RESUMO

A descriptive study is presented on a case series of 3 patients, with IgM an IgG serology diagnosis of Chikungunya in whom other metabolic, autoimmune and infectious diseases were ruled out. They presented with other ocular manifestations with vascular changes and inflammatory characteristics. They were treated at the Retinovascular and Retinal and Vitreous clinics at the "Dr. Rodolfo Robles Valverde Hospital" and "Benemérito Comité Pro-ciegos y Sordos" in Guatemala City during 2019. Guatemala is a country with a high prevalence of vector-borne diseases, including chikungunya. In recent years cases of this disease has increased in recent years, and complications are currently seen more frequently. Within these complications, there have been reports of cases of optic neuritis, iridocyclitis, episcleritis, retinitis, uveitis, and blood dyscrasia that can affect the retina. It is important to consider these as an important differential diagnosis of ocular vascular alterations.

OBJECTIVE: Describe the ocular findings of a case series of 8 patients with a diagnosis of dengue. MATERIALS AND METHODS: Review of clinical records and interviews with patients during outpatient visits, after informed consent was obtained and following the ethical standards of the Helsinki declaration. The patients were diagnosed with diagnosis by IgM / IgG dengue serology in whom ocular involvement was identified, between January and October 2017 in Retinal-vascular clinic of the "Dr. Rodolfo Robles Valverde Hospital", Guatemala. RESULTS: The 8 patients, 5 men and 3 women from rural areas in Guatemala, were diagnosed with dengue by IgM / IgG serology with associated ocular involvement. The mean age was 32.3 years, with the oldest being 45 years old and the youngest being 20 years old. The ocular manifestations identified were, 4 neuroretinitis, 3 venous obstructions, 3 maculopathies, 2 serous detachments, 1 episcleritis, and 1 vasculitis. Two patients developed optic atrophy after resolving the neuroretinitis, and 1 developed peri-foveal scarring after the maculopathy. The ocular involvement was resolved in all patients after treatment, with an improvement in visual acuity, although in some cases damage to the visual field developed as a sequela of neuroretinitis. CONCLUSION: It is necessary to consider dengue as an important differential diagnosis in an endemic country such as Guatemala. There are a large number of ocular manifestations due to direct involvement, as well as by an immune system reaction, and thus avoid considering the different ocular manifestations as idiopathic, or due to a different condition.

The objective of this experiment was to determine the feeding value of a mechanically extracted nontoxic variety of oil (JCO) as source of energy for feedlot lambs. Twenty Pelibuey × Katahdin lambs were individually fed a dry-rolled-corn-based finishing diet supplemented with 0%, 2%, 4%, or 6% JCO (diet dry matter basis). Supplemental JCO replaced dry rolled corn in the basal diet. Fatty acid composition of JCO was C16:0, 14.0%; C18:0, 8.2%; C18:1, 26.0%; C18:2, 50.3%, and C18:3, 0.4%. Daily intakes of JCO averaged 24.7, 51.1, and 77.3 g/day or 0.57, 1.08, and 1.62 g/kg LW for the 2%, 4%, and 6% levels of supplementation, respectively. Supplemental JCO did not affect ( = 0.33) dry matter intake (DMI), but tended to increase (linear effect, = 0.06) average daily gain, efficiency of gain (linear effect, < 0.01), and dietary net energy (linear effect, < 0.01) and decreased (linear effect, < 0.01) the ratio of observed/expected DMI. At low levels (20 g/kg diet dry matter) of supplementation, the net energy (NE) value of JCO corresponds closely (0.99) to the NE value assigned by current standards (), and this NE value decreased linearly as the inclusion level of JCO increased. There were not treatment effects on plasma metabolites. Across treatments, the concentrations of hemoglobin (11.64 ± 1.08 g/dL), hematocrit (39.15 ± 3.67%), glucose (85.2 ± 17.64 mg/dL), creatinine (1.43 ± 0.28 mg/dL), and urea (20.70 ± 4.35 mg/dL) were within normal (9-15 g/dL, 27%-40%, 50-90 mg/dL, 1.0-1.8 mg/dL, and 15-50 mg/dL, for hemoglobin, hematocrit, glucose, creatinine, and urea, respectively) ranges for healthy lambs. Based on DMI, performance and plasma metabolites observed in this study, nontoxic JCO is a suitable source of energy in finishing diets for lambs.

