RESUMO

The extraction of Rhizostoma pulmo discarded off the Goa coast, India resulted in obtaining remarkably higher yield of 47% (based on lyophilized weight) type I collagen. The amino acid composition showed presence of glycine, hydroxyproline and proline and other amino acids in consistent with mammalian collagens. Interestingly, it also possessed essential amino acid tryptophan that makes this collagen superior than other commercially available collagen products. The dose and time response anti-oxidant activity (DPPH assay) of jellyfish collagen showed increase in the percentage activity with its efficiency better than marine fish collagen. In the dose response, IC50 was found to be 11.0 mg/ml. The percentage DPPH activity gradually increased from 47.58 to 81.11% with time 1-8 h, respectively at concentration of 7 mg/ml of jellyfish collagen. It was noteworthy to observe that the anti-oxidant activity remained 80% even after 24 h of analysis. The EDX analysis showed presence of minerals like Cl, Na, Mg, K, Cu, Fe, Zn etc. essential for healthy bones. The mass assisted laser desorption ionization-time of flight mass spectrometric (MALDI-TOF MS) data showed several precursor peaks of different peptides which has been presented here for the first time. The finding showed higher production of tryptophan containing anti-oxidant collagen that will certainly enhance its benefit in neurotransmission and cognitive function.

To elucidate the individual and multiple roles of physiological bacterial groups involved in biogeochemical cycles of carbon, nitrogen, phosphorus and sulfur, the changes in the abundance of aerobic bacteria (heterotrophs, methane oxidizers, ammonia oxidizers, sulfur oxidizers, phosphate solubilizers, phosphate accumulators) and anaerobic bacteria (total anaerobes, nitrate reducers, denitrifiers and sulfate reducers) were investigated in a biosecured, zero-exchange system stocked with whiteleg shrimp, Litopenaeus vannamei for one production cycle. Key water quality parameters during the 96-day production cycle fell within the normal range for L. vannamei culture. Results of Spearman's correlation matrix revealed that different sets of variables correlated at varying levels of significance of the interrelationships between bacterial abundances and water quality parameters. The three nitrogenous species (ammonia, nitrite and nitrate) strongly influenced the physiological bacterial groups' abundance. The strong relationship of bacterial groups with phytoplankton biomass and abundance clearly showed the trophic interconnections in nutrient exchange/recycling. Canonical correspondence analysis performed to assess the total variation revealed that the three dissolved nitrogen species followed by salinity, temperature, phytoplankton biomass and pH collectively accounted for as much as 82% of the total variation. In conclusion, the results of the study revealed that the major drivers that interweaved biogeochemical cycles are the three dissolved nitrogen species, which microbially mediated various aerobic-anaerobic assimilation/dissimilation processes in the pond ecosystem. Considering the pond microbial ecology becoming an important management tool where applied research could improve the economic and environmental sustainability of the aquaculture industry, the findings of the present study are practically relevant.

Considering the magnitude of pollution caused by marine plastics, the present study assessed their abundance, distribution, surface morphology and polymer type in ten sandy beaches spread across three states (Maharashtra, Karnataka and Goa) along the west coast of India (WCI). The total abundance of plastics (∼1-100 mm) in the studied beaches ranged from 4.1 to 23.4% (19±1-346 ± 2 items/m2). Location-wise, the abundances of both micro (43.6 ± 1.1-346 ± 2 items/m2) and macroplastics (21.6±3-195 ± 6 items/m2) were relatively higher in beaches along the Maharashtra coast. Surface morphology-wise, fragments were predominantly abundant in both micro (76±2-346 ± 2 items/m2) and macroplastics (50.6 ± 1.5-195 ± 6 items/m2) followed by pellets (43.3 ± 2.5-245.6 ± 2 items/m2). Fourier-transform infrared spectroscopy (FT-IR) analysis of plastics revealed a dominance of polyethylene (PE) followed by polypropylene (PP). IR spectra of the collected plastics at absorption band at 1750-1700 cm-1 reflect minimal surface oxidation. White-colored plastics were observed most frequently, followed by pale-yellow, dark-brown, green, blue, transparent and red. A short-term (72 h) experimental study to assess the toxicity of PE microbeads (∼1 mm) in a commercially important shrimp species, Litopenaeus vannamei revealed toxicological changes. An elevated level of lipid peroxidation (LPX)-the tagged biochemical marker, was recorded only at the maximum dose (0.15 mg/L) of PE microbeads. A moderate increase in the levels of enzymatic antioxidants (catalase and glutathione S-transferase) was also recorded at the same dose. Comprehensive information on marine plastics, including ecotoxicity provided in this study, would help in evolving strategies in minimizing plastic pollution along the WCI.

Nonlinear phenomena in animal vocalizations fundamentally includes known features, namely, frequency jump, subharmonics, biphonation, and deterministic chaos. In the present study, the multifractal detrended fluctuation analysis (MFDFA) has been employed to characterize the phase couplings revealed in the feeding clicks of Hippocampus kuda yellow seahorse. The fluctuation function Fq(s), generalized Hurst exponent h(q), multifractal scaling exponent τ(q), and the multifractal spectrum f(α) calculated in the procedure followed were analyzed to comprehend the underlying nonlinearities in the seahorse clicks. The analyses carried out reveal long-range power-law correlation properties in the data, substantiating the multifractal behavior. The resulting h(q) spectrum exhibits a distinct characteristic pattern in relation to the seahorse sex and size, and reveals a spectral blind spot in the data that was not possible to detect by conventional spectral analyses. The corresponding multifractal spectrum related width parameter Δh(q) is well clustered, defining the individual seahorse clicks. The highest degree of multifractality is evident in the 18 cm male seahorse, signifying greater heterogeneity. A further comparison between the seahorse body size and weight (wet) with respect to the width parameter Δh(q) and the second-order Hurst exponent h(q=2) underscores the versatility of MFDFA as a robust statistical tool to analyze bioacoustic observations.

