RESUMO

The main objective of this study was to evaluate the in vitro antiproliferative effects of isoalantolactone against liver cancer cells (Hep-G2) and also monitor its mechanism of action. The MTT assay was involved in proliferation assessments and phase contrast microscopy was used to check cellular morphology. Acridine orange/ethidium bromide staining along with western blotting was used to evaluate proapoptotic effects of isoalantolactone. DCFH-DA staining was used in ROS measurements. Transwell migration and invasion assay were executed to check the effects of isoalantolactone on migration and invasion of Hep-G2 cells. Western blotting was used to check the expressions of Ras/Raf/MEK signalling pathway in Hep-G2 cells. Results demonstrated that isoalantolactone significantly (*p<0.05 and **p<0.01) inhibited the proliferation of Hep-G2 cells in a concentration and time-reliant fashion. The IC50 value of the tested isoalantolactone molecule was found to be 71.2 µM and 53.4 µM at 12 h and 24 h time intervals respectively. Moreover, the antiproliferative effects of isoalantolactone were mediated through induction of caspase-dependent apoptosis and oxidative stress (ROS mediated). The proapoptotic effects of isoalantolactone were evident from morphological assessments and improved expressions of caspase-3, -8, and -9 and Bax while antiapoptotic Bcl-2 was reduced significantly. Additionally, antiproliferative and proapoptotic effects of isoalantolactone were found to be a consequence of blocking of Ras/Raf/MEK signalling in Hep-G2 cells. Furthermore, isoalantolactone significantly (*p<0.05) targeted the migration and invasion of Hep-G2 cells. In conclusion, these results validated that isoalantolactone shows strong antiproliferative activity against Hep-G2 liver cancer cells. Therefore, it could prove as a leading candidate in liver cancer research, drug discovery and design.

RESUMO

Hydroxylamine (NH2OH) has been verified to efficiently strengthen pollutants oxidation in Fe(II)/peroxydisulfate (PDS) and Fe(II)/H2O2 systems. However, the different effects of hydroxylamine salts types were rarely recognized. Herein, the effects of two commonly used hydroxylamine salts (i.e. NH2OH·HCl and (NH2OH)2·H2SO4) on oxidation kinetics and reactive species composition were compared in Fe(II)/PDS and Fe(II)/H2O2 systems for the first time. Pseudo first order kinetics could only describe benzoic acid (BA) oxidation well in Fe(II)/NH2OH/H2O2 system, which was related to the different concentration changes of Fe(III) determined by [Formula: see text] . Hydroxylamine salts types influenced not kinetic rules, but reaction rates of target compounds. The empirical reaction rate constant of BA in Fe(II)/NH2OH·HCl/PDS system was 141.5% of that in Fe(II)/(NH2OH)2·H2SO4/PDS system under the same concentration of NH2OH (1.4 mM), while the apparent reaction rate constant in Fe(II)/NH2OH·HCl/H2O2 system was 68% of that in Fe(II)/(NH2OH)2·H2SO4/H2O2 system. This opposite effect resulted from the differences in primary reactive species compositions and their interactions with Cl-. Reactive species identification indicated that Cl- would decrease the contribution of ferryl ion (Fe(IV)) and transform sulfate radical (SO4·-) to hydroxyl radical (·OH) in Fe(II)/NH2OH/PDS system, while it competitively consumed the only reactive species ·OH in Fe(II)/NH2OH/H2O2 system. This study highlights the importance of reductants types on strengthening Fenton oxidation and offers a reference for reasonable construction of the relevant systems.

RESUMO

Ultra-performance liquid chromatography-quadrupole-electrostatic field Orbitrap mass spectrometry(UHPLC-Q-Exactive Orbitrap MS/MS) was used for rapid identification of the chemical components in Kaixin San substance benchmark. The gradient elution was performed through a Waters ACQUITY~(TM) BEH C_(18) column(2.1 mm×150 mm, 1.7 µm) with water-acetonitrile as mobile phase, a column temperature of 30 ℃, a flow rate of 0.3 mL·min~(-1), and a sample size of 1 µL. The scanning was performed in the negative ion mode. The complex component groups in Kaixin San substance benchmark were quickly and accurately identified and clearly assigned based on the comparison of the retention time and MS data with those of the reference substance as well as the relative molecular weight of the same or similar components in the mass spectrum database and literature. A total of 77 compounds were identified, including 26 saponins, 13 triterpenoid acids, 20 oligosaccharide esters, 5 xanthones, and 13 other compounds. The qualitative method established in this study can systematically, accurately, and quickly identify the chemical components in Kaixin San substance benchmark, which can provide a basis for the further analysis of its active components in vivo and the establishment of its quality control system.