Recent findings have shown that microbial nitrogen flow and digestible energy of diets are increased when urea is combined with a slow-release urea (SRU) in diets with a starch to acid detergent fibre ratio (S:F) 4:1. This affect is attributable to enhanced synchrony between ruminal N availability for microbial growth and carbohydrate degradation. To verify the magnitude of this effects on lamb performance, an experiment was conducted to evaluate the effects of combining urea and a SRU in diets containing S:F ratios of 3:1, 4:1, or 5:1 on performance, dietary energetics and carcass characteristics of finishing lambs. For that, 40 Pelibuey×Katahdin lambs (36.65±3 kg) were assigned to one of five weight groupings in 20 pens (5 repetition/treatments). The S:F ratio in the diet was manipulated by partially replacing the corn grain and dried distiller's grain with solubles by forage (wheat straw) and soybean meal to reach S:F ratios of 3:1, 4:1 or 5:1. An additional treatment of 4:1 S:F ratio with 0.8% urea as the sole source of non-protein nitrogen was used as a reference for comparing the effect of urea combination vs. conventional urea at the same S:F ratio. There were no treatment effects on dry matter intake (DMI). Compared the urea combination vs urea at the same S:F ratio, urea combination increased (p<0.01) average daily gain (ADG, 18.3%), gain for feed (G:F, 9.5%), and apparent energy retention per unit DMI (8.2%). Irrespective of the S:F ratio, the urea combination improved the observed-to-expected dietary ratio and apparent retention per unit DMI was maximal (quadratic effect, p≤0.03) at an S:F ratio of 4:1, while the conventional urea treatment did not modify the observed-to-expected net energy ratio nor the apparent retention per unit DMI at 4:1 S:F ratio. Urea combination group tended (3.8%, p = 0.08) to have heavier carcasses with no effects on the rest of carcass characteristics. As S:F ratio increased, ADG, G:F, dietary net energy, carcass weight, dressing percentage and longissimus thoracis (LM) area increased linearly (p≤0.02). Combining urea and a slow-release urea product results in positive effects on growth performance and dietary energetics, but the best responses are apparently observed when there is a certain proportion (S:F ratio = 4:1) of starch to acid detergent fibre in the diet.

Four Holstein steers with ruminal and duodenal cannulas were used in a 4 × 4 Latin square design to examine the effect of daily intake of 0, 2, 4 or 6 g/steer of standardized plant extract containing a mixture of quaternary benzophenanthridine alkaloids and protopine alkaloids (QBA+PA) on the characteristics of ruminal fermentation and characteristics of digestion. The basal diet consisted of a steam-flaked corn-based finishing diet that contained 62% corn and 12% sudangrass hay and the rest of diet was composed of mainly dried distillers grains, molasses, fat, and minerals. The source of QBA+PA used was Sangrovit-RS (Phytobiotics Futterzusatzstoffe GmbH, Eltville, Germany) and supplementation levels of 2, 4, and 6 g Sangrovit-RS∙steer∙d, which represented a net daily ingestion of approximately 6, 12, and 18 mg of QBA+PA compounds, respectively. Inclusion of QBA+PA linearly increased ( = 0.04) flow to the duodenum of nonammonia N and linearly decreased ( < 0.01) duodenal flows of ammonia N. Ruminal microbial efficiency (duodenal microbial N; g/kg OM fermented in the rumen) and protein efficiency (duodenal nonammonia N; g/g N intake) were increased ( < 0.05) as the level of QBA+PA increased. There were no effects of QBA+PA supplementation on ruminal, postruminal, and total tract digestion of OM, starch, and NDF, but postruminal and total tract digestion of N increased ( < 0.01) as the level of QBA+PA increased. Digestible energy of the diet tended to increase (linear affect, = 0.09) with QBA+PA supplementation. Ruminal pH and total VFA molar concentrations were not different between treatments. Ruminal NH-N concentration linearly decreased ( = 0.02) with QBA+PA supplementation. Ruminal molar proportion of acetate increased ( = 0.04) as the supplementation level of QBA+PA increased. It is concluded that QBA+PA supplementation enhances efficiency of N utilization in feedlot steers fed a steam-flaked corn-based finishing diet. This effect was due, in part, to enhanced ruminal microbial efficiency, decreased ruminal degradation of dietary nonammonia N, and enhanced postruminal N digestion.