Elucidation, through molecular analyses, of bacterial afflictions in commercially important aquaculture-reared shrimps is pivotal for the prevention and/or control of disease outbreaks. In this study, we examined the phylogenetic relatedness and compared the possible immune-related functional roles of both translationally controlled tumour protein (TCTP) and ferritin genes with previous studies. Both TCTP and ferritin genes were substantially upregulated in the Indian white prawn, Fenneropenaeus indicus (H. Milne Edwards), post-larvae following bath challenge with the virulent strain of bacteria, Vibrio harveyi D3. Full-length cloning of these genes by rapid amplification of complementary DNA ends -polymerase chain reaction (RACE-PCR) yielded 727-base pair (bp)-long TCTP and 1212-bp-long ferritin gene sequences. Their open reading frames (ORFs) were 507 and 510 bp, respectively. The TCTP-ORF coded for 168 amino acids with three substitutions at positions 37, 141, 155, and the ferritin ORF coded for 170 amino acids with no species-specific substitutions. Phylogenetic analysis suggested the closest relatedness of both TCTP and ferritin from F. indicus to Chinese white prawn, Fenneropenaeus chinensis (Osbeck). In addition to reporting the full-length sequences of these immune-relevant genes, this study highlighted their conserved natures, which perhaps make them important defence-related proteins in the innate immune system of F. indicus.

We identified 38 genes and eight hypothetical proteins by sequencing of 1200 clones from a Vibrio harveyi challenged Fenneropenaeus indicus subtracted cDNA library. Based on physiological roles and functions these genes were categorized into 10 groups with ∼29% of the sequences having no matches in the databases. Immune related transcripts in the library were carboxypeptidase B, ferritin, translationally controlled tumor proteins (TCTP), hemocyanin, chitinase and serine carboxy peptidase. Remarkably, qPCR results imply 4.15, 3.45 and 1.86-fold rises in expression of ferritin, TCTP and hemocyanin transcripts respectively. Additionally, minor upregulation of other immune relevant genes lectin, penaeidin, crustin, MnSOD was observed in the challenged postlarvae.

A subtracted cDNA library was constructed and analyzed to elucidate the response of Penaeus monodon postlarvae challenged with Vibrio harveyi. As many as 960 randomly selected cDNA fragments generated through suppression subtractive hybridization were single pass sequenced. Forty five genes and 20 hypothetical proteins were identified, a few being first reports from shrimps. The most abundant immune relevant genes were ferritin, hemocyanin, and TCTP (translationally controlled tumor protein) indicating their upregulation as also confirmed through qPCR. Post-infection qPCR analyses confirmed 2.04, 2.09, 3.28, 5.49, 6.47, and 11.63 fold rise respectively in ferritin, penaeidin, MnSOD, lysozyme, TCTP, and hemocyanin genes. These genes may be involved in the regulation of the host defense against V. harveyi.

The HOBAS aeration system was tested to compare changes in environmental and bacteriological parameters in ponds growing Penaeus monodon during a single production cycle. The stocking density in the aerated pond was doubled to 12 post-larvae (PL) m(-2) in contrast to the non-aerated pond with 6 (PL) m(-2). Microbial abundance in the ponds ranged between 10(5-6) cells ml(-1). Among the physiological groups of bacteria enumerated, the heterotrophs dominated with an abundance of 10(4) CFU ml(-1). Of the nitrogen and sulfur cycle bacteria, the nitrifiers flourished in the aerated pond and could maintain ammonia-N concentration within permissible levels. Bacterial activity also maintained sulfide concentrations at < 0.03 mg l(-1). Non-aerated conditions promoted denitrification maintaining nitrate concentration between 0.32 and 0.98 microM NO(3)(-)-N l(-1). However, a marked increase in ammonium content was observed in the non-aerated pond at the end of the culture period. Thus in high-density ponds, the aerators served to stimulate bacterial growth and activity which consequently maintained the quality of the water to match that of low-density ponds. Accordingly, these aerators could be effectively used to sustain higher yields. The effluent from the aerated pond is less likely to alter the redox balance of the receiving waters.

Endosulfan, a broad-spectrum non-systemic organochlorine (OC) pesticide is extensively used to control a wide variety of pests in agriculture, horticulture and public health programmes. Biochemical changes occurring in the metabolically active tissues of gills (GL), hepatopancreas (HP) and muscle (MU) of the penaeid shrimp, Metapenaeus monoceros (Fabricius) on exposure to two sublethal doses (40 and 60 ng L(-1)) of endosulfan were studied for 23 days of exposure (DoE). Sublethal doses of endosulfan significantly (P<0.05) altered the levels of the total protein (TP), the total carbohydrates (TC), the glycogen (GLY), the total free sugars (TFS) and the total lipids (TL) in test shrimps. Concentrations of biochemical components significantly varied with the DoE but were dose-independent (P<0.05). Percent decrease in all biochemical components increased with the progress of the DoE irrespective of the exposure concentrations. The order of percent decrease in the concentrations of the TP, TC, GLY, TL and TFS in different tissues at the end of 23 DoE was found to be MU>GL>HP, HP>GL>MU, MU>HP>GL, HP>MU>GL and MU>GL>HP, respectively. The results of the study revealed that sublethal doses of endosulfan significantly alters the proximate composition of major tissues, particularly the TP levels in the MU tissues thereby reducing the nutritive value of this economically important penaeid shrimp. Since M. monoceros exhibits significant biochemical changes on exposure to endosulfan, this species could possibly be used as biosensor of coastal marine and estuarine pollution by OCs.