RESUMO

Variations in iodinated aromatic disinfection byproducts (DBPs) in the presence of I- and organic compounds as a function of reaction time in different molar ratios (MRs) of HOCl:NH3-N were investigated. Up to 17 kinds of iodinated aromatic DBPs were identified in the breakpoint chlorination of iodide (I-)/organic (phenol, bisphenol S (BPS) and p-nitrophenol (p-NP)) systems, and the possible pathways for the formation of iodinated aromatic DBPs were proposed. The reaction pathways include HOCl/HOI electrophilic substitution and oxidation, while the dominant iodinated DBPs were quantified. In the I-/phenol system (pH = 7.0), the sum of the concentrations of four iodinated aliphatic DBPs ranged from 0.32 to 1.04 µM (triiodomethane (TIM), dichloroiodomethane (DCIM), diiodochloromethane (DICM) and monoiodoacetic acid (MIAA)), while the concentration of 4-iodophenol ranged from 2.99 to 12.87 µM. The concentration of iodinated aromatic DBPs remained stable with an MR = 1:1. When the MR was 6:1, iodinated aromatic DBPs decreased with increasing reaction time, in which the main disinfectant in the system was active chlorine. This study proposed the formation mechanism of iodinated aromatic DBPs during the breakpoint chlorination of iodide-containing water. These results can be used to control the formation of hazardous iodinated aromatic DBPs in the disinfection of iodine containing water.

RESUMO

Esophageal squamous cell carcinoma (ESCC) increases at fast rate of all cancer types in China, which urges the investigations of its potential mechanism. In this research, a highly expressed kinesin superfamily protein 22 (KIF22) was founded both in ESCC tissues and cancer cell lines. The following experiments pointed out that down-regulation of KIF22 remarkably restrained the malignant progression of ESCC cells. Besides, KIF22 knockdown promoted ESCC cells apoptosis and arrested cells in G0/G1 phase, while KIF22 also regulated the expression of cell cycle- and EMT-related proteins. Previous research revealed that the aberrant expressions of microRNAs (miRNAs) are related to tumors development. Based on the predict result, KIF22 was considered as the target of miR-122, which was demonstrated by luciferase reporter assay. miR-122 inhibitor could significantly reverse the function of KIF22 knockdown, including cell proliferation, migration and invasion. Furthermore, down-expressed miR-122 altered the function of KIF22 knockdown on cell cycle- and EMT-related proteins. In a word, this work illustrated the regulatory function of KIF22/miR-122 axis in ESSC and provided potential targets for potential targets for ESSC treatment.

RESUMO

ß-thalassaemias are inherited hemoglobin disorders caused by defects in the ß-globin gene. In recent years, researches have re-mentioned the therapeutic significance of drug-induced fetal hemoglobin (HbF), which can reduce the imbalance of α and ß chains and improve the severity of anemia by increasing the expression of γ chain. Drug trials, such as hydroxyurea, thalidomide and desitabine have shown elevated hemoglobin, decreased blood transfusion dependence, and reduced symptoms other than anemia after treatment. In addition, in vitro experiments suggested that HbF can also induce by other drugs, which providing important clues for safe and effective HbF inducers. Therefore, this article reviews the current research progress so as to expect beneficial to clinical treatment.

RESUMO

Dabigatran is an orally active direct thrombin inhibitor, initially approved by FDA for the prophylaxis of stroke and systemic embolism in the setting of non-valvular atrial fibrillation (NVAF). Major bleeding is its most common adverse event which is of great concern. However, other types of adverse events such as esophagitis, esophageal ulcer, exanthem and pustular eruptions were reported increasingly in recent years. We present a case of immune hemolytic anemia (IHA) due to dabigatran use in a 72-year-old male with NVAF. This new and rare reported type of adverse event associated with dabigatran suggests that dabigatran may be a new cause of drug-induced immune hemolytic anemia (DIIHI).