Forty-eight Pelibuey×Katahdin male intact lambs (23.87±2.84 kg) were used in an 84-d feeding trial, with six pens per treatment in a 2×2 factorial design arrangement. The aim of the study was to evaluate the interaction of two dietary energy levels (3.05 and 2.83 Mcal/kg ME) and two dietary protein levels (17.5% and 14.5%) on growth performance, dietary energetics and carcass traits. The dietary treatments used were: i) High protein-high energy (HP-HE); ii) High protein-low energy (HP-LE); iii) Low protein-high energy (LP-HE), and iv) Low protein-low energy (LP-LE). With a high-energy level, dry matter intake (DMI) values were 6.1% lower in the low-protein diets, while with low-energy, the DMI values did not differ between the dietary protein levels. Energy levels did not influence the final weight and average daily gain (ADG), but resulted in lower DMI values and higher gain efficiencies. No effects of protein level were detected on growth performance. The observed dietary net energy (NE) ratio and observed DMI were closer than expected in all treatments and were not affected by the different treatments. There was an interaction (p<0.03) between energy and protein level for kidney-pelvic and heart fat (KPH), KPH was higher in lambs fed high energy and high protein diet but not in high energy and low protein diet. The KPH was increased (20.2%, p = 0.01) in high-energy diets, while fat thickness was increased (21.7%, p = 0.02) in high-protein diets. Therefore, it is concluded that dietary energy levels play a more important role in feed efficiency than protein levels in finishing lambs with a high-energy diet (>2.80 Mcal/kg ME). Providing a level of protein above 14.5% does not improves growth-performance, dietary energetics or carcass dressing percentage.

As a result of the cost of grains, the replacement of grains by co-products (i.e. DDGS) in feedlot diets is a common practice. This change produces diets that contain a lower amount of starch and greater amount of fibre. Hypothetically, combining feed grade urea (U) with slow release urea (Optigen) in this type of diet should elicit a better synchrony between starch (high-rate of digestion) and fibre (low-rate of digestion) promoting a better microbial protein synthesis and ruminal digestion with increasing the digestible energy of the diet. Four cannulated Holstein steers (213±4 kg) were used in a 4×4 Latin square design to examine the combination of Optigen and U in a finishing diet containing different starch:acid detergent fibre ratios (S:F) on the characteristics of digestive function. Three S:F ratios (3.0, 4.5, and 6.0) were tested using a combination of U (0.80%) and Optigen (1.0%). Additionally, a treatment of 4.5 S:F ratio with urea (0.80% in ration) as the sole source of non-protein nitrogen was used to compare the effect of urea combination at same S:F ratio. The S:F ratio of the diet was manipulated by replacing the corn grain by dried distillers grain with solubles and roughage. Urea combination did not affect ruminal pH. The S:F ratio did not affect ruminal pH at 0 and 2 h post-feeding but, at 4 and 6 h, the ruminal pH decreased as the S:F ratio increased (linear, p<0.05). Ruminal digestion of OM, starch and feed N were not affected by urea combination or S:F ratio. The urea combination did not affect ADF ruminal digestion. ADF ruminal digestion decreased linearly (p = 0.02) as the S:F ratio increased. Compared to the urea treatment (p<0.05) and within the urea combination treatment (quadratic, p<0.01), the flow of microbial nitrogen (MN) to the small intestine and ruminal microbial efficiency were greater for the urea combination at a S:F ratio of 4.5. Irrespective of the S:F ratio, the urea combination improved (2.8%, p = 0.02) postruminal N digestion. As S:F ratio increased, OM digestion increased, but ADF total tract digestion decreased. The combination of urea at 4.5 S:F improved (2%, p = 0.04) the digestible energy (DE) more than expected. Combining urea and Optigen resulted in positive effects on the MN flow and DE of the diet, but apparently these advantages are observed only when there is a certain proportion of starch:ADF in the diet.

