We studied the effects of wintertime undernutrition on the fatty acid composition of bone marrow triacylglycerols (TAGs) of legs in freely-ranging reindeer calves (<1 year) and adult hinds by comparing reindeer in poor condition slaughtered in February with reindeer in good condition slaughtered in October. Significant reductions were found in the proportions of the major monounsaturated fatty acid, or oleic acid, and in linoleic and alpha-linolenic acids in the femur TAGs of the undernourished reindeer as compared with the reindeer in good condition. As a result of these changes, the unsaturation index (UI) of the femur TAGs was reduced by 11% both in the calves and hinds. Similarly, there were also significant reductions in the proportions of oleic and linoleic acid in the metatarsal TAGs in the undernourished hinds, but only in linoleic acid in the calves. The UI of the metatarsal TAGs of the hinds was reduced by 7%, but that of the calves remained unchanged. The results suggest selective mobilization of oleic acid and the principal C18-polyunsaturated fatty acids from bone marrow TAGs in the undernourished reindeer during winter. These changes decrease the unsaturation degree of bone marrow fats, and, if advanced, may impair their fluidity and the functioning of the legs in the cold.

We examined the effects of undernutrition on lipid metabolism in reindeer (<1 year) during mid-winter and spring, with particular focus on the proportions of polyunsaturated fatty acids (PUFAs) in major serum lipids. The reindeer (n=8) were fed their winter feed, lichen, ad libitum for 5 weeks, followed by 40% restriction of energy for 8 weeks and refeeding to normal for 6 weeks. The concentrations of major serum lipids, cholesterol and phospholipids decreased significantly during the ad libitum period (by 50 and 44%, respectively). The proportion of major PUFA, linoleic acid in serum cholesteryl esters, decreased from 48.2 to 38.4% during the ad libitum period (P < 0.01), and to 29.2% during the restriction period (P < 0.001). The proportion of linoleic acid in phospholipids decreased from 27.9 to 15.6% during the ad libitum period (P < 0. 001), and to 13.0% during the restriction (P < 0.01). Also alpha-linolenic acid in the major lipids decreased significantly during the ad libitum and restriction periods. The decreases in the major lipids and linoleic acid were reversed during the refeeding. The control group (n=8) which was fed high-quality concentrates ad libitum gained weight most of the spring but showed similar although slower decreases in the major serum lipids and PUFAs than the lichen group. Our results indicate that feeding reindeer on lichen during winter leads to the retardation of growth and reductions in major serum lipids and their principal C18-PUFA proportions. The decreased proportions of the principal PUFAs most probably reflect their low dietary intake but may have been modified also by seasonal factors.

We examined serum fatty acid composition in reindeer during the close postnatal period (from < 8 h to 3 weeks) by using maternal serum as a reference point and focusing on the proportions of polyunsaturated fatty acids (PUFAs) in serum lipids. A striking dissimilarity was found in the serum PUFAs between the neonatals and their mothers. In particular, the proportions of linoleic acid (18:2) and alpha-linolenic acid in serum cholesteryl esters and phospholipids of the newborn reindeer were significantly lower than those of the mothers. Furthermore, serum phospholipids of the newborns had lower arachidonic acid and docosapentaenoic acid but higher docosahexaenoic acid proportions than the maternal phospholipids. Although the proportions of the principal C18 PUFAs were low in reindeer milk, they increased sharply in serum cholesteryl esters and phospholipids of the calves during the first few days after birth. In particular, there were significant positive correlations in the proportions of 18:2 between serum and milk lipids. We conclude that the proportions of the serum C18 PUFAs are low in the newborn reindeer, but they are increased during the close perinatal period by a rate which suggests an efficient mechanism for selective retention of these fatty acids from milk lipids.

Captive reindeer were fed four different rations which supplied equal energy but varying amount of crude protein: lichens (3%), mixed hay plus lichens (10%) and two feed concentrates with medium (12%) or high (18%) protein content. Kinetics of total body water were measured with tritiated water from February to March 1985, and thermal energy cost of daily water intake was estimated. Biological half-time of water was shorter, and daily water intake significantly higher on the hay plus lichens diet and on the medium- or high-protein concentrates diet than on the lichens diet during March 1985. Similar differences were found between reindeer on medium-protein concentrate and on lichens at the end of the corresponding feeding period during April 1986. Daily water inflow was positively correlated with a dietary supply of digestible crude protein (r = 0.916). Thermal energy costs of daily water intake were highest 1.9 MJ (3.7 l-1) in reindeer on high-protein concentrate during March 1985, and 2.0 MJ (3.9 l-1) in reindeer on medium-protein concentrate during April 1986. The reindeer fed on lichens had minimal and nearly twice as small thermal energy cost of daily water intake (mean 1.1 MJ 2.1 l-1) as on either of the concentrates. Our results show that even a moderate feed protein ration can significantly increase free water intake and its thermal energy cost in reindeer as compared to dominant natural feed (lichens) during winter. Careful protein supplementation is recommended to support body condition with a concomitant addition of easily soluble carbohydrates to compensate for increased thermal costs of water intake.

