RESUMO

The presence of ultraviolet filters (UVFs) and stabilizers (UVSs) was evaluated for the first time in the common bottlenose dolphin (Tursiops truncatus). UVFs and UVSs are compounds of growing concern because their effects on the environment are not completely known. UVFs and UVSs are added to personal care products (PCPs), such as cosmetics and products related to sun care and once released to the aquatic ecosystem, marine organisms can bioaccumulate these substances. This work aimed to determine the presence of 12 UVFs and UVSs in cetacean blubber samples to assess the pollution to which these animals of the highest trophic chain levels are exposed due to human activity. Analytical determinations were carried out using a method based on microwave-assisted extraction combined with ultrahigh-performance liquid chromatography and tandem mass spectrometry detection. The developed method was successfully applied to determine the target compounds in the blubber tissues of five necropsied common bottlenose dolphins. Three of the 12 studied compounds, namely 2-ethylhexyl 2-cyano-3,3-diphenylprop-2-enoate (octocrilene, OC), 2-hydroxy-4-methoxybenzophenone (benzophenone 3, BP3) and 3-methylbutyl (E)-3-(4methoxyphenyl) prop-2-enoate (IMC), were detected in several samples. Of the identified compounds, OC was present in all the samples and at the highest concentration within the range from 52.61 ± 18.59 to 108.0 ± 11.32 ng·g-1.

Seafood plays an important role in diet because of its health benefits. However, the fact that chemical compounds such as high production volume chemicals may be present in seafood means that its consumption can be a potential risk for population. To assess the occurrence of HPVs and estimate the exposure and risk associated with their consumption, specimens of the most consumed seafood species in Catalonia and the Canary Islands, Spain, were collected and analysed. Results showed higher levels of HPVs in samples from Catalonia and a prevalence of phthalate esters and benzenesulfonamides over the other target compounds in samples from both locations. Multivariate analysis showed spatial differences between the mean concentration profiles of HPVs for the samples from Catalonia and the Canary Islands. Exposures were higher for the samples from Catalonia, although the intake of HPVs via seafood was not of any real concern in either of the locations.

Marine environment pollution has increased in recent decades as a result of anthropogenic activities. Macrophytes can assimilate the compounds dissolved in the water and respond to changes in surround conditions, for that, they can be used as bioindicators of pollution in aquatic environments. Currently organic ultraviolet (UV) filters have shown ever-increasing in pollution levels in marine ecosystems. The anthropogenic pollution produced by eight organic ultraviolet (UV) filters in coastal macrophytes was studied. A microwave-assisted extraction (MAE), followed by ultrahigh-performance liquid chromatography with tandem mass spectrometry (UHPLC-MS/MS) was applied to 76 macrophyte (seaweeds and seagrass) samples from three different beaches on the Gran Canaria Island (Spain), collected for 6 months. All studied UV filters were found with different detection frequencies from 16% to 100% in macrophyte samples. Octocrylene (OC) was detected in all the analysed samples throughout the sampling period. The highest concentration, 19,369 ng·g-1 dry weight (dw), was for this compound in the seagrass Cymodocea nodosa. The bioconcentration ratio was determined for several seaweed groups (red, brown, green). Different bioconcentration grades were obtained. Those above 1000 indicated significant accumulation, which increases the possibility of chronic effects on seaweed and at upper tropic levels.