Forty-eight crossbred heifers (378.1±18 kg) were used in a 56-d feeding trial (four pens per treatment in a randomised complete block design) to evaluate the influence of ionophore supplementation on growth performance, dietary energetics and carcass characteristics in finishing cattle during a period of heat stress. Heifers were fed a diet based on steam-flaked corn (2.22 Mcal NEm/kg) with and without an ionophore. Treatments were: i) control, no ionophore; ii) 30 mg/kg monensin sodium (RUM30); iii) 20 mg/kg lasalocid sodium (BOV20), and iv) 30 mg/kg lasalocid sodium (BOV30). Both dry matter intake (DMI) and climatic variables were measured daily and the temperature humidity index (THI) was estimated. The maximum THI during the study averaged 93, while the minimum was 70 (THI average = 79.2±2.3). Compared to controls, monensin supplementation did not influence average daily gain, the estimated NE value of the diet, or observed-to-expected DMI, but tended (p = 0.07) to increase (4.8%) gain to feed. Compared to controls, the group fed BOV30 increased (p≤0.03) daily gain (11.8%), gain to feed (8.3%), net energy of the diet (5%), and observed-to-expected DMI (5.2%). Daily weight gain was greater (7.6%, p = 0.05) for heifers fed BOV30 than for heifers fed MON30. Otherwise, differences between the two treatments in DMI, gain to feed, and dietary NE were not statistically significant (p>0.11). Plotting weekly intakes versus THI, observed intake of controls was greater (p<0.05) at THI values ≤77 than ionophore groups. When THI values were greater than 79, DMI of control and MON30 were not different (p = 0.42), although less than that of groups fed lasalocid (p = 0.04). Variation in energy intake was lower (p>0.05) in the ionophores group (CV = 1.7%) than in the control group (CV = 4.5%). Inclusion of ionophores in the diet resulted in relatively minor changes in carcass characteristics. It is concluded that ionophore supplementation did not exacerbate the decline of DM intake in heat-stressed cattle fed a high-energy finishing diet; on the contrary, it stabilised feed intake and favoured feed efficiency. Ionophore supplementation reduced estimated maintenance coefficients around 10% in finishing cattle during a period of heat stress. This effect was greatest for heifers supplemented with 30 mg lasalocid/kg of diet.

Four male lambs (Katahdin; average live weight 25.9±2.9 kg) with "T" type cannulas in the rumen and proximal duodenum were used in a 4×4 Latin square experiment to evaluate the influence of supplemental dry distillers grain with solubles (DDGS) levels (0, 10, 20 and 30%, dry matter basis) in substitution for dry-rolled (DR) corn on characteristics of digestive function and digestible energy (DE) of diet. Treatments did not influence ruminal pH. Substitution of DR corn with DDGS increased ruminal neutral detergent fiber (NDF) digestion (quadratic effect, p<0.01), but decreased ruminal organic matter (OM) digestion (linear effect, p<0.01). Replacing corn with DDGS increased (linear, p≤0.02) duodenal flow of lipids, NDF and feed N. But there were no treatment effects on flow to the small intestine of microbial nitrogen (MN) or microbial N efficiency. The estimated UIP value of DDGS was 44%. Postruminal digestion of OM, starch, lipids and nitrogen (N) were not affected by treatments. Total tract digestion of N increased (linear, p = 0.04) as the DDGS level increased, but DDGS substitution tended to decrease total tract digestion of OM (p = 0.06) and digestion of gross energy (p = 0.08). However, it did not affect the dietary digestible energy (DE, MJ/kg), reflecting the greater gross energy content of DDGS versus DR corn in the replacements. The comparative DE value of DDGS may be considered similar to the DE value of the DR corn it replaced up to 30% in the finishing diets fed to lambs.

Four Holstein steers (137 ± 2 kg) with cannulas in the rumen and proximal duodenum were used in a 4 × 4 Latin square design to evaluate the influence of processing method on comparative digestion of white corn. Treatments consisted of a basal finishing diet containing 80% corn grain (DM basis) as 1) dry-rolled white corn (DRWC), 2) steam-flaked white corn (SFWC) with 0.36 kg/L flake density (SFWC36), 3) SFWC, 0.31 kg/L flake density (SFWC31), and 4) steam-flaked yellow corn (SFYC) with 0.31 kg/L flake density (SFYC31). Characteristics of ruminal, postruminal, and apparent total tract digestion of OM, starch, and N were similar (P ≥ 0.08) for SFYC31 and SFWC31 treatments. Decreasing flake density of white corn (from 0.36 to 0.31 kg/L) did not affect (P = 0.22) ruminal OM digestion, but increased (1.9%, P = 0.07) apparent total tract OM digestion. Compared with dry rolling, steam flaking white corn increased ruminal (9.4%, P = 0.05), postruminal (14.4%, P < 0.01), and apparent total tract OM digestion (8.2%, P < 0.01), reflecting corresponding increases in ruminal (13.3%, P < 0.01), postruminal (43%, P < 0.01), and apparent total tract (12.3%, P < 0.01) starch digestion. Apparent postruminal and apparent total-tract N digestion also were greater (6.5 and 5.6%, respectively, P = 0.04) for SFWC than for DRWC. The DE value of SFWC and SFYC diets was similar, averaging 3.39 Mcal/kg. The DE value of SFWC was greater (8.1%, P < 0.01) than that of DRWC. Ruminal pH (4 h postprandial) averaged 5.74 and was not affected (P ≥ 0.48) by dietary treatments. Compared with dry rolling, steam flaking markedly enhances the feeding value of white corn, with optimal flake density being less than 0.36 kg/L. Although white corn has greater vitreous endosperm content, characteristics of ruminal starch digestion and undegradable intake protein are similar to conventional yellow dent corn when processed to a similar flake density (0.31 kg/L). However, postruminal and apparent total tract starch digestion tends to be slightly less for flaked white corn than for yellow corn.