RESUMO

The aim of this study was to observe pathological response and change in serum vascular endothelial growth factor (VEGF) in esophageal carcinoma (EC) during chemoradiotherapy (CRT).Eighty-nine patients diagnosed with EC were treated with radiotherapy at the Department of Radiotherapy of the Second People's Hospital of Changzhou between May 2008 and December 2014, including 65 patients with CRT. Gastroscopy and pathological examination were conducted 4 weeks afterwards. The pathological responses were classified as complete response (CR) and non-CR. Serum samples were collected from the patients before radiotherapy, during week 4 of radiotherapy, and 1 week after radiotherapy. The VEGF changes were classified as increase, stable, and decrease. The median overall survival (OS) and median progression-free survival (PFS) in the pathological CR group was significantly longer than that of the non-CR group (P < .001). The 1-, 3-, and 5-year OS rates in the non-CR group were lower than that in the CR group (P < .05). Moreover, the 1-, 3-, and 5-year PFS rates in the non-CR group were lower than that in the CR group (P < .05). VEGF serum level was decreased during and after radiotherapy compared with pre-radiotherapy, and the differences were statistically significant (P < .05). The 1-, 3-, and 5-year OS rates in the increased group were lower than that in the decreasing group (P < .05). Moreover, the 1-, 3-, and 5-year PFS rates in the increasing group were lower than that in the decreasing group (P < .05). Pathological response (P < .05), serum VEGF trend (P < .05), and tumor-node-metastasis stage (P < .05) in response to CRT were factors that influenced patient prognosis.Pathological response and serum VEGF change during CRT can predict prognosis of nonsurgical patients with EC. Monitoring these changes is of significance in individualized treatment.

RESUMO

Circular RNA (circRNA) is a key regulator in the development and progression of various types of carcinomas. However, its role in gastric cancer (GC) tumorigenesis is not well understood. The present study aimed to investigate the expression profile and potential modulation of circRNAs on GC carcinogenesis. Human circRNA microarray was performed to screen for abnormally expressed circRNA in GC tissue. Results showed that a decrease in the circPVRL3 expression level was associated with the presence of GC, and also with higher TNM stage and lower overall survival rates compared with that in adjacent noncancerous tissues. In vitro assays of the GC cell lines MKN-45 and MGC-803 demonstrated that knockdown of circPVRL3 promoted cell proliferation significantly. Prediction and annotation revealed circPVRL3 was able to sponge to 9 miRNAs and may be also able to have a binding with AGO2, FUS, LIN28A, PTB, and EIF4A3. In addition, based on the structure of internal ribosomal entry sites, open reading frame, and m6A modification, circPVRL3 may have the potential ability to encode proteins. Taken together, our study indicated that down-regulation of circPVRL3 could promote the proliferation in gastric carcinoma and have potential to encode protein.

RESUMO

The 2014-2015 Ebola epidemic was considered to be the largest and most complex outbreak, which caused 11,310 reported deaths. The epidemic disease can cause a mental health crisis, however, there is only a small amount of scientific literature available related to this health issue so far. We evaluated the psychological symptoms of 161 participants including Ebola survivors and healthcare workers in Sierra Leone, analyzed the impact of job classification, education level on psychological status. We found that the order of total general severity index (GSI) scores from high to low was EVD survivors, SL medical staff, SL logistic staff, SL medical students, and Chinese medical staff. There were 5 dimensions (obsession-compulsion, anxiety, hostility, phobic anxiety, and paranoid ideation) extremely high in EVD survivors. GSI were associated with university education negatively. We believed our information is necessary to develop the comprehensive emergency response plan for emerging infectious disease outbreak.

RESUMO

In order to explore the effects of cold-shock on the growth and flower bud differentiation of tomato seedlings under high temperature, tomato seedlings were subjected to cold-shock treat- ments every day with 10 °C for 10 minutes in. an artificial climate chamber. Tomato seedlings were treated with cold-shock at the first true leaf stage and the treatment lasted for 15 days. Tomato seed- lings without cold-shock were used as control. At the fourth true leaf period of tomato seedlings, five plants were randomly sampled and the growth characteristics and the ultrastructure changes of meso- phyll cell of tomato seedlings were examined. The flower bud differentiation process of tomato seed- lings was observed at the periods of the second, fourth and sixth true leaves respectively. Flowering and fruiting of tomato seedlings were also investigated after transplanting. The results showed that the stem diameter and health index of tomato seedlings with cold-shock were enhanced by 7.2% and 55.5% compared with seedlings without cold-shock. Mesophyll cells of the seedlings with cold-shock arranged loosely and various organelles such as chloroplasts and mitochondria were morphologically integrated, while chloroplasts and mitochondria of seedlings mesophyll cells without cold-shock swelled up and thylakoids vacuolized apparently. The flower bud differentiation process of seedlings with cold-shock could be advanced significantly at the early seedling stage compared with the control and the advancement was weakened with the seedling growing. Fruit set number and percentage on the first and second inflorescence of tomato plants transplanted by seedlings with cold-shock were enhanced significantly compared with those of the control. These results indicated that the injury of membrane structure of various organelles, especially chloroplast and mitochondria could be allevia- ted by cold-shock treatment under high temperature tress. Cold-shock treatment could not only im- prove the seedling growth under high temperature stress, but also advance the process of early flower bud differentiation and improve the fruit set.