Organic ultraviolet (UV) filters are used in personal care products, but they are also added to industrial products and are constantly released to the environment. This study analyses the occurrence of 8 widely used organic UV filters in seawater from three beaches on the Gran Canaria Island (Spain) and in three wastewater treatment plants (WWTPs) by taking samples from influents and effluents. It also discusses the target compounds' post-treatment removal efficiencies. Sampling was carried out for 6 months and analytes were extracted by solid phase extraction with Sep-pak C18 cartridges. They were determined by ultra-high performance liquid chromatography coupled to mass spectrometry in tandem. The potential environmental hazard associated with the found concentrations was also assessed for marine organisms. Different target compounds were detected on the analysed beaches and in the wastewater. Benzophenone-3 (BP3) was the most recurrent compound in the seawater samples (frequency detection of 83%) and also in wastewater influents and effluents (measured in all the samples). However, the highest concentrations for seawater (172 µg L-1) and influent wastewater (208 µg L-1) corresponded to octocrylene, while methylene bis-benzotriazolyltetramethylbutylphenol was the compound most concentrated in secondary treatment effluent (34.0 µg L-1) and BP3 in tertiary treatment effluent (8.07 µg L-1). All the analysed samples showed that at least one target UV filter was present. Regarding the removal efficiencies of these compounds in the studied WWTPs, consistent differences between the target compounds were observed in influent concentration terms, where the average removal rates were higher than 50% for most of the compounds. Conventional treatment is unable to completely remove many studied compounds, while tertiary treatment acts as an additional elimination for some of them. An environmental hazard quotient above 1 was found for octocrylene, benzophenone-3 and 4-methylbenzylidene camphor, which indicates a potential high hazard for living species if these compounds are present.

Microplastics (MPs) in oceans adsorb different types of pollutants, which can negatively impact the food chain. The extensive use of personal care products (PCPs) has led to their ubiquitous environmental presence, and their partition between plastic matrices and surroundings is determined by their physico-chemical characteristics and environmental conditions. This work develops and applies a methodology to determine 12 UV filters (UVFs) and UV stabilisers (UVSs) in MPs collected in beach sand. The analyses were carried out by ultrasound-assisted extraction and ultrahigh-performance liquid chromatography with tandem mass spectrometry detection. The validated procedure was applied to MPs samples taken in sand samples from 13 beaches on the Canary Islands (Spain). The results showed the presence of 10 UV filters and UV stabilisers at concentrations between 1 and 4031 ng·g-1, where octocrylene was the most frequently found. The target analytes were present in all the sampling beaches.

Hormones in edible matrices, such as milk, are a subject of concern because of their adverse effects on the endocrine system and cell signaling and the consequent disruption of homeostasis in human consumers. Therefore, the assessment of the presence of hormones in milk as potential endocrine-disrupting compounds is warranted. However, the complexity of milk as a sample matrix and the ultra-low concentration of hormones pose significant analytical challenges. Fabric phase sorptive extraction (FPSE) has emerged as a powerful analytical technique for the extraction of emerging pollutants from complex aqueous matrices. FPSE allows for substantially simplified sample handling and short extraction and desorption times, as well as the decreased use of organic solvents. It is considered a green alternative to traditional extraction methodologies. In this work, the FPSE technique was evaluated to perform the simultaneous extraction of 15 steroid hormones from raw milk without employing any sample pretreatment steps. Clean and preconcentrated hormone solutions obtained from FPSE of raw milk were analyzed using ultra-high-performance liquid chromatography-tandem mass spectrometry to achieve low detection limits, which ranged from 0.047 to 1.242 ng·mL-1. Because of the presence of many interferents in milk, such as proteins, lipids, and sugar, the effect of fat content on the extraction procedure was also thoroughly studied. Additionally, for the first time, the effect of lactose on the extraction of steroid hormones was evaluated, and the results showed that the extraction efficiencies were enhanced in lactose-free samples. Finally, the optimized methodology was applied to commercial samples of cow and goat milk, and no measurable concentrations of the studied hormones were detected in these samples.