Two trials were conducted to evaluate the interaction of the maceration process and surfactant (Tween 80) supplementation on feeding value of rice straw. Treatments were steam-flaked, corn-based diets containing 14% forage (DM basis), which was 1) Sudangrass hay; 2) ground rice straw; 3) ground rice straw plus 0.22% Tween 80; 4) macerated rice straw; and 5) macerated rice straw plus 0.22% Tween 80. In the maceration process, rice straw was passed through 2 sequentially placed pairs of corrugated rolls set at zero tolerance under a ram pressure of 62,050 millibars, similar to a conventional grain roller mill, except that the opposing rolls operated at different speeds (12 and 14 rpm, respectively). Sudangrass hay and rice straw (native and macerated) were ground through a 2.6-cm screen before incorporation into complete mixed diets. In trial 1, 125 Holstein steers (292 +/- 1.7 kg of BW) were used in a 188-d evaluation of the treatment effects on growth performance and carcass characteristics. In trial 2, 5 Holstein steers (224 +/- 3.5 kg of BW) with cannulas in the rumen and proximal duodenum were used in a 5 x 5 Latin square design to evaluate the treatment effects on digestion. There were no interactions between maceration and surfactant on growth or carcass characteristics. Tween 80 did not influence the feeding value of rice straw. Compared with grinding alone, maceration of rice straw increased the carcass-adjusted ADG (6%, P < 0.10), G:F (6%, P < 0.05), and dietary NE (5%, P < 0.05); DMI was similar across treatments. Assuming NE(m) and NE(g) of Sudangrass hay are 1.18 and 0.62 Mcal/kg, the NE(m) and NE(g) were 0.61 and 0.13 Mcal/kg for ground rice straw and 1.21 and 0.65 Mcal/kg for macerated rice straw. There were no treatment interactions on characteristics of digestion. Tween 80 did not influence ruminal or total tract digestion of OM, starch, NDF, or N. Compared with grinding alone, maceration of rice straw increased ruminal digestion of OM (7.7%, P < 0.10) and NDF (30.8%, P < 0.05), and total tract digestion of OM (2.3%, P < 0.10), NDF (21.1%, P < 0.01), and N (3.7%, P < 0.05). Total tract digestion of OM, NDF, starch, and N for the Sudangrass diet corresponded closely with that of the macerated rice straw diets. Maceration increases the feeding value of rice straw to a level similar to that of good-quality (flag stage of maturity) Sudangrass hay, which is attributable to increased OM and NDF digestion. Effects of surfactant supplementation on growth performance and digestion are not appreciable.

The study presented is a multicentric, prospective, open and comparative study designed with the objective of evaluating the performance of an antibacterial hydrocolloid dressing with hydroactivated silver (Comfeel Plata), when used to activate the healing process in wounds with high bacterial load, clinical signs of infection or malodour. Additionally, once the wound bed was appropriately prepared, a comparison in terms of efficacy was made between, on the one hand, continued treatment with the antibacterial hydrocolloid dressing, and, on the other hand, continued treatment with other dressings specifically designed for the proliferative phase of healing. Included into this study were 43 patients with chronic ulcers who were divided into two parallel treatment groups: In one group, Comfeel Plata (Coloplast AIS) was used until complete wound healing or for a maximum of 10-12 weeks, and in the second group Comfeel Plata (Coloplast A/S) was used until a clean wound bed was obtained and until the wound showed signs of positive evolution, at which moment the treatment was continued until complete healing or for a maximum of 10-12 weeks with dressings without silver designed especially for the proliferative phase of healing [Alione, Comfeel or Biatain (Coloplast AIS)]. The results obtained from the various study parameters indicate that the use of Comfeel Plata in the treatment of infected or colonized wounds prepares the wound bed and facilitates more rapid healing, and that the use of Comfeel Plata effectively reduces pain and malodour. The results indicate that once a clean wound bed is obtained, the use of a dressing without silver specifically for the proliferative phase will facilitate healing.

