed in erythrocyte membranes. It is also stated, that dinitrosyl iron complexes clearly normalized erythrocyte energy metabolism already by the 3rd day after trauma. In addition, infusions of dinitrosyl iron complexes caused marked stimulation of aldehyde dehydrogenase catalytic activity in burned rats via mechanism, associated with enzyme detoxification properties.

The aim of this work is complex estimation of the nitric oxide action on whole blood of healthy people. We tested the reaction of whole human blood (n=14) to the processing of it with cold NO-containing plasma. We performed direct sparging of blood samples by gaseous flow with NO in a special plant. Cold NO-containing plasma was generated by apparatus "Plazon" (Russia). We tested lactate dehydrogenase activity in direct and reverse reactions, aldehyde dehydrogenase and superoxide dismutase activity, total protein and lactate level, acid-base balance and the partial pressure of gases in blood. For integral assessment of energy metabolism changes a number of derivative parameters (coefficients of energy reaction balance and substrate provision) were used. Our experiments showed, that the processing of whole human blood with NO-containing gas flow (NO concentration--800 ppm) results in significant changes of its physical and chemical parameters. This exposure leads to inhibition of erythrocytes energy metabolism, decreasing plasma antioxidant reserves, developing moderate ionic disorders and acid-base disbalance in blood samples in vitro.

RESUMO

We studied the kinetics of liver aldehyde dehydrogenase in rats with cold injury. Low-temperature exposure was followed by a decrease in activity and catalytic efficiency of aldehyde dehydrogenase in rat liver mitochondria. Atypical changes in kinetic characteristics of aldehyde dehydrogenase were found in the cytoplasmic fraction during cold injury.

The study was designed to examine catalytic properties of non-specific aldehyde dehydrogenase from rat liver and erythrocyte as the main markers of endogenous intoxication after burn. Enzymatic activity was assayed from changes in the rate of NADH synthesis during acetaldehyde oxidation. Burn was shown to decrease it both in the liver and in erythrocytes which resulted in the accumulation of toxic aldehydes and the development of intoxication. Simultaneous fall in alcohol dehydrogenase and lactate dehydrogenase activities is supposed to contribute to the decrease of aldehyde dehydrogenase activity as a result of thermal injury.

Viruses of genus Carlavirus encode a small cysteine-rich protein (CRP) of unknown function. To investigate the role of CRP of carlavirus chrysanthemum virus B (CVB), a recombinant potato virus X (PVX) genome was constructed, which carried the CVB CRP gene. Expression of CVB CRP in the PVX genetic background drastically changed the PVX symptom phenotype in N. benthamiana. Instead of symptomless infection and mild mosaic, which are characteristic of PVX in this plant host, the recombinant virus expressing CVB CRP induced formation of necrotic local lesions on inoculated leaves and necrosis of the apical leaves. In N. tabacum, the infection pattern depended on the host genotype: the recombinant PVX was able to spread systemically only in N gene-carrying plants. In agroinfiltration-mediated transient expression assay, CVB CRP did not exhibit the properties of avirulence factor in N. benthamiana and was unable to suppress post-transcriptional gene silencing. Thus, CVB CRP is the viral pathogenicity determinant controlling the virus interaction with plant hosts in a manner which depends on plant defense mediated by resistance genes such as the N gene.

Examination of 42 first-year students from the Pomor State University in Arkhangelsk has shown that 61.91% of them had obvious signs of social disadaptation and high reactive and personality anxiety and 38.09% had no social disadaptation with low and moderate anxiety. Examination was performed in the intersession period. Students with phenomena of social disadaptation displayed reduced functioning of basic cognitive mental processes: memory, thinking, attention. The individual properties predisposing to disadaptation were as follows: instability of central nervous system performance and the personality features were hypersensitivity, sensitiveness, emotional liability, some demonstrativeness in behavior, high anxiety, inclination to phobias. The detected changes in cognitive mental processes show it necessary to develop and to introduce comprehensive sociopsychological rehabilitative programmes for students of higher educational establishments for early assessment and adequate correction of social disadaptation phenomena.

Diverse behavioral disorders and the intensity of lipid peroxidation (LPO) of biological membranes were estimated in different rat tissues after the 7-day administration and subsequent withdrawal of morphine or promedol. 24 hours after the withdrawal of the analgetics the demonstrated a high initial level of motor activity in the open field. Naloxone, an antagonist of opiate receptors, potentiated motor activity and the intensity of withdrawal syndrome (by 160%) in rats with morphine rather than promedol dependence. The behavioral disorders in dependent animals were accompanied by LPO activation in liver and brain membranes.