RESUMO

BACKGROUND: A Chinese medical team managed Ebola virus disease (EVD) patients in Sierra Leone from October 2014 to March 2015 and attended to 693 suspected patients, of whom 288 had confirmed disease. METHODS: A retrospective study was conducted of the 288 patients with confirmed disease. Clinical symptoms, manifestations, and serum viral load were analyzed and compared among the different groups for mortality and survival time. RESULTS: Among the 288 confirmed EVD patients (149 male and 139 female, median age 28 years, and median log viral load 6.68), 98 died, 36 recovered, and 154 were lost to follow-up. Common symptoms were fever (77.78%), fatigue (64.93%), abdominal pain (64.58%), headache (62.85%), and diarrhea (61.81%). Compared to patients aged<18 years, those who were older than 40 years had a higher probability of death (odds ratio 2.855, p=0.044). Patients with a viral load of >10(6) copies/ml had a higher case fatality rate than those with <10(6) copies/ml (odds ratio 3.095, p=0.004). Cox regression showed that age, viral load, and the presence of diarrhea correlated with mortality. CONCLUSION: Patients with a high viral load, of older age, and with diarrhea had a higher mortality and shorter survival time.

RESUMO

To explore the dwarfing mechanism of tomato seedlings induced by cold-shock treatment followed by high temperature, tomato seedlings were subjected to cold-shock treatment once a day at 8:00 with temperature of 5, 10 and 15 °C for 10, 20 and 30 min, respectively, and ethylene production rate was measured. Plant height, ethylene production and gibberellin (GA3) content of the seedlings treated with T10 °C D10 min (cold-shock with 10 °C for 10 min), coupled with utilization of growth regulators, were also evaluated. The results showed that the release of ethylene was increased with the decrease of cold-shock temperature and extension of treatment time. The cold-shock treatment of 5 °C and 30 min had the highest ethylene production rate of 60.3 nL h-1 . g-1, which was 6.5 times of the control. None of ethephon (ETH), silver thiosulphate (STS), GA, or paclobutrazol (PP333) could completely block high ethylene production induced by cold-shock treatment. Tomato seedlings with cold-shock treatment (T10 °C D10 min ) resulted in reduction in GA3 content by 38.1% compared with the value of control (130.6 µg . g-1). Neither ethephon nor STS had significant effect on the dwarfing induced by cold-shock. However, GA3 weakened the dwarfing effect induced by cold-shock treatment (T10 °C D10 min), while PP333 greatly enhanced it. The dwarfing effect by cold-shock treatment of T10 °C D10 min was equivalent to that of application of 4.0 mg . L-1 PP333 based on the seedling height as an evaluation indicator. It was concluded that cold-shock treatment stimulated shoot ethylene production and blocked GA3 synthesis. GA3 played a vital role in dwarfing effect on tomato seedling induced by cold-shock treatment. Cold-shock with 10 °C and duration of 10 min could promote the growth of tomato seedlings with shorter stem and higher dry mass accumulation.