The structural effects of titanium citrate on the human erythrocyte membrane were studied through its interaction with intact erythrocytes and isolated unsealed human erythrocyte membranes (IUM). The studies were carried out by scanning electron microscopy and fluorescence spectroscopy, respectively. Titanium citrate induced shape changes in erythrocytes, which were damaged and ruptured leaving empty and retracted membranes. Fluorescence spectroscopy measurements in IUM indicated a disordering effect at both the polar head group and the acyl chain packing arrangements of the membrane phospholipid bilayer. Titanium citrate also interacted with molecular models of the erythrocyte membrane consisting in bilayers of dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylethanolamine (DMPE), representing classes of phospholipids located in the outer and inner monolayers of the erythrocyte membrane, respectively. X-ray diffraction indicated that titanium citrate induced structural perturbation of the polar head group and of the hydrophobic acyl regions of DMPC, while the effects on DMPE bilayers were negligible. This conclusion is supported by fluorescence spectroscopy measurements on DMPC large unilamellar vesicles. All these findings indicate that the structural perturbations induced by titanium to human erythrocytes can be extended to other cells, thereby affecting their functions.

In order to compare the antihypertensive effectiveness and tolerability of a standardized extract from Hibiscus sabdariffa with captopril, a controlled and randomized clinical trial was done. Patients from 30 to 80 years old with diagnosed hypertension and without antihypertensive treatment for at least 1 month before were included. The experimental procedure consisted of the administration of an infusion prepared with 10 g of dry calyx from H. sabdariffa on 0.51 water (9.6 mg anthocyanins content), daily before breakfast, or captopril 25 mg twice a day, for 4 weeks. The outcome variables were tolerability, therapeutic effectiveness (diastolic reduction > or = 10 mm Hg) and, in the experimental group, urinary electrolytes modification. Ninety subjects were included, 15 withdrew from the study due to non-medical reasons; so, the analysis included 39 and 36 patients from the experimental and control group, respectively. The results showed that H. sabdariffa was able to decrease the systolic blood pressure (BP) from 139.05 to 123.73mm Hg (ANOVA p < 0.03) and the diastolic BP from 90.81 to 79.52mm Hg (ANOVA p < 0.06). At the end of the study, there were no significant differences between the BP detected in both treatment groups (ANOVA p > 0.25). The rates of therapeutic effectiveness were 0.7895 and 0.8438 with H. sabdariffa and captopril, respectively (chi2, p > 0.560), whilst the tolerability was 100% for both treatments. A natriuretic effect was observed with the experimental treatment. The obtained data confirm that the H. sabdariffa extract, standardized on 9.6mg of total anthocyanins, and captopril 50 mg/day, did not show significant differences relative to hypotensive effect, antihypertensive effectiveness, and tolerability.

We present a new method for protein secondary structure prediction, based on the recognition of well-defined pentapeptides, in a large databank. Using a databank of 635 protein chains, we obtained a success rate of 68.6%. We show that progress is achieved when the databank is enlarged, when the 20 amino acids are adequately grouped in 10 sets and when more pentapeptides are attributed one of the defined conformations, alpha-helices or beta-strands. The analysis of the model indicates that the essential variable is the number of pentapeptides of well-defined structure in the database. Our model is simple, does not rely on arbitrary parameters and allows the analysis in detail of the results of each chosen hypothesis.