Tourism is an economic sector of great importance worldwide. In coastal areas, this activity is associated with the use of personal care products, such as ultraviolet (UV) filters and stabilizers. Therefore, assessing their presence and the exposure of living organisms to the impact of this kind of pollutant in such areas could be especially important. The Canary Islands (Spain) are considered an outermost region, and their main economic activity is based on tourism, both national and international. Thus, this area could be remarkably vulnerable to this kind of pollution, and its characterization could be useful to infer conclusions for other similar regions. With this aim, the occurrence of organic UV filters and stabilizers in different environmental matrices in Gran Canaria Island is presented in this work. Six benzotriazole compounds, UV-P, UV-326, UV-327, UV-328, UV-329 and UV-360, were found in wastewater, seawater, sludge, sediment, seaweed and fish samples. The numerous studies devoted to establishing the distribution of these target compounds in many different matrices on a touristic and particularly overcrowded island such as Gran Canaria can be used to understand the pollution situation in similar locations. The works in which determination procedures using different extraction techniques were optimized and validated for the analysis of liquid and solid samples are summarized. They are critically discussed regarding their characteristics and analytical parameters. This research is of interest to environmental managers specializing in the conservation of coastal areas where tourism is an important industry since the active components of UV filters and stabilizers can bioaccumulate and biomagnify in the trophic chain.

In the last decade, microplastics (MPs) have become an increasing cause for concerning. These particles are scattered throughout seas and oceans and have the capability of transporting adsorbed pollutants as pharmaceutical compounds, which can cause toxic effects and be transferred along the food chain. The development, optimization and validation of a sensitive and reliable analytical procedure for the extraction and determination of ten common pharmaceuticals adsorbed on MPs is reported in this study. This method involves ultrasound-assisted extraction coupled with ultra-high-performance liquid chromatography and tandem mass spectrometry. All of the variables included in the extraction process, such as the extraction time and type and solvent volume, were studied and optimised. Under optimal conditions, good reproducibility and repeatability, with relative standard deviations lower than 15% in most cases, were obtained while limits of detection between 0.25 and 15.8 ng g-1 were achieved. Last, the method was applied to the analysis of samples collected from beaches in the Canary Islands (Spain). The results indicated the presence of several analytes adsorbed on MPs in concentrations ranging from 34.0 to 111 ng g-1.

Fish species can bioaccumulate different pollutants present in the marine environments and incorporate them into the trophic chain. In this work, the occurrence of organic ultraviolet (UV) stabilizers and filters in different species of fishes of high consumption has been studied. A multiresidue method based on microwave-assisted extraction and ultra-high performance liquid chromatography with mass spectrometry detection was developed and then it was applied to nine fish species from markets in the Canary Islands and Catalonia (Spain). Three UV filters (BP-3, OC and BM-DBM) and two stabilizers (UV-328 and UV-329) were found in some of the studied species, in concentrations ranging between 0.067 and 0.683 µg g-1 dry weight (dw). BP-3 (UV filter) was the most frequently detected compound, followed by UV-329 (UV stabilizer). Thunnus thynnus was the most heavily polluted species, with a concentration of 1.201 µg g-1 dw as the sum of all measured compounds.

Chemicals added in personal care products are of emerging concern because their fate and their effect on the environment is not completely known. Benzotriazole UV stabilizers (BUVSs) are compounds used in different cosmetic products, which may reach the marine environment through marine discharge from treated waters or directly from bathing areas. Once released into the aquatic ecosystem, BUVSs can be bioaccumulated by aquatic organisms. To identify the human exposure risk, it is important to have suitable analytical methods to determine the presence of BUVSs in these organisms. Because of the complexity of such a biological matrix, selective extraction and detection techniques are required to isolate and quantify these kinds of pollutants at trace levels. In the present work, we optimized a method based on microwave-assisted extraction combined with ultra-high performance liquid chromatography and mass spectrometry detection to determine six benzotriazole compounds in fish samples. The absolute extraction yields provided by the proposed method were higher than 40% for most compounds, with intra-day and inter-day relative standard deviations ranging from 0.27 to 6.06 and 1.12-21.3%, respectively. The limits of quantification were in the range of 1.13-9.66 ng g-1 (dry weight). The method was applied to the study of three species of fish (Boops boops, Sphyraena viridensis, Sphoeroides marmoratus) that were collected close to three marine outfalls of treated waters on the Gran Canaria Island (Spain) for two years. Four of the six studied compounds, UV-326, UV-328, UV-329 and UV-360, were found with concentrations ranging from 1.34 ng g-1 to 45.6 ng g-1 (dry weight).

