RESUMO

The authors investigated how treatment of infertile marriages is affected by the introduction of assisted reproduction. They compared the five-year period 1978-1982--485 couples with group II (1983-1987)--787 married couples. In group II successively all methods of assisted reproduction were introduced. The authors found that group II (1983-1987) had by 18.3% more deliveries, as compared with the first period. The increase is due to assisted reproduction in 10.6% and due to other therapeutic methods in 7.7%. The increase occurred in all investigated indication groups using methods of assisted reproduction within the range of 6.9-14.4%.

Four periods of microsurgery of tubal sterility and present approach to individual operation techniques in respect to assisted reproduction are evaluated. The evaluation concerns 240 operated women, their average was 31.8 years, 68.8 per cent women were operated for the first time. The pregnancy rate arrived at 90 pregnancies (40.8 per cent), 80 women delivered (33.3 per cent), the abortion occurred in 7 cases (2.9 per cent) and ectopic pregnancies occurred in 11 cases (4.6 per cent). By adhesiolysis we achieved 56 per cent pregnancies, by fimbrioplastics 34 per cent pregnancies and by neostomies only 27 per cent pregnancies. Tubal anastomosis (11 operations) and tubal implantations (9 operations) had no significant influence upon evaluation. Deflection from tubal implantations, anastomoses (except refertilizing ones) or neostomies (especially iterative ones) in favour of in-vitro-fertilization is evident.

The kinetics of SOS system induction in Escherichia coli PQ37 cells by gamma-irradiation has been studied by the SOS chromotest technique. It was shown that the synthesis of constitutive alkaline phosphatase is not immediately stopped in cells that suffered lethal damages from gamma-irradiation. The production of DNA damages inducing the SOS system was 0.021/Gy per genome. The SOS system was switched off approximately 200 min after gamma-irradiation. A correction is proposed to the calculation of the SOS system induction factor.

The authors evaluate the results of the programme of transfer of gametes into the oviduct or of embryos into the uterus in conjunction with microsurgery of sterile women. During the period between August 1981 and December 1988 a total of 211 microsurgical operations were made in stimulated cycles. The ovaries were stimulated with clomiphene citrate or in combination with human menopausal gonadotropin. The operation was made 36 hours after administration of human choriogonadotropic hormone. In 146 women at least one oocyte was obtained. A total of 72 gametotransfers into the oviduct were made and in eight, i. e. 11.1%; pregnancy was achieved. Of 43 embryotransfers five pregnancies were achieved, i. e. 11.6%. A total of 13 pregnancies were achieved with four spontaneous deliveries, seven miscarriages and two extrauterine pregnancies. The combination of selected cases of microsurgery of sterility with the IVF programme increases the rate of successful operations without a further burden for the patient, and it extends the possibilities of sterility treatment.

A study was made of the SOS induction of the gene sulA of Escherichia coli K12 in relation to the gene dosage of the gene recA. In experiments the sulA::lacZ fusion strain PQ37 and derivatives of PQ37 with the multi-copy plasmids pDR1453 or pBR322 were used. The SOS response was induced with nitrofurantoin, SOS induction of the gene sulA was determined on the basis of the amount of beta-galactosidase synthesized, i.e. by the SOS chromotest (Quillardet et al., 1982a). It was found in this work that cells with the plasmid pDR1453, which contain the gene recA of E. coli K12 (Sancar and Rupp, 1979), have a decreased SOS induction of the gene sulA. Cells with the plasmid pBR322 do not exhibit this decrease. Inactivation of the gene recA in the plasmid pDR1453 with preservation of the functional gene recA in the chromosome leads to a restoration of 'standard' SOS induction of the gene sulA. The results show that the amount of the gene product of the gene recA affects the SOS induction of the gene sulA.