RESUMO

BACKGROUND: To investigate the relationship between pathologic tumor response to concurrent chemo- radiotherapy and variation of serum VEGF in patients with esophageal cancer. MATERIALS AND METHODS: Forty six patients with esophageal cancer who were treated with concurrent chemo-radiotherapy were enrolled. Endoscopic and pathologic examination was conducted before and four weeks afterwards. Serum level of VEGF was documented before, four weeks later and after chemo-radiotherapy. The relationship between pathologic response and the variation of serum level of VEGF and its influence on the prognosis were investigated. RESULTS: Serum level of VEGF decreased remarkably during and after chemo-radiotherapy in patients whose pathologic response was severe (F=5.393, 4.587, P(0.05). There were no statistical differences of serum VEGF level before, during and after chemo-radiotherapy for patients whose pathologic response was moderate or mild. There were 18 (85.7%), 7 (53.8%) and 6 patients (50.0%) whose serum VEGF level dropped in the severe, moderate and mild group, respectively, with significant differences among these groups (p=0.046). Two year survival rates of patients with severe, moderate and mild pathologic response were 61.9%, 53.8% and 33.3% respectively, and no statistically difference between severe and mild group regarding OS (p=0.245) was tested. CONCLUSIONS: Tumor pathologic response during chemo-radiotherapy and the changes of serum VEGF lever could predict curative effects of chemo-radiotherapy in patients with esophageal cancer.

RESUMO

AIM: To investigate the short-term efficacy and tolerability of radiotherapy plus thalidomide in patients with esophageal cancer (EC). METHODS: Serum samples from 86 EC patients were collected before, during, and after radiotherapy, and the vascular endothelial growth factor (VEGF) level was examined by ELISA. According to the change in serum VEGF level during radiotherapy, the patients were divided into two groups: in the drug group, VEGF level was increased or remained unchanged, and thalidomide was administered up to the end of radiotherapy; in the non-drug group, VEGF level was decreased and radiotherapy was given alone. Thirty healthy volunteers served as controls. The efficacy and safety of radiotherapy plus thalidomide therapy were investigated. RESULTS: The 86 EC patients had a significantly higher level of VEGF compared with the 30 healthy controls before radiotherapy (P < 0.01), and the VEGF level was significantly correlated with primary tumor size, lymph node metastasis, histopathologic type, and clinical stage (P < 0.01). Of 83 evaluable cases, VEGF level was significantly decreased after radiotherapy in 32 patients in the drug group (P < 0.05), with an effective rate of 71.88%. The incidence of dizziness and/or burnout in the drug group and non-drug group was 62.50% and 15.69%, respectively (P = 0.000), and the incidence of somnolence was 12.50% and 0%, respectively (P = 0.019). No significant differences were observed. CONCLUSION: Thalidomide can down-regulate serum VEGF level in EC patients, and combined with radiotherapy may improve treatment outcome. Thalidomide was well tolerated by EC patients.

RESUMO

AIM: To study the cancer stem cell population in esophageal cancer cell lines KYSE-150 and TE-1 and identify whether the resulting stem-like spheroid cells display cancer stem cells and radiation resistance characteristics. METHODS: A serum-free medium (SFM) suspension was used to culture esophageal cancer stem cell lines and enrich the esophageal stem-like spheres. A reverse transcription polymerase chain reaction assay was used to detect stem cell gene expression in the spheroid cells. Radiosensitivity of stem-like spheres and parental cells were evaluated by clonogenic assays. Furthermore, different cells after different doses of irradiation were tested to evaluate the change in sphere formation, cell cycle and CD44(+)CD271(+) expression of tumor stem-like spheroid cells using flow cytometry before and after irradiation. RESULTS: The cells were observed to generate an increased number of spheres in SFM with increasing cell passage. Radiation increased the rate of generation of stem-like spheres in both types of cells. The average survival fraction (SF2) of the cultured KYSE-150 compared with TE-1 stem-like spheres after 2 Gy of radiation was 0.81 ± 0.03 vs 0.87 ± 0.01 (P < 0.05), while the average SF2 of KYSE-150 compared with TE-1 parental cells was 0.69 ± 0.04 vs 0.80 ± 0.03, P < 0.05. In the esophageal parental cells, irradiation dose-dependently induced G2 arrest. Stem-like esophageal spheres were resistant to irradiation-induced G2 arrest without significant changes in the percentage population of irradiated stem-like cells. Under irradiation at 0, 4, and 8 Gy, the CD44(+)CD271(+) cell percentage for KYSE150 parental cells was 1.08% ± 0.03% vs 1.29% ± 0.07% vs 1.11% ± 0.09%, respectively; the CD44(+)CD271(+) cell percentage for TE1 parental cells was 1.16% ± 0.11% vs 0.97% ± 0.08% vs 1.45% ± 0.35%, respectively. The differences were not statistically significant. Under irradiation at 0, 4, and 8 Gy, the CD44(+)CD271(+) cell percentage for KYSE-150 stem-like spheres was 35.83% ± 1.23% vs 44.9% ± 1.67% vs 57.77% ± 1.88%, respectively; the CD44(+)CD271(+) cell percentage for TE1 stem-like spheres was 16.07% ± 0.91% vs 22.67% ± 1.12%, 16.07% ± 0.91% vs 33.27% ± 1.07%, respectively. The 4 and 8 Gy irradiated KYSE-150 and TE-1 stem-like spheres were compared with the 0 Gy irradiated group, and the differences were statistically significant (P < 0.05). CONCLUSION: The KYSE-150 and TE-1 stem-like spheres are more radioresistant than their parental cells which may suggest that cancer stem cells are related to radioresistance.