Cytostatic compounds are an important group of micro-pollutants since they are used to kill cells or stop cell division. For this reason, they are also considered mutagenic. Several cytostatic compounds have been detected in hospital effluents, in the influents and effluents of wastewater treatment plants and even in river water. However, their detection in solid matrices is very scarce. In this work, we have developed a new procedure based on microwave-assisted extraction (MAE) for the extraction of cytostatic compounds from sludge and sediment before determination by ultra-high-performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS). To develop this procedure, we have chosen a group of eight widely used cytostatic compounds and carried out a systematic experimental design to optimize the extraction conditions. Under these optimal conditions, the studied cytostatic compounds are extracted with good sensitivity, with recoveries ranging from 65 to 122% in sludge and recoveries varying between 49 and 109% in sediment, with the exception of etoposide, which has a lower recovery from these types of samples. The limits of detection were from 0.42 to 79.8 ng g-1 in sludge and from 0.10 to 87.5 ng g-1 in sediment. Intraday and interday relative standard deviations (RSDs) were below 15% and 18%, respectively, in both matrices at the tested concentrations. The total procedure was applied to samples of sludge taken from the main wastewater treatment plant (WWTP) of the island of Gran Canaria (Spain) and for sediment samples obtained close to the marine outfalls of different wastewater treatment plants for the same island. Pharmaceuticals and personal care products (PPCPs) constitute a group of chemicals of concern because of their potential toxicity when reaching aquatic environments. Wastewaters are one of the main pathways of introduction into the environment of the chemical compounds used in PPCPs because, in most cases, wastewater treatment facilities are not 100% efficient in their removal. This problem is accentuated in rural zones and isolated communities where conventional treatment systems are too expensive to build and operate. Waste-stabilization ponds and constructed wetlands (CWs) are natural wastewater treatment systems which are used to improve the quality of sewage from small communities because of their low cost and easy maintenance. There is growing interest in combining the two technologies to make a more robust system, taking into account their respective strengths and weaknesses. In this work, a combined macrophyte pond-CW system was evaluated for the presence at three sampling points (influent, pond effluent and CW effluent) of fifteen steroid hormones and six benzotriazole ultraviolet stabilizers (BUVSs). None of the targeted BUVS compounds were detected in either the influent or effluent, probably because of the particular characteristics of the population served by the wastewater system. In contrast, eight different steroid hormone compounds were detected at concentrations ranging from 17.3 to 247.7 ng·L-1 in influent samples and from 8.1 to 22.1 ng·L-1 in final effluent samples. The pond-CW system showed high elimination rates of steroid hormone residues with average removal efficiencies of over 77%. This efficacy was confirmed in the ecological risk assessment evaluation that was performed. Final effluents showed a low ecological risk associated with steroid hormones in contrast to the medium-high ecological risks found in the influent samples.

Benzotriazole ultraviolet stabilisers (BUVSs) are emerging compounds used in personal care products and in other products, such as plastics, to absorb UV light. BUVSs have been described as bioaccumulative, persistent and toxic, so it is of great interest to understand their presence in the environment. Some marine organisms, such as seaweeds, have been used as bioindicators of contamination in the environment because they are able to accumulate metals and organic compounds. We have selected seaweeds to develop a novel method to extract, identify and determine six BUVSs (UV P, UV 326, UV 327, UV 328, UV 329, UV 360) based on microwave assisted extraction (MAE) and ultra-high-performance liquid chromatography with diode array (UHPLC-DAD) and mass spectrometry confirmation (UHPLC-MS/MS). Under optimum conditions, recoveries ranging from 49.8 to 92.3% were obtained, while intra-day and inter-day precision values were lower than 10% for most of the compounds. Limits of detection in the ranges 1.79-4.58 and 0.89-1.76 ng g-1 dry weight (dw) were obtained for UHPLC-DAD and UHPLC-MS/MS, respectively. The optimised method was applied for the analysis of twelve species of seaweed sampled during four months in 2018 from Las Canteras beach (Gran Canaria, Spain), with the results confirmed by UHPLC-MS/MS. UV 360 was found in concentrations between 42.5 and 115 ng g-1 (dw) in five of the twelve species. Although the highest concentrations were found in Asparagopsis taxiformis, the presence of UV 360 in other species could suggest that seaweeds can act as potential bioindicators of the occurrence of these compounds in the coastal environment.