Gramicidin incorporation to DPPC or lecithin-PC large unilamellar vesicles (LUVs) leads to pore formation that, under hyper-osmotic conditions, produces a noticeable increase in the rate of trans-membrane water flow. This pore formation is more efficient in the more fluid lecithin-PC LUVs. Exposure of these vesicles to peroxyl radicals generated in the aerobic thermolysis of 2,2'-azo-bis(2-amidinopropane) (AAPH), changes the physical properties of the bilayer (as sensed employing fluorescent probes), modifies gramicidin molecules (as sensed by the decrease in Trp fluorescence) and notably reduces the transbilayer rate of water outflow. In order to evaluate if this reduced water-transport capacity is due to changes in the membrane due to lipid-peroxidation and/or direct damage to gramicidin channels, results obtained in the oxidable vesicles (lecithin-PC) were compared to those obtained in DPPC vesicles. The data obtained show that most of the water transport efficiency loss can be ascribed to a direct disruption of gramicidin channels by AAPH derived peroxyl radicals.

An experimental protocol based on a mathematical epidemiology model was developed to study the transmission, virulence, and recovery rates of White Spot Syndrome Virus (WSSV). Two modes of transmission were compared for WSSV in Litopenaeus vannamei. We compared transmission by ingestion of infected cadavers to transmission by cohabitation with infected animals. In addition, we compared the ingestion transmission of WSSV in L. vannamei and in L. setiferus. Finally, we compared the virulence and recovery rates of WSSV in L. vannamei and L. setiferus. The transmission rate of WSSV to L. vannamei by cohabitation was 0.01. The transmission rate by ingestion of infected cadavers was over an order of magnitude larger at 0.46, suggesting that cohabitation is a much less important mode of transmission for WSSV. A statistically significant difference was detected between the estimates of ingestion transmission of L. vannamei (0.46) and those of L. setiferus (0.84), yet no differences in the virulence or recovery rates were detected between hosts. The overall estimated virulence rate was 0.34, and the overall estimated recovery rate from a WSSV infection was 0.007 for both species. According to epidemiological theory the threshold density of hosts necessary for an epidemic to occur is directly related to the virulence and recovery rates and inversely related to the transmission rate. Therefore, the epidemic threshold density may be lower for ingestion transmission than cohabitation transmission and lower for L. setiferus than for L. vannamei.

Shrimp viruses can remain infectious in frozen shrimp tissue and have been found in frozen commodity shrimp. Therefore, the threat of viral outbreaks in wild and cultured shrimp via frozen commodity shrimp exists. Because frozen shrimp are imported with and without the cephalothorax, more knowledge is needed concerning the infectivity of a cephalothorax relative to that of an abdomen. We compared the mortality rates from shrimp exposed to a WSSV-infected cephalothorax, abdomen, or whole shrimp cadaver. Estimates of transmission coefficients from the exposures to the infected cephalothorax, abdomen, or whole shrimp were also calculated because the transmission coefficients account for differences in the initial doses. In addition, we compared the variability in infectivity of pieces of shrimp by feeding 24 equal-sized pieces of cephalothorax and abdomen to 24 individually isolated shrimp. In Expt 1, susceptible shrimp did not completely consume the infected abdomen, and a significant difference was detected among shrimp exposed to the abdomen (mortality rate = 0.40), cephalothorax (mortality rate = 0.75), and whole shrimp cadaver (mortality rate = 0.67). The calculated transmission coefficients were 0.95 from an infected cephalothorax, 0.59 from an infected abdomen, and 0.69 from an infected whole shrimp cadaver. In Expt 2, susceptible shrimp were starved to ensure complete ingestion of each dose. No significant difference was observed in the estimated mortality rates from an infected cephalothorax (0.58), abdomen (0.63), or whole shrimp (0.67). The calculated transmission coefficients were 0.84 from an infected cephalothorax, 0.83 from an infected abdomen, and 0.60 from an infected whole shrimp cadaver. In Expt 3, no difference was observed in the mortality rates resulting from exposures to pieces of infected cephalothorax (0.57) or abdomen (0.58). Our results suggested that there was no difference in the viral loads of a WSSV-infected cephalothorax or abdomen, but that the cephalothorax was more infectious, probably because it was more palatable. In addition, our results are inconsistent with some assumptions of pathogen transmission used in epidemiological models. Some shrimp may be less aggressive feeders; therefore, susceptible shrimp are differentially contacting the dead infected shrimp in the exposure tanks, violating the random mixing assumption. Moreover, virus is probably not homogeneously distributed throughout an infected shrimp, suggesting that contacts between susceptible and infected shrimp are not equally likely to result in transmission.

The selected oligomers are afterwards ordered following a simple statistical approach. The use of capillary electrophoresis for the analysis is emphasized.