RESUMO

High temperature stress (HTS) is one of the major limiting factors that affect the quality of intensively cultured seedlings in protected facilitates during hot season. Increasing the cross adaptive response of plant induced by temperature stress is an effective way to improve plant stress resistance. In order to explore the alleviating effect of cold-shock intensity on tomato seedlings under HTS, tomato seedlings were subjected to cold-shock treatments every day with 5 °C, 10 °C, and 15 °C for 10 min, 20 min, and 30 min, respectively, in an artificial climate chamber. The effect of single appropriate cold-shock on the gene expression of small heat shock proteins LeHSP 23.8 and CaHSP18 was investigated. The results showed that hypocotyl elongation and plant height of tomato seedlings were restrained by cold-shock treatment before HTS was met. The alleviating effect of tomato seedlings under HTS by cold-shock varied greatly with levels and durations of temperature. The membrane lipids in the leaf of tomato seedlings were subjected to peroxidation injury in the cold-shock treatment at 5 °C, in which the penetration of cell membrane was increased and the activities of antioxidant enzyme was inhibited. The alleviating effect to HTS by cold-shock was decreased with the increasing cold-shock duration at 10 °C, however, a reverse change was found at 15 °C. The results indicated that cross adaptive response of tomato seedling could be induced with a moderate cold-shock temperature for a proper duration before HTS was met. The optimum cold-shock treatment was at 10 °C for 10 min per day, under which, the dry mass, healthy index, activities of protective enzymes (including SOD, POD and CAT) in leaves of tomato seedlings were significantly increased, the contents of proline and soluble protein were enhanced, relative conductivity and malondialdehyde concentration were significantly decreased, and the expression levels of Le-HSP23.8 and CaHSP18 were increased compared with that under normal temperature condition.

RESUMO

AIM: To investigate the effects of the WWOX gene on the human hepatic carcinoma cell line SMMC-7721. METHODS: Full-length WWOX cDNA was amplified from normal human liver tissues. Full-length cDNA was subcloned into pEGFP-N1, a eukaryotic expression vector. After introduction of the WWOX gene into cancer cells using liposomes, the WWOX protein level in the cells was detected through Western blotting. Cell growth rates were assessed by methyl thiazolyl tetrazolium (MTT) and colony formation assays. Cell cycle progression and cell apoptosis were measured by flow cytometry. The phosphorylated protein kinase B (AKT) and activated fragments of caspase-9 and caspase-3 were examined by Western blotting analysis. RESULTS: WWOX significantly inhibited cell proliferation, as evaluated by the MTT and colony formation assays. Cells transfected with WWOX showed significantly higher apoptosis ratios when compared with cells transfected with a mock plasmid, and overexpression of WWOX delayed cell cycle progression from G1 to S phase, as measured by flow cytometry. An increase in apoptosis was also indicated by a remarkable activation of caspase-9 and caspase-3 and a dephosphorylation of AKT (Thr308 and Ser473) measured with Western blotting analysis. CONCLUSION: Overexpression of WWOX induces apoptosis and inhibits proliferation of the human hepatic carcinoma cell line SMMC-7721.

RESUMO

Magnetic tweezers were employed to investigate the interactions between DNA and cisplatin (transplatin). Cisplatin shortened DNA and reduced its persistence length more significantly than transplatin due to the formation of many more diadducts than those formed by transplatin. Interestingly, the presence of transplatin could enhance the ability of cisplatin in shortening DNA. An optimal concentration ratio of transplatin to cisplatin existed at which cisplatin showed the highest ability in shortening DNA. Moreover, abrupt length changes were also observed when DNA was treated with a mixture of cisplatin and transplatin at high concentrations. A model was proposed to qualitatively explain well these results.