Benzotriazole UV stabilizers (BUVSs) are emerging contaminants that are widely used in personal care products, such as cosmetics and sunscreens, to absorb ultraviolet light. These compounds have been described as bioaccumulative, pseudo-persistent and toxic; therefore, it is of great importance to investigate them and determine their presence and distribution in the environment. In this research, we performed a two-year monitoring study to identify six BUVS compounds (UV-P, UV-326, UV-327, UV-328, UV-329 and UV-360) in different environmental compartments from Gran Canaria (Canary Islands, Spain): influent and effluent from five wastewater treatment plants (WWTPs) and seawater and sediment samples from three marine areas influenced by sewage outfalls discharges from WWTPs. Two methods based on on-line solid-phase extraction and microwave-assisted extraction coupled with ultra-high-performance liquid chromatography with mass spectrometry detection were applied to quantify the analytes in liquid and solid samples, respectively. The target BUVSs were measured in sewage, coastal seawater and sediment samples in concentrations in the ranges of 13.12-1933 ng L-1, 67.01-2419 ng L-1 and 4.42-2162 ng kg-1 dry weight, respectively. The studied compounds exhibited different trends of occurrence in aqueous and solid samples due to their different coefficients of hydrophobicity. The majority of the positive samples belonged to the most touristic sampling location of the island. The estimated hazard quotient, HQ, revealed no risk from the target compounds at the measured concentrations levels.

Contamination of the aquatic environment caused by multiple human activities may exert a negative impact on all living organisms. Several contaminants of emerging concern such as personal care products (PCPs) are continuously released into the aquatic environment where they are biologically active and persistent. This work reviews the current knowledge, provided by papers published after 2010 and indexed by SciFinder, Scopus, and Google search engines, about the determination and occurrence of PCPs in marine biota. Analytical methodologies have been critically reviewed, emphasizing the importance of green and high-throughput approaches and focusing the discussion on the complexity of the solute-matrix interaction in the extraction step, as well as the matrix effect in the instrumental determination. Finally, the worldwide distribution of PCPs is surveyed, taking into account the concentrations found in the same organism in different marine environments. Differences among various world areas have been highlighted, evidencing some critical aspects from an environmental point of view.

Eight wastewater samples from a university campus were analysed between May and July of 2014 to determine the concentration of 14 natural and synthetic steroid hormones. An on-line solid-phase extraction combined with ultra-high performance liquid chromatography coupled with mass spectrometry (on-line SPE-UHPLC-MS/MS) was used as extraction, pre-concentration and detection method. In the samples studied, three oestrogens (17ß-estradiol, estrone and estriol), two androgens (boldenone and testosterone), three progestogens (norgestrel, progesterone and norethisterone) and one glucocorticoid (prednisone) were detected. The removal of hormones was studied in primary and secondary constructed wetland mesocosms. The porous media of the primary constructed wetlands were palm tree mulch. These reactors were used to study the effect of water flow, i.e. horizontal (HF1) vs vertical (VF1). The latter was more efficient in the removal of 17ß-estradiol (HF1: 30%, VF1: 50%), estrone (HF1: 63%, VF1: 85%), estriol (100% both), testosterone (HF1: 45%, VF1: 73%), boldenone (HF1:-77%, VF1: 100%) and progesterone (HF1: 84%, VF1: 99%). The effluent of HF1 was used as influent of three secondary constructed wetland mesocosms: two double-stage vertical flow constructed wetlands, one with gravel (VF2gravel) and one with palm mulch (VF2mulch), and a mineral-based, horizontal flow constructed wetland (HFmineral). VF2mulch was the most efficient of the secondary reactors, since it achieved the complete removal of the hormones studied with the exception of 17ß-estradiol. The significantly better removal of BOD and ammonia attained by VF2mulch suggests that the better aeration of mulch favoured the more efficient removal of hormones.