1. The localization and cellular morphology of adipose tissue was studied by light, fluorescence and electron microscopy in reindeer between 2 weeks pre partum and 4.5 months post partum during calving, and the subsequent growth period. The respiratory capacity of the adipose tissue was examined in terms of morphometric mitochondrial volume and cytochrome-c oxidase or succinate dehydrogenase activity. 2. Adipose tissue was located at specific anatomical sites in the newborn reindeer (from 0 to 2 days of age). The perirenal-abdominal depot was the largest location (32%) followed by the inter(pre)scapular (18%) and sternal (12%) depots. Internal depot dominated over external or peripheral depots (66-34%). The locations of adipose tissue were largely similar in foetal, newborn and young reindeer. 3. The adipose tissue of the newborn reindeer had all the typical cell morphological characteristics of brown adipose tissue: abundant mitochondria, multilocular fat, high vascularization and a dense spot-like sympathetic innervation between the adipocytes. In the young reindeer, however, it resembled white adipose tissue, being almost totally unilocular with few mitochondria. 4. There was a significant correlation between morphometric mitochondrial volume and cytochrome-c oxidase activity (r = 0.848) in the adipose tissue. Mitochondrial volume, cytochrome-c oxidase and succinate dehydrogenase activity were highest after birth and decreased to almost an undetectable level during the first month. A parallel decrease occurred in the amount of brown adipose tissue from birth (1-2%) to the age of about one month (0.3%). 5. It is concluded that the distinct cell morphological features and high respiratory capacity of the adipose tissue indicate the presence of brown adipose tissue at specific anatomical locations in newborn reindeer. A marked progression towards the characteristics of white adipose tissue then takes place at the same locations during the first month. The results suggest the fundamental significance of brown adipose tissue for non-shivering thermogenesis in newborn reindeer.

Reindeer inhabit a severe arctic or subarctic environment, with the young born in early spring under adverse weather conditions. The extreme northern climate imposes a major thermal challenge to the newborn, and in the present study we have examined fetal, neonatal, and young (from 2 wk before birth to 16 mo postpartum) semidomesticated reindeer from northern Finland for the presence of thermogenic brown adipose tissue. Adipose tissues were removed, mitochondria were prepared, and the proteins were separated by molecular weight and blotted onto nitrocellulose membranes. The membranes were then probed for the presence of the 32,000-relative molecular weight mitochondrial uncoupling protein (UCP) unique to brown fat by use of a rabbit anti-(ground squirrel UCP) serum. Immunoreactivity at the molecular weight characteristic of UCP was present in perirenal, abdominal, inter(pre)scapular, sternal, intralumbar, vertebral, tracheal, inguinal, and omental-mesenteral adipose tissues of newborn reindeer (0-2 days of age). No immunoreactivity was detected in coronary adipose tissue. UCP was found at high levels in interscapular and perirenal adipose tissues of fetal reindeer at 2 wk before birth. Although the protein was present during the first few days postpartum, little immunoreactivity was found at 1 mo of age, and none was evident by 2 mo. UCP and its mRNA were also apparent in perirenal adipose tissue of the newborn of another species of Cervidae, the red deer. It is concluded, on the basis of the immunologic identification of UCP, that most adipose tissues of newborn reindeer represent functional brown fat but that there is a subsequent conversion to white adipose tissue by the 2nd mo of life.(ABSTRACT TRUNCATED AT 250 WORDS)

Thermoregulatory capacities of 51 reindeer calves (Rangifer tarandus tarandus L.) aged 1-35 days were studied at -26.5 to +35.0 degrees C ambient temperatures at Kaamanen reindeer research station, Finland (69 degrees 10' N) during calving periods in May 1981 and May-July 1982. The newborn calves aged 1-4 days maintained a high body temperature (Tre) (mean +40.2 degrees C) even at the lowest experimental temperature of -22.5 degrees C by increasing their metabolic rate five-fold above the level at +11.0 degrees C. Heat production of the new-born calves was largely based on the metabolism of brown adipose tissue, stimulated by cold-induced discharge of the sympathetic nervous transmitter, noradrenaline (NA). Sensitivity of the calves to exogenous NA disappeared during the first 3-4 weeks of life. Thermal conductance of the calves was low at low ambient temperatures, but rose strongly as Ta increased above +10 degrees C. The extensive peripheral cooling, especially in the feet, was demonstrated in the calves aged 1-10 days. The lowest foot temperature (+10.5 degrees C) was measured in a 4-day-old calf at -14.5 degrees C. Slight shivering thermogenesis was recorded in the calves aged 1-4 days and occasionally in the older calves at low values of Ta. Shivering appears to be a reserve mechanism against severe cold. At about +20 degrees C and above the calves increased their Tre (approximately 1 degree C), oxygen consumption and heart rate. In the newborn calves oxygen consumption rose four- to five-fold and in 1-month-old calves about two-fold. Fast growing calves (maximum 400 g/day) appear to be more stressed by heat than by cold exposure.