Every year, hundreds of tons of organic pollutants reach the environment through effluents released from wastewater treatment plants worldwide, and many of these compounds have harmful effects on the aquatic ecosystem. A new class of emerging pollutants of high concern is cytostatic drugs, which are designed to treat different types of cancers by attacking cells. Environmental concentrations of cytostatic drugs are known to be in the range of ngL-1, and for this reason, it is imperative to develop analytical methods of extraction and preconcentration to allow for subsequent instrumental analysis of these drugs. In this work, a rapid, simple and green method for the analysis of seven cytostatic drug compounds that are commonly used in anti-cancer therapies was developed using a novel extraction process based on a powerful miniaturized technique, fabric phase sorptive extraction (FPSE) coupled to ultra-high performance liquid chromatography tandem mass spectrometry (UHPLC-MS/MS). The major parameters that affect the extraction process were optimized. The new method shows good linearity, with a relative standard deviation (RSD) of less than 12%. Relative recoveries higher than 40% were obtained for the studied compounds, and the detection limit of the method was within the values at which these compounds are usually found in environmental water (0.20ngL-1 to 80ngL-1). The Limit of Quantification ranged from 0.68 to 267ngL-1. Significant suppression of the signal due to the matrix effect, a common shortcoming attributed to interference from the extraction process as well as the use of ionization mode, was not observed. Subsequently, the method was applied to real wastewater samples from an effluent obtained from a hospital area and three wastewater treatment plants located in Gran Canaria Island, Spain.

Steroid hormones produce adverse effects on biota as well as bioaccumulation in fish and seafood, making it necessary to develop methodologies to evaluate these compounds in samples related to the food chain. This work presents an analytical method for evaluating 15 steroid hormones in fish tissue. It is based on microwave-assisted extraction and solid-phase extraction coupled to ultra-high-performance liquid chromatography tandem mass spectrometry (MAE-SPE-UHPLC-MS/MS). The proposed method shows appropriate detection limits (0.14-49.0ngg-1), recoveries in the range of 50% and good repeatability. After optimization, the method was applied to different tissues from two small fishes of the Canary Islands that constitute an important level of the food web (Boops boops and Sphoeroides marmoratus) and were exposed to the outfall of the Las Palmas de Gran Canaria wastewater treatment plant. The concentrations of eight detected compounds ranged from below the quantification limits to 3.95µgg-1.

Benzotriazole UV stabilizers (BUVSs) are a group of compounds added to personal care products such as sunscreens, hair dyes, make up formulations, soaps or shampoos, among others. Direct input from beaches or another aquatic recreational areas is the main source of BUVSs incorporation to the environment, where they can be mutagenic, toxic, pseudo-persistent and bioaccumulative. Due to the low levels of concentration of these compounds found in environmental samples, an extraction process is required prior to their determination. Fabric phase sorptive extraction integrates the advanced material properties of sol-gel hybrid inorganic-organic sorbents with flexible, permeable and functionally active fabric substrates, being a highly responsive, efficient and cheap device that also can be reused. In this paper, we applied fabric phase sorptive extraction methodology to analyse six BUVSs in twenty-four seawater samples from different coastal areas of Gran Canaria Island (Spain). It was coupled to ultra high performance liquid chromatography with tandem mass spectrometry in order to achieve a fast, reliable and sensitive separation and determination of the analytes from different simple matrices, regardless of its complexity and composition. Under the optimum conditions, the proposed method provided enrichment factors of 25 times with limits of detection from 1.06 to 8.96 ng L-1 and limits of quantification from 3.54 to 29.9 ng L-1 for the analytes under study in spiked samples. Intra and inter-day relative standard deviations were between 3.97 and 20.8% for all compounds. The application of the optimized methodology to non-spiked seawater samples allows detecting and quantifying the UV 360 in the range from 41.12 to 544.9 ng L-1.
